1.Apoptotic mechanism of leukemic K562 cells induced by mangiferin

Objective To study the apoptosis mechanism of K562 cell lines induced by mangiferin. Methods The mRNA expression levels of apoptosis-related genes including bcl-2, bax, survivin of K562 cells treated by mangiferin (25—200 ?mol/L) for 24, 48, 72, and 96 h were determined by RT-PCR; the BCR/ABL protein P210 level was detected by Western blotting. Results Mangiferin up-regulated bax gene of K562 cells significantly and down-regulated bcl-2 gene slightly, resulting in an enhancement of the ratio of bax/bcl-2. Mangiferin down-regulated the expression levels of P210 in K562 cells in a time-and concentration-dependent manner and so is the expression level of survivin mRNA in K562 cells. ConclusionThe mechanism of mangiferin-induced apoptosis in K562 leukemic cells might be involved in up-regulating the gene expression of bax and down-regulating the mRNA expression of BCR/ABL protein P210, bcl-2, and survivin.
2.Influence of ischemia/reperfusion on function of vascular endothelial cells and effect of intervention with drug

Objective:To explore the influence of ischemia/reperfusion (anoxia/reoxygenation)[FK(W16*2。142mmZQ1mm]on immunofunction of endothelial cells(ECs) and effect of intervention with 2-hydroxymethyl-3,5,6-trimethylpyrazine on it.Methods:Model of ECs induced by anoxia/reoxygenation was established to mimic ECs ischemia/reperfusion injury in vivo with human umbilical vein endothelial cell line ECV304.Then 2-hydroxymethyl-3,5,6-trimethylpyraxine was used to intervene the anoxia/reoxygenation process.Nuclear transcriptional factor-?B(NF-?B) was exhibited by fluorescent staining,HLA-ABC,HLA-DR,CD86 and CD54 were detected by flow cytometry.Mixed endothelial cell-lymphocyte reaction(MELR) was conducted to examine the proliferation of lymphocyte,production of IL-2 and percentage of apoptotic lymphocyte.Results:Anoxia/reoxygenation made the ECV304 cell became round,shrunk and abscised,with increased plasma NF-?B,and changed from positive cytoplasm to positive nucleus.HLA-ABC,HLA-DR and CD86 on surface of cells increased but CD54 showed unchanged.MELR showed the incorporation of ~3H-TdR and production of IL-2 increased significantly and the percentage of apoptotic lymphocyte decreased.After 2-hydroxymethyl-3,5,6-trimethylpyrazine intervention,the ECV304 cell shapes recovered,NF-?B expression didn’t down-regulated,but the percentage of positive cells decreased,changes to positve dominant.Besides,reversal changes were shown in other parameters.Conclusion:Anoxia/reoxygenation influences some important immune related molecules in ECV304 cells.2-hydroxymethyl-3,5,6-trimethylpyrazine could antagonize these influences to maintain the immune function of endothelial cells in a relative normal manner.
3.STUDIES ON TREATMENT OF ODL FIELD WASTEWATER BY OIL-DEGRADING BACTERIA

A mixed bacteria culture F6 isolated from oil field wastewater can degrade petroleum hydrocarbons efficiently. The bacteria were suitable to treat oil-polluted wastewater of oil field. Simulated result treating oil-polluted wastewater in laboratory showed that after "XingyiLian" wastewater of Liaohe Oil Field was treated by fluidized-bed bioreactor system with the vehicle of activated carbon , the amount of oil and CODcr of the flow out water were decreased from 45mg/L to 4. 1mg/L and 470mg/L to 42mg/L separately , according with first class standards of Chinese Wastewater Discharge Regulation.
4.Effect of transplantation of umbilical cord blood mesenchymal stem cells on traumatic brain injury in rats

