1.AN COMPUTERIZED 3D RECONSTRUCTION METHOD BASED ON RAT'S CERVICAL CORD SERIAL SECTIONS

Objective With personal computer(PC) as the platform,to explore an computerized 3D reconstruction method based on rat's cervical cord serial sections. Methods Normal cervical cord segment of SD rat was embeded in paraffin block,four locating marks were made at four corners around the specimen,the specimen was sectioned serially with Leica paraffin microtome,after each three sections,the images of specimen and outer locating marks were taken with CANON digital camera and the serial sections were obtained.The obtained images were ordered,cut,turned into gray scale images and background uneven corrected.The edges of white matter,grey matter and locating marks in each image were extracted,the locating marks were used for automated registration of serial sections with 3D-DOCTOR software,the 3D surface reconstruction of cervical cord segment was observed,cut and measured freely in PC. Results The grey matter,white matter of rat cervical cord segment were reconstructed from serial transverse sections,the reconstructed segment was measured to obtain the surface area and volume data.Conclusion The paraffin embedding and "hard" locating marks techniques can be used to obtain the images of serial transverse sections of rat's cerival cord segment,to make 3D reconstruction which can be observed and measured freely.
2.Blocking effects of anti-sense peptide of C5a on the adhesion between pulmonary vascular endothelial cell and neutrophil

Objective:To study the blocking effects of anti-sense peptide of C5a on the adhesion between pulmonary vascular endothelial cell(PVEC) and neutrophil resulting from C5a anaphylatoxin.Methods:It was determined by Flowcytometry that the change of adhesion molecule expression on PVEC and the activity of MPO in PMN was determined after adhesion.Results:In response to C5a after interactions with several concentrations of anti-sense peptide R4, the expression of P-Selectin on PVEC decreased significantly and reached the minimum at the concentration of 5 000 ng/ml, and the activity of MPO in PMN reduced by 40% at the concentration of 5 000 ng/ml of anti-sense peptide R4.Conclusion:The results suggested that anti-sense peptide of C5a has significant blocking effects on C5a anaphylatoxin.
3.Acupuncture Treatment for Labyrinthine Tinnitus Based on Intergrated Traditional Chinese Medicine & Western Medicine

Based on the differentiation of deficiency (hyporeactivity) and excess (hyperreactivity) syndromes according to the Traditional Chinese Medicine (TCM) logic, 65 labyrinthine tinnitus cases of the example was classified into three groups, the excess constitutional type (18 cases), the deficiency constitutional type (37 cases) and the intermediate type (10 cases). Acupuncture treatment was carried out under the guidance of the basic principles of "seeking the fundamental of a disease in treatment"based on TCM.
The effect was evaluated using two parameters of subjective tinnitus assessment score in a special questionnaire (STS) and objective tinnitus loudness level in dB SL (OTL). Based on the changes in STS and OTL before and after the end of all therapeutic courses, the gross effective rate (GER) of 65 cases containing effective STS or OTL was 72.3%, while the strict effective rate where both STS and OTL showed effective (BER) was 47.7%. Among the three TCM syndromes differentiation groups, the excess constitutional type had the best therapeutic effect (66.7%of BER), fellows the deficiency constitutional type (48.7%of BER), and the intermediate type was the worst (only 10%of BER). Statistically, there was a significant difference among each syndrome differentiation group (p<0.05).
Meanwhile, the entire GER and BER went down to 55.4%and 38.5%respectively 2 months after the end of treatment to show a decreasing tendency of therapeutic effectiveness compared with those treatment rates immediately after the end of treatment. Especially, GER of the deficiency constitutional type showed a significant decrease (P<0.05).
We concluded that the TCM differentiation of deficiency and excess syndromes is a prerequisite for the determination of a correct acupuncture treatment for labyrinthine tinnitus. Through a thorough analysis and treatment on 65 clinical cases, the tinnitus of the excess constitutional type showed a higher reactivity for the treatment comparing to the other two types, the deficiency and the intermediate.
4.Ventilator-associated Pneumonia Following Severe Craniocerebral Trauma and Its Clinical Characteristics

