1.Structure, receptor and pathophysiological function of natriuretic peptide
Chinese Journal of Pathophysiology 2000;0(08):-
Five natriuretic peptides (NP) have been identified: A-type, B-type, C-type, D-type and V-type. All of them have ringed-structures. The actions of the natriuretic peptides are modulated through their cognate receptors, which are named as NPR-A,NPR-B and NPR-C. The receptors are involved in the cGMP-dependent signaling cascades, which take part in many different physiological regulation, such as diureses, vasodilatation, depressing blood pressure, improving immunity, promoting development of bone, regulating nerve system and so on. At present, ANP and BNP have become a marker in diagnostic some cardiovascular diseases.

To study the antiviral effect of Hypericum perforatum L. extract (HPE) on influenza A virus (IAV) (H1N1) in vitro and in vivo. Cytopathic effect (CPE) and neutral red (NR) dye uptake were used to examine the antiviral effect of HPE on Madin Darby Canine Kidney (MDCK) cells which were infected with IAV in vitro. HPE was effective against influenza A virus (IAV) in vitro, with a 50% effective concentration (EC50) of 40 μg/mL. The mean 50% cytotoxic concentration (CC50) in the MDCK used in these experiments was 1.5 mg/mL. Ribavirin was run in parallel with EC50 values of 5.0 μg/mL; the mean CC50 for ribavirin was 520 μg/mL. Oral gavage administrations of HPE or ribavirin to mice infected with the IAV were highly effective in preventing death, slowing the decline of arterial oxygen saturation, inhibiting lung consolidation and reducing lung virus titers. The minimum effective dose of HPE in these studies was 31.25 mg/kg/day, which was administered twice daily for 5 d beginning 4 h prior to virus exposure. Below a dosage of 2000 mg/kg/day, almost all treated mice survived, which suggests that HPE is of low toxicity. Ribavirin's minimum effective dose was 40 mg/kg/day with the LD50 determined to be 200 mg/kg/day. Delay of the initiation of either HPE or ribavirin therapy, using approximately 1/3 LD50 dose each time, could still be protective as late as 48 h after exposure to the IAV. While both agents appeared to have similar efficacy against IAV infections, HPE was considered to be less toxic and may warrant further evaluation as a possible therapy for influenza.

Objective: To detect the damage of excessive iodine on the activities and mRNA of cathepsin B and D and observe the effects of selenium intervention. Method: Thyroid follicular epithelial cell line FRTL was cultured in Kaighn’s modified Ham’s F-12 medium. ① The cells were divided into 8 groups for detection of activities of cathepsin B and D (CB and CD). The medium of NI group did not contain selenium or iodine, while EI group contained 50 mmol/L KI, and IS group contained 50mmol/L KI and different dosages of 0.1,0.2,0.5,1.0,2.0 and 5.0 ?mol/L selenium (sodium selenite). ②The mRNA levels of cathepsin B and D were detected by real-time PCR. The medium of NI group and EI group was same as above, but IS group only contained 0.1?mol/L selenium besides 50mmol/L KI. Results: The activities of CB decreased in EI group (50mmol/L KI treated 12 h), while CD decreased after 24 h and both activities were improved in IS groups with 0.1~0.5?mol/L selenium. The mRNA levels of CB and CD in EI group decreased, while in IS group were improved. Conclusion: Excessive iodine resulted in goiter through decreased levels of activities and mRNA of CB and CD. The intervention of selenium on goiter mayinduce its amelioration effect on decrease of CB and CD activities and the optimal dosage ranges from 0.1?mol/L to 0.5?mol/L.