Objective To investigate the feasibility of human umbilical cord blood mesenchymal stem cells (CB-MSCs) in differentiating into neural-like cells and the effect of CB-MSC transplantation on traumatic brain injury in rats. Methods Healthy Wistar rats were induced into model with experimental traumatic brain injury by drilling and hitting their brain tissue, and then, they were randomized into 3 groups (n=18): model group, control group (injured models + injecting 1.25 μL saline) and CB-MSC transplantation group (injured model + injecting CB-MSC suspension). CB-MSC were derived from separated umbilical cord blood, cultured, marked with BrdU and injected into injured area of rats in the CB-MSC transplantation group. The motor function scale was performed 3 and 10 d after the transplantation, and Y maze test was employed to observe the rat's learning and memory abilities 2 and 4 w after the transplantation. CB-MSC in the CB-MSC transplantation group was detected by immunohistochemistry 2 and 5 w after the transplantation; BrdU-GFAP and BrdU-NSE positive cells were observed. Results Significant differences between the 3 groups were observed in motor function scores on the 10th day of transplantation and in rat's learning and memory abilities with Y maze test 2 and 4 w after the transplantation. BrdU-GFAP and BrdU-NSE positive cells were found in the area of transplantation in the CB-MSC transplantation group only by the end of 2 and 5 w after transplantation. Conclusion CB-MSC transplantation can help the recovery of brain injury in rats and improve the learning and memory abilities; CB-MSC transplanted into the rats' brain tissue can differentiate into neurons-like cell in vivo.
5.Microsurgical treatment of petroclival tumors

Objective To discuss the microsurgical treatment and outcome of tumors in the petroclival region. Methods A total of 48 patients with tumors in the petroclival region, had operation between Jan 2002 and Dec 2009, were chosen in the study; their data (their imaging, operation methods and complications) were retrospectively analyzed. Results Total resection of the tumors was achieved in 30 cases, subtotal resection in 11 and largely partial resection in 7. A 4-6-year follow up was performed on 46 patients; except 1 with permanent facial paralysis, the other patients got improvement to varying degrees. Conclusion Total resection rate of tumor in the petroclival region can be increased and complications and mortality can be decreased through full preparation of the operation, proper operation approaches and skillful manipulation of microneurosurgical technique.
6.Effects of endovascular embolization treatment on brain damage and recovery of neurological function in patients with posterior communicating artery aneurysm combined with oculomotor nerve palsy

Objective To explore the effects of endovascular embolization treatment on brain damage and recovery of neurological function in patients with posterior communicating artery aneurysm combined with oculomotor nerve palsy.Methods Eighty-four patients with posterior communicating artery aneurysm combined with oculomotor nerve palsy,admitted to our hospital from January 2007 to January 2011,were chosen in our study; 42 of them used intravascular interventional therapy (group A)and other 42 used microsurgical treatment (group B); the operation time,intraoperative blood loss,time of postoperative recovery respiratory and postoperative extubation time between the groups were compared; and the levels of serum neuron specific enolase (NSE),glucose and lactic acid at the end of the operation,postoperative recovery of the oculomotor nerve palsy and postoperative complications at discharge of the hospital were observed and compared between the two groups.Results As compared with group B,group A had significantly shorter operation time and postoperative extubation time (P<0.05),and significantly less intraoperative bleeding (P<0.05); NSE,glucose and lactic acid levels in group A were significantly lower than those in group B at the end of the operation (P<0.05); the incidence of cerebral vasospasm and cerebral edema in group A were obviously lower than that in group B (P<0.05).The complete recovery of oculomotor nerve paralysis showed no significant difference between the two groups (P>0.05).Conclusion As compared with microsurgical treatment,intravascular interventional therapy has good effects (no bleeding,rapid recovery and few complications) in patients with posterior communicating artery aneurysms combined with oculomotor nerve palsy.
7.Scanning electron microscopic study of titanium-hydroxyapatite, the functionally graded-material implanted in rabbits.

OBJECTIVEThe purpose of this study was to observe the tissue tolerance of Ti-HA functionally graded-material (FGM) and the form of the material-bone interface.
METHODSThe sintered Ti -HA FGM, pure HA and pure Ti were respectively implanted into the parietal bone of rabbits. The specimens were observed by SEM at 2, 4, 8 postoperative weeks.
RESULTSIn the early stage, the new bone surrounding the Ti -HA FGM formed earlier with larger amount and better maturity than the pure Ti. The condition was similar to the pure HA. Two months after the operation, direct bonding of material-bone interface was formed between the Ti -HA FGM and the new bone as an integral body. However, there was a little space left between the new bone and the pure Ti.
CONCLUSIONSThe Ti -HA FGM has good tissue tolerance. Its early integration with bone is similar to pure HA and better than pure Ti.

8.Clinical features and microsurgical management of rare tumors in the sellar region.

