1.Study on the Anti-inflammatory Effect of Biota orientalis

AIM The purpose is to investigate the Anti-inflammatory mechanism of Biota orientalis. METHODS Using rat leukocytes and rabbit platelets as experimental material the biosynthesis of leukotriene, 12-hydroxyheptadecatrienoic acid(12-HHT) and 12-hydroxyeicosatetraenoic acid(12-HETE) in the cells was determined with HPLC. RESULTS Biota orientalis was shown to inhibit the biosynthesis of leukotriene B4(LTB4) and 5-hydroxyeicosatetraenoic acid(5-HETE) with IC50 of 0.40 and 0.41 mg/ml crude herb, respectively. It was also displayed to inhibit biosynthesis of 12-HHT in the platelets. CONCLUSION Biota orientalis leaves contain anti-inflammatory components and the anti-inflammatory mechanism was related with inhibiting metabolism of arachidonic acid.
2.THE BREEDING STUDY BY UV-INDUCING PROTOPLASTS OF LENTINUS EDODES

The experiment was carried out to inquire about the possibility of strain selection and improvement in Lentinus edodes through protoplast formation and mutagenesis by UV. Mycelial growth rate, yield, and period of fructification of 105 selected regenerants from UV-treated protoplasts have been measured and compared with their parents. Several selected regenerants achieved some better qualities such as higher yield and earlier fruiting. Several repeated cultivations of these regenerants reconfirmed their new capacities. The results indicated that protoplast formation and mutagenesis are the potential methods for strain selection and improvement in edible fungi.
3.Study on the anti-inflammatory effect of proanthocyanidins and its mechanism

Objective To investigate the anti-inflammatory effect of proanthocyanidins and its mechanisms.Methods Inflammation models such as dimethylbenzene-indueed ear swelling and carrageenan-induced hind paw edema in mice and rats were prepared.The contents of PGE2 in exudate from edema paws of rats were measured by ultraviolet spectrophotography and the protein expression of COX-2 in edema paws of rats by Western-blot and immunohistoehemistry(IHC)assay.Results Pro-anthocyanidins remarkably inhibited the ear edema induced by dimethylbenzene in mice at the dose of10 mg/kg and 20 mg/kg;paw edema of rats induced with carrageenan was significantly inhibited byproanthocyanidins 5,20 mg/kg ip from 2 to 5 h;proanthoeyanidins 5,20 mg/kg ip reduced the contents of PGE2 in exudate from edema paws of rats induced by carrageenan;proanthocyanidins 5,20 mg/kg ip inhibited the protein expression of COX-2.Conclusion Proanthocyanidins has an anti-inflammatory effect in vivo which may be related to inhibition of protein expression of COX-2 and downregutation of PGE2 biosynthesis.
4.Quantitative analysis of the 5-lipoxygenase mRNA in the rat peritoneal macrophages cultured in vitro by capillary electrophoresis

Objective To quantify the mRNA of 5-lipoxygenase (5-LO) in the rat peritoneal macrophages cultured in vitro by capillary electrophoresis (CE). Methods The rat peritoneal macrophages were isolated and cultured for indicated time. The mRNA of 5-LO was detected by RT-PCR, and the products of RT-PCR were quantified by CE. Results The DNA fragments in the 100 bp DNA marker and the products of the RT-PCR were separated successfully by CE with the sieving buffer containing 1.8% hydroxypropylmethylcellulose (HPMC). It proved that there were no significant changes on the expressions of 5-LO mRNA when the cells were cultured for 72 h quantified by CE. The mRNA of 5-LO significantly decreased by almost 80% by CE with the cells cultured for 120 h in vitro.Conclusions The products of RT-PCR could be separated and quantified by CE directly.The 5-LO mRNA could express normally in the rat peritoneal macrophages for 72 h in vitro.
5.Effects of navigation and robot technology on percutaneous ultrasound-guided liver tumor ablation: Accuracy and feasibility of clinical application

