1.Lumbar disc degeneration is associated with local transforming growth factor beta1 and inflammatory cytokines

BACKGROUND:With the research development of disc degeneration at molecular level, transforming growth factor β1 gene has a role in the proliferation and differentiation of intervertebral disc cels, and it is involved in the repair of intervertebral disc injury. But whether transforming growth factor β1 is also involved in pathophysiological process of disc degeneration has not been determined. OBJECTIVE:To discuss the relationship between transforming growth factor β1, inflammatory cytokines and degree of lumbar disc degeneration. METHODS: Twenty-two patients with lumbar disc degeneration (22 mild, 26 moderate and 24 severe cases) were enroled as observation group, and another 30 patients without lumbar disc degeneration served as control group. Levels of transforming growth factorβ1, interleukin-6, interleukin-8, tumor necrosis factor α were detected and compared among patients with mild, moderate and severe lumbar disc degeneration as wel as between the observation and controlgroups. RESULTS AND CONCLUSION: Levels of transforming growth factor β1, interleukin-6, interleukin-8, tumor necrosis factor α were significantly higher in the observation group than the control group (P < 0.01). In the observation group, the above-mentioned indicators were ranked as folows: severe cases > moderate cases > mild cases (P < 0.01). There was a positive correlation between these indicators and the severity of lumbar disc degeneration (r=0.198, 0.312, 0.356, 0.275, 0.724,P < 0.01). These findings indicate that the levels of transforming growthfactor β1, interleukin-6, interleukin-8 and tumor necrosis factor α are increased with the severity of lumbar disc degeneration.
2.Influence of Invigorating Spleen and Restoring Virility to the SS Level and SSR1 Gene Expression in Brain of Spleen Insufficiency Model of Rat

Objective To compare on the influence of invigorating spleen and restoring virility to the SS level and SSR1 gene expression in brain. Method The spleen insufficiency model of rat was established by adopting bitter decline diarrhea, wild eating and overworking. SS level and SSR1 gene expression change of hypothalamus ventral kernel, hippocampi CA1 area and forehead corte were detected by immunohistochemistry and in situ hybridization techniques. Result In the model group, the immunoreaction masculine substance of SS and SSR1 mRNA, hypothalamus ventral kernel, hippocampi CA1 area and forehead cortex decreased obviously. The immunoreaction masculine substance of SS and SSR1 mRNA on above mentioned parts in the group of invigorating spleen and restoring virility increased distinctly. Conclusion In the model group, the SS level and SSR1 gene expression in brain to promoting study memory ability changed. Invigorating spleen and restoring virility may affect study memory ability by regulating the SS level and SSR1 gene expression.
3.Hepatocyte growth factor protects cultured rat cardiomyocytes against apoptosis induced by gamma ray irradiation

Objective Hepatocyte growth factor (HGF) has been proposed to be used as an endogenous cardioprotective agent. The aim of the present study was to investigate the anti-apoptosis effect of HGF on rat cardiomyocytes after being irradiated by single dose gamma ray. Methods The study was performed using primary cell cultures of cardiomyocytes isolated from hearts of new-born rats. After being cultured for 72h in vitro, cardiomyocytes were irradiated with gamma ray in a single dose of 20Gy. In the HGF treated groups, cells were incubated with HGF in the dose of 20 and 40ng/ml 3h prior to irradiation, respectively. At 48h post-irradiation, the concentration of LDH in the supernatant of cell culture was determined. Apoptosis and cell cycle were determined with flow cytometry. 4.Predictive value of trauma index on the prognosis in patients with traumatic spinal cord injury

Objective To investigate the predictive value of trauma index on the prognosis in pa-tients with traumatic spinal cord injury.Methods Enrolled 168 cases of patients with traumatic spinal cord injury,the traumatic index score and the American Spinal Injury Association (ASIA)grade were as-sessed on admission,then followed up for 6 months,the Spearman correlation analysis was performed to a-nalysis the relationship between trauma index and ASIA grade,and Receiver operating curve (ROC)anal-ysis was used to evaluate the relationship between trauma index and prognosis.Results There was a sig-nificant negative correlation between trauma index score and ASIA grade(r =-0.68,P <0.01).A total of 157 cases (93.5%)completed follow-up,the trauma index score had a good predictive value on the traumatic death in these patients,it's sensitivity was 71.4%,specificity was 76.1%,the best cut-off point was 25 points(P <0.01 );Compared with the low score group (<25 points),the high group (≥25 points)showed a lower ASIA grade and higher mortality,the differences were statistically significant (P <0.01).Conclusion The trauma index score was related with the spinal cord injury,and it is a predictive factor of poor prognosis in patients with traumatic spinal cord injury.
5.Adventitium-derived relaxing factor may be a protein factor secreted by adipocytes with non-species-specificity and not limited to periadventitial fat