OBJECTIVE To explore the characteristics of the pathogens in ventilator-associated pneumonia (VAP) following severe craniocerebral trauma. METHODS The 220 cases with severe craniocerebral trauma were analyzed retrospectively. RESULTS The incident rate of VAP was 24% in these patients. A total of 169 pathogens were isolated,including 146 ( 86%) strains of bacteria and 23 (14%) fungal isolates. The main Gram-negative bacteria (114 strains) were Pseudomonas aeruginosa,Escherichia coli,Klebsiella pneumoniae,etc. The Gram-positives (32 strains) were Staphylococcus aureus and twenty-four (75%) of them were meticillin-resistant. CONCLUSIONS The incident rate of VAP is high following severe craniocerebral trauma. Most of the isolated pathogens are drug resistant. These cases have mixed infection occurred commonly. To prevent and treat VAP,we should pay attention to disinfection and aseptic manipulation,and make the rational use of antimicrobial agents.
5.Revision for infected total hip arthroplasty

Objective To discuss operative treatment of the postoperative infection after total hip arthroplasty. Methods 11 cases of infected hip arthroplasty, 6 males and 5 females, with a mean age of 63 years old (from 54 to 71 years), were treated operatively in our department. Their preoperative mean Harris score was 33 (from 25 to 40), and their mean erythrocyte sedimentation rate (ESR) was 61 mm/h (from 34 to 80 mm/h). Bacterial cultures and drug susceptibility tests were carried out before and during operation. One- stage revision was performed for 6 cases, and two- stage revision was performed for 5 cases. Of the 11 cases, 2 were classified as early postoperative infection and 9 as late chronic infection. Preoperative microorganism cultures of joint aspiration or pus in sinus were positive in 8 cases but negative in 3 cases. 5 had staphylococcus epidermidis infection, 2 had escherichia coli infection and 1 had staphylococcus aureus infection. Results No recurrence of infection was found after a mean follow- up period of 17 months (from 12 to 31 months). ESR and blood tests after operation revealed nothing abnormal. 6.The collateral ligaments: normal and pathologic appearances on MRI

Objective To evaluate the MR findings of normal and abnormal collateral ligament. Methods The MR findings of 56 normal and 36 surgically confirmed injured collateral ligaments were retrospectively reviewed and compared. Results The normal tibial and fibular collateral ligaments were hypointense string on both T 1 and T 2 coronal MR images, with an average length of 6.8 cm for tibial collateral ligament and 5.7 cm for fibular collateral ligament. 36 collateral ligaments were injured, including 18 tibial and 12 fibular collateral ligaments. Three cases (6 ligaments) had both tibial and fibularcollateral ligaments injuries. The injury of collateral ligament was classified as: Grade I (18 ligaments), Grade Ⅱ (11 ligaments), and Grade Ⅲ (7 ligaments). The complications of anterior cruciate ligament sprains (13 cases), posterior cruciate ligament sprains (9 cases), meniscal tears (12 cases), bony contusion and fracture (7 cases), and joint effusion (20 cases) were found in 25 patients(78%). Conclusion Coronal MR imaging can clearly demonstrate the structure of the knee collateral ligaments, accurately diagnose and classify the ligament injury, and correctly detect the complications. MRI should be used as a routine examining method for knee trauma.
7.Analysis of Hashimoto disease and its concurrent thyropathy

