1.Effects of Neuromuscular Activation Training on Balance and Walking in Children with Hemiplegic Cerebral Palsy

Objective To observe the effects of neuromuscular activation (Neurac) training on balance and walking in children with hemiplegic cerebral palsy. Methods 30 children with hemiplegic cerebral palsy from March to October, 2015 were divided into observation group (n=15) and control group (n=15). Both groups received routine rehabilitation, while the observation group received 30-minute Neurac training in addition, 5 times a week, for 3 months. They were assessed with D and E domains of Gross Motor Function Measure 88 (GM-FM-88), balance of Fugl-Meyer Assessment and footprints analysis before and after treatment. Results The scores of D and E domains of GMFM-88 and balance of Fugl-Meyer Assessment, and the step length, step width and velocity improved in both groups after training (t>7.31, P<0.001), especially in the observation group (t>2.08, P<0.05). Conclusion Neuroac training can further promote the recovery of gross motor function, balance and walking in children with hemiplegic cerebral palsy.
2.Clinical feature of pulmonary manifestation in mixed connective tissue disease:analysis of 112 patients

Objective To study the clinical manifestations,diagnosis,treatments and prognosis of the pulmonary change in mixed connective tissue disease(MCTD).Methods From January 1996 to June 2008,112 patients with definite MCTD in Second Affiliated Hospital of Harbin Medical University were retrospectively studied,and the clinical feature about pulmanary manifestations,echocardiography feature,radiology features and treatments were reviewed and analyzed.44 patients were followed up.Results 48(5 male)had pulmonary manifestation.The median age of onset of symptoms was 39 years and the median duration of disease was three years.The most common manifestation was pleural effusion and(or)pachynsis pleurae(21 cases,18.8%).16 patients(14.3%) had interstitial lung disease,and 8 cases(7.1%)had pulmonary hypertension.The cardinal symptom was short breath.Thirteen patients(11.6%)had dyspnea,4 cases(3.6%)had chest pain,and 4 cases(3.6%)had cough.Reductions of pulmonary function or abnormalities of ultrasonographic image were noted in 25(52.1%)of 48 patients with pulmonary involvement of MCTD who had not any complaint of respiratory symptoms.Six patients had reductions in diffusing capacity of the lung for carbon monoxide(DLCO),17 patients had pulmonary abnormality on chest X ray or CT,and two patients had pulmonary hypertension on doppler echocardiography.Conclusion Pulmonary involment of MCTD may contribute to prognosis seriously.DLCO was the most sensitive parameter.Pulmonary hypertension was the most common cause of death.
3.THE EFFECT OF PEROXIREDOXIN Ⅱ ON IN VITRO DEVELOPMENT OF MOUSE PREIMPLANTATION EMBRYOS AND ITS MECHANISM

Objective To study the effect of Peroxiredoxin Ⅱ on development of mouse preimplantation embryos in vitro and to investigate the possible action of Peroxiredoxin Ⅱ on preimplantation embryonic development. Methods One-cell embryos collected from the oviduct of the superovulated mice were cultured in microdrops of medium for 48?h.The effect of peroxiredoxin Ⅱ antibody on the development of embryo in vitro was observed,and the percentage of embryos developing to 2-and 4-cell stage of embryos was used as evaluation criteria.With redox-sensitive fluorescence probe 2′,7′-dichlorodihydroflurescin diacete(DCFH-DA),reactive oxygen species(ROS) induced by Peroxiredoxin Ⅱ antibody was monitored in mouse embryos after 24?h culture by laser confocal scanning microscopy. Results The embryonic development rate from 2-cell stage to 4-cell stage was decreased significantly by Peroxiredoxin Ⅱ antibody in the dilution of 1∶100 and 1∶200,but Peroxiredoxin Ⅱ antibody had no inhibitive effect on development from 1-cell to 2-cell stage.Moreover,Peroxiredoxin Ⅱ antibody in the dilution of 1∶200 induced increase in the production of reactive oxygen species in mouse embryos from 24?h culture of one-cell embryo.Conclusion Peroxiredoxin Ⅱ antibody induced the generation of “2-cell block” by increasing the production of reactive oxygen species(ROS) in mouse embryos.These results also indicated that Peroxiredoxin Ⅱ may reduce or eliminate ROS in preimplantation embryos and promote the development of preimplantation embryos.;
4.STUDY ON THE LOCALIZATION OF PEROXIREDOXIN II mRAN IN FOLLICLES AT DIFFERENT STAGES IN MOUSE OVARIES BY IN SITU HYBRIDIZATION