AIM To test if "adventitium-derived relaxing factor"(ADRF) possesses species- and tissue-specificity and make preliminary research on proteins separated from the bath solution. METHODS Record the tension of aortic ring with and without periadventitial fat, induced by phenylephrine(Phe) and analyze the proteins extracted from the bath solution with SDS-PAGE electrophoresis. RESULTS ① In Sprague-Dawley rats, the concentration-response curve of Phe to rings without the periadventitial fat shifted to rightward, as compared to the curve of the intact aortic rings, which means periadventitial fat can reduce the contraction induced by Phe. The same phenomena as the above could be found in aortic ring of Wistar rats, guinea pigs, and rabbits. ② Moreover, the contraction induced by Phe was obviously reduced by moving adipose tissue from greater omentum into the bath solution. ③ The release of ADRF was strongly reduced by 10 μmol·L-1 genistein (tyrosine kinase inhibitor). But the effect of existed ADRF could not be counterposed by genistein. ④ Five protein bands were separated from the bath solution, with relative molecular mass 74.0, 59.8, 54.4, 28.7 and 13.8 ku. CONCLUSION ① ADRF is a non-species specific factor. ② The entire name of ADRF should change from "adventitium-derived relaxing factor" to "adipocyte-derived relaxing factor". ③ Some proteins which may include ADRF are separated from the bath solution.
6.Identification and characterization of partner proteins interacting with fatty acid activation enzyme Slr1609 in Synechocystis sp. PCC 6803.

To understand molecular modules related to polyunsaturated fatty acids (PUFA) synthesis and eventually produce PUFA at high efficiency, we developed a protein complex analysis technology in Synechocystis sp. PCC 6803, and applied it to identify possible partner proteins interacting with the key enzymes that catalyze PUFA biosynthesis. We first constructed a recombinant expression of protein of slr1609 encoding the fatty acid activation enzyme, by fusing 3xFLAG tag with the target protein. Then we verified its expression by Western blotting targeting 3xFLAG tag. To maximize purification of Slr1609 protein complex, we optimized the protein expression conditions of Slr1609 in Synechocystis in a 5 L fermenter by monitoring its gene expression using RT-qPCR. The purification of the Slr1609 protein complexes was demonstrated by a Native-PAGE analysis. Finally, LC-MS/MS proteomic analysis allowed identification of the possible partner proteins interacting with Slr1609.

7.Low frequency transcranial magnetic stimulation for children with Tourette's syndrome

Objective To investigate the effects of low frequency repetitive transcranial magnetic stimulation (rTMS) applied to the supplementary motor area (SMA) of children with Tourette's syndrome (TS). Methods Thirty TS subjects less than 16 years old were treated with 1 Hz rTMS to the SMA at 110% of the resting motor threshold (RMT) in 20 daily sessions,receiving 1200 pulses/day.Clinical assessment and physiological measures of the left and right RMT were conducted at different time points during the treatment. ResultsAfter 4 weeks of treatment,statistically significant reductions were observed in assessments with the Yale global tic severity scale (YGTSS) and in terms of clinical global impression (CGI).Symptomatic improvement was correlated with dramatic increases in both right and left RMTs. ConclusionApplication of 1 Hz rTMS to the SMA can improve the clinical symptoms of TS children.
8.Research on real-time data collection and demonstration system for acupuncture manipulation

OBJECTIVE: To apply the real-time data collection and demonstration system for acupuncture manipulation to quantitative analysis of acupuncture manipulation for acupuncture teaching and research. METHODS: The real-time observation and analysis of parameter changes of acupuncture manipulation were implemented by this system. RESULTS: During the course of lifting and thrusting manipulation, the needle body moved in a uniform variable motion, and the action force was still kept while the arrival of Qi was achieved. During the course of twirling manipulation, the variations of twirling number and radian caused the changes of resistance moment, and this reaction was related to the body response. During the course of rotating and shaking manipulation, the radius for rotating and shaking was related to such factors as the force, the action point and the direction. CONCLUSION: The real-time data collection and demonstration system for acupuncture manipulation can provide a new experimental method for quantitative data analysis, standardized research and teaching demonstration of acupuncture.
9.Effect of platelet-rich plasma on wnt3 gene and klotho gene of adipose-derived stem cells of rabbit

Objective To investigate the effect of platelet-rich plasma (PRP) on starting of wnt3 gene and klotho gene of adipose-derived stem cells (ADSCs) of rabbit.Methods Epididymal adipose tissue stem cells were obtained from New Zealand white rabbits,and the cells identified by morphology and inducing differentiation,and the cells were cultured to the fourth generation,PRP and PPP (platelet-poor plasma) were prepared by traditional centrifugal method from abdominal aortic of rabbit; ADSCs were cultured in culture medium containing PRP (experimental group),PPP (control group) and all medium (blank group) for each 5% for 24h,48h and 72h.Cells of each group were dissociated and total RNA extracted.Effects of the starting of wnt3 gene and klotho gene were detected by RT-PCR.Results Primary ADSCs of rabbit grew in the way of long spindle swirly.The results of oil red O and alizarin red staining of the ADSCs were positive.Expression of wnt3 gene and klotho gene in the experimental group significantly increased from the results of RT-PCR (P<0.05).Conclusions PRP can promote proliferation of the ADSCs of rabbit and increase the expression of wnt3 gene and klotho gene significantly.
10.Angiography Features and Clinical Significance of Pelvic Tumor

Objective To study angiography features and clinical significance of "multiple tumor feeding embolization".Methods Twelve patients with pelvic tumor were reviewed retrospectively.The lesions included fibrosarcoma(n=1),chondrosarcoma(n=2),osteosarcoma(n=2) and metastatic tumor(n=7).Results Angiography features were:(1)multiple origin and branch of tumor-feeding vessels.(2)large vascular mass with dense tumor stain.(3)larger size of tumor stain comparing with that of bone destraction.After treatment,symptom relief and tumor shrink were obviously using"the multiple tumor-feeding embolization".Conclusion Angiography features of pelvic tumor are great value in guiding the clinical therapy.
