1.The effects of tanshinone inhibiting calmodulin signal system on malignant arrhythmias after myocardial infarction

Objective To observe the effects of tanshinone on malignant arrhythmias which occured in rabbit model of myocardi-al infarction,and explore the relationship between tanshinone induced calmodulin(CaM)signal system changes and the incidence of malignant arrhythmias after myocardial infarction.Methods A total of 90 healthy big ear rabbits were selected and randomly divid-ed into 3 groups:myocardial infarction model rabbits(non-intervention group)and myocardial infarction model rabbits with tanshi-none intervention(intervention group)and sham operated rabbits(control group),30 rats in each group.The incidence of malignant arrhythmias,calmodulin(CaM),calmodulin kinaseⅡ(CaMKⅡ),transmural dispersity of repolarization(TDR)and the concentra-tion of Ca2 + in myocardial cells were statistically analysed and compared among the 3 groups.Results The control group did not undergo malignant myocardial infarction,in non-intervention group and intervention group,malignant arrhythmia incidence was higher than that in control group(P <0.05);in non-intervention group,malignant arrhythmia rate(60.00%)was higher than that in intervention group(10%)(P <0.05 ).The CaM concentration in non-intervention and intervention group rabbits were 1.483 and 1.321 times of control group rabbits respectively,the difference was statistically significant(P <0.05);CaM protein levels of non-intervention group was higher than that of intervention group(P <0.05).No statistical significance of CaMKⅡ activity among the 3 groups were observed(P >0.05).Serum Ca2 + concentration in non-intervention group was higher than that in control group and intervention group(P <0.05);serum Ca2 + concentration in control group was lower than that in intervention group(P <0.05).The TDR of intervention group is lower than that of non-intervention group,the two groups′TDR were both higher than that of control group(P <0.05).At single cell level,the intracellular calcium concentration of intervention group was lower than that of non-inter-vention group (P <0.05).Conclusion The relevant signal factors of CaM signal transduction system have implications on malig-nant arrhythmia after myocardial infarction.Tanshinone can inhibit CaM signaling system and play an important role in the preven-tion of malignant arrhythmia after myocardial infarction.
2.Research progress of chimeric antigen receptor T cell and T cell antigen coupler modified T cell technology

At present,malignant tumor has become one of the public problems that seriously threaten human health. In addition to surgery,radiotherapy,chemotherapy,targeted therapy and other methods,with the development of molecular biology,immunotherapy has also developed rapidly,becoming an emerging method of cancer treatment. The most commonly used immune cells in clinical treatment are DC,NK,CIK,CTL and chimeric antigen receptor T cell(CAR-T). Among them,CAR-T technology is the initial technology for global research,while due to its off-target,neurotoxicity,transfection vector defects and other problems,it also has certain limitations in clinical application. T cell antigen coupler modified T cell(TAC-T)technology is a new technology developed on the basis of CAR-T,which uses natural T cell receptor(TCR)to modify T cells and retarget the antigen of cancer cells. In this paper,the research status of CAR-T technology and the research progress of TAC-T technology are reviewed in order to provide reference for further study on the mechanism of TAC-T technology and its safety of clinical application.
3.Detection the MLH3 gene C2531T mutation in oligospermia and azoospermia by 4P-ARMS-PCR

Objective Analysing the potential relationship of mismatch repair protein MLH3 gene C2531T mutation with oligospermia and azoospermia cases. Methods Using MLH3 C2531T as an example to establish a method of tetra-primer amplification refractory mutation system-PCR (4P-ARMS-PCR); We chose 81 cases of oligospermia, azoospermia patients and 80 normal controls, detecting MLH3 C2531T both using 4P-ARMS-PCR and PCR-RFLP; randomly to select 20% samples to sequence. Results The results suggested higher prevalence of CT + TT genotypes in patients as compared to the controls and association of T allele with disease (P <0. 01); PCR-RFLP results , 4P-ARMS-PCR results and sequence results are fully consistent. Conclusion 4P-ARMS-PCR can be a fast, easy-operating and accurate technology for detection of gene mutation of single nucleotide polymorphism (SNP) ; MLH3 C2531T polymorphism is related to oligospermia and azoospermia.

