AIM: To study the expression of endostatin in ischemic myocardium of myocardial infarction(MI) rats in various periods and the correlation with VEGF expression and microvascular density(MVD).METHODS: Thirty-two male Sprague-Dawley rats after myocardial infarction were randomly divided into 7,14,21 and 28 days group.The sham group was normal control group(eight rats in each group).The expression of endostatin,VEGF and MVD in ischemic myocardium were observed by immunohistochemistry.RESULTS: The expression of endostatin significantly increased in the ischemic myocardium after MI,peaked at 7 days,then gradually decreased at 14,21 and 28 days.The endostatin level at 28 days was the same as the shams.The changing trends of expression of endostatin in ischemic myocardium after MI were similar to that of VEGF and were significantly correlated with the MVD.CONCLUSION: The expression of endostatin increased in ischemic myocardium of myocardial infarction rats.The changing trends of endostatin were similar to that of VEGF and positively correlated with the MVD.These data suggest that endostatin may modulate ischemic myocardium angiogenesis after myocardial infarction.

To investigate the influence of Bufei Jianpi Granule (BJG) on immune function in mice. Spleen cellular proliferation and the activities of IL-1 and IL-2 in mice were observed. The differences of spleen cellular proliferation stimulating by Con A, the secretion of IL-2 from spleen cells and the secretion of IL-1 from celiac macrophage were significant among BJG group, the control group and the water-drinking group. [Conclusion] BJG can promote the function of cellular immune and the secretion of IL-1 and IL-2 from immune active cells in mice.

Objective To discuss the feasibility of developing psychological pressure assessment tools coping with hospitalized patients based on Neuman systems model. Methods We analyzed the fea-sibility of developing assessing tools specifically by theoretical analysis, literature review and case study.Results It was proved that Neuman systems model was suitable for guiding the development and applica-tion of assessment tools for evaluating the psychological pressure. Conclusions Neuman Systems Model could effectively evaluate the psychological pressure of non-psychiatric settings. Therefore, it can be con-cluded that the model is suitable for developing the psychological assessment tools for hospitalized patients.

Objective To investigate the pathogenic features and antibiotic resistance profile of nosocomial and community-acquired spontaneous bacterial peritonitis (SBP) in liver cirrhosis patients.Methods Two hundred and twenty-six cirrhotic patients with SBP who were admitted to Beijin Ditan Hospital from January 2001 to December 2008 were recruited into this study. The bacterial identification and drug susceptibility were performed. The data were analyzed by Chi square test and t test. Results Eighty-six(38.0% ) patients were diagnosed with nosocomial SBP and 140 (62.0%)were diagnosed with community-acquired SBP. The proportion of Child-Pugh Class C cases in patients with nosocomial SBP was higher than patients with community acquired SBP (97.7% vs. 82.8%; x2= 11. 489, P=0.001). Mortality rate in patients with nosocomiat SBP was also higher than patients with community acquired SBP (50. 0% vs. 30. 0%; x2 =9. 081,P=0. 003). Total 28 species (232strains) of bacteria were isolated from these patients. 77.5 % (69/89) of the nosomial SBP cases and 76.9% (110/143) of community-acquired SBP cases were caused by Gram-negative bacteria (mainly were Escherichia coli and Klebsiella pneumoniae). 19.1% nosocomial SBP cases and 21. 8%community-acquired SBP cases were caused by Gram-positive bacteria. Fungus infections accounted for 3.4% and 1.4% of these two population, respectively(P＞0.05). In patients with nosocomial SBP,19 out of 32 Escherichia coli stains and 5 out of 14 Klebsiella pneunmoniae strains were extended spectrum β-lactamase (ESBL) positive, while among 60 Escherichia coli stains and 32 Klebsiella pneunmoniae strains, only 11 Escherichia coli stains were ESBL positive (P＜0.05). The resistance rates of Gram-negative strains to cephalosporin and quinolone in nosocomial SBP patients were both higher than those in community-acquired SBP patients(P＜0. 05), but all Gram-negative isolates were sensitive to imipenem (P＞ 0. 05). No Gram-positive isolates resistant to vancomycin were found.Conclusions The liver cirrhosis patients with Child-Pugh Class C are vulnerable to nosocomial SBP and the prognosis is poor. Although the pathogenic spectrum are similar in cirrhotic patients with nosocomial and community-acquired SBP, which mainly are Escherichia coli and Klebsiella pneumoniae, the percentage of ESBL producing strains is higher in nosocomial SBP patients compared to that in community-acquired SBP patients.