Objective To sum up the experience on diagnosis and treatment of Hashimoto disease and its concurrent thyropathy. Methods Clinical data were analyzed on 52 cases of Hashimoto disease(HD) surgically found to have concurrent thyropathy from Jan 1984 to Jun 2001 out of 138 HD cases undergoing surgery. Results Thyroid nodules were found in all 52 HD cases, among which 17 were with thyroid cancer, 2 with malignant lymphoma, 5 with thyroid adenoma, 2 with hyperthyroidism, and 26 with nodular goiter. Thyroidectomy of different extent was performed. Postoperatively all cases were given thyroxine. On follow-up a patient with malignant lymphoma died, and one patient suffered hypothyroidism. Conclusion HD is a ubiquitous disease. The clinical diagnosis of is still difficult. HD cases complicated with thyroid nodules should be treated surgically to exclude malignant tumors. Intraoperative frozen section is a reliable diagnostic method and may guide the surgical modality.
8.Fibronectin supports endotoxemia mice survival

AIM: To study the preventive effect of fibronectin on hepatic failure induced by endotoxin in mice.METHODS: The survival rate was observed in endotoxemia mice injected with fibronectin from human plasma. The tissue damage and expression of TNF?, IL-1?, IL-6 mRNA in hepatocyte were detected by the methods of histology, ultrastructure, DNA fragementation and RT-PCR. RESULTS: ①Fibronectin obviously reduced the mortality of endotoxemia mice sensitized by D-galactosamine(GalN). ②Histopathology showed that less necrosis occurred on the hepatocyte of endotoxemia mice injected with fibronectin, compared with saline control. ③Ultrastructure and DNA fragmentation showed fibronectin suppressed hepatocyte apoptosis induced by LPS. ④Fibronectin down-regulated the overexpression of TNF?, IL-1?, IL-6 mRNA on hepatocyte induced by LPS. CONCLUSION: Fibronectin supports endotoxemia mice surrival by down-regulating the expression of TNF?, IL-?, IL-6, it may be a potent therapy for endotoxemia.
9.Clinical Study of the Effects of Traditional Chinese Acupuncture Treatment on Tinnitus with Age-related Hearing Loss

Traditional Chinese acupuncture was used in four cases of intractable tinnitus associated with presbyacusis, showing greater losses at higher frequencies while lower frequencies were only slightly below the normal threshold on a pure tone audiogram. Further audiologic evaluation by means of a speech discrimination test and short-increment sensitivity index (SISI) showed that cases 1 and 2 were the labyrinthine impairment type, case 3 was the mixed type, and case 4 was the retrolabyrinthine type. However, the etiology and pathogenesis of all patients were diagnosed as constitutional deficiency of kidney essence failing to nourish both the ear and brain, the so-called “sea of marrow” based on TCM differentiation. Following the principle of invigorating kidney essence and nourishing “Kidney Qi, ” acupuncture treatment was given around the ear as well as at distal points on the extremities once a week, with 10 sessions constituting one therapeutic course.
In cases 1, 2 and 3, a temporary minor reduction effect of tinnitus lasting half a day to a few days could usually be achieved by each acupuncture session, and long-term effects, with significant improvement of subjective intensity and loudness of tinnitus (which always dropped to 1/2-1/3 of the pretreatment levels) was possibly obtained after 3-4 treatment sessions. Acupuncture was less effective in case 4, in which some fluctuation of tinnitus loudness values was reported within the period of treatment, but there was no remarkable stable improvement, either in terms of loudness or disturbance of the tinnitus after 10 treatment sessions.
The results indicate that traditional Chinese acupuncture is a recommendable therapy in treating age-related tinnitus, especially the labyrinthine type or mixed type rather than the retrolabyrinthine type.
10.Analysis of gene expression profile of multidrug resistant MCF/DOX cell line after benflumetol derivative LY980503 treatment

AIM: To investigate the effect of LY980503(a benflumetol derivative)on multidrug resistance of tumor cell line using DNA microarray. METHODS: Total RNA was extracted from multidrug resistant MCF/DOX cell line. cDNA microarray containing 320 cDNAs was used to detect the gene expression profile. RESULTS: 9 down-regulated genes and 1 up-regulated gene were identified after multidrug resistant MCF/DOX cells were treated with LY980503. CONCLUSION: LY980503 can effectively reverse the resistance of MCF/DOX to DOX in vitro by adjusting the expression of multi-genes.
