3.Determination of Stilbence Glucoside in Polygonum Multiflorum by HPLC

Objective:To determine the content of stilbene glucoside in polygonum multiflorum by HPLC. Methods: An Agilent Eclipse Plus C18(100 mm ×4.6 mm,3.5 μm)column was adopted with acetonitrile-water(18∶82) as the mobile phase at a flow rate of 1. 0 ml·min-1 . The detection wavelength was set at 320nm and the injection volume was 8 μl. Results:The linear range of stilbene glucoside was 38-237. 5 μg·ml-1(r=0. 999 4)with the average recovery of 98. 32%(RSD=1. 59%). Conclusion: The method is simple, sensitive and specific in the content determination of stilbene glucoside in polygonum multiflorum.
4.Application of placental mesenchymal stem cells

BACKGROUND:Placental mesenchymal stem cels are becoming a new source of seed cels because of wide range of sources, low immunogenicity and not involving ethical issues.
OBJECTIVE:To elaborate the sources, biological characteristics and latest application of placental mesenchymal stem cels.
METHODS:Literature search was performed in PubMed, ScienceDirect, OvidSP, CNKI databases for relevant literatures published from 2003 to 2015. The key words were “placenta, mesenchymal stem cels, placenta mesenchymal stem cels, cel transplantation, application mechanism” in Chinese and English, respectively. Then, 57 papers were further analyzed and reviewed in line with the theme.
RESULTS AND CONCLUSION:Placental mesenchymal stem cels have been isolated and cultured successfuly, and confirmed to have multi-differentiation potential. A large number of placental mesenchymal stem cels have been used in the experimental animal and clinical researches, and they have a great potential in bone tissue engineering, revascularizaion and nerve repair. However, the specific mechanism underlying the application of placental mesenchymal stem cels is not clear. In order to ensure the safety and effectiveness, there are stil many problems to be further studied before placental mesenchymal stem cels are widely used in clinic.
5.Prenatal Sonographic Characteristics of Clubfoot and Relationship with Chromosomal Abnormalities

[Objective]To evaluate the incidence of chromosomal abnormalities and associated abnormalities in prenatally diag?nosed clubfoot,and to determine the prognostic factors as well.[Methods]A total of 89 fetuses with clubfoot diagnosed during Janu?ary 2010 to October 2015 in prenatal ultrasound scan and confirmed postnatally or by autopsy,were selected,within which 16 (18.0%)cases were without other abnormalities and 73(82.0%)cases were with other abnormalities. The associated abnormalities were identified ,the correlation with chromosomal abnormalities were analyzed with Fisher analysis and the factors affecting the outcomes were determined with Logistic regression analysis.[Results]Among associated abnormalities ,the skeletal abnormalities besides the clubfoot were the most frequently associated anomalies (35 ,47.9%),and the central nervous abnormalities followed secondly(30,41.1%). A chromosomal abnormality,with trisomy 18 being the most frequently detected,was identified in 34%(17/49)of the clubfoot fetuses with other anomalies ,whereas none of chromosomal abnormality was identified in 11 fetuses without other anomalies,a significant different rate of aberrant chromosome noted(P < 0.001). The survival rate of clubfoot fetuses without other anomalies was higher than that of clubfoot fetuses with other anomalies(50.0%vs 1.3%,P=0.03). The conditions of with or without associated anomalies were the independent prognostic factors (P = 0.01),the clubfoot fetuses associated with other anomalies had poor outcomes[OR=11.9(95%CI:1.8,80.1)].[Conclusion]Skeletal abnormalities besides the clubfoot were the most frequently associated anomalies. The condition of with or without associated anomalies is the independent prognostic index for fetuses with clubfoot. Aneuploidy were more commonly in clubfoot fetuses with associated abnormalities than in those without other abnormalities. No indication for karyotyping suggests for the clubfoot fetuses without other abnormalities due to the low incidence of associate chromo?somal anomalies.

