Objective To establish the relationship of serum TNF-? and IL-6 contents with myocardial cell apoptosis in the chronic heart failure (CHF)rats, and to explore the therapeutic mechanism of Anxin Granules (AXG)in treating CHF.Methods Fifty female SD rats were randomly divided into five groups:normal control group (A),model group (B),captopril group(C),low-dosage AXG group (D)and high-dosage AXG group (E).CHF models were established by intraperitoneal injection of small-dose of adriamycin, once per week for continuous 6 weeks.Meanwhile, group C was given captopril injection (6.25mgkg-1d-1),and groups D and E received gastric gavage of low-dosage AXG (0.8 gkg-1d-1)and high-dosage AXG (1.6gkg-1d-1)respectively.After 6 weeks of modeling, serum levels of TNF-? and IL-6 of different groups were tested by ELISA,the apoptosis of myocardial cells was tested by TUNEL,and then the apoptosis index(AI)was counted. Results Serum levels of TNF-? and IL-6 were increased in the model group (P
2.Intervention Effect of Retinoic Acid on Hyperoxic Lung Fibrosis in Neonatal Rat
Journal of Applied Clinical Pediatrics 1992;0(06):-
Objective To explore intervention effect of retinoic acid(RA) on hyperoxic lung fibrosis in neonatal rat,and to observe the role of transforming growth factor-?1(TGF-?1)and ?-smooth muscle actin(?-SMA)in hyperoxic lung fibrosis. Methods The SD neonatal rats were randomly divided into 4 groups:air-exposed control group(group Ⅰ),air-exposed and RA-treated group(group Ⅱ),hyperoxia-exposed control group(group Ⅲ),hyperoxia-exposed and RA-treated group(group Ⅳ).The rats of group Ⅲ,Ⅳ were kept in chambers containing 850 mL/L oxygen, the other 2 groups were exposed to air.The rats of group Ⅱ,Ⅳ were intraperitoneally injected with RA [500 ?g/(kg?d)],group Ⅰ,Ⅲ were intraperitoneally given the same dose of oleum lini.At the end of exposure, the lung histophatholoical changes and radical alveolar counts(RAC) were observed by HE staining under light microscope, the degree of pulmonary fibrosis was evaluated by Masson trichrome method and fibrosis score. The protein expression of TGF-?1 and ?-SMA were determined by immunostaining. Results At 14 d of exposure, group Ⅲ resulted in a significant increase in fibrosis score and expressions of TGF-?1 and ?-SMA compared with group Ⅰ,Ⅱ and Ⅳ(Pa0.05).Conclusion TGF-?1 and ?-SMA may have important role in hyperoxic induced lung fiborsis injury, the earlier period intervention of RA can reduce lung fibrosis regeneration and exceptional alveolus development.

Objective To investigate the clinical features and treatment of a group of patients of mitochondrial encepha-lomyopathy with actic acidosis and stroke (MELAS) with onset of status epileptics. Methods Clinical features, EEGs, image ifndings, and therapeutic data of 4 cases with onset of status epileptics patients ifnally diagnosed as MELAS were retrospectively reviewed. Results Four Patients were onset with status epileptics. The levels of serum lactic acid, ammonia, myocardial enzymes were increased, and the serum sodium level was reduced, and accompanied with metabolic acidosis. EEG found corresponding paroxysmal and interictal activities. Brain images showed basal ganglia calciifcation, brain atrophy, and acute cortex edema. Genetic detection found mtDNA3243 mutation. Conclusions The status epilepticus was commonly present in MELAS. The treatment of epileptic attack in this disease was dififcult, which needs early diagnosis. Appropriate anti-leptic drugs and relevant treatment to symptoms are important to alleviate cerebral injury.

Objective To evaluate the distribution of neuropeptide Y and its receptors in the cardinal ligament and uterosaeral ligaments in women with and without pelvic organ prolapse(POP).Methods Sixteen patients with pelvic organ prolapse entered the study. All patients were evaluated by pelvic organ prolapse quantitation(POP-Q).Group A consisted of six patients with grade Ⅰ,Ⅱ POP,and group B comprised ten patients with grade Ⅲ,Ⅳ POP.Eight nonfunctional ovarian tumor patients without POP were recruited as control subjects.Biopsies of cardinal ligament and uterosacral ligament were obtained from each woman during surgery.Immunohistochemical study with polyclonal antibody against a general nerve marker S-100 and neuropeptide Y was performed on paraffin-embedded sections of all the samples. In addition,mRNA levels of the human NPY-Y1 and NPY-Y2 receptors were assessed in both patients and controls. Results (1)NPY immunoreactivities were identified in both cardinal ligament and uterosacral ligament. NPY immunoreactive nerve fibers were insignificantly lower in POP patients(P>0.05).The distribution pattern of NPY was similar in cardinal ligament and uterosacral ligament ( P>0. 05 ). (2)mRNAs encoding the NPY-Y1 and NPY-Y2 receptors were detected in the pelvic supporting tissues. Besides the expected NPY-Y1 PCR products, an additional 97 bp long amplicon originating from an alternative splicing event was found in most tissues studied. (3)In cardinal ligaments, mRNA encoding NPY-Y1 receptor had a significant difference between group A(3.9±1.0)and B (6. 0±1.5), and between control (3.4±0.9) and group B (P = 0. 019,P = 0. 004), while there was no significant difference between group A and controls(P =0. 082). In uteresacral ligaments, mRNA encoding NPY-Y1 receptor had no significant difference between Group A(6. 0±1.1) and B (6. 3±0. 7), or between group A and controls(4. 8±0. 7;P = 0. 151 ,P = 0. 690);while there was a significant difference between group B and controls (P = 0. 016).(4) mRNA encoding NPY-Y2 receptor had no significant difference between controls (0. 49±0. 34, 0. 61±0. 15 ), group A (0. 56±0. 21,0. 67±0. 13) and group B (0. 85±0. 43, 0. 69±0. 21 ) patients in cardinal ligament and uterosacral ligaments ( P>0. 05 ). (5) mRNA encoding NPY-Y1 ( P = 0. 084 ) and NPY-Y2 (P=0.470) receptors had no significant difference between cardinal ligament and uterosacral ligament. Conclusions There are NPY and NPY receptors in cardinal and uterosacral ligaments. The increased expression of NPY Y1 receptor may be related to local blood flow reduction and structural changes of pelvic supporting tissue.

Acta Pharmaceutica Sinica 2014;49(5):622-626
Hypoxia-inducible factor-1 (HIF-1) is a key transcription factor on hypoxia responses in mammalian tissues. HIF-1 plays as a positive factor in solid tumor and leads to hypoxia-driven responses that enhance its downstream gene expression for tumor growth and survival. LXY6099 was obtained by the structural modification and optimization of manassantin A (MA) as a high potent HIF-1 inhibitor. Antitumor activity of LXY6099 was observed in this study. LXY6099 with an IC50 value of 2.46 x 10(-10) mol x L(-1) showed more sensitive inhibition activity to HIF-1 than that of MA detected by reporter gene assay (> 100 folds). It showed strong inhibition on the growth of human solid tumor cell lines. Furthermore, LXY6099 exhibited significant antitumor activity against established human tumor xenografts in nu/nu mice with treatment of MX-1 breast cancer. Thus, LXY6099 as a novel HIF-1 inhibitor could be further developed into anti-cancer agents.
