1.Repair of spinal cord injuries in mice using peripheral nerve and activited macrophages transplantation

[Objective]To explore the effect of spinal cord injury(SCI) repair by peripheral nerve combined with activitied macrophages graft.[Method]Fistly the preparation of the activated macrophages and fabrication of the peripheral nerve were done.81 BALB/C mice were grouped in A,B,C,D groups randomly after their spinal cord were hemi-resected.20 mice of group A were dealed with activated macrophages graft;20 of group B were dealed with peripheral nerve graft;21 of group C were transplanted both two graftes;20 of group D were contral group.Each of the 4 groups was respectively divided into 3 subgroups randomly.7 of subgroup 1 were investigated by their restoration of the motor function and improvement of the somatosensory evoked potential(SEP).7 of subgroup 2 were tabken for histological examination.The spinal cords were fixed and cut into longitudinal frozen sections,and were immuneostained and stained with hematoxylin and eosin(HE).The numbers of cells or neurofilaments were counted under microscope.6 of subgroup 3(7 of C3)were used as complement ones.All the results were recorded from 1 to 12 weeks postoperative and analyzed statistically using the SPSS 12.0 software package.[Result]The BBB scale and the SEP wave:there was a significant difference between group C1 and the other three groups,there was a significant difference between group A1,B1 and D1,too(ONE WAY ANOVA,P
2.Determination of Four Sulfa Antibiotics in Groundwater, Soil and Excreta Samples Using High Performance Liquid Chromatography

A method was developed for the determination of four sulfa antibiotics in groundwater, soil and excreta using solid phase micro extraction disks coupled with high performance liquid chromatography. The influence of eluent, different solid phase micro extraction membranes on the recovery of sulfa antibiotics in groundwater was investigated and it was found that when using the mixture of methyl alcohol and 1 . 0% formic acid as eluent, HLB ( divinyl benzene-N-vinyl pyrrolidone polymer ) as extraction membranes, an optimal enrichment effect was obtained. Different pretreatment methods for the 3 kinds of samples abovementioned were also examined. It was found that the signal response values obtained by using mixture of methyl alcohol and 1 . 0% formic acid as base solution of standard or sample solution was higher 8-10 times than that by using methyl alcohol only. Under the optimal conditions, good linear relationships were obtained in the sulfa antibiotics concentrations of 0 . 005-10 . 0 mg/L with the correlation coefficients>0 . 9999;The detection limits of sulfathiazole ( ST ) , sulfadiazine ( SM ) , sulfamethazine ( SM2 ) , sulfamethoxazole ( SMX ) were 1 . 08 , 3. 56, 4. 63 and 1. 84 ng/L(S/N=3), respectively. The enrichment factors for four sulfa antibiotics were 4000 times with solid phase micro extraction disks. The RSD of matrix spiked samples were 0. 1%-0. 4%(n=7). The proposed method was applied to the determination of the four sulfa antibiotics in groundwater, soil and excreta with spiked recoveries of the four sulfa antibiotics in the range of 69 . 80%-117 . 60%.
3.Genetic diversity and genetic relationship of Jatropha curcas L. in Sichuan and Yunnan evaluated by cpSSR markers.

OBJECTIVEThe genetic diversity and genetic relationship of Jatropha curcas resources in Sichuan and Yunnan were studied in order to provide a theoretical basis for breeding fine varieties and protecting germplasm resources.
METHODTen J. curcas populations were studied by 12 cpSSR primers in this paper. On the base of amplified bands, genetic diversity parameters were analyzed by POPGENE version 1.32. Furthermore, UPGMA tree of 10 J. curcas populations established from pairwise population distance by NTSYSpc version 2.10.
RESULTTwenty-two polymorphic bands were detected, and the percentage of polymorphic loci (P) was 76.28%. Among of the 10 J. curcas populations, the average percentage of polymorphic loci of YNSB was higher than that of the other populations, and it reached 95.45%; On the other hand, that of YNLS was the lowest in all populations, and it was 45.45%. Nei's gene diversity index(H(e)), Shannon information index(I), Effective Num of alleles(A(e)) were respectively 0.4020, 0.576 7, 1.713 6. The total gene diversity (H(T)), the gene differentiation coefficient (G(st)), the gene flow (N(m)) and the gene diversity within populations (H(s)) were 0.443 3, 0.080 2, 3.058 5, 0.405 1, 0.035 7, respectively. The highest gene diversity ratio was showed within populations and the lowest among populations. The results by AMOVA analysis showed that 91.02% of genetic variation existed within populations while 8.98% of genetic variation existed among populations. On the base of the results, the conclusion was extracted that variation existed mainly within populations, and the variation within populations was bigger than that among populations. The result was consistent with that of the gene differentiation coefficient. The order of the genetic diversity was YNLS population < XSBN population < SCHPZ population < SCHD population < SCJH population < YNPR population < SCLB population < YNSB population < YNFY population < SCHL population. The range of Nei's genetic identity and genetic distance of 10 respectively populations were respectively 0.812 7-0.979 8, 0.020 4-0.207 3. All these showed the similarity was higher and there was a close relationship among the 10 respectively populations; Results based on the cluster analysis showed that 10 respectively populations were divided into 2 groups: one was SCJH population and CHPZ population, the other was SCHL population, SCHD population, SCLB population, YNSB population, YNFY population, YNPR population, XSBN population and YNLS population.
CONCLUSIONSignificant genetic diversity was observed among respectively resources in Sichuan and Yunnan. On the other hand, genetic relationship was close between populations.