Objective To summarize the clinical experience of nasal endoscopic treatment for refractory epistaxis. Methods .A retrospective analysis was made on 258 patients with refractory epistaxis treated with intranasal endoscopic exploration and hemostatic treatment from January 1998 to December 2004 in this hospital. Results .Hemorrhage was found on the nasal septum and the lateral nasal wall in 118 and 47 patients, respectively. Nose bleeding resulted from intranasal tumor in 8 patients. The site of bleeding was not clearly identified in 85 patients. Of the 258 patients, 257 responded to the treatment (99.6%) and 1 died ( 0.4%).Recurrent bleeding occurred in 43 patients. The incidence of recurrent bleeding was significantly higher in patients with unknown bleeding sites than in clearly diagnosed patients (?2=17.689,P=0.000). Conclusions .The emphasis of nasal endoscopic treatment for refractory epistaxis should be laid on the initial management, during which the bleeding site should be carefully identified. For patients with repeated nose bleeding, a combination treatment, including endoscopic filling-up of the anterior nasal cavity, endoscopic embolization of anterior and posterior nose, or embolization of the internal maxillary artery, is necessary.

Objective RNA interference (RNAi) expression vector was constructed to inhibit the focal adhesion kinase(FAK)expression in colon carcinoma HT29 cells, and then the sensitivity of the cells to 5-fluorouracil (5-FU) was determined. Methods One specific pair of oligonucleotides with short hairpin and its negative control sequence were designed and synthesized based on FAK cDNA sequences, then they were inserted into pGenesil-l vector to generate the recombinant plasmids. After identification by the restriction endonuclease and DNA sequencing, the recombinant plasmids were transfected into HT29 cells by lipofectamine TM 2000. The stable transfected cells were selected in a medium containing geneticin G418. The change in FAK expression in HT29 cells before and after RNA interference was detected by reverse transcription and polymerase chain reaction (RT-PCR) analysis and SP immunocytochemistry technique. Sensitivity of HT29 cells to 5-FU was determined by MTT assay. Results The recombinant plasmids coincided completely with the designs in the restriction map and the sequence analysis. After FAK being targeted by RNA interference, immunocytochemistry showed the protein expression of FAK was reduced dramatically, and RT-PCR revealed FAK mRNA expression was down-regulated by 76.94% compared to that of untransfected cells. Objective To investigate the antitumor activity of Gefitinib, a selected epidermal growth factor receptor-tyrosine kinase inhibitor, on human colorectal cancer cell lines in vitro, and to explore the relationship between the inhibitory effect of Gefitinib on cancer cells and the expression of epidermal growth factor receptor (EGFR). Methods The growth inhibitory effects of Gefitinib, which expressed as the half growth inhibition dose IC50, on colorectal cancer cells were assessed by MTT assay. EGFR mRNA expression was detected by reverse transcriptional PCR (RT-PCR). Western blot was used to determine the expression of EGFR protein as well as its phosphorylated forms (p-EGFR). Results Gefitinib inhibited growth of all the six colorectal cancer cell lines in vitro with an IC50 range from 6.5 to 172.7?mol/L. Lovo cell line, with an IC50 value less than 10?mol/L, was the most sensitive one to Gefitinib, HT29 and SW480 were moderate sensitive to 10?mol/L