OBJECTIVETo investigate the clinical characteristics and microsurgical managements of rare tumors in the sellar region.
METHODSSix rare cases of tumors in the sellar region treated by microsurgery from Jan 2000 to Jan 2010 were reviewed retrospectively. Subsequent treatments were according to the status of preoperative alpha fetal protein (AFP) and human chorionic gonadotropin (HCG) measurement as well as confirmed by histopathological examination in all six patients.
RESULTSTotal resection of the tumor was achieved in 2 cases and subtotal resection in 4 cases. Postoperative histopathology confirmed that the lesions were tumors in 5 cases and fungal pseudotumor in 1 case. Moreover, variety of histological types were observed in the present series, including leiomyosarcoma, malignant yolk sac tumor, mixed germ cell tumor, embryonal carcinoma, pilocytic astrocytoma and fungal pseudotumor, respectively. The serum levels of AFP and HCG were elevated to some extent in the patients with malignant yolk sac tumor, mixed germ cell tumor or embryonal carcinoma. Follow-up was conducted in all patients for 1 month to 3 years. The patients with malignant yolk sac tumor and embryonal carcinoma as well as leiomyosarcoma died in 5, 6, 10 months after operation, respectively. Subarachnoid hemorrhage occurred in the case of fungal pseudotumor at 2 months after surgery. The other two patients were surviving well.
CONCLUSIONSRare non-germinomatous malignant germ cell tumors are predominantly susceptible to the sellar region. Furthermore, High misdiagnosis rate and poor prognosis are characteristic in the present study. Dynamic AFP and HCG detection may play an important role in the diagnosis of those non-germinomatous malignant germ cell tumors located in the sellar region. The importance of awareness of the presence of such rare lesions in the sellar region is emphasized.

9.Expression of p53 in breast cancer and regulatory effects of 5-aza-2′-deoxycytidine on p53

Objective Tumor suppressor gene p53 can inhibit tumor cell growth, arrest cell cycle, and promote apoptosis.Howev-er, the effects of p53 on the pathogenesis of breast cancer have not been fully elucidated.The aim of this study was to explore the expression of p53 protein and the correlation with clinical pathologic features in breast cancer.Furthermore, the regulatory effects of 5-aza-2′-deoxycyti-dine on p53 in breast cancer cell line were also studied. Methods The expression of p53 protein in 80 cases of breast cancer and normal and adjacent tissue were determined by the immunohistochemical staining .The expressions of p53 mRNA and p53 protein in breast cancer cell line were determined by RT-PCR and Western blotting. Results The positive rate of p53 in breast cancer (41.25%) was higher than that in the normal and adjacent tissue (22.5%) (P<0.01).The expression of p53 was not significantly correlated with age, grade, stage and lymph node metastasis (P>0.05).The low expression of p53 both in mRNA and in protein levels were found in breast cancer cell line of MCF-7.The expres-sion of p53 increased after 5-aza-2′-deoxycytidine administration . Conclusion p53 is highly expressed in breast cancer , which may play an im-portant role in the development and progression of breast cancer. 5-aza-2′-deoxycytidine, up-regulating the p53 expression in breast cancer cell line, which provides the evidents for the development of therapeutic drugs for the patients with low expression of p53 breast cancer.
10.Study on mismatch repair genes of chronic myeloid leukemia.

OBJECTIVETo investigate the expression and regulation mechanism of mismatch repair (MMR) genes in chronic myeloid leukemia (CML).
METHODSExpression of MMR genes hMSH2, hMSH3, hMSH6, hMLH1 and hPMS2 mRNAs in 62 CML patients and K562 cell line were detected by semi-quantitative reverse transcription polymerase chain reaction (RT-PCR). Expression of bcr-abl mRNA and MMR genes mRNA were detected by RT-PCR in 26 CML patients with allogeneic peripheral blood stem cell transplantation (allo-PBSCT) and 4 CML patients on imatinib treatment. Expression of bcr-abl mRNA was detected by RT-PCR and tyrosine phosphorylation of BCR-ABL fusion protein by Western blot.
RESULTSExpression of hMSH2, hMSH3 and hMLH1 mRNA was significantly lower in CML and K562 cells than in normal control (P < 0.05). In 26 CML with allo-PBSCT and 4 CML patients on imatinib treatment, expressions of hMSH2, hMSH3 and hMLH1 mRNA was enhanced while expression of bcr-abl mRNA decreased. In CML MNC after imatinib treatment and in K562 cells, expression of hMSH2, hMSH3 and hMLH1 mRNA was enhanced while tyrosine phosphorylation of BCR-ABL fusion protein decreased.
CONCLUSIONExpressions of hMSH2, hMSH3 and hMLH1 mRNA were down-regulated by bcr-abl fusion gene.
