1.Effect of sevoflurane pretreatment on expression of cystathionine β-synthase and heme oxygenase-1 during myocardial ischemia-reperfusion injury in rats

Objective To investigate the effect of sevoflurane pretreatment on the expression of cystathionine β-synthase(CBS)and heme oxygerase-1(HO-1)during myocardial ischemia-reperfusion(I/R)injury in rats.Methods Thirty adult male SD rats,weighing 180-220 g,were randomly divided into 3 groups(n =10each):sham operation group(group S),I/R group and sevoflurane group(group Sev).Myocardial I/R injury was produced by ligation of the left anterior descending branch of coronary artery for 30 min followed by 2 h reperfusioo.In group Sev,sevoflurane was inhaled before ischemia,the end-tidal concentration was 1.5%-1.7% and myocardial I/R was produced 60 min later.The rats were sacrificed at 2 h of reperfusion and myocardial tissues were taken for determination of the contents of MDA,GSH,H2S and CO,SOD activity and expression of CBS mRNA and HO-I mRNA.The ultrastructure of myocardium was examined with electron microscope.Results Compared with group S,the contents of MDA,H2S and CO were significantly increased,the expression of CBS mRNA and HO-1 mRNA was up-regulated,and SOD activity and GSH content were significantly decreased in groups I/R and Sev(P < 0.05).Compared with group I/R,the contents of MDA,H2S and CO were significanfly decreased,the expression of CBS mRNA and HO-1 mRNA was down-regulated,SOD activity and GSH content were significantly increased in group Sev.The damage to mitochondrial structure induced by I/R was mitigated by pretreatment with sevoflurane.Conclusion The mechanism by which sevoflurane pretreatment reduces myocardial I/R injury is related to down-regulation of the expression of CBS and HO-1 and decrease in the activities in rats.
2.The effects of implant morphology on stress distribution in implant-bone interface

Objective:To investigate the effects of different shapes of implant on stress distribution in implant-bone interface and to select the most appropriate shape for orthodontic anchorage. Method:3-dimesional finite element analysis method was used to analyze the stress and strain in implant-bone interface of knife-edged threaded, square threaded and smooth type implants.Results: The first,second and third main stress (MPa) of knife-edged threaded implant were 6.67, 1.47 and 0.52 respectively with 7.72 MPa of Von Mises stress;those of square threaded 13.00,2.51,0.57 and 11.8;smooth type 10.50,2.50,0.67 and 12.60,respectively. The variables of alveolar bone at the cervix of knife-edged threaded,square threaded and smooth type implants were (0.11×10-3), (0.13×10-3) and (0.94×10-4) mm respectively. Conclusion: The stress in knife-edged threaded implant-bone interface and alveolar bone deformation at the cervix of the implants is minimal,therefore the implants are more suitable for orthodontic anchorage.
3.Establishment of three-dimensional finite element model for implant as orthodontic anchorage

Objective:To establish a 3-dimensional finite element model for implant in molar deprived region of mandible. And to lay basis for precise analysis for the biomechanic characteristics of implant anchorage system. Methods:The cross section outline drawn from the molar deprived region of mandible was input into a computer, and ANSYS 5.5 (Swanson Analysis Systems, Inc. Houston, USA) finite element analysis software was used to perform the finite element modeling of the mandible.Results:The cross section outline of the mandible,imitated by a spline curve was smooth and lifelike. This cross section outline was extruded into a three dimensional model. The three dimensional models of the implant and mandible could be smartly meshed to obtain the finite element model of mechanics. Conclusion: An effective 3-dimensional finite element biomechanical analysis model for implant as orthodontic anchorage was established.
4.Effects of high insulin on chemotherapeutic sensitivity in MCF-7/ADR cells