Objective In this study, we investigated the distribution of Peroxiredoxin II mRNA in follicles at all stages in mouse ovaries and mouse secondary oocytes(MII eggs) basing on our previous researches, to provide the morphological basis for exploring the effect of Peroxiredoxin II on oogenesis and oocyte maturation in the mouse. Methods To observe the distribution of Peroxiredoxin II mRNA in follicles at all stages in ovaries and the localization of Peroxiredoxin II mRNA in Germinal-Vesicle intact oocytes(GV oocytes) and MII eggs by in situ Hybridization. Results In situ hybridization of ovary section revealed that the signals for Peroxiredoxin II mRNA were undetectable in oocytes of primordial follicles, and moderate signals for Peroxiredoxin II mRNA were observed in oocytes of primary follicles. Moreover, strong signals for Peroxiredoxin II mRNA were evident in antral follicles. The signals for Peroxiredoxin II mRNA also existed in GV oocytes and MII eggs in vitro. The hybrid signals were stronger in GV oocytes than in MII eggs. In addition, the weak but consistent signals for Peroxiredoxin II mRNA were detected in follicular cells from primordial follicles to large antral follicles. Peroxiredoxin II mRNA was located in cytoplasm of oocytes and follicular cells, but not in nuclei.Conclusion These results suggested that Peroxiredoxin II might be involved in the regulation of oogenesis and oocyte matruation in the mouse.;
5.Effects of promoter region 5′CpG island demethylation on biological phenotype in human colorectal cancer cells

AIM: To explore the relationship between methylation status of promoter region 5′CpG island and the biological phenotype in human colorectal cancer RKO cell lines. METHODS: RKO cells were treated with selective DNA methyltransferase (DNMTs) inhibitor, 5-Aza-2′-deoxycytidine (5-Aza-CdR), for 72 h. Methylation-specific PCR (MSP), T-A clone and DNA sequence analysis were used to detect 5′CpG island methylation status of p16/CDKN2 tumor suppresor gene. Cell growth, cell cycle arrest and apoptosis were analyzed by MTT, flow cytometry (FCM), fluorescent dye staining and transmission electron microscope. 7.Antimicrobial Effects of Nano-Silver Gauze against Common Bacterial Isolates.

BACKGROUND: Silver has been used for disinfection and sterilization. We aimed to confirm the in-vitro antibacterial effects of nanocrystalline silver-coated gauze. METHODS: Fourteen clinical isolates each of Escherichia coli and Acinetobacter baumannii were used. Bacterial suspensions made in tryptic soy broth were exposed to Ordinary and silver-coated gauze. Bacteria were then harvested from the gauze immediately and after 24 h incubation, cultured on blood agar plates and eunmerated for viable counts. The number of colonies was converted into common logarithms for comparison. RESULTS: The number of colonies recovered from silver-coated gauze was significantly lower than those recovered from ordinary gauze when harvested immediately after exposure (E. coli, 3.06 vs 1.73; A. baumannii, 3.13 vs 1.98; P<0.001). After 24 h incubation of exposed gauze, silver-coated gauze produced less than 1 CFU/mL, whereas ordinary gauze produced a number of colonies significantly higher than it did immediately after exposure (E. coli, 4.13; A. baumannii, 4.46; P<0.001). Conclusion: Compared with ordinary gauze, silver-coated gauze was shown to have 99.99% antibacterial effect.

