ObjectTo study the adsorptive properties of spherical activated carbon on some effective ingredients from Chinese materia medica (CMM) and to approach the application feasibility to CMM purification. Methods The spherical activated carbon with high strength has been successfully prepared by the carbonation and activation of polymeric resin. The apparent and theoretical adsorption capacities under the condition of static operation were investigated by Langmuir monolayer adsorption model. Elution by 75% alcohol was studied. Results The static adsorption capacities of the berberine hydrochloride and dioscin were 35.46 mg/g and 47.12 mg/g, and theoretical adsorption capacities were 96.16 mg/g and 102.04 mg/g, respectively. The apparent adsorption amount of rutin was 40.88% mg/g. The elution ratio were 83.71%, 91.45%, and 87.69%, respectively. Conclusion The spherical activated carbon adsorbent shows better comprehensive adsorption properties and can be used to purification.

AIM: To explore the effect of physcion (P) on the level of IL-1? and expression of ICAM-1 and caspase-3 during cerebral ischemia-reperfusion injury. METHODS: The 91 healthy adult SD rats were selected, and were randomly divided into normal group, sham-operated group, cerebral ischemia-reperfusion group (model), low-dose physcion (PLD) and high-dose physcion (PHD) treatment group. The level of IL-1? was detected by radioimmunoassay. The expression of ICAM-1 and caspase-3 was detected by immunohistochemistry. The changes of tissue pathology were also investigated. RESULTS: The level of IL-1? reached the peak at 6 h after ischemia-reperfusion (IR). The protein expression of ICAM-1 and caspase-3 reached the peak at 24 h after IR. The level of IL-1? and the protein expression of ICAM-1 and caspase-3 in PHD group decreased obviously compared with those in model group (P

Objective To study the impact of hepatitis B virus (HBV) infection on the activity of cord hematopoietic stem cells. Methods CD34+ cells were isolated from healthy human cord blood by miniMACS. Cells were cultured in IMDM complete culture medium containing stem cell factor (SCF),fms-like tyrosine kinase 3 ligand (FL), thrombopoietin (TPO), interleukin-3 (IL-3) and 10% fetal bovine serum. High copies HBV were added to the culture system. The proliferation of stem ceils and virus replication were observed. Following the proliferation, dendritic cells (DCs) were induced by adding granulocyte-macrophage colony-stimulating factor and IL-4. Morphous of stem cells and DCs were observed by microscope and the cell surface molecules were detected. Results The proliferation of stem cells infected with HBV was significantly lower than that of healthy stem cells (P<0.01),and enhanced after adding cytokines (P<0.01). At the same time, HBV replication was increased after adding cytokines in the culture system (P<0.01), but the proliferation was still lower than that of healthy stem cells with cytokines in the culture medium (P<0.05). Dane particles were found in the cytoplasma of stem cells infected with HBV by electron microscope. The expression of CD80,CD86 ,CD1a and HLA-DR on DCs derived from HBV infected stem cells were all lower than those on DCs from non-infected stem cells (P<0.01). Conclusions HBV could infect CD34+ stem cell and the proliferation of the stem cell could enhance the virus replication. HBV could not only inhibit the proliferation of stem cells, but also down-regulate the immuno-phenotype expression of DCs derived from CD34+stem cells.