Objective To analyse the effects of high insulin on the expression and function of P-glycoprotein (P-gp), and preliminarily investigate the influence of insulin on chemotherapeutic sensitivity in MCF-7/ADR cells. Methods MCF-7/ADR cells were cultured with different concentrations of insulin(0.001, 0.005, 0.01, 0.05 and 0.1μmol/L). Real-time PCR was used to detect the expression of P-gp mRNA. Western blot assay was used to detect the expression level of P-gp. Rhodamine 123 was used to detect the efflux function level of P-gp. Cell viability and chemotherapeutic sensitivity were detected by MTT assay. Results High concentration of insulin (0.1 μmol/L) promoted the proliferation of MCF-7/ADR cells. The concentration of insulin (0.05 and 0.1 μmol/L) accelerated P-gp mRNA and protein expression, which also augmented the efflux function of P-glycoprotein and reduced the chemotherapeutic sensitivity to epirubicin. Conclusion High concentration of insulin may influence the drug resistance of breast cancer cells by promoting the expression and function of P-glycoprotein of MCF-7/ADR cells.
6.Negative immune regulatory molecule TIPE2 for treating SLE mice through regulating macrophage subtype

Objective To investigate the role of tumor necrosis factor (TNF)-alpha-induced protein 8-like 2 (TIPE2) for regulating the macrophage polarization in systemic lupus erythematosus and its curative effects on experimental SLE mice.Methods The mice were treated with activated lymphocytes derived DNA (ALD-DNA) for inducing mice model,randomly divided into AAV-scr control group and AAV-TIPE2 experimental group,and injected with AAV-TIPE2 or AAV-scr virus solution from the tail vein of mice.The expression of TIPE2 mRNA and protein in polarized macrophages,serum dsDNA antibody titer,urine protein and renal pathological index were detected.Results (1) The TIPE2 expression level of TIPE2 mRNA and protein in AAV-TIPE2-transfected cells was 13.5±1.6 times and 10.8±1.6 times of AAV-scr control group respectively.(2) M2 macrophage specific molecule MGL+ was 59.6% in AAV-TIPE2 group and MGL + cells in the AAV-scr group was 8.4%.M2/M1 odds ratio of AAV-TIPE2 experimental group to AAV-scr control group was 16.(3) The recombinant TIPE2 adenovirus related vector could stably expressed in transfected HEK-293.In vitro and in vivo experiments confirmed that AAV-TIPE2 was able to induce M2 polarization of macrophages in ALD-DNA-induced lupus mice.(4) The serum anti-dsDNA antibody,urinary protein and renal pathology in the AAV-TIPE2 group were significantly lower than those in the AAV-scr group(P<0.01).Conclusion TIPE2 alleviates the disease condition of ALD-DNA induced SLE mice through induction of macrophage polarization to M2 phenotype,which may be used as a promising therapeutic method for ALD-DNA induced SLE mice.
7.Genetic cloning and sequence analysis of farnesyl pyrophosphate synthase in Panax notoginseng

Objective To clone and sequence the cDNA encoding farnesyl pyrophosphate synthase(FPS) from Panax notoginseng.Methods The cDNA,encoding FPS in P.notoginseng,was amplified by RACE strategy with the total RNA of root as the template.The fragment of FPS was cloned and sequenced.Results The analysis results revealed that the full-length cDNA had(1 409) bp with an open reading frame encoding 343 amino acids of protein.The FPS sequence had 95%,87%,and 86% amino acid sequence homology to the FPS sequence of Centella asiatica,Parthenium argentatum,and Artemisia annua,respectively. Conclusion The cDNA encoding FPS from P. notoginseng is cloned and reported.This works provide a foundation for exploring the mechanism of saponins biosynthesis and application to the other medical plants.
8.Clinical observation of preventive hepatoprotective effect of traditional Chinese medicine on drug -induced liver injury induced by chemotherapy of breast cancer

Objective To assess the efficacy and significance of hepatic protection by observing the incidence of liver injury in patients with chemotherapy who were treated and untreated with traditional Chinese medicine (TCM).Methods A retrospective analysis was applied to compare the changes in liver function tests and incidence rate of drug -induced liver injury between the observation group (treated with TCM)and the control group (untreated with TCM)after chemotherapy.Results The incidence rates of liver injury in the observation group and control group was 4.7%(2 /43)and 23.9%(11 /46)respectively.The difference between the two groups was statisti-cally significant(χ2 =6.611,P =0.01).The ALT,AST,TBIL,ALP between the two groups had statistically signifi-cant differences after treatment (all P <0.05).Conclusion Using TCMin hepatic protection can improve liver tests and reduce the incidence of drug -induced liver injury from chemotherapy.
9.Effect of vascular endothelial growth factor on apoptosis of endothelial cells induced by hypoxia