OBJECTIVE: To assess the effects of navigation and robot technology on percutaneous ultrasound-guided liver tumor ablation. METHODS:The liver model tumors were placed inside excised calf livers, and the needle was guided into the tumor using either conventional ultrasound-guided method or two new methods-3D navigation and a method based on a robotic assistant. Totally 40 experiments were performed, and the distance (all methods) from the tip of the needle to the planning point in the tumors was measured.RESULTS: Based on the measurements, the robotic assistant performed better than both the 3D navigation and the conventional ultrasound-guided method. While, the 3D navigation performed better than the conventional ultrasound-guided method.CONCLUSTON: This study demonstrates navigation combined with robot technique is feasible to improve the precision of deploying ablation needles, future studies are warranted to optimize the system to meet requirements in clinical practice.
6.Effect of the Nursing Intervention on Type 2 Diabetes in Communities

Objective To observe the effect of nursing intervention on type 2 diabetes in communities. Methods During Jul. 2007 to Jul. 2008, 256 cases with type 2 diabetes were chosen and intervened with non-medication treatment, direction for rational application of medication and management of the follow-up by community nurses. Results The awareness rates about blood presure, blood glucose, blood lipids and diabetes prevention & treatment among the patients were 47% , 43.8% , 60.7% and 91.5% . The compliance rates for medication, the treatment of non-medication and the attainment rate of self monitoring were 89%, 77.6% and 68%, respectively, and all of these results were significantly increased. Conclusion Nursing intervention on patients with Type 2 Diabetes in communities promotes the comprehensive treatment of diabetes.

Randomized control study of gemfibrozil and pantethine was made in 39 NIDDM patients to compare their effect on anti-lipid-peroxidation.The patients were divided into two groups at random. One group took gcmfibrozil 900 mg/d for 4 weeks, the ether group took pantethine 540 mg/d for 4 weeks. 9.Growth, reproduction and blood physiological and biochemical indexes in an atopic dermatitis mouse model

BACKGROUND:The skin lesions, pathological and immunological characteristics of Nc/Nga mice are consistent with human atopic dermatitis, and as an atopic dermatitis animal model, it has great research value. But there are no reports on the physiological and biochemical parameters of Nc/Nga mice in China. OBJECTIVE:To observe the growth and reproduction and blood physiological and biochemical parameters of atopic dermatitis model of NC/Nga mice METHODS:The data of reproductive performance of NC/Nga mice from the first to the third generation was analyzed, including mean litter size, weaning rate, pregnancy rate and generation interval. The body mass of 60 Nc/Nga mice with 1-56 days old was measured, 30 mice of female and male, then the growth curve was draw. The blood samples from the infraorbital vessels were col ected to detect the blood physiological and biochemical parameters in the mice. RESULTS AND CONCLUSION:The mean delivery interval of Nc/Nga mice was (25.8±3.1) days, mean litter size was (7.5±2.5) mice, mean weaning rate was (97.2±1.2)%, the mean pregnancy rate was (97.0±1.4)%, and there was no significant difference among the three generations of these mice (P>0.05). The body mass of Nc/Nga mice was increased with the time increasing of days, the body mass of the mice was maximal within 1-2 weeks post-weaning, and there was no significant difference in body mass between males and females at 6 weeks post-weaning (P>0.05). Comparison of the blood physiological and biochemical parameters of mice at the same age between males and females showed that the levels of red blood cells and hemoglobin in female mice were significantly higher than those in the male mice;the platelet count in the male mice was significantly higher than that in the female mice (P<0.05);the triglycerides and albumin levels in the female mice were higher than those in the male mice (P<0.05). The results indicate that the gender and age may influence the blood physiological and biochemical parameters of Nc/Nga mice.
10.Retroperitoneal Laparoscopy Deroofing for Parapelvic Cyst:Report of 16 Cases

Objective To explore the efficacy of retroperitoneal laparoscopy in the treatment of parapelvic cyst.Methods From August 2005 to December 2008,16 cases of parapelvic cyst were treated by retroperitoneal laparoscopic deroofing and decompressing in our hospital.Under general anaesthesia,an operational space was established via retroperitoneal route.And then,along the urethra,the parapelvic cyst was explored.Afterwards,the cyst was unroofed 3 to 5 mm from the normal renal tissues.Results The operation was completed in 15 of the patients with a mean operation time of(75?20) minutes and mean blood loss of(28?8) ml.No injuries to the kidney pedicle or pelvis occurred.Another patient was converted to open surgery because of difficulty in exposure of the cyst.This series were followed up for a mean of 13 months(range,6 to 20 months).No one had recurrence during the period.Conclusions Retroperitoneal laparoscopic deroofing is minimally invasive,safe and effective or parapelvic cyst.It should be the first choice for the disease.