8.Responses of Patients and Guardians to Phlebotomists' Wearing Gloves and Disinfecting Their Hands.

BACKGROUND: A hand hygiene policy has been introduced by the Joint Commission International Accreditation Standards for Hospitals (JCI); the purpose of this policy is to ensure that systematic standards are continuously practiced at hospitals in order to improve the quality of medical care and the safety of patients and health care workers. This policy requires that phlebotomists wear new gloves and disinfect their hands before attending a patient to prevent nosocomial infections and to protect the phlebotomists. This study aimed to assess the responses of patients and guardians regarding the hand hygiene procedures. METHODS: Between January and February 2010, a questionnaire survey was performed at our hospital for 310 outpatients, 93 inpatients, and 189 guardians. RESULTS: In all, 70.8% of respondents answered that phlebotomists did not require considerable time to remove and wear gloves, and 69.9% responded that phlebotomists could readily detect veins even with their gloves on. Besides, 81.9% respondents thought that it was sanitarily important for phlebotomists to remove their gloves and disinfect their hands after a venipuncture, whereas only 2.8% thought that this practice caused discomfort. CONCLUSION: The patients and guardians recognized that the hand hygiene procedures were important for their own safety and encouraged their application rather than considering them uncomfortable or inappropriate. Introduction and maintenance of the hand hygiene policy of JCI would not be difficult because patients or guardians recognized its importance.

9.Preparation of a kind of G418 resistance fibroblast feeder layers

Objective:To prepare a kind of G418 resistance fibroblast feeder layers.Methods: In order to prepare a stock primary embryonic fibroblast (EMFI) cells, the pregnant Smad3ex8+/- mice containing heterozygous neo gene were killed 14 days after coitus to get the embryo. EMFI cells were tested by PCR to identify their gene types. Only one EMFI cell which contained heterozygous neo gene was chosen and treated with mitomycin C.The mitomycin C-treated EMFI feeders were stored at -70℃. Results: The EMFI feeder layers could support the growth of embryonic stem cell line TC-1 efficiently and could live in G418 resistance medium for 1-2 weeks.
10.Clinical effect of two targeted drugs in treatment of advanced hepatocellular carcinoma: A comparative analysis

ObjectiveTo investigate the clinical effect of the targeted drug sunitinib in the treatment of advanced hepatocellular carcinoma (HCC). MethodsA total of 48 patients with advanced HCC who were admitted to Chang′an Hospital form September 2015 to September 2016 were enrolled and divided into control group and observation group, with 24 patients in each group. The patients in the control group were given oral sorafenib, and those in the observation group were given oral sunitinib. The patients were followed up for 1 year, and the clinical outcome, improvement in clinical symptoms, and adverse events were recorded. The t-test was used for comparison of continuous data between groups, and the Mann-Whitney U test was used for comparison of categorical data between groups. ResultsThe disease control rate (DCR) in the observation group was 68.42% (13/19), and among these 13 patients, 4 achieved effective remission and 9 had stable disease; the DCR in the control group was 76.19%, and there was no significant difference in DCR between the two groups (P>0.05). There was a significant difference in the incidence rate of hand-foot skin reaction between the observation group and the control group (0% vs 12.5%, U=31.07, P<0.05). The observation group had a significantly higher incidence rate of neutropenia than the control group (25% vs 16.67%, U=29.87, P<0.05). Both groups experienced digestive tract reactions (nausea and vomiting) and abnormal liver function; since the digestive tract reactions were tolerable, no special treatment was given, and the patients with abnormal liver function were given liver-protecting drugs, so all adverse events were effectively alleviated. ConclusionSunitinib has a good clinical effect in the treatment of advanced HCC, and further studies with a larger sample size are needed to investigate the long-term clinical effect of sunitinib and combined treatment.
