1.Empirical study of furazolidone-induced dilated cardiomyopathy model in rats

Objective:To determine the efficacy and reliability of furozolidone-induced dilated cardiomyopathy model in rats and to evaluate it by transthoracic echocardiography.Methods:Wistar rats were fed with furazolidone for eight weeks.Left ventricu- lar end-diastolic diameter(LVDD),left ventricular end-systolic diameter(LVSD),fraction shortening(FS)and left ventricular e- jection fraction(LVEF)were determined by echocardiogram.Myocardial cell morphology was observed by histopathologic slides. Results:Compared with the normal control group,the body weights,FS and LVEF were decreased obviously,whereas the ra- tios of heart weight and body weight and LVSD were increased significantly.The HE and VG staining displayed that Myocardial cells developed hypertrophy and degeneration,whereas the interstitial collagen fibers got hyperplasia.Conclusion: The DCM rats model can be established successfully hy feeding with furozolidone for long time.This method is simple with high success rate and low death rate.The transthoracic echocardiography can be applied in monitoring the animal model's formation and effect.

3.CT and MRI Features of Pulmonary Carcinoma of Mediastinal Type

Objective To study CT and MR imaging features of pulmonary carcinoma of mediastinal type.Methods CT and MRI data (including anatomic site,size,shape,margin of the tumors and the relationship between tumors and mediastinum) of 15 patients with pulmonary carcinoma of mediastinal type confirmed by surgery and pathology were analyzed retrospectively.All patients underwent plain CT scans,and 11 of them also underwent contrast-enhanced CT scans,and 3 of them underwent plain MR scans and Gd-DTPA enhanced MR scans.Results On CT and MRI,the single mass was located subpleural of mediastinum with wide base.There were 5 cases of small cell lung carcinoma,8 squamous carcinoma,1 adenocarcinoma and 1 adenosquamous carcinoma.The majority of tumors manifested as ellipse.The tumors were lobulated in 12 and spicular margins in nine.8 tumors located in superior mediastinum,5 in medium mediastinum,2 in infra mediastinum and 12 in the borderline between the anterior mediastinum and medinum mediastinum.The tumors and enlarged lymph nodes of hilar and mediastinum connected closely in 7 cases.The bronchus were stenosed or obstructed by the tumors in 9 cases,and metastatic bone destruction of thoracic cage in 4 cases.Conclusion The pulmonary carcinoma of mediastinal type is easy misdiagnosed,CT and MRI findings in combination with clinical data can improve its diagnostic rate.
4.Preparation and Characterization of Eugenol-β-cyclodextrin Inclusion Complex

OBJECTIVE:To optimize the inclusion technology of Eugenol-β-cyclodextrin (β-CD) inclusion complex,and to identify and characterize it. METHODS:With the molar ratio of eugenol to β-CD,inclusion temperature and inclusion time as fac-tors,using the yield of inclusion compounds as index,the inclusion technology was optimized by orthogonal test. The formation of inclusion compound was identified by the spectra change of FT-IR,XRD and 1H NMR. Its structure was characterized by 1H RO-ESY NMR. RESULTS:The optimized inclusion conditions were that the molar ratio of eugenol to β-CD was 1.0:1;inclusion tem-perature was 60 ℃;inclusion time was 2.0 h. And the yield of inclusion compound was 73.86%(RSD=0.17%,n=3). 1H NMR results ofβ-CD and its inclusion complex indicated that the optimum qualitative ratio of the inclusion complex was 1.0:1. The inter-molecular interaction between eugenol and β-CD was confirmed by the spectrum analysis of FT-IR and XRD. 1H ROESY NMR re-sults indicated the structure of inclusion complex mainly was that the phenyl of eugenol was in the cavity of β-CD,the vinyl was outside. CONCLUSIONS:The inclusion technology is reasonable and feasible,and can be used for the inclusion of eugenol andβ-CD. The formation of inclusion compound is confirmed by the spectrum analysis.
5.Effects of Compound Catechu Anti-Diarrhea Ointment on Expression of Aquaporin3 in Colonic Tissues of Diarrhea Model Rats