Objective To explore the effect of hypoxia on the apoptosis of cultured human umbilical vein endothelial cells(HUVECs) and the role of vascular endothelial growth factor(VEGF) in inhibition of apoptosis. Methods ①Culture and identification of HUVECs.②Establishment of hypoxic model(0,12,24,48 h)in HUVECs.③Incubation of HUVECs with VEGF(0 ng, 100 ng) under hypoxic condition for 24 h. ④Detection of apoptosis of HUVECs with TUNEL method. Results The percentages of apoptosis were different under different hypoxic conditions. The longer hypoxic time was,the higher apoptosis percentage was.VEGF reduced the apoptosis of HUVECs induced by hepoxia. Conclusion Over-apoptosis EVCs in one of the important factors for the impairment of endothelial function. HEGF inhibits the apoptosis of HVCs and having a pretive function on them.
10.Analysis of prevalence and risk factors of pelvic organ prolapse of women underwent gynecologic health care in Peking Union Medical College Hospital

Objective To investigate prevalence and risk factors of pelvic organ prolapse in women underwent routine gynecologic health care in Peking Union Medical College Hospital (PUMCH).Methods From Jan.2008 to Aug.2009,972 women underwent gynecological health care in PUMCH Were enrolled in this study.Questionnaires and pelfic examinations were given.The pelvic organ prolapse quantitive examination(POP-Q)system was used as the assessment tool.Results (1)Among all participants,the mean ages were(42±10)years(range 22 to 78 years),the mean height were(162±5)cm(range 142 to 180 cm),and the mean weight were(59±8)kg(range 42 to 91 kg).83.8%(815/972)of women were multipara.The mean total vagihal length(TVL)of 972 women was 8.20 cm.No women met the standard of pelvic organ prolapse, while 35.5% (345/972) of women presented mild posterior vaginal descent and 96. 7% (940/972) presented mild anterior vaginal descent, all of them were asymptomatic. (2) The length of genital hiatus (gh), TVL and C, D proximal to the hymen in nullipara were (2.26 ±0. 32), (8.08 ±0. 30), ( - 7.08 ± 0. 24) and ( - 8. 08 ± 0. 30) cm, which were significantly less than ( 2. 33 ± 0. 39 ),( 8. 22 ± 0. 35 ), ( - 7. 14 ± 0. 28 ) and ( - 8. 22 ± 0. 35 ) cm in multipara ( P < 0. 05 ). Ap and Pb proximal to the hymen of ( - 2. 87 ± 0. 22) and ( - 2. 87 ± 0. 22 ) cm in nullipara were significantly larger than ( -2.81 ±0.25) and ( -2.81 ±0.25) cm in multipara (P<0.05). When compared with nullipara, the incidence of posterior and anterior vaginal wall protrusion were increased ( OR = 1. 819). (3) The index of POP-P were compared among women at groups of 22 -34 years, 35 -49 years and more than 50 years (P <0. 05 ). Those index did not show statistical difference between women at group of 22 - 34 years and group of 35 -49 years (P >0. 05). However, those in women at group of 22 -34 years and 35 -49 years showed statistical difference when compared with women at group of more than 50 years ( P < 0. 05 ). When compared with women at group of 22 - 34 years, the incidence of posterior and anterior vaginal wall protrusion were increased ( OR = 1. 713, 3. 765). (4) Menopause status was associated with severities of all kinds of descent ( P < 0. 05 ) and presence of posterior vaginal protrusion ( OR = 3. 354 ). Conclusions Mild anterior and posterior vaginal descent by POP-Q were common among women in China. The risk of anterior vaginal descent is relatively higher than posterior vaginal descent. However, most of the women with descent are asymptomatic and need no treatment. The most important factors associated with the severity and detectable ratio of descent is parity and age.
