1.Evaluate regional myocardial function of hypertrophic cardiomyopathy patients by quantitative tissue velocity imaging

Objective To explore value of quantitative tissue velocity imaging(QTVI) in the regional systolic and diastolic myocardial function of hypertrophic cardiomyopathy(HCM) patients and healthy persons. Methods By Doppler myocardial imaging 20 HCM patients and 18 healthy subjects were collected,then the regional velocity were measured in the mid and basal segment in systolic,early diastolic and later diastolic period respectively. Results The parameters as systolic velocity(Vs),early diastolic velocity(Ve) and early systolic velocity/later diastolic velocity (Ve/Va) of the HCM group were significantly lower than those of the normal group,while later diastolic velocity(Va) showed no statistic difference between two groups. Conclusions The systolic and diastolic myocardial functions of hypertrophic cardiomyopathy patients are reduced. The abnormality of heart function in HCM patients could be accurately detected by QTVI.
2.Application of chitosan and its derivatives in bone tissue engineering

Chitosan, deacetylated derivative of chitin, is a kind of natural polysaccharide polymer. It has advantages of rich source, good biocompatibility and biodegradation. Chitosan can be processed into porous scaffolds used for cell transplantation and tissue regeneration in bone tissue engineering, control-released carrier of growth factors and delivery vector of exogenous genes. Meanwhile, it can also be processed into injectable scaffolds used in bone tissue engineering in the form of microspheres or hydrogel. Chitosan and its derivatives will have broad application prospects in the research field of bone tissue engineering. However, chitosan composite scaffold has poor mechanical function, difficult accuracy control of In vivo degradation, and low efficiency for genetic carrier, chitosan research stays in in vitro tests and in vivo animal experiments. With the development of materials science and life science, chitosan will be widely used for clinical application of bone tissue defect.
3.The expression and clinical significance of MyD88 in laryngeal cancer.

OBJECTIVE:
To investigate the myeloid differentiation factor 88 (MyD88) expression in laryngeal carcinoma and its clinical significance.
METHOD:
Fifty-one patients with laryngeal carcinoma were collected, and all patients were confirmed by pathological diagnosis results. The expression of MyD88 protein was detected by immunohistochemical method in laryngeal cancer and its adjacent tissues to investigate the correlation among MyD88 expression, clinicopathological characteristics and prognosis of patients.
RESULT:
The positive expression rate of MyD88 in laryngeal cancer tissues was 68.6%, significantly higher than that in normal tissues adjacent to carcinoma of which positive expression rate was 11.8%; MyD88 positive rate had nothing to do with laryngeal cancer patients age, sex, differentiation and tumour location (all P > 0.05), but correlated with clinical stage (P < 0.01) and lymph node metastasis (P < 0.05). In addition, the study also found that the expression of MyD88 quantity was inversely proportional with the five-year survival rate. The survival rate of patients with higher expression of MyD88 was significantly lower than that of patients with lower expression (P < 0.05).
CONCLUSION
MyD88 may be an important participant in the pathogenesis of laryngeal carcinoma, MyD88 targeted therapy may improve the prognosis of patients with laryngeal cancer.

5.Evaluation of regional myocardial function in hypertrophic cardiomyopathy patients and healthy persons by strain imaging

Objective To evaluate the regional myocardial systolic function in the patients with hypertrophic cardiomyopathy(HCM) by strain imaging technique.Methods Doppler myocardial imaging of 20 HCM patients(HCM group) and 22 healthy subjects(control group) was collected,and their regional peak systolic strain in the mid and basal segment was measured respectively.The data were compared and analyzed between the two groups.Results In the healthy subjects,the strain value showed no difference between regional myocardium.In the interventricular septum,anterior and inferior wall of left ventricle,the strain value of HCM group was significantly lower than that of control group.Conclusions Strain imaging could detect the abnormal regional myocardial function of HCM patients accurately, and provide a new quantitative parameter for evaluating the regional myocardial functions.
6.Discovery of potential nicotinic acid receptor agonists from Chinese herbal medicines based on molecular simulation.

Nicotinic acid could increase high density lipoprotein and reduce serum total cholesterol, low density lipoprotein cholesterol and triglycerides in human bodies, thus is frequently applied in treating low high-density lipoprotein cholesterol and hypertriglyceridemia in clinic. However, according to the findings, nicotinic acid could also cause adverse effects, such as skin flush, beside its curative effects. In this study, bioisosterism, fragment-based search and Lipinski's Rule of Five were used to preliminarily screen out potential TCM ingredients that may have similar pharmacological effects with nicotinic acid from Traditional Chinese medicine database (TCMD). Afterwards, homology modeling and flexible docking were used to further screen out potential nicotinic acid receptor agonists. As a result, eleven candidate compounds were derived from eight commonly used traditional Chinese medicines. Specifically, all of the candidate compounds' interaction with nicotinic acid receptor was similar to nicotinic acid, and their docking scores were all higher than that of nicotinic acid, but their druggability remained to be further studied. Some of the eight source traditional Chinese medicines were used to lower lipid according to literature studies, implying that they may show effect through above means. In summary, this study provides basis and reference for extracting new nicotinic acid receptor agonists from traditional Chinese medicines and improving the medication status of hyperlipidemia.

7.A review of detection methods for human bocaviruses.