4.Test and analysis of radioactivity levels in main agricultural production around a uranium mine in Northern Guangxi

Objective To investigate radioactivity levels in the main agriculture products around a uranium mine in Northern Guangxi.Methods The agriculture products and soil samples were collected and analyzed by using HPGe gamma ray spectrometer.Results The specific activity of 226Ra in radish (including leaf),radish leaves and radish,collected in one place,were 45.0,66.7 and 32.3 Bq/kg,respectively.Those of 226Ra and 23SU in the radish soil collected in the same place were 19 672 and 85 917 Bq/kg,respectively.The transfer coefficients of soil-to-radish and soil-to-leaves were 1.61 × 10-3 and 3.40 × 10-3,consistent with those reported in relevant literature.Radioactivity levels in agricultural products in another survey was in consistence with those in the national survey for food products.Radioactivity levels in soil elsewhere near the radish site was consistent with the results of the national soil radioactivive background survey.Conclusions The soil in this place has been contaminated by the nearby uranium mine.It is important to investigate this place further and take the necessary measures.
5.Expression of thyroid stimulating hormone β splice variant in thyroid of mouse with autoimmune thyroiditis

Objective To investigate the expression of marrow-derived thyroid stimulating hormone β (TSHβ) splice variant in thyroid of mouse with autoimmune thyroiditis induced by thyroglobulin(Tg) immunization,and to analyze whether TSHβ splice variant participated in the pathological process of autoimmune thyroiditis.Methods Using random number table,forty-eight mice(24 females and 24 males) of 7 to 8 weeks old with body mass 20 to 25 g were randomly divided into 4 groups(12 females and 12 males in each group) based on body weight and gender.The control group:fed with deionized water; the Tg-treated group(TG):fed with deionized water,and immunized subcutaneously with 0.1 mg Tg at 8 weeks old,boost immunized twice at 11 and 15 weeks old,respectively; the high iodine-treated group (HI):fed with deionized water containing 0.05% sodium iodide (NaI); the Tg combined with HI group (TG + HI):fed with deionized water containing 0.05% NaI,and immunized the same way as the TG group did.Peripheral blood was collected after 8 weeks treatment,which was used for determination of total tetraiodothyronine (TT4),free tetraiothyronine (FT4),total triiodothyronine (TT3) and free triiodothyronine (FT3) with chemiluminescence immunoassay (CIA); thyroid glands were collected to examine the expression of TSHβ splice variant with SYBR Green fluorescent quantitative real-time PCR,and frozen sections were HE stained for observation of histopathological changes of thyroid cells under light microscopy.Results Under naked eyes,the thyroid gland enlarged significantly,and looked dark red in HI and TG + HI groups.Under an optical microscope,thyroid follicular epithelial cells presented cuboidal,with abundant cytoplasm,presented abundant glial in follicular cavity,without lymphocyte infiltration in the control group; in TG group,the thyroid follicular epithelial cells presented cuboidal,with some single scattered lymphocytes; in HI group,colloid volume expansion appeared in thyroid follicles,thyroid follicular epithelial cells presented low cuboidal or flat,with few single scattered lymphocytes; in TG + HI group,most colloid accumulative large follicles presented in thyroid,thyroid follicular epithelial cells presented flat,some destructive thyroid follicular structure and infiltrating lymphocytes appeared.The differences of FT3,TT4,FT4 and TSHβ splice variant between groups were statistically significant(F =4.00,12.54,31.92,214.29,all P < 0.05).Compared to the control group,the serum TT3(nmol/L:0.92 ± 0.07 vs.1.30 ± 0.33,t =-2.24),TT4(nmol/L:1.30 ± 0.33 vs.95.60 ± 14.10,t =-3.02),FT4(pmoL/L:54.07 ± 3.67 vs.154.80 ± 0.01,t =-54.87) and the thyroids' TSHβ(× 10-3:4.11±0.32 vs.8.38 ± 0.22,t =-19.11) were higher in TG group(all P < 0.05) ; the serum TT4(nmol/L:67.75 ± 11.91 vs.45.50± 3.85,t =3.55,P < 0.05) was lower in HI group; the serum FT4(pmol/L:54.07 ± 3.67 vs.139.46 ± 30.00,t =-5.65) and the thyroids' TSHβ splice variant (× 10-3:4.11 ± 0.32 vs.5.33 ± 1.47,t =-5.95) were higher in TG + HI group (all P < 0.05).Conclusions High iodine has aggravated thyroiditis of BALB/c mice induced by Tg immunization; the level of thyroid TSHβ in mice with autoimmune thyroiditis is higher; all of these results indicated that TSHβ is involved in the pathogenesis of autoimmune thyroiditis.

