1.Investigation of amino acid sequence of specific ouabain conjugated peptides

AIM: The purpose of the study was to investigate the gene and amino acid sequence of specific ouabain conjugated peptides (OCP) in order to get an experimental bases to block or antagonist the actions between endogenous ouabain(EO) and sodium pump in hypertension. METHODS: Screening the phage displayed 12-peptide library by biopaning for OCP. The sequence of each selected peptide was determined and the sequence was analyzed through internet. The bioactivity was determined by erythrocyte [ 86 Rb] uptaking. RESULTS: Three kinds of peptides were screened out. Peptide A (12 peptide) was occupied in 66 7%(8/12), peptide B (8 peptide) 16 7% (2/12) and peptide C (12 peptide) 8 3% (1/12). There was only one case without insertron. The analysis of protein showed that there were no homogenous between peptide A, B, C and sodium pump. The amino acid sequence of specific OCP was Leu-Leu-Ala-Asp-Thr-Thr-His-His-Arg -Pro-Trp-Thr. CONCLUSIONS: Determination of the sequence of OCP supplies an important experimental foundation for ouabain research. The results also show that phage display peptide library is an effective, simple and efficient method to select specific steroid receptors. [
2.Extraction and Determination of Polysaccharide from Discarded Fi brous Roots of Radix Panacis Quingueforlii

Objective:To establish a feasible method for extracti ng polysaccharide from the discarded fibrous roots of Radix Panacis Quingueforli i afterpanaquilon had been extracted, and determine the polysaccharide content. Methods:Enzymolysis technique and alcohol was applied for decolorization, and ph enol-sulfuric-acid method was used to determine the active polysaccharide conte nt.The content of trace element and heavy mental was measured by element-analyze r and atomic fluorescence photometer respectively. Results: The yield of polysac charide from the fibrous roots was close (about 11.7%) to the main root.And the content of heavy metal can match the national standard.Conclusion:I t is valuable to extract the polysaccharide from the discarded fibrous root of R adix Panacis Quingueforlii.

4.Preparation and preliminary application of monoclonal antibody against Staphylococcal enterotoxin C2

Objective:To obtain monoclonal antibodies(McAb) against Staphylococcal enterotoxin C2(SEC2) and establish method for detecting SEC2.Methods:The secreted SEC2 from staphylococcus aureus was used as antigen to immune BALB/c mice. Monoclonal antibodies against SEC2 were prepared by normal hybridoma technique. By identifying the characters of McAbs, the quantitative detection ELISA test method were established and were preliminarily applied.Results:Four hybridmas producing antibodies against SEC2 were obtained. IgG isotypes of four McAbs were IgG1. Their binding site was different except two McAbs that shared the same binding site.The McAbs were proved to be specific for SEC2.The sandwich ELISA method had good specificity, sensitivity and reproducibility, and it was founded to be able to detect SEC2 at concentration from 0.5 to 20 ng/ml. Its recovery ranged between 97.8% and 101%,and CV value ranged between 2% and 5%.Conclusion:The prepared McAb against SEC2 can be used for SEC2 immunoassay. This work provides a basis for controlling the quality of JINPUSU and researching staphylococcal enterotoxin.
5.The collateral ligaments: normal and pathologic appearances on MRI

Objective To evaluate the MR findings of normal and abnormal collateral ligament. Methods The MR findings of 56 normal and 36 surgically confirmed injured collateral ligaments were retrospectively reviewed and compared. Results The normal tibial and fibular collateral ligaments were hypointense string on both T 1 and T 2 coronal MR images, with an average length of 6.8 cm for tibial collateral ligament and 5.7 cm for fibular collateral ligament. 36 collateral ligaments were injured, including 18 tibial and 12 fibular collateral ligaments. Three cases (6 ligaments) had both tibial and fibularcollateral ligaments injuries. The injury of collateral ligament was classified as: Grade I (18 ligaments), Grade Ⅱ (11 ligaments), and Grade Ⅲ (7 ligaments). The complications of anterior cruciate ligament sprains (13 cases), posterior cruciate ligament sprains (9 cases), meniscal tears (12 cases), bony contusion and fracture (7 cases), and joint effusion (20 cases) were found in 25 patients(78%). Conclusion Coronal MR imaging can clearly demonstrate the structure of the knee collateral ligaments, accurately diagnose and classify the ligament injury, and correctly detect the complications. MRI should be used as a routine examining method for knee trauma.

Objective To observe the effects of neuromuscular activation (Neurac) training on balance and walking in children with hemiplegic cerebral palsy. Methods 30 children with hemiplegic cerebral palsy from March to October, 2015 were divided into observation group (n=15) and control group (n=15). Both groups received routine rehabilitation, while the observation group received 30-minute Neurac training in addition, 5 times a week, for 3 months. They were assessed with D and E domains of Gross Motor Function Measure 88 (GM-FM-88), balance of Fugl-Meyer Assessment and footprints analysis before and after treatment. Results The scores of D and E domains of GMFM-88 and balance of Fugl-Meyer Assessment, and the step length, step width and velocity improved in both groups after training (t>7.31, P<0.001), especially in the observation group (t>2.08, P<0.05). Conclusion Neuroac training can further promote the recovery of gross motor function, balance and walking in children with hemiplegic cerebral palsy.
8.Apoptosis in heart of multiple organ dysfunction syndrome rats at high altitude

AIM: To study apoptosis in heart of multiple organ dysfunction syndrome (MODS) rat at high altitude. METHODS: The two-hit model of MODS rat was used at two different altitude(1 510 m, 3 900 m). Hemorrhage was induced in Wistar rats by catheterizing the femoral artery until a mean arterial pressure was 35 mmHg and maintained for 1 hour. Rususcitation was performed with lactated Ringer′s solution at 24 h after hemorrhage, cecal ligation and puncture(CLP) was performed .Then rats were killed at 3 h,6 h,12 h and 24 h after CLP, and myocardium sample was excised and stored in liquid nitrogen. Apoptosis in heart was determined by DNA agarose gel electrophoresis, flow cytometry(FCM), transmission electron microscope(TEM) and terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling(TUNEL). AIM: The purpose of this study was to eva lu ate the effect of tetracycline combined with anti-inflammation drug on the serum antibody level against porphyromonas gingivalis (Pg) in patients with early-ons et periodontitis (EOP). METHODS: The study population consisted of 12 individuals with e arly-onset periodontitis, 16 patients with adult periodontitis (AP) and 12 perio dontally healthy subjects (HS). Clinical examination and recordings of probing p ocket depth (PPD), periodontal attachment level (PAL) and bleeding on probing (B OP) were performed prior to treatment and at three months following the periodontal treatment. The serum antibody titer against Pg in patients with EOP was measured at diagnosis and at the termination of treatmeat. RESULTS: A significant improvement was observed in probing pocke t depth, periodontal attachment level and gingival bleeding index post treatment for three months. The serum IgG antibody titer against porphyromonas ging ivalis was decreased significantly after the treatment. CONCLUSION: Our results demonstrate that mechanical periodontal treatment combined with tetracycline and anti-inflammation drug provides clinica lly favorable results in patients with early-onset periodontitis.
