Objective To investigate the effect of different delivery modes and related obstetric factors on the short-term function of the pelvic floor.Methods One hundred and twenty healthy primiparae women were interviewed at 6-8 weeks postpartum,with 72 women in the vaginal delivery group and 48 women in the elective cesarean section group.Questionnaire on stress urinary incontinence and measurement of diastolic and contractive function of the pelvic floor muscles by electromyogram(EMG)were used for the evaluation and comparison.Results The prevalence of stress urinary incontinence in the vaginal delivery group and the elective cesarean section group was 21% and 10%(P=0.134),respectively.The values of the right act,right work,and average work surveyed by EMG in vaginal delivery group were significantly lower than those in cesarean section group(right act,12.9?0.8 vs 17.3?1.7,P
2.Preparation and Identification of High Immunogenic A/PR/8/34 Maternal Strain HA Protein for Influenza Virus Classical Reassortment.

Preparation of maternal strain A/PR/8/34 HA antiserum for influenza virus classical reassortment. A/PR/8/34 virus was digested by bromelain after inactivation and purification. 5%-20% sucrose continuous density gradient centrifugation method was used to purify HA protein. SIRD method was used to select the target protein. SDS-PAGE method was used to identified HA protein. High Immunogenic A/PR/8/34 HA protein was successfully prepared and HI titer reached 10240. High purity HA antiserum was identified by SIRD method. The key reagent in the classical reassortment of influenza virus was prepared, and the complete set of technical methods were explored, which laid the foundation for the independent research and development of seasonal influenza vaccine strains of China.

3.Analysis on application effect of scale score of treatment compliance of pulmonary tuberculosis patients in Guizhou province

Objective To verify the accuracy of ARS scale.Methods A cohort study was conducted in 10 counties (city)the in-troduction of the ARS scale from 2013 November to 2014 May (6 months),the registrations were undertake score,and score < 4 were compliance group,more than 4 cent were nonadherence group.Follow-up observation include review at the end of 2nd month after treatment and medicine compliance.Results There was no significant difference in age,gender,nationality and whether the floating population between the compliance group and non compliance group (P >0.05),initial treatment and retreatment was sta-tistically significant (P <0.05).1 824 patients had a mean score of 3.40±1.40,including compliance group mean score was 2.34± 0.73,is not in compliance with group mean score was 4.69±0.81.Mainly concentrated in the 2-5 scores accounted for 86%.The ARS score of 2 at the end of the packet and judge review the results of test,the Kappa coefficient was 0.403 (P <0.05).The sensi-tivity was 71.1%,specificity was 70.2%,the correct index 0.41.Multivariate Logistic and review the compliance of patients re treatment ARS score,at the beginning of the results of regression analysis showed that ARS score,under the same conditions,re-treatment review compliance was initially treated patients with difference (OR=0.41,P <0.05);in the initial treatment and retreat-ment premise under the same circumstances,review of compliance with the ARS score less than 4 points.It′s 5.65 times greater than or equal to 4 points (P <0.05).Conclusion The ARS scale can accurately judge the patient compliance,with high sensitivity and specificity,suitable for application in Guizhou Province,but we should consider other interference factors of promotion.
4.Effect of Sinomenine on Expression of Purinergic Receptors A2A and P2X7 in Mouse Model and In-vitro Macrophages Stimulated by Lipopolysaccharide

Objective To investigate the effect of sinomenine on the purinergic receptors A2A and P2X7 in endotoxemia mouse model and RAW264.7 macrophage model stimulated by lipopolysaccharide(LPS). Methods BALB/c mice were randomly divided into blank control group, model group and sinomenine group. Thirty minutes after the rats of sinomenine group were pretreated with intraperitoneal injection of sinomenine (40, 80, 160 mg/kg), the mice were given intraperitoneal injection of 15 mg/kg LPS to induce endotoxemia model. The serum levels of tumor necrosis factor-alpha(TNF-α) and interleukin-6(IL-6) were measured by enzyme-linked immunosorbent assay (ELISA). The expression levels of purinergic receptor A2A and P2X7 in the liver and spleen were detected by reverse transcription-polymerase chain reaction(RT-PCR). RAW264.7 macrophages were divided into blank control group, LPS group and sinomenine group. Sinomenine group was firstly treated with sinomenine(300μmol/L) for 2 h, and then LPS group and sinomenine group were treated with LPS (100 ng/mL) for another 8 hours. TNF-α in the cell supernatant was measured by ELISA, and the expression levels of A2A and P2X7 in RAW264.7 cells were detected by RT-PCR. Results Stimulation with LPS could induce the increase of the mouse serum levels of TNF-α and IL-6 as well as the expression of A2A and P2X7 in mouse liver and spleen, and sinomenine had a counteraction on the above indexes(P<0.05) . In-vitro stimulation with LPS could induce the increase of the content of TNF-α and the expression of A2A and P2X7 in RAW264.7 cells , and sinomenine decreased TNF-α content and P2X7 expression (P<0.05) , but had an effect on enhancing A2A expression. Conclusion Sinomenine suppresses the expression of purinergic receptor P2X7 in mouse spleen and liver as well as in RAW264.7 macrophages, but its effect on the expression of A2A in various tissues and cells varies, whose related mechanism is needed further study.
5.Clinical analysis of fallopian tube prolapse after hysterectomy

