Objective To construct expressing recombinant of recF and recO from Deinococcus radiodurans R1and express the target protein in E.coli BL21(DE)3.Methods recF and recO gene were amplified by PCR from genomic DNA of Deinococcus radiodurans R1 and inserted into expression plasmid vector pET30b(+) to construct pET30b(+)-recF and pET30b(+)-recO.The recombinant plasmids were transformed into E.coli BL21(DE)3 and the recombinant proteins were expressed by the isopropyl-?-D-thiogalactopyranoside(IPTG) and were analyzed with SDS-PAGE.The changes in the survival rate of bacteria in each group before-and after UV-irradiation were calculated.Results The recombinant plasmid pET30b(+)-recO and pET30b(+)-recF was obtained and the recombinant protein could be highly expressed in E.coli BL21(DE)3.Conclusion This study has provided a foundation for further studies and applications of the recombinant RecF and RecO,and initial detection shows that recO and recF gene can increase the resistant ability of E.coli.

Objective To investigate the pathophysiological changes in irradiation-induced heart injury in rats in order to provide preventive and therapeutic strategies for irradiation-induced heart disease(RIHD).Methods Experiments were performed using female Wistar rats with body weight of approximately 220g.12 female Wistar rats were randomly assigned to two groups(sham-irradiated group and irradiated group).Heart of rats was locally irradiated with 60Co ?-ray at a dosage of 20 Gy.During the post-radiation period,the intake of food and activities of the animals were observed daily and the body weight was recorded monthly.On the 120th day after irradiation,myocardial perfusion was tested using real time myocardial contrast echocardiography(RTMCE).On the 180th day after irradiation,the hearts were isolated from the rats of the both groups,and they were perfused with a Langendorff apparatus.Hemodynamic measurements,including peak systolic LV pressure(LVSP),maximal rate of LV pressure rise(LV dp/dtmax)and fall(LV dp/dtmin)were studied.Then the hearts were examined pathologically with HE staining.Results Animals irradiated with single dose of 20Gy locally were apparently in a good condition during the observation period of 180 days.No differences in food intake,physical activities and body weight were observed between irradiated and control animals.However,myocardial blood velocity(beta)of irradiated rats was significantly lower than that of sham irradiated group(P

Objective:To identify the function of extracellular soluble vimentin that promotes proliferation, activation and chemotaxis of inflammatory cells. Methods: The proliferation of rat splenocytes stimulated with vimentin was evaluated in vitro. The lymphocyte counts and vimentin-antibody levels of peripheral blood in mice immunized with vimentin were detected in vivo. Peritoneal macrophages were collected and cultured with different concentrations of vimentin to detect the effect on phagocytosing chicken red blood cells. The chemotaxis of NIH3T3 fibroblast towards vimentin was observed in transwell chamber. Results:In vitro vimentin dose dependently promoted the proliferation of splenocytes. The proliferation indexes of primed and naive splenocytes cultured with 16μg/ml vimentin were reached up to ( 196. 0 ± 9. 7 )% and ( 208. 9 ± 4. 6 )% respectively without significant difference. In vivo vimentin significantly enhanced the lymphocytes number(109 L-1)of peripheral blood(5. 74±0. 51 vs. 1. 69±0. 13)and the levels of vimentin specific antibody( OD value 2. 31 ± 0. 06 vs. 0. 19 ± 0. 08 ) that shown no significant difference from immunization with vimentin plus CFA. In vitro vimentin dose dependently stimulated phagocytic ability of macrophages and performed the chemotactic effects on NIH3T3 fibroblasts. Conclusion:Extracellular soluble vimentin promotes the proliferation,activation and chemotaxis of concerned inflammatory cells and possesses the functions as an alarmin.

Objective: To study the nutritional status and problems of Chinese elite athletes for instructing training scientifically. Method: 599 elite athletes were investigated by means of dietary survey, physical measure and biochemical detection. Results: (1) The daily average energy intake reached the adequate intake (AI) recommended for athletes, but the energy ratio both of protein and fat higher, carbohydrate lower, being 18.9%, 38.6% and 42.5% respectively. Except vitamin A and B1, B2 the intakes of other vitamins and minerals were sufficient. (2) The average BMI and body fat percentage were 23.0?3.0 and (12.1?3.2)% in men and 21.9?3.0 and (20.5?3.9)% in women respectively. (3) The average hemoglobin levels were (145.7?13.3)g/L in men and (130.6?11.8)g/L in women. The rates of anemia and iron deficiency anemia were 12.6% and 5.2% respectively, female higher than male. (4)The hyperlipidemia rate was 22.3%, including 12.6% high TG, 9.7% high TC and 1.2% high LDL, female higher than male. (5) The rates of VB1 and VB2 insuficiency were 46.2% and 32.7% respectively, both including 9.6% deficiency. Conclusion: The nutritional status of Chinese elite athletes was good, but still with anemia, vitamin insufficiency and hyperlipidemia.