Object To study the influence of main active principles of ZHIGANCAO DECOCTIDN (ZD), glycyrrhizic acid (GA), ginseng total saponin (GTS) and Ophiopogon total saponin (OTS) on electrophysiology of isolated rat myocardium Methods The influence of GA, GTS and OTS on the automaticity, excitability and functional refractory period of isolated rat atrium and papillary muscle were studied in comparison with ZD made free of the above said active ingredients Results The combine use of GA, GTS and OTS significantly decreased the automaticity, inhibited excitaibility and prolonged the functional refractory period of isolated rat atrium; decreased the automaticity and arrhythmia of papillary muscle induced by epinephrine, while ZD made free of GS, GTS and OTS showed much less effect than the intact decoction Conclusion GA, GTS and OTS proved to be the main effective ingredients responsible for the antiarrhythmic activity of ZD
7.An overview of correlations between intestinal flora imbalance and common connective tissue diseases in dermatology

More and more studies have found that intestinal flora is associated with connective tissue diseases. This review summarizes characteristics of intestinal flora and its mechanism of action in connective tissue diseases, and mainly elaborates correlations between intestinal flora imbalance and 3 common connective tissue diseases (lupus erythematosus, systemic scleroderma and Sjogren′s syndrome) , its possible mechanism of action and related hypothesis. Probiotics can regulate the intestinal flora imbalance, and serve as one of treatments for these common connective tissue diseases.
8.Application of narrative medicine courses in standardized residency training in municipal hospital

Objective:To explore the effect of narrative medicine course on the empathy ability and comprehensive ability training of residents in the standardized residency training in municipal hospitals.Methods:A narrative medicine group was set up in a municipal hospital, and a anarrative medical course of 2 months of narrative theory teaching and 4 months of clinical practice education was carried out among 42 residents. The Chinese version of the Interpersonal Response Index Scale (IRI-C) was used to measure the empathy ability at 4 time points (before intervention, 2 months after theory education, 2 months after practice, and 4 months after practice). The effect of narrative medicine courses was evaluated through the satisfaction questionnaire survey, parallel medical record writing and the first-time pass rate of Medical Practitioner Qualification Examination. SPSS 25.0 was used to analyze the variance of one-way repeated measurement.Results:The empathy ability of 42 residents was significantly improved. The empathy score after 2 months after practice was higher than that before intervention and after the theoretical course ( P < 0.05), while the empathy score after 4 months of practice was higher than that before intervention and after the theoretical course ( P < 0.05). The most popular theoretical teaching forms among residents were interactive theoretical teaching (64.29%, 27/42), video viewing (59.52%, 25/42), and situational simulation (52.38%, 22/42). The comprehensive abilities of parallel medical record writing and podium performance among residents were improved. Passing the Medical Practitioner Qualification Examination is a necessary condition for passing the residency training program. The first-time pass rate of the Qualification Examination for the residents was 72.00% (18/25), which was higher than that of the residents who were not recruited during the same period (64.71%, 11/17). Conclusion:Carrying out narrative medicine education is helpful to improve the empathy ability and comprehensive ability of training residents in municipal hospitals.
9.Effect of Euphorbia helioscopia Aqueous Extract on Chronic Obstructive Pulmonary Disease and Precancerous Lesion-associated Proteins of Lung Tissues in Mice Induced by Cigarette Smoke