Objective To investigate the effect of compound catechu anti-diarrhea ointment on the expression of aquaporin3 (AQP3) in the colonic tissues of diarrhea rat model, and the mechanism thereof. Methods Forty SD rats were randomly divided into four groups:blank control group, model control group, positive control group and compound catechu anti-diarrhea ointment group. Rats were given 6 g blank matrix cream in model control group, 2 mL suspension of berberine in positive control group and compound catechu anti-diarrhea ointment in compound catechu anti-diarrhea ointment group, two times/d for 7 d. The rat model of diarrhea was established by using senna intragastric administration. The water content of feces was measured. The expression of AQP3 in colonic tissues was detected by immunohistochemistry assay. Results The water contents of feces were significantly higher in model control group (64.09±0.41)%than those of other three groups (F=53.879,P<0.05). There was no significant difference in the water content of feces between compound catechu anti-diar-rhea ointment group (48.83 ± 1.08)%and positive control group (46.87 ± 2.19)%. The AQP3-positive cells were mainly ex-pressed in the intestinal mucosa. The dyeing index was significantly lower in model control group (0.85±0.18) than that of other three groups (F=14.971,P<0.05). There was no significant difference in the dyeing index between compound catechu anti-diarrhea ointment group (1.30±0.18) and positive control group (1.37±0.14). Conclusion Compound catechu anti-di-arrhea ointment can significantly reduce the water contents of feces, which may be related to the increased AQP 3 expression in colonic tissues.

We investigated health condition and evaluate the viability of Barthel's measuring scale for 1297 elder patients at 17 nursing homes.We nursed all patients by standards and performed a comparison study.The nursing percentages of highest and lowest levels were higher than the previous ones ( P < 0.01 ),while the nursing percentage of normal level was lower than the previous one ( P < 0.01 ).The cooperation between doctors and nurses became optimized through progressive patient care.Ideal nursing standard should be based upon the doctors' judgment and nurses' evaluation.The new standard will be more suitable for patients and easier to control for nurses.
7.Preparation and biocompatibility of panax notoginseng saponins-hydroxyapatite/chitosan scaffold

BACKGROUND:Panax notoginseng saponins promotes bone repair by improving vascular proliferation. Therefore, the scaffolds carrying panax notoginseng saponins were supposed to be used to improve bone repair at the bone defect region. However, the biocompatibility of scaffolds remains unclear.
OBJECTIVE:To evaluate the biocompatibility of panax notoginseng saponins-hydroxyapatite/chitosan scaffolds.
METHODS:A new bone repair scaffold has been generated by thoroughly mixtures of 0, 0.1, 1, 10 mg panax notoginseng saponins and chitosan/hydroxyapatite using in-situ composite technique and freeze-drying technique. Morphology and mechanical property of the scaffold were observed under a scanning electron microscope. (1) Cel proliferation test:rabbit bone marrow mesenchymal stem cel s of passage 3 were cultured in four kinds of drug loaded hydroxyapatite/chitosan composite material leaching liquor. Cel s normal y cultured were considered as controls. 3-(4, 5-Dimethylthiazol-2-yl) 2, 5-diphenyl tetrazolium bromide was used to measure absorbance value of cel s in each group. (2) Hemolysis test:Rabbit anticoagulated blood was added with four kinds of drug loaded hydroxyapatite/chitosan composite material leaching liquor. Absorbance values were measured using a microplate reader in each group. (3) Pyrogen test:The four kinds of drug loaded hydroxyapatite/chitosan composite material leaching liquor and saline were respectively injected into ear vein of rabbits, and the increase of rabbit body temperature was detected.
RESULTS AND CONCLUSION:Drug loaded hydroxyapatite/chitosan composite material contained three dimensional porous structure of 110μm in diameter. Drug loading process of panax notoginseng saponins did not significantly affect the porosity, pore size and density of the composite material, but decreased its breaking strength and elastic modulus. The larger amount of drug loading showed a more obvious effect. Simple hydroxyapatite/chitosan composite material had good cel ular compatibility. The composite material after drug loading obviously suppressed cel proliferation, and the larger amount of drug loading showed a more obvious effect. The composite material had good blood compatibility before and after drug loading. The composite material had good pyrogen effects before and after drug loading, but accorded with acceptable quality level of pyrogen test.