Objective: To explore the effects of Polygonatum odoratum on body based on metabonomics. Methods: The ultra performance liquid chromatography coupled with mass spectrometry (UPLC-MS) and principle component analysis(PCA) and orthogonal partial least squares discriminant analysis(OPLS-DA) were used to evaluate the changes of endogenous substances in rats after intragastric administration of Polygonatum odoratum. Results: Significant differences between the control group and administration group were observed in PCA and OPLS-DA model. Five potential biomarkers between control group and administration group were identified. The relative content of Alpha-Tocotrienol, PC(14:1(9Z)/14:1(9Z)), Stearic acid, Theasapogenol A, Docosahexaenoic acid increased. Conclusion: The biomarkers had many beneficial activities, so the Mongolian medicine Polygonatum odoratum had the function of health care. Background The dosage of verteporfin photodynamic therapy (PDT) for central exudative chorioretinopathy(CEC) depends on the calculation formula of Treatment of Age-related Macular Degeneration with Photodynamic Therapy Study Group(TAP)and Verteporfin in Photodynamic Therapy Study Group(VIP).Some local adverse responses or normal tissue damage have been noted under the standard dose of verteporfin during the treatment of CEC.So it is necessary to explore an appropriate therapeutic dosage of verteporfin.Objective This clinical study aimed to observe and compare the clinical efficacy and safety of different doses of verteporfin PDT for CEC.Methods Ninety eyes of 90 patients with CEC were enrolled in this study with the approval of the Ethic Commission of Affiliated Second Hospital of Nanchang University.Written informed consent was obtained before PDT.The patients were randomly divided into standard dose group,half dose group and 1/3 dose group.All of the patients received PDT.Standard dose(6 mg/m2),3 mg/m2 or 2 mg/m2 of verteporfin was applied in the three groups respectively,with the laser intensity 50 J/cm2.The patients were followed-up for 3 months.Repeat treating regimen was performed in the same way in the patient with fundus fluorescein leakage after initial PDT.Results The BCVA(logMAR) value was 0.44±0.36 after PDT in the standard dose group.Visual acuity improved in 20 patients (66.67％),stabilized in 8 patients(26.67％)and decreased in 2 patients (6.67％).The BCVA showed a significant difference between the preoperative and postoperative BCVA (t =6.719,P =0.000).CNV disappeared in 19 patients (63.33％),obviously improved in 7 patients (23.33％),partially disappeared in 3 patients (10.00％) and unchanged in 1 patient (3.33％).The central fovea thickness (CFT)value was significantly declined in postoperation compared with preoperation (t =5.758,P =0.000).Eight patients received secondary PDT with the recurrence rate 26.67％.In the half dose group,visual acuity improved in 18 patients(60.00％),stabilized in 11 patients(36.67％)and decreased in 1 patient (3.33％).CNV disappeared in 16 patients (53.33％),obviously improved in 8 patients (26.67％),partially disappeared in 5 patients (16.67％) and unchanged in 1 patient (3.33 ％).Significant difference was seen between the preoperative BCVA and postoperative BCVA(t=8.294,P =0.000).The decrease of C FT was significant in postoperation(t =8.493,P =0.000).Ten patients received secondary treatment with the recurrence rate 33.33％.In 1/3 dose group,visual acuity improved in 8 patients (26.67 ％),stabilized in 12 patients (40.00％),decreased in 10 patient (33.3％).CNV disappeared in 8 patients (26.67％),obviously improved in 8 patients (26.67％),partially disappeared in 6 patients (20.00％) and unchanged in 8 patient (26.67％).There was no significant difference between the preoperative BCVA and postoperative BCVA (t =0.536,P =0.596).The difference between preoperative CFT and postoperative CFT was insignificant(t =0.942,P=0.354).Fourteen patients received secondary PDT and 8 patients received three times with the recurrence rate 73.33％.Conclusions 3 mg/m2 verteporfin PDT for CEC shows a similar clinical efficiency and safety to 6 mg/m2 verteporfin.3 mg/m2 verteporfin PDT can decrease cost,but the effect of 2 mg/m2verteporfin PDT is not satisfacted.

Objective To compare efficacy and side effects of intravenous versus oral indomethacin treatment for symptomatic patent ductus arteriosus (PDA) in preterm infants.Methods Fourty-nine preterm infants were reviewed retrospectively who were diagnosed as having symptomatic PDA confirmed by echocardiography.According to the using type and approach that were divided into 2 groups (intravenous group,n=21;oral group,n=28) and their doses and intervals were same.The rates of ductal closure and side effects were compared in 2 groups.Results There were no significantly different between 2 groups in single ductal closure and complicating other diseases. Objecttive To observe the repaired effect of distinct source olfactory ensheathing cells (OECs) on spinal cord injury (SCI) rats. Methods These OECs were dissociated from olfactory bulb and olfactory mucosa of SD rats and transplanted to the injuried region of spinal cord injury rats. The function of nerve, motor evoked potential of hind legs and the histopathlogical diversities of injuried spinal cord were observed. Results The OECs grafts into the SCI area could survive longer time. The BBB scale, incubation stage of EP and histopathologic manifestations showed that the group with transplanted OECs regained more improvement in hindlimb than the control group. Conclusion The OECs of two sources have the same ability to regain and improve the axonal function which can promote axons regeneration of SCI.