In recent years, with the changes of human living environment, the morbidity of respiratory tiact allergic diseases such as allergic rhinitis, infant wheezing, allergic asthma, and other diseases increased rapidly, thereby affecting the quality of children′s life, and increasing heavy burdens to families and society.Microbial infection, including virus, bacteria, mycoplasma pneumoniae/chlamydia, fungi and parasites, can regulate the occurrence and development of respiratory tract allergic diseases through the immune mechanism.In order to deepen the understanding of the relationship between respiratory allergy and microbial infection and its related mechanism, the research progress on the relationship between respiratory allergy and microbial infection was reviewed in this paper.
6.Comparison of different chemiluminecence immunoassay analyzers for determination of serum thyroid stimulating hormone in patients with suspicious subclinical hypothyroidism

Objective To evaluate the differences of serum TSH of suspicious subclinical hypothyroidism determined by four automatic biochemical analyzers and the impact on clinical diagnosis and treatment.Methods Taking results of Roche Cobas e601 laboratory test as a reference, 103 serum samples with TSH 2.50-10.00 mU/L(90 with TSH≥4.27 mU/L) and normal FT3, FT4 were selected.Four different automatic biochemical analyzers (Cobas e601, Immulite2000, Centaur XP, I2000) were used to measure TSH of the serum samples at the same time.Wilcoxon signed rank test, Spearman correlation analysis were used for data analysis.Results TSH (M(P25, P75)) measured by 4 methods were 5.20(4.73, 6.40), 2.95(2.59, 3.48), 3.30(2.94, 4.15) and 4.10(3.43, 4.75) mU/L, which varied significantly from one assay to another (z values:-8.78,-8.41,-7.64,-8.09,-8.50, all P<0.05).The correlations between methods were of great differences (rs ranged from 0.45 to 0.92).Significant differences existed in each other for subclinical hypothyroidism diagnosis based on TSH cutoff respectively.Conclusion Results from different automatic immunoassay analyzers in patients with TSH of 2.50-10.00 mU/L varied widely, hence, it is indeterminate to diagnose subclinical hypothyroidism only relies on a single serum TSH test.
7.Analysis on the effect of entecavir dispersible tablets on the treatment of patients with decompensated hepatitis B cirrhosis

Objective To study and analyze of entecavir dispersible tablets on the treatment of patients with decompensated hepatitis B cirrhosis. Methods From February 2014 to September 2016, 100 patients with decompensated hepatitis B cirrhosis were randomly divided into the control group and the experimental group, 50 patients in each group. The control group were received routine treatment, while the experimental group were received entecavir dispersible tablets treatment on the basis of conventional treatment. The therapeutic effects in the two groups were compared and analyzed. Results The effective rate in the experimental group was 92.0%, and 70.0% in the control group. The effective rate in the experimental group was significantly higher than that in the control group, the difference has statistically significant (P<0.05). The Child-pugh score in the experimental group was (6.12±1.43) points, and the score in the control group was (7.23±1.95) points. The score in the control group was significantly higher than that in the experimental group, with statistical difference (P<0.05). The negative rate of HBeAg in the experimental group was 22.0%, while 86% in the control group the difference has statistically significant (P<0.05). Conclusion The clinical effect is better which entecavir dispersible tablets is used in the treatment of the patients with decompensated hepatitis B cirrhosis, which can improve the treatment efficiency to a great extent, recover the liver function, has the further promotion and application significance.
8.Content determination of irbesartan in drugs by HPLC