Objective To investigate the clinical diagnosis,treatment and prevention of fallopian tube prolapse(FTP)after hysterectomy.Methods A total of 7949 patients received hysterectomy from 1983 to Aug 2005 in Peking Union Medical College Hospital,including 6229 cases of trans-abdominal hysterectomy(TAH),780 cases of transvaginal hysterectomy(TVH),and 940 cases of laparoscopic assisted vaginal hysterectomy(LAVH).Nine cases(including 1 case from other hospital)of FTP after hysterectomy were analyzed retrospectively for their symptoms,diagnosis and treatment.All of them were diagnosed according to the results of histology and follow-up.Results The overall incidence of FTP after hysterectomy was 0.11%(9/7949).Incidence of FTP after trans-abdominal hysterectomy was 0.08% (5/6229),after vaginal hysterectomy 0.51%(4/780),and after laparoscopic assisted vaginal hysterectomy 0(0/940).There were no symptoms in 3 cases,but the other 6 cases had symptoms.The pelvic examination revealed the typical prolapsed fimbrial end of a fallopian tube in 3 cases and red granulation tissue in the other 6 cases.All of them were excised vaginally and cauterized.The results were confirmed by histological examination.No recurrent cases were reported in follow up.Conclusions FTP is a rare complication after hysterectomy.The prognosis is well after proper diagnosis and treatment.Salpingectomy or fixation of accessories into the pelvic wall and complete peritonealisation at the time of hysterectomy are important methods to prevent FTP after hysterectomy.

Objective To establish the chick embryo chorioallantioc membrane(CAM)as a model for in vivo research on endometriosis.The model was used to investigate the mechanism of anti-vascular endothelial growth factor(VEGF)antibody for treatment of endometriosis.Methods Human endometrial fragments were explanted onto the CAM.Then anti-VEGF antibody was used for the endometriosis-like lesions after transplantation of human endometrial fragments.The CAM models were treated respectively as control groups and experimental groups.The terminal deoxynucleotidyl transferase-mediated biotin- deoxyuridine triphosphate(dUTP)nick end labeling(TUNEL),proliferating cell nuclear antigen(PCNA) and microvessel density(MVD)were used in vivo for analysis of anti-angiogenesis.Results The apoptosis intensity of anti-VEGF antibody treated groups(6.7?0,9,6.9?0.8)was significantly higher than that of the control groups(5.0?0.9,5.4?1.1;P
7.Postoperative evaluation of tension-free vaginal tape procedure.

OBJECTIVETo study the outcome of tension-free vaginal tape (TVT) for the treatment of stress urinary incontinence (SUI) in women with cystocele.
METHODSForty-two patients with SUI confirmed by urodynamics underwent the TVT procedure under local anesthesia. A prolapse repair was done simultaneously.
RESULTSMean TVT operation time was 26.29 minutes. Mean blood loss was 29.86 mL. Eighty-eight percent of the
PATIENTSwere able to micturate spontaneously within 12 hours and residual urine was less than 100 mL. And 12% of the patients had to use indwelling catheter for 3-11 days. Average hospital stay was 2.91 days. Eighty-eight percent of patients were discharged within 2 days. All patients were followed up (an average of 10.26 months). According to subjective and objective assessment of the outcome, 39 patients (93%) were cured, another 3 patients (7%) were significantly improved and none was failed. There were no major complications such as bladder injury occurred.
CONCLUSIONTVT procedure is a minimal invasive, effective, and safe surgery for treatment of SUI.

8.Estrogen receptors in pelvic floor for female stress urinary incontinence.

