2.Promising drugs to treat Alzheimers disease——?-secretase inhibitors

Mutations in presenilins caused most of the autosomal dominant familial forms of Alzheimers disease by altering ? secretase activity. The ? secretase is a large multiprotein complex including presenilin heterodimers,nicastrin,PEN 2 and additional unidentified components.? secretase cleaves beta amyloid precursor protein,Notch,E cadherin,ErbB 4 receptor tyrosine kinase and other membrane proteins.The development of various ? secretase inhibitors not only provides a tool for investigating the structure, function and mechanism of ? secretase,but also presents a therapeutic strategy to slow progression of Alzheimers disease pathology. Threre are various classes of ? secretase inhibitors: compounds containing a difluoro ketone or a difluoro alcohol group, (hydroxyethy) urea peptidomimetics,compounds possessing a hydroxyethylene dipeptide isostere, short helical peptides, non peptidic inhibitors derived from 4 chloro isocoumarin synthon, compounds containing alanyl moiety.Among these species of ? secretase inhibitors,those which selectively affect Abeta production are especially the most promising drugs to treat Alzheimers disease.
3.Comparision Among Four Species Puhuang on Blood Coagulation System in Vivo and Vitro

Test on coagulation time,prothrombin time and platelet aggregation of mice and rats showed that all four species of Puhuang had effects on blood coagulation, in which Typha angustata Bory et Chaub. was the strongest followed by T. davidiana Hand. -Mazz., T. latifolia L,and T. angustifolia L. Their mechanism of coagulation were not the same,in which T. angustata Bory et Chaub. mainly acted on thrombogen,while T.davidiana Hand. -Mazz. mainly acted on platelet aggregation.
4.Effect of insulin-like growth factor 1 receptor ?-strain interrupted on lung adenoma cell line SPC-A-1

Purpose:To study the effect of insulin-like growth factor 1 receptor ?-strain(IGF1R-?) interrupted by a special antibody (IGFⅠR-?Mab)on lung adenoma cell line SPC-A-1.Methods:IGFⅠR-?Mab was extracted by hybrid technology. SPC-A-1 cells were separated into 2 groups,the IGFⅠR-?Mab and the blank control. The IGFⅠR-?Mab cells were interfered by different densities of IGFⅠR-?Mab, including 20,40,60,80,100,120,140,160,180 and 200 ng/ml. The MTT curve line, morphology, ultrastructure and cell cycles were observed at 0,24,48,72 hours after the intervention respectively. Results:Compared with the control, apoptosis in IGFⅠR-?Mab group was significant(P= 0.009)and proliferation rate was decreased obviously within 160 ng/ml. However, the proliferation rate was no significant when the special antibody density was more than 200 ng/ml.Conclusions:The affinity of IGFⅠR-?Mab at IGF1R ?-strain is high. The interruption of IGF1R ?-strain by IGFⅠR-?Mab shows the obvious biological effects in vitro ,with inclusion of promoting apoptosis and suppressing proliferation, which indicate the interruption targeting IGF1R ?-strain is prospective for non-small-cell lung carcinoma.
5.Progress in new drugs targeting reverse cholesterol transport

Numerous epidemiologic studies have shown that there is a strong inverse relationship between high-density lipoprotein cholesterol levels and artherosclerosis. The first atheroprotective mechanism of HDL is the RCT(reverse cholesterol transport). Recently,considerable pharmacological trials have focused on targeting critical proteins and enzymes in RCT including apoAⅠ, ATP binding cassette transporter A1 and cholesteryl ester transport protein etc. A number of new drugs have demonstrated conspicuous protective effects on artherosclerosis. Therefore RCT will become an attractive target for prevention and cure of dyslipidamia and artherosclerosis.
6.Lentivirus vector-mediated enhanced green fluorescence protein transfection on in vitro corneal epithelial cells of rabbits

Objective To observe the effect of lentivirus vector-medicated enhanced green fluorescence protein (EGFP) transfection on in vitro corneal epithelial cells.Methods Primary corneal epithelial cells of rabbits were cultured and transfected with lentivirus vector into different multiplicity of infection (MOI) in order to find the best transfection dose.After 24,48,72,and 96 h of transfection,expression of EGFP was observed under microscope and detected by RT-PCR.The transfection rate of corneal epithelial cells was calculated in different MOI.Results EGFP was expressed in corneal epithelial cells 48 h after transfection,which increased with the increasing transfection time.The transfection rate of corneal epithelial cells was 4.5%?0.6%,30.4%?0.9%,51.9%?1.4%,and 75.4%?0.7%,respectively,when MOI was 1,10,50,and 100(P

Alzheimers disease (AD) is a neurodegenerative disorder severely affecting the aged population. Its pathological hallmarks include amyloid plaques, neurofibrillary tangles, loss of synapes and neurons as well as brain inflammation. This review focuses on multiple roles of p38MAPK, one of the members of mitogen-activated protein kinases(MAPK) family, in the pathogenesis and progression of AD. The review discusses the evidences of p38 activation in AD models, its connection with each pathological mark of AD and the interaction between p38 and significant pathophysiological process, such as inflammation, neuronal apoptosis and hyperphosphorylation of tau. Moreover, a discussion is also made to address the basis of p38 as the target for therapeutic intervention in AD. All in all, p38 is considered to be a new promising target for drugs to control the progression of AD.
9.Association of the E-selectin polymorphism with Systemic Lupus Erythematosus in the Ethnic Zhuang population from Guangxi Province

Objective To investigate the distribution of E-selectin polymorphisms in Guangxi ethnic Zhuang population and the association of E-selectin gene polymorphism in exon 2 and in exon 4 with systemic lupus erythematosus(SLE). Methods Polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) was used to study the E-selectin polymorphisms in 82 patients with SLE and 95 healthy controls of ethnic Zhuang population in Guangxi Province. Results The frequencies of GG?GT?TT genotypes in the exon 2 of E-selectin were 86.6%,12.2%, and 1.2% in SLE group; 90.5%, 8.4%, and 1.1% in the control group, respectively. The frequencies of AA?AC?CC genotypes in the exon 4 and E-selectin were 81.7%,15.9%, and 2.4% in SLE group; 92.6%,7.4%, and 0.0% in the control group, respectively. 10.Surgical treatment of esophageal and cardiac cancer in elderly patients

Purpose:To study the surgical treatment of the cases with cancer of esophageal and gastroesophageal conjunction by thoracotomy. Methods:Total of 43 patients were selected. Squamous cell carcinoma of esophagus were found in 26 patients,among them 23 received radical resection and 3 failed due to massive invasion of the tumor.The rest were diagnosed as adeno carcinoma of cardia,13 of them undergoing radical resection and 4 failed.Results:The results of the whole group were surgical resection 93.0 %,complication 41.9% and operative mortality 4.7%.Conclusions:The authors conclude that selecting patients with precise indications as well as treating complication correctly was very important for surgical procedures of elderly patients.The pre-operative preparation,nutrition and approach for resection are also important.