Objective To study the inhibition effects of various gastrin-shNAs on gastrin expression in gastric cancer cell line BGC-823. Methods Four nucleotide sequences of shRNA were designed corresponding to various sites of gastrin gene.Four shRNAs were synthesized by in vitro transcription and transfected into gastric cancer cell line BGC-823 at the final concentration of 10nmol/L,20nmol/L,40nmol/L and 80nmol/L respectively.In situ hybridization and immunohistochemistry techniques were applied to investigate the inhibition of gastrin expression and screen the most effective shRNA.The inhibitory effect on gastrin mRNA of screened shRNA was further identified by RT-PCR.MTT assay was used to determine the inhibitory effect of 4 shRNAs at various final concentrations on the growth of BGC-823 cells. Results The gastrin mRNA and protein exression were suppressed distinctly 24,48,and 72hours after transfection,and exhibited time-and concentration-dependent tendency.The highest suppression efficiency on both mRNA(54.27?0.042)% and protein(41.69?0.038)% level occurred 72 hours later in the cells transfected with shRNAs.The RT-PCR result showed that the inhibitory ratio of shRNA3 on gastrin mRNA of BGC-823 was 48.1%.MTT displayed a proliferative inhibition of the BGC-823 cells after transfection of shRNAs with a concentration-denpendent tendency except the shRNA4 treated cells.Conclusion Four gastrin-shRNAs showed a significant inhibition effect on gastrin expression of gastric cancer cell BGC-823 on mRNA and protein level.shRNAs might be the most effective gastrin-shRNA.Inhibited gastrin expression by shRNAs resulted in a significant decrease of proliferative ability of BGC-823 cells.

10.Diagnosis and surgery for intramuscular hemangioma of skeletal muscle.

OBJECTIVETo explore effects of color Doppler ultrasound and magnetic resonance imaging (MRI) in diagnosis intramuscular hemangioma of skeletal muscle.
METHODSFrom December 2000 to January 2013, 54 patients treated by operation confirmed as intramuscular hemangioma of skeletal muscle by pathology postoperatively, there were 19 males and 35 females aged from 11 to 59 years old (averaged 33.6); the courses of disease ranged from 2.5 to 15 years with an average of 5.2 years. Thirty-eight patients were checked by color Doppler ultrasound, and 14 patients were inspected by MRI. All patients were treated by operation. Postoperative operative time, blood loss in operation, and complications and pathology postoperatively were observed, and IMH clinical effective evaluating standard were used to evaluate clinical outcomes.
RESULTSForty-three patients were followed up from 7 to 49 months with an average of 28.4 months. Operative time was (53 to 187) min with average of 76.3 min, blood loss was (70 to 350) ml with an average of 223.6 ml. No infections and death occurred. Thirty-five patients were diagnosed by color Doppler ultrasound and 13 patients were confirmed by MRI. Twenty patients were capillary type, 22 patients were spongy vascular type and 12 patients were mixed type according to Brown pathological type. In accordance with IMH clinical effective evaluating standard, 29 cases obtained excellent results, 8 moderate and 4 dissatisfaction and 2 poor.
CONCLUSIONColor doppler ultrasound and MRI get a high rate diagnosing patients with intramuscular hemangioma and have an significant valuable in clinical application, and surgical operation which has advantages of relieve symptoms obviously, improve life quality and reduce recurrence rate, could receive good curative effect.