OBJECTIVETo observe the statuses of estrogen receptor (ER) alpha and beta in the pelvic floor and its relation with stress urinary incontinence (SUI).
METHODSThe ERa and ERP in levator ani muscle and pelvic floor structure of premenopausal and postmenopausal SUI patients were detected by immunohistochemical staining and Western blot.
RESULTSThe positive biopsy rates in the premenopausal and postmenopausal SUI groups were 11% and 0, and the corresponding results in the control group were 35% and 33%. No ERalpha and ERbeta expression was detected in the levator ani muscle. ERalpha and ERbeta located in pelvic fasia tissue around the levator ani muscle. ERbeta was negatively stained in some segments of pelvic fasia tissue. The positive expression rates of ERbeta in the pelvic fasia tissue of premenopausal and postmenopausal SUI group were 57% and 33%, which were significantly lower than those in the control groups (P < 0.05). The positive expression rate of ERP in the pelvic fascia was significantly lower in postmenopausal group than in the premenopausal group (P < 0.05). The positive expression rates of ERa in the pelvic fasia tissue in the premenopausal and postmenopausal SUI groups were (4.43 +/- 2.64)% and (5.14 +/- 1.79)%, which were significantly lower than (9.61 +/- 5.48)% and (10.88 +/- 2.90)% in control group (P < 0.05). Western blot showed that ERbeta expression in pelvic fasia tissue was less than the expressions of ERalpha, ERalpha and ERbeta in the SUI group was also lower than that of the control group.
CONCLUSIONERalpha and ERbeta do not express in the levator ani muscle of women. Pathogenesis of premenopausal SUI correlates with lower serum estrodiol level and the expression of ER correlates SUI.

9.Correlation analysis between meteorological factors, biomass, and active components of Salvia miltiorrhiza in different climatic zones.

In this study, the growth and accumulation of active components of Salvia miltiorrhiza in twenty two experimental sites which crossing through three typical climate zones. The S. miltiorrhiza seedlings with the same genotype were planted in each site in spring, which were cultivated in fields with uniform management during their growing seasons till to harvest. The diterpene ketones (dihydrotanshinone, cryptotanshinone, tanshinone I and tanshinone II(A)) in S. miltiorrhiza root samples were determined by using high-performance liquid chromatography (HPLC) method. The biomass of root (root length, number of root branches, root width and dry weight) was also measured. The results showed that tanshinone II(A) in all samples of each site were higher than the standards required by China Pharmacopoeia. It has been found there is a relationship between root shape and climate change. The correlation analysis between active components and meteorological factors showed that the accumulation of tanshinones were effected by such meteorological factors as average relative humidity from April to October > average vapor pressure from April to October > average temperature difference day and night from April to October > annual average temperature and so on. The correlation analysis between root biomass and meteorological factors exhibited that root shape and accumulation of dry matter were affected by those factors, such as average annual aboveground (0-20 cm) temperature from April to October > annual average temperature > average vapor pressure from April to October > annual active accumulated temperature > annual average temperature > average vapor pressure from April to October. The accumulation of tanshinones and biomass was increased with the decrease of latitude. At the same time, the dry matter and diameter of root decreased if altitude rises. In addition, S. miltiorrhiza required sunlight is not sophisticated, when compared with humid and temperature. To sum up, S. miltiorrhiza can adapt to a variety of climatic conditions and the southern warm humid climate is more conducive to its growth and accumulation of active components.

10.Estrogen induced rat model of uterine leiomyoma.

OBJECTIVETo establish an appropriate animal model of uterine leiomyoma and to understand the pathogenesis of this disease.
METHODSMature female rats were intramuscularly injected with estradiol benzoate at 200 μg or 300 μg twice a week. After injection for 8 or 10 weeks, the rats were sacrificed. We measured the serum levels of estrogen (E(2)) and progesterone (P), evaluated ER and PR expression, and calculated the leiomyoma forming rate and mortality of the rats. Histological changes were compared between rat uterine leiomyoma and human uterine leiomyoma with HE staining. The optimal dose and duration of E(2) for induction of uterine leiomyoma in rat were determined.
RESULTSIn the rats treated with estradiol benzoate 200 μg for 8 weeks ìn the serum E(2) level increased significantly (P<0.01). Uterine nodules were visible in some of the tested rats. Based on the pathohistological Results , the uterine leiomyoma developed in the treated rats demonstrated similar features as in human uterine leiomyoma. The expressions of ER and PR were increased in the leiomyoma tissues.
CONCLUSIONThe rat model of uterine leiomyoma can be established by intramuscular injection of estradiol benzoate at 200 μg twice per week for 8 weeks, with similar features as those of human uterine leiomyoma. The high concentrations of ER and PR in uterine tissue might be related with the development of uterine leiomyoma in animal.
