2.Clinical Study on the Effect of Pre-intervention with Electroacupuncture on Oxidative Stress and Inflammatory Reaction in Hepatic Ischemia-reperfusion
Shanghai Journal of Acupuncture and Moxibustion 2017;36(2):175-179
Objective To observe the effect of pre-intervention with electroacupuncture on oxidative stress (OS) and inflammatory reaction after hepatectomy under portal occlusion.Method Thirty patients going to receive hepatectomy under Pringle's portal occlusion were randomized into an electroacupuncture group and a sham electroacupuncture group, 15 cases each. Three days prior to the surgery, the patients started to receive corresponding acupuncture or sham electroacupuncture treatment by selecting bilateral Riyue (GB24), Qichong (ST30) and Yanglingquan (GB34), once a day. The anesthesia method was general (inhalational and intravenous) anesthesia. Central venous blood was drawn before the surgery, and respectively 6 h, 24 h, 48 h and 72 h after the surgery to examine the OS-relatedparameters, superoxide dismutase (SOD) and malondialdehyde (MDA), as well as the inflammatory mediators, including tumor necrosis factor-α(TNF-α), interleukin (IL)-1, IL-6, and IL-10.Result Compared to the sham electroacupuncture group, the SOD activity significantly increased (241±11.4 U/mL,180±12.6 U/mL,P<0.05) and the MDA level significantly dropped in the electroacupuncture group at 6 h and 24 h after the surgery(11.0±0.8 mmol/L,10.0±0.9 mmol/L,P<0.05); there were no significant between-group differences in the rest indexes (P>0.05).Conclusion Pre-intervention with electroacupuncture can up-regulate the activity of SOD and down-regulate the level of MDA in patients after hepatic portal occlusion, and thus partially reduce OS, but it doesn't shownoticeable effecton inflammatory response.
3. Study on decompression extraction of Sansheng Hehuan Decoction
Chinese Traditional and Herbal Drugs 2013;44(14):1918-1924
Objective: To explore the advantages of decompression extraction applied in the preparation of Sansheng Hehuang Decoction (SHD). Methods: The process parameters of decompression extraction of SHD were optimized using synephrine and magnolol as indexes, the active components content, extract yield, extract powder characteristics, and volatile components by decompression extraction and atmospheric extraction were compared, and the microcosmic structures of synephrine and magnolol were observed. Results: The optimum extraction parameters were as follows: vacuum degree of 0.08 MPa (60°C), extracting twice for 30 min each time, and solid-liquid ratio of 1:10. Compared with the extraction at normal pressure by water, the content of synephrine increased by 13%, the extracting rate of magnolol increased by 10% while the extracting rate of timosaponin BII was almost the same, and the yield of the dried extract decreased by about 33%. The kinds of volatile components by decompression extraction were almost the same as those by atmospheric extraction, but the yield was lower than that by atmospheric extraction. Besides, the powder characteristics including flowability and hygroscopicity of the extract prepared by decompression extraction were similar to those by water extraction and alcohol precipitation at normal pressure. Scanning electron microscopy showed that the decompression extraction did not break the cell wall, and the cells presented arrangement disorder and shrinkage, while the cell integrity was better than that extracted at normal pressure. Conclusion: In regard to energy consumption, extraction effectness, and convenience to the subsequent preparation, the decompression extraction method is superior to the traditional atmospheric extraction method, and it could be applied in the extraction process for Chinese materia medica preparations.

Modern Clinical Nursing 2013;(10):16-19
Objective To explore the sub-health and TCM physical constitution of stroke patients’families.Method Four hundred and twelve family members of stroke patients hospitalized during September 2001 to May 2013 were involved in the survey by filling out TCM physical constitution scale(TCMPC)and the sub-health self-rating scale(SSS)for the purpose of investigating the sub-health and conditions of TCM physical constitution. Results Of all the patients,189 were in sub-health,accounting for 45.87%. There were 7 main clinical symptoms related to sub-health,including poor sleep, less activeness, proneness to upsets and irritation, memory loss, depression, tension in interpersonal relations and appetite loss, with the incidences ranging from 64.55%to 48.15%. In TCM,51.85%of the family members were balanced in physical constitution,10.58%of them had Yang-deficiency constitution, 14.29%of them had Yin-deficiency,23.28%of them other kind of constitution. Conclusions The family members of stroke patients have a high rate of sub-health.The members in sub-health hold a balanced constitution,Yang-deficiency or Yin deficiency. In order to eliminate their sub-health,improving the quality of care for patients with stroke,we should guide the families of stroke patients to develop good living habits, strengthen the TCM mental nursing, help them to establish a good social support system.
