1.The efficacy and safety of HCPT combined with 5-fluorouracil in treatment of bladder cancer

Objective To investigate the efficacy and safety of HCPT combined with 5-fluorouracil(5-Fu) in treatment of bladder cancer .Methods Selected 92 patients with advanced bladder cancer in our hospi-tal from February 2011 to August 2015 , equally divided into observation group and control group of 46 patients based on the random number table ,the control group were received 5-Fu intravesical chemotherapy ,The obser-vation group were given the HCPT combined with 5-Fu chemotherapy ,the chemotherapy infusion were once a week,continuous perfusion were observed for 3 months.Results The response rates in the observation group was 95.7%and the control group was 76.1%,the observation group were significantly higher than the control group (P<0.05).The occurrence of bladder irritation adverse reaction ,blood abnormalities,liver and kidney dysfunc-tion,bone marrow suppression in the observation group during chemotherapy compared with the control group were no significant difference(P>0.05),all adverse reactions after symptomatic treatment were improved .All patients were followed up for 1 year,The mortality and recurrence rates in the observation group were 8.7%and 4.3%re-spectively,while the control group were 30.4%and 17.4%,the mortality and recurrence rates of the observation group were significantly lower than the control group (P<0.05).Conclusion HCPT combined with 5 -Fu chemotherapy for bladder cancer has strong anti -cancer effect and without increase the incidence of adverse re-actions,it can promote the improvement of the long -term prognosis,so it is a safe and effective topical bladder chemotherapy drugs .
2.Research in reliability and validity of the Chinese version of job satisfaction scale

Objective To evaluate the reliability and validity of the Chinese version of job satisfaction scale in order to supply quantitative tool for evaluation of nurses" job satisfaction degree. Methods 336 nurs-es from 14 general hospitals were tested with Chinese version of job satisfaction scale and analysis was carried out for reliability and validity according to the test result. Results The total Cronbach's α coefficient of Chi-nese version of job satisfaction scale was 0.901, Cronbach's α coefficient range of every dimension was 0.732~ 0.921, re-test reliability was 0.832, 9 factors were extracted through factors analysis and they explained 59.57%of the overall variance. Conclusions Chinese version of job satisfaction scale has good reliability and validi-ty, it can be used in evaluation of nurses' satisfaction degree preliminarily.
3.Role of P21 protein and XIAP on apoptosis-inducing effect by mevastatin against A549 cell line

Objective To investigate the role of P21 protein and XIAP on apoptosis-inducing effect by mevastatin against A549 cell line and its mechanisms. Methods The cells proliferation was detected by MTT assay. The cell cycle distribution and apoptosis induction were evaluated with flow cytometer and electron microscopy. The mRNA expression of P21 and XIAP was measured with reverse transcription-polymerase chain reaction.The protein expression of P21 and XIAP was tested with flow cytometer and Western Bolt respectively. Results Mevastatin inhibited A549 cell survival in a time-dependent and concentration-depended manner. Flow cytometry showed that mevastatin induced G0/G1 phase arrest and caused apoptosis. Mevastatin did not affect P21 expression at both mRNA and total protein level.Concomitantly,P21 protein localized on cellular membrane decreased.Both mRNA and protein expression in XIAP were down regulated by mevastatin. All these effects were reversed by mevalonate.Conclusion Mevastatin can inhibite proliferation,induce apoptosis and interfere with cell progression of A549 through mevalonate pathway. Its mechanisms are explained by decreasing the expression of XIAP mRNA and protern.Targeting HMG-CoA reductase,Mevastatin blocks the isoprenylation of P21 protein which affects its anchorage on the cellular membrane.
4.Role of P21 protein and survivin on apoptosis-inducing effect by mevastatin against NSCLC cells