Objective To establish a HPLC method for the content determination of irbesartan in drugs.Methods The diamonsil C18 column (150 mm ×4.6 mm,5μm) was used, with the mobile phase of phosphoric acid solution-acetonitrile(adjusted to pH 3.2 with triethylamine)(62:38).The column temperature was 35℃, with the UV detection wavelength of 245 nm and a flow rate of 1.0 mL/min.Results Irbesartan exhibited a good linear relationship with the peak area in the concentration range of 0.04-0.40 mg/mL with the correlation coefficient of 0.999 99.The average recovery was 98.37% with RSD of 0.7%(n=9).Conclusion This method is simple and accurate with wide linear range and good reproducibility,and can be used to determine the content of irbesartan in drugs.
9.Feasibility of medical glue for open wound of the frontal plane in children

BACKGROUND:Medical glue as a biological hemostatic material shows a good diffusivity in body fluids and blood, and has a rapid polymerization reaction, which plays an ideal hemostatic effect and promotes wound healing. OBJECTIVE:To investigate the clinical value of medical glue as a hemostatic material in the open wound of the frontal plane in children. METHODS:Totaly 1 218 children with open wounds of the frontal plane treated with medical glue were enroled as observation group, and another 600 children with open wounds of the frontal plane treated with silk sutures as control group. Wound healing time, postoperative wound bleeding, suture reaction, pigmentation and scar hyperplasia were compared between the two groups. RESULTS AND CONCLUSION:Compared with the control group, the operative time, blood loss and wound healing time were al less in the observation group (P < 0.05), and the effective bleeding rate was higher in theobservation group (P < 0.05). Stage I wound healing occurred in 1 215 cases of the observation group, and 586 cases of the control group. At the end of 12-month folow-up, healed scar formed with no knot scar and needle scar, the skin color was similar and cosmetic results were good in the observation group; in the control group, there were more obvious scars, visible knot and needle scars, and erythema, induration and hypertrophic scars were found around the most wounds. The medical glue for open wounds of the frontal plane in children has good biocompatibility and cosmetic results, which is safe and convenient in clinical use.
10.Repairing experimental femoral head necrosis of goat with BMP-2 gene medicine

Objective To repair the experimental femoral head necrosis with the implantation of tricalcium phosphate (TCP) loaded with human bone morphogenetic protein-2(hBMP-2) gene transfected bone marrow derived mesenchymal stem cells(BMSCs). Methods Established the animal models of femoral head necrosis in 22 goats by the ligation of the lateral and medial circumflex femoral arteries and delivery of liquid nitrogen into femoral head. 4 goats served as the control group without any treatment. 3 weeks after surgery, necrotic bone in the other 18 goats were removed and TCP loaded gene transfer cells were implanted into the femoral head with BMP-2 gene transfected BMSCs in right side and ?-gal gene transfected BMSCs in left side. BMP-2 concentrations in femoral head were tested by the ELISA at 1st, 2nd, 3rd week after implantation. Histological observation was done before and at 6th, 10th, 16th week after implantation. New bone volumes (NBV) were measured at 16th week after implantation by the histomorphometry method. X-ray was taken at 16th week after operation in untreated group and after treatment in BMP-2 group and ?-gal group. Results The concentration of BMP-2 in femoral head of BMP-2 group was higher than that of ?-gal group at different time point. Empty lacunae and fibrotic marrow were demonstrated before implantation. New bone was evident in the implantation field of BMP-2 group while fibrous tissues were evident in that of ?-gal group at 6th, 10th and 16th week after treatment. Histomorphology analysis indicated that the NBV in BMP-2 group was significantly higher than that in ?-gal group. Articular surface collapse were observed in the X-ray photograph of untreated group. Regular shape and normal density of femoral head in BMP-2 group compared with the irregular shape and low density of femoral head in ?-gal group could be demonstrated in the X-ray photograph 16 weeks after treatment. Conclusion Implantation of the TCP loaded with human BMP-2 gene transfected BMSCs could repair early-stage experimental femoral head necrosis.