5.Effects of substance P on the activity of intestinal mucosal mast cells in rats with multiple organ failure
Chinese Journal of Digestion 1996;0(05):-
Objective Multiple organ failure (MOF) has been regarded as a continuous, uncontrolled inflammatory response. Intestinal mucosal mast cells (IMMC) may be involved in MOF. Substance P (SP), one of gut peptides, is an important regulator in the neuro-endocrine-immune network. However, the effects of SP on IMMC, especially in the case of MOF, remain unclear. This study was aimed to investigate the effects of SP on IMMC in the development of MOF. Methods The rat model of MOF was established by injecting zymosan. After thirty minutes of the injection, SP was given through tail veins at the dose of 20 nmol/kg weight and 0.2 nmol/kg weight. The concentrations of histamine and tumor necrosis facfor-?(TNF-?) in plasma and intestine tissues were measured. The pathological alterations of essential organs including intestine, liver, kidney and lung were examined under light microscope. Their corresponding functions were reflected with ALT, Cr and PO 2. The ultrastructure of the IMMC was also observed under a transmission electronic microscope. Results Compared with the controlled rats, those injected with SP showed much more serious inflammatory response under light microscope. Both ALT and Cr increased by about 50%, but PO 2 decreased by about 40%. Histamine level in the intestinal tissue of the rats treated with SP remarkably decreased, whereas the plasma histamine level did not show any significant changes. The level of TNF-? was higher in the intestinal tissue of the rats treated with SP but no change in plasma, and the degranulation of IMMC under transmission electronic microscope was more obvious. Conclusions SP may trigger MOF through acting on IMMC which may release inflammatory mediators such as histamine and TNF-?.
6.Somatostatin suppressed the activity of intestinal mucosal mast cells in rats with multiple organ failure
Chinese Journal of Digestion 2001;0(08):-
Objective To investigate the effect of somatostatin(SST) on the activity of intestinal mucosal mast cells(IMMC) and its pathological significance in the development of multiple organ failure(MOF). Methods The rat model of MOF was established by the peritoneal injection of zymosan. Thirty minutes after the injection of zymosan, SST at 2.300 ng?kg -1 ?h -1 or 0.023 ng?kg -1 ?h -1 was injected respectively through tail veins. The concentration of histamine and tumor necrosis factor-? (TNF-?) in plasma and intestinal tissue were measured. The pathological alterations of essential organ including intestine, liver, kidney, lung and heart were studied under light microscope. Their corresponding functions were reflected with alanine aminotransferase (ALT), cretinine (Cr) and oxygen pressure (PO 2). In addition, the ultra structure of the IMMC was observed under a transmission electronic microscope. Results Compared with the controlled rats, the rats injected with SST (2.300 ng?kg -1 ?h -1 ) showed less serious inflammatory response under light microscope. ALT and Cr were decreased 53% and 60% respectively. However, the lung ventilation was improved and PO 2 was increased by 50%. The histamine level in the intestinal tissue from rats treated with SST remarkably increased( ( 8.60? 0.50 ) ng/g protein to ( 14.50? 1.08 ) ng/g protein), whilst the plasma histamine level did not show any significant changes. Exogeneous SST also resulted in lower level of TNF-? in intestine but no changes in plasma. Furthermore, degranulation of IMMC from the rats treated with SST was less obvious. Conclusion SST may prevent from or arrest the development of MOF through suppression of the release of inflammatory mediators, such as histamine and TNF-?.
7.Effects of gut peptides on the activation of mast cells from rat intestinal mucosa in vitro
Chinese Journal of Immunology 1999;0(12):-
Objective:To investigate the effects of substance P(SP),somatostain(SST) and vasoactive intestinal peptide(VIP) on the activation of rat intestinal mucosal mast cells(IMMC) in vitro. Methods:IMMC isolated and purified from the whole intestines of normal rats were incubated with gut peptides at various concentrations. The histamine concentration in IMMC and their supernate were determined.Furthermore, the ultrastructure of the incubated IMMC was observed under a transmission electronic microscope.Results:①The spontaneous release rate of histamine was (22.86?3.22)%.②SP significantly increased the histamine release rate from IMMC(P0.05)④At the concentration from 1?10 -1 mol/L to 1?10 -8 mol/L,the higher concentration of VIP was used, the lower histamine release rate was observed(P