OBJECTIVE:To supply the reference for benchmarking framework of unit dose dispensing(UDD).METHODS:To introduce the running module and the training mode for UDD pharmacists of the UDD management mode,and the working quality was controlled using 6? management pattern.RESULTS & CONCLUSIONS:The benchmarking framework of UDD is feasible in practice and it can help guarantee dispensing accuracy,improve the quality of pharmaceutical care.
7.Pathogen distribution and drug resistance in pneumonias among immunocompromised hosts

Objective To provide evidence for diagnosis and treatment by investigating the pathogens and drug resistance of immunocompromised host (ICH)pneumonias.Methods Statistical method was used to analyze retrospectively the data of pathogens and drug resistance of ICH pneumonia treated in our hospital in 2014. Results We confirmed 187 cases of ICH pneumonia by pathogen study of 324 patients.Gram-negative bacillus (68.42%)was the main pathogen of ICH pneumonia.Acinetobacter baumanii (AB)accounted for the first place. Staphylococcus aureus infection was still the first of Gram-positive coccus.Conclusion Gram-negative bacillus was the main pathogen of ICH pneumonia in our hospital and the isolated bacteria show strong antibiotic resistance. Therefore,it is necessary to strengthen dynamic monitoring of pathogens in the secretions from ICH pneumonia patients’ lower respiratory tract and drug resistance.It is suggested that clinicians make anti-infection treatment cover drug-resistant bacteria.
8.A case of AIDS complicated with Pneumocystis jirovecii pneumonia and disseminated infection of Talaromyces marneffei and Cryptococcus neoformans

Abstract: AIDS combined with Pneumocystis jirovecii pneumonia (PJP) and disseminated infections of Talaromyces marneffei and Cryptococcus neoformans are rare. This paper summarizes and analyzes the diagnosis and treatment of an AIDS patient with multiple fungal infections for reference. A 79-year-old male patient was admitted to the hospital with "stool habit change for more than 20 days". The white blood cell count was 4.57×109/L, the percentage of neutrophils was 81.8%, the absolute count of CD4+ lymphocytes was 6/μL, and the CD4/CD8 ratio was 0.17. HIV antibody positive was confirmed by CDC. The cerebrospinal fluid and alveolar lavage fluid were positive for Cryptococcus neoformans capsular antigen, and Pneumocystis jirovecii was found by the bronchoalveolar lavage fluid stained with hexamine silver. The cerebrospinal fluid culture was positive for Cryptococcus neoformans, and the blood culture was positive for Cryptococcus neoformans and Talaromyces marneffei. CT showed that bronchovascular bundles in both lungs were more thick, patchy and cable-like high-density shadows were seen in both lungs, and the edges were blurred. Nodular and cable-like high-density shadows were seen in the posterior apical segment of the left upper lobe, with clear margins. Infection of both lungs was considered, and secondary pulmonary tuberculosis occurred in the left upper lobe. After admission, the patient was treated with various anti-bacterial and fungal drugs due to recurrent fever, but the effect was not effective. The fever symptoms of the patient could not be significantly improved, and his condition continued to worsen, and he eventually died. The patient with AIDS complicated with bacterial and fungal infection, especially PJP infection in serious condifiton and has a poor prognosis for rapid development, so clinical attention should be paid to.
9.Study on Acute Toxicity of Xiaobai Capsules in Mice

Objective: To study the acute toxicity of Xiaobai capsules in mice after intragastric administration .Methods: The mice were randomly divided into two groups , the treatment group and the control group .The treatment group was given Xiaobai cap-sules by gavage, 3 times daily.The acute toxicity was recorded, and the median lethal dose (LD50) and the maximum dose were deter-mined.Results:The maximum daily dose of Xiaobai capsules was 141.6 g· kg-1(equivalent to 211.3 times of the clinical dose).At the dose, the mice showed no toxicity without death in 14 days or changes in organs after the dissection .Conclusion:Xiaobai capsules have very low acute toxicity in mice after intragastric administration with high security .
10.Effect of ginkgolide B on high glucose -induced human lens epithelial cell apoptosis

AIM: To study the effect of ginkgolide B on high glucose-induced human lens epithelial cell (HLEC) apoptosis.
METHODS: The high glucose-induced HLEC model was established. Different concentrations of ginkgolide B were intervened. Cell viability was assayed by MTT assay, morphology of cell apoptosis was observed by hochest33258 staining. Cell ultrastructural changes were detected by transmission electron microscopy. Expressions of apoptosis - related factors caspses - 3 and caspase-9 were tested by colorimetric detection.
RESULTS: High glucose inhibited the activity of HLECs, induced apoptosis reaction of HLECs, caused high expression of apoptosis factors in cells; while ginkgolide B inhibited the decrease of cell viability induced by high glucose, decreased the HLEC apoptosis and reduced the expression of apoptosis factors Caspase-3 and Caspase-9.CONCLUSION: Ginkgolide B may inhibit the expression of caspses-3 and caspase-9 then effectively inhibited high glucose-induced apoptosis in HLECs.