Human bocavirus (HBoV) 1-4 have been detected both in respiratory and stool samples since the first HBoV was discovered in 2005. HBoV-1 is mostly associated with respiratory infection, while HBoV 2-4 are usually associated with intestinal tract infection. A variety of signs and symptoms have been described in patients with HBoV infection, including cough, wheezing, pneumonia, and diarrhea, but the research on pathogenic mechanism of HBoV is limited because HBoV cannot be cultured in vitro due to the lack of appropriate host cells. Three-dimensional epithelial cell culture, reverse genetics, and viral metagenomics are identified as novel tools that may promote the research on pathogenic mechanism of HBoV and the discovery of new viruses. This review summaries currently available diagnostic approaches such as electron microscopy, cell culture, PCR, and immunoassay in order to provide a method reference for indepth research on HBoV.

8.Identification and characterization of marker chromosome in Turner syndrome

Objective To analyze the karyotypes of 11 cases of Turner syndrome with marker chromosome,and study the phenotypic effects resulting from the abnormal karyotype.Methods Eleven Turner syndrome patients had a mosaic karyotype and carried a marker chromosome,and 6 marker chromosomes were ring chromosomes.Their karyotypes were showed as mos.45,X/46,X,+mar or mos. 45,X/46,X,+r.Fluorescence in situ hybridization(FISH)technique with X/Y centromere probes was performed to determine the origin of the marker chromosome.Reverse chromosome painting technique was used to identify the breakpoints of two largest markers.Phenotype effects with different chromosome breakpoints were compared.Results All the 11 marker chromosomes were ring X chromosomes.The breakpoints of the r(X)were involved in Xp22,Xq22,Xq24 and Xq26,etc.Conclusions The marker chromosomes in Turner syndrome mainly originate from X chromosome and form ring chromosome X.Each r (X)in our patients was mosaic,indicating it was originated from mitosis error during early embryo development.To analyze the origin of the marker chromosome and the breakpoint of r(X)will provide guidance for the therapy and prognosis of the Turner syndrome patient.
9.Study on anti-hyperlipidemia mechanism of high frequency herb pairs by molecular docking method.

Traditional Chinese medicine (TCM) has definitely clinical effect in treating hyperlipidemia, but the action mechanism still need to be explored. Based on consulting Chinese Pharmacopoeia (2010), all the lipid-lowering Chinese patent medicines were analyzed by associated rules data mining method to explore high frequency herb pairs. The top three couplet medicines with high support degree were Puerariae Lobatae Radix-Crataegi Fructus, Salviae Miltiorrhizae Radix et Rhizoma-Crataegi Fructus, and Polygoni Multiflori Radix-Crataegi Fructus. The 20 main ingredients were selected from the herb pairs and docked with 3 key hyperlipidemia targets, namely 3-hydroxy-3-methylglutaryl coenzyme A reductase (HMG-CoA reductase), peroxisome proliferator activated receptor-α (PPAR-α ) and niemann-pick C1 like 1 (NPC1L1) to further discuss the molecular mechanism of the high frequency herb pairs, by using the docking program, LibDock. To construct evaluation rules for the ingredients of herb pairs, the root-mean-square deviation (RMSD) value between computed and initial complexes was first calculated to validate the fitness of LibDock models. Then, the key residues were also confirmed by analyzing the interactions of those 3 proteins and corresponding marketed drugs. The docking results showed that hyperin, puerarin, salvianolic acid A and polydatin can interact with two targets, and the other five compounds may be potent for at least one of the three targets. In this study, the multi-target effect of high frequency herb pairs for lipid-lowering was discussed on the molecular level, which can help further researching new multi-target anti-hyperlipidemia drug.

10.Plasma omentin level and related factors in type 2 diabetes mellitus combined with fatty liver disease

Objective To assay the plasma omentin level in patients of type 2 diabetes mellitus (T2DM) combined with non-alcoholic fatty liver disease (NAFLD) and investigate the relationship between plasma omentin level,glucose and lipid metabolism,insulin resistance and NAFLD.Methods The plasma omentin level was assayed by enzyme-linked immunosorbent assay(ELISA) in all subjects,including patients of T2DM controls with NAFLD (group A,50 cases),T2DM without NAFLD (group B,50 cases),simple with NAFLD(group C,51 cases) and normal controls (group D,49 cases).Meanwhile,blood glucose,glycosylated hemoglobin(HbA1c),lipids and insulin levels were also measured.Body mass index (BMI) and waist-to-hip ratio were evaluated.Insulin sensitivity was assessed by HOMA-IR.Results The plasma omentin level was (17.85 ±3.68),(13.89 ±10.68),(26.05 ±7.26) and (22.92 ±2.71)μg/L in group A,B,C and D respectively.The plasma omentin level of group A and group B was significantly lower than that of group C and group D(P < 0.05).The plasma omentin level of group A was higher than that of group B (P < 0.05).The plasma omentin level of group C was higher than that of group D (P< 0.05).Correlation analysis showed that the plasma omentin level was negatively correlated to weight,BMI,waist,triglyceride,fasting blood glucose,fasting insulin (FINS) and HOMA-IR (P <0.05 or <0.01),and positively correlated to high-density lipoprotein cholesterol (HDL-C)(P <0.01).Multiple stepwise regression analysis showed that BMI,HOMA-IR and FINS was independent variable of omentin.The concentration of omentin was 24.82 μ g/L which could predict the risk of NAFLD in people with normal glucose regulation.Conclusions The plasma omentin level is closely correlated with glucose,lipid metabolism and insulin resistance.Plasma omentin may play an important role in the pathogenesis of T2DM and NAFLD.