Objective To investigate the role of P21 protein and survivin on the cell apoptosis of non-small cell lung cancer (NSCLC) induced by mevastatin. Methods The inhibitory effect of mevastatin on A549 and NCI-H520 cell line was evaluated by MTT assay. The cell cycle distribution and apoptosis induction were determined with flow cytometer and transmission electron microscope. The expression of p21 protein was assessed with flow cytometer. The mRNA expression of p21 and survivin was assessed with RT-PCR. Results Flow cytometry showed that mevastatin induced G_0/G_1 cell arrest in NSCLC cell lines. The results indicated that mevastatin caused apoptosis in concentration-dependent manner. Mevastatin produced no change in expression of P21 mRNA and total P21 protein. Concomitantly, P21 protein localized on cellular membrane was decreased. It was also found that mevastatin suppressed the expression of survivin mRNA in NSCLC cell lines. All these effects were reversed by mevalonate. Conclusions Mevastatin inhibited the proliferation of NSCLC cell lines. Mevastatin exerts growth inhibitory effect and induces apoptosis effect by inhibiting mevalonate synthesis. Its mechanisms might involve blockade of the isoprenylation of p21 protein and down-regulation of the expression of survivin mRNA.
5.The expression and clinical signiifcance of miR-221 in plasma of patients with small cell lung cancer

Background and purpose:MicroRNAs (miRNAs, miR) are directly involved in cancer initiation, progression and metastasis. Alterations of miRNAs expression in cancer tissue may be reflected in circulation. We attempted to investigate the expression and clinical signiifcance of plasma miR-221 in patients with small cell lung cancer (SCLC). The plasma and tissues levels of miR-221 in 51 SCLC patients and 20 controls were evaluated and compared among various clinicopathological characteristics. Methods: Confirmation of higher miR-221 levels in primary SCLC tissues than normal lung tissues. Evaluation of plasma miR-221 concentrations by comparing results from 51 consecutive SCLC patients and 20 healthy volunteers. Evaluation of the assay for monitoring tumour dynamics in SCLC patients. Results:Expression of miR-221 was signiifcantly higher in SCLC tissues than in para-carcinoma tissues and normal lung tissues. Plasma miR-221 concentrations were signiifcantly higher in SCLC patients than those in normal people (P<0.05). The expression of plasma miR-221 was not associated with gender, and age (P>0.05), correlated with significantly chemosensitivity, overall survival and clinical stage (P<0.05). Cox regression analysis indicated that plasma miR-221 expression and disease stage were found to be significantly independent prognostic factors for the SCLC patients. Conclusion: Plasma miR-221 could be a useful biomarker for cancer detection, monitoring tumour dynamics and predicting malignant outcomes in SCLC patients, and may contribute to clinical decision making in treatments.
6.24 h esophageal pH monitoring in patients with gastroesophageal reflux-induced cough treated with omeprazole

To monitor the 24 h esophageal pH in patients with gastroesophageal reflux-induced cough (GERC) before and after omeprazole treatment. Methods The prospective study was conducted in Department of Gastroenterology, People's Hospital of Wuhan University from February 2007 to April 2008. Twenty patients with GERC received 40 mg of omeprazole daily and underwent 24 h esophageal pH monitoring before and 8 weeks after omeprazole treatment. Six parameters by Johson and Demeester were adopted for evaluation: pereentage of total time with pH <4, percentage of uptight reflux time with pH <4, percentage of supine reflux time with pH < 4, the number of reflux episodes with pH < 4, the number of reflux episodes longer than 5 min and the percentage of longest reflux duration. Results The values of six parameters before treatment were 13.5±8.5, 12.2±4.0, 15.8 ±4.1, 56 ± 13, 4. 15 ± 1.87 and 26.2 ± 16. 5 respectively; while those after treatment were 2.7± 1.3, 3.9 ± 1.9, 1.9 ± 1.0, 18 ± 11, 1.24 ±0.65 and 7.4 ± 2. 1. There were significant differences in these six parameters between pre-treatment and post-treatment (P<0.01 ). Conclusion The 24 h esophageal pH monitoring shows that omeprazole is effective in treatment of GERC by decreasing esophageal pH.
7.The Effect of the Replantation and Full-crown Prosthesis in the Treatment of Refractory Molar Periapical Disease