ObjectiveTo investigate the protective effect and mechanism of Euphorbia helioscopia aqueous extract (EHE) on mice with chronic obstructive pulmonary disease (COPD) and its influence on precancerous lesion-associated proteins in lung tissues induced by cigarette smoke (CS). MethodThe COPD model was induced by CS in 60 mice and the model mice were randomly divided into control group, model group, positive drug group (dexamethasone, 2 mg·kg-1), and low-, medium-, and high-dose EHE groups (1.875, 3.75, 7.5 g·kg-1). The high-performance liquid chromatography (HPLC) method was used to determine the related components in EHE. The changes in end-expiratory pause (EEP), airway resistance (Penh), expiratory flow at 50% vital capacity (EF50), and other pulmonary function indexes were detected by the spirometer. The levels of inflammatory factors, such as interleukin (IL)-2, IL-5, IL-18, IL-17A, and IL-27 in bronchoalveolar lavage fluid (BALF) of mice were detected by high-throughput liquid protein chip technology. Hematoxylin-eosin (HE) staining was used to detect the pathological changes in lung tissues in mice. The content of malondialdehyde (MDA), myeloperoxidase (MPO), and glutathione peroxidase (GSH-Px) in lung tissues was determined by the colorimetric method. The mRNA relative expression of tumor necrosis factor-α (TNF-α), transforming growth factor-β (TGF-β), matrix metalloproteinase-2 (MMP-2), matrix metalloproteinase-9 (MMP-9), and matrix metalloproteinase-12 (MMP-12) was detected by Real-time fluorescence quantitative polymerase chain reaction (Real-time PCR). Immunohistochemistry (IHC) was used to detect the expression of tumor protein (P53) and cell proliferation-associated antigen (Ki67) in lung tissues, and Western blot was used to detect the relative expression of tumor suppressor protein (P16), DNA (cytosine-5)-methyltransferase 1 (DNMT1), and fragile histidine triad (FHIT) in lung tissues. ResultThe results showed that the main compounds in EHE included phenols (gallic acid and protocatechuic acid) and flavonoids (such as hyperoside, rutin, myricetin, naringenin, quercetin, luteolin, kaempferol, and licorice chalcone A), among which gallic acid and rutin were the highest in content. Compared with normal group, model group showed increased levels of EEP, EF50, and Penh (P<0.05), and showed increased MDA and MPO levels (P<0.01) and decreased GSH-Px (P<0.01), and the model group displayed increased levels of IL-2, IL-5, IL-18, IL-17A, IL-27, TNF-α, TGF-β, MMP-2, MMP-9, and MMP-12 (P<0.05). And the model group exhibited up-regulated expression of P53, Ki67, and FHIT in lung tissues (P<0.01) and down-regulated expression of DNMT1 and P16 (P<0.01). Compared with model group, the EHE groups showed decreased EEP and EF50 levels (P<0.05). The pathological injury of lung tissues in mice of the model group was observed under HE staining, and the pathological injury of basal cell hyperplasia of lung tissues was gradually improved after treatment with EHE. The EHE groups showed reduced levels of MDA and MPO (P<0.01) and increased GSH-Px (P<0.01). The EHE groups displayed decreased levels of IL-2, IL-5, IL-18, IL-17A, IL-27, TNF-α, TGF-β, MMP-2, MMP-9, and MMP-12 (P<0.05). And the EHE groups showed down-regulated Ki67 and FHIT in lung tissues (P<0.05) and up-regulated expression of P53 and DNMT1 (P<0.05). ConclusionEHE can protect mice from COPD and inhibit precancerous lesions, and the mechanism may be related to the inhibition of inflammation and oxidative stress response, regulation of protease and antiprotease imbalance, and regulation of epithelial cell growth.

BACKGROUNDBased on the excellent medical care and management system for Chinese veterans, as well as the detailed medical documentation available, we aim to construct a Chinese Veteran Clinical Research (CVCR) platform on non-communicable diseases (NCDs) and carry out studies of the primary disabling NCDs.
METHODSThe Geriatric Neurology Department of Chinese People's Liberation Army General Hospital and veterans' hospitals serve as the leading and participating units in the platform construction. The fundamental constituents of the platform are veteran communities. Stratified typical cluster sampling is adopted to recruit veteran communities. A cross-sectional study of mental, neurological, and substance use (MNS) disorders are performed in two stages using screening scale such as the Mini-Mental State Examination and Montreal cognitive assessment, followed by systematic neuropsychological assessments to make clinical diagnoses, evaluated disease awareness and care situation.
RESULTSA total of 9 676 among 277 veteran communities from 18 cities are recruited into this platform, yielding a response rate of 83.86%. 8 812 subjects complete the MNS subproject screening and total response rate is 91.70%. The average participant age is (82.01±4.61) years, 69.47% of veterans are 80 years or older. Most participants are male (94.01%), 83.36% of subjects have at least a junior high school degree. The overall health status of veterans is good and stable. The most common NCD are cardiovascular disorders (86.44%), urinary and genital diseases (73.14%), eye and ear problems (66.25%), endocrine (56.56%) and neuro-psychiatric disturbances (50.78%).
CONCLUSIONWe first construct a veterans' comprehensive clinical research platform for the study of NCDs that is primarily composed of highly educated Chinese males of advanced age and utilize this platform to complete a cross-sectional national investigation of MNS disorders among veterans. The good and stable health condition of the veterans could facilitate the long-term follow-up studies of NCDs and provide prospective data to the prevention and management of NCDs.