Objective This paper will study the effect of the replantation and full crown prosthesis in the treatment of refractory molar periapical disease as well as their related factors .Methods The retrospective analysis will be used in this paper.There were 98 patients who were diagnosed as refractory molar periapical disease in our dental clinic from 2012 to 2015.These 98 cases are divided into two groups:the study group and the control group .This paper will make a comparison of the study group which represents the replantation and full crown prosthesis in the treatment and the control group which re -presents endodontic therapy .Results Through the comparison , the average effective rate of the study group is 93.9%while the control group 65.3%.There is a significant difference between two groups (P<0.01) and the rate of the study group is higher than that of the control group .Conclusion Through the analysis , this paper finds that the joint optimization of replan-tation and full crown prosthesis can provide an unconventional approach to treat refractory periapical of teeth and effectively preserve the tooth .

9.Analysis of 27 Cases of Anaphylactoid Reactions Caused by Magnesium Isoglycyrrhizinate Injection

OBJECTIVE:To explore the regularity and characteristics of anaphylactoid reactions induced by Magnesium iso-glycyrrhizinate injection,and provide reference for clinical rational drug use. METHODS:27 ADR reports induced by Magnesium isoglycyrrhizinate injection from Hunan Center for ADR Monitoring from Jan. 2014 to Mar. 2015 were retrospectively analyzed. RE-SULTS:In the 27 cases,15 were male (55.56%) and 12 were female (44.44%),with percentage of 1.25∶1;ADR mainly oc-curred in patients with 41-60 years old(51.85%);involved organ/system were mainly skin and its appendages(35.85%)and sys-temic damage(26.42%),the main clinical manifestations were rash,itching,fever,chills,vomiting and dizziness;there were 7 severe ADR reports(25.92%),mainly showed chills,fever and other systemic symptoms;most ADR can be cured(40.74%)or relieved(55.56%)by related processing. CONCLUSIONS:There are no risk tips for severe allergic reactions in instruction,it is necessary to strengthen post-marketing surveillance and further improve the drug instructions to reduce the risk of severe ADR in-duced by Magnesium isoglycyrrhizinate injection.
10.Study on the Extraction Technology of Polysaccharides from Antlion and Its Immune Activity in vivo

OBJECTIVE:To optimize the extraction technology of polysaccharides from antlion and explore its effect on im-mune functions of mice. METHODS:Using content of polysaccharides as investigation index,the effects of extracting polysaccha-rides from antlion by water extraction method protease hydrolysis extraction(optimized by orthogonal test using extraction tempera-ture,enzyme dosage,extraction time as indexes),and diluted alkali extraction(optimized by orthogonal test using alkali concentra-tion,extraction temperature,extraction time as indexes)were compared. 128 KM mice were randomly divided into 4 groups,then randomly divided into control group(normal saline),polysaccharides low-dose,medium-dose,high-dose groups(20,40,80 mg/kg),8 in each group,iv in tail vein,0.2 mL/10 g,once a day,for 1 week,which were respectively used to determine the phago-cytosis percentage and phagocytic index of peritoneal macrophages,spleen and thymus index,lymphocyte transformation rate and serum hemolysin levels. RESULTS:The contents of polysaccharides by 3 methods were 14.48%,38.66%,30.62%,respectively. The content of polysaccharides by protease hydrolysis extraction was the highest,the optimal extraction technology were as follows as using 100 μg/g papain extracting 3 h under 40 ℃. Compared with control group,phagocytosis percentage,phagocytic index, spleen index in polysaccharides low-dose,medium-dose,high-dose groups were significantly increased (P<0.05),thymus index was significantly decreased(P<0.05),while lymphocyte transformation rate had no significant changes(P>0.05);serum hemoly-sin in polysaccharides medium-dose group was significantly increased (P<0.05). CONCLUSIONS:Protease hydrolysis extraction is suitable for the extraction of polysaccharides from antlion,the optimal technology is reliable. Polysaccharides from antlion show activity in enhancing mice non-specific immunity and humoral immunity.
