Objective:To study the blocking effects of anti-sense peptide of C5a on the adhesion between pulmonary vascular endothelial cell(PVEC) and neutrophil resulting from C5a anaphylatoxin.Methods:It was determined by Flowcytometry that the change of adhesion molecule expression on PVEC and the activity of MPO in PMN was determined after adhesion.Results:In response to C5a after interactions with several concentrations of anti-sense peptide R4, the expression of P-Selectin on PVEC decreased significantly and reached the minimum at the concentration of 5 000 ng/ml, and the activity of MPO in PMN reduced by 40% at the concentration of 5 000 ng/ml of anti-sense peptide R4.Conclusion:The results suggested that anti-sense peptide of C5a has significant blocking effects on C5a anaphylatoxin.

AIM: To study the immunity modulation mechanism of achyranthes bidentata polysaccharides (ABPS). METHODS: (1) PBMC from patient with asthma or lung cancer were cultured with 800 mg/L ABPS for 18 h. AIM: To investigate the relationship between morphologic changes in neuron or neuroglial cells and expression of tumor necrosis factor ? (TNF-?) and c-Myc in cortex after focal cerebral ischemia/reperfusion in MCAO rats. METHODS: The focal cerebral ischemia/reperfusion model was established by intraluminal thread occlusion of the middle cerebral artery (MCAO). The middle cerebral arteries of rats were occluded for 2 hours and reperfused for 1, 3 and 7 days. Using the techniques of immunohistochemical staining and optical microscopy, the morphologic changes in neuron or neuroglial cells were observed in the cortex of frontal or parietal lobe; the cell types which dynamicaly expressed TNF-?, c-Myc in the different period were also observed. RESULTS: The degeneration or necrosis of neuron or neuroglial cells were observed at the center of infarction, it was very serious at 3 d after reperfussion. Astrocyte and microglial cell proliferation were observed at the broder of infarction. TNF-? and c-Myc positive cells, most of which were astrocytes and microglial cells, increased significantly at 3 d after reperfusion. CONCLUSION: TNF-? and c-Myc may play an important role in the regulation of neuron or neuroglial cells after focal cerebral ischemia with reperfusion. [

AIM: To observe the effect of antisense bcl-2 oligodeoxynucleotides(AS-PS-ODN) on bcl-2 mRNA and protein expression, cell proliferation,viability and apoptosis in a small-cell lung cancer cell line NCI-H446. METHODS: Semi-quantitative RT-PCR was performed to detect the bcl-2 mRNA expression, the Bcl-2 protein was determined by immunocytochemistry and flow cytometry analysis, and the effect of bcl-2 AS-PS-ODN on cell proliferation, viability and apoptosis were investigated by colony assay , cell count, DNA content analysis and TUNEL. RESULTS: ① 1 ?mol/L bcl-2 AS-PS-ODN significantly down-regulated the expression of bcl-2 mRNA and protein. The inhibition rate of mRNA and protein were 69.5% and 62.7%, respectively. ② bcl-2 AS-PS-ODN decreased cell proliferation and viability , induced cell apoptosis.The apoptosis rate was 22.3%-32.7% in cells treated with 1?mol/L bcl-2 AS-PS-ODN. CONCLUSION: bcl-2 AS-PS-ODN down-regulated expression of bcl-2 mRNA and protein, inhibited cell proliferation and induced apoptosis in a small cell lung cancer cell line, NCI-H446.

OBJECTIVE To analyze the homology of carbapenem-resistant Klebsiella pneumoniae(KPN),offering help for clinical therapy and nosocomial infection control.METHODS The antimicrobial-resistant phenotype of forty carbapenem-resistant K.pneumoniae strains was analyzed by the WHONET 5.4 soft and the resistant genotypes were determined by plasmid profile analysis and pulse-field gel electrophoresis(PFGE).RESULTS Analyzing antimicrobial-resistant phenotype to usual eighteen clinical drugs,the main drug resistant profiles were pan-resistant and only sensitive to tobramycin among the eight antimicrobial-resistant profiles(72.5%).Additionally,the main strains were type Ⅰ and type Ⅱ among the five strains analyzed by plasmid profile(82.5%).When analyzed by PFGE,five types were identified and among these strains type Ⅰ was predominant in 34 strains(85.0%).CONCLUSIONS The strains used in this study exhibit higher homology.Therefore,clinical departments and nosocomial infection departments should pay more attention to these strains to avoid outbreak.

OBJECTIVE To understand the drug resistance genes in Klebsiella pneumoniae (KPN) isolate resistance to 18 kinds of antibacterials which included imipenem and meropenem. METHODS We detected 36 kinds of drug resistance genes for the strain of KPN by PCR method,included the beta-lactamases genes,blaTEM,blaSHV,blaLEN,blaOKP,(blaCTX-M-1,2 and 9) groups,(blaOXA-1,2 and 10) groups,CARB,PER,VEB and GES; the genes of metallo-beta-lactamases genes,IMP,VIM and KPC; the AmpC genes,DHA,ACT,MOX and LAT; the aminoglycosides resistant genes,aac(3)-Ⅰ,aac(3)-Ⅱ,aac(6')-Ⅰb,aac(6')-Ⅱ,ant(2″)-Ⅰ and ant(3″)-Ⅰ; the quinolones resistant gene qnr; the TMP resistant genes,dfrA1 and dfrA17; the disinfectant-sulfanilamide resistantgene,qacE△1-sul1;the integron genes,intⅠ-1,intⅠ-2 and intⅠ-3; and the transposon genes,tnpA and merA.RESULTS We found 9 kinds of drug resistance genes in this KPN isolate. They were the beta-lactamases genes,blaTEM and blaSHV; the metallo-beta-lactamasesgene blaKPC-2; the aminoglycosides resistant genes,aac(3)-Ⅱ and ant(3″)-Ⅰ; the quinolones resistant gene qnr; the TMP resistant gene dfrA17; the disinfectant-sulfanilamide resistant gene qacE△1-sul1 and the integron genes intⅠ-1. CONCLUSIONS We discovere multiple drug resistant genes (some in the chromosome,some are plasmid-mediated) in this isolate. We also find the infrequent plasmid-mediated drug resistant gene blaKPC-2. We think it's concerned with the pan-resistant and the multi-drug resistant genes in this KPN,and we must pay highly attention to this isolate in clinic.

OBJECTIVEWe aimed to construct a recombinant adenovirus Ad-APN-EGFP and to evaluate the transfection efficiency and effect of the adenovirus promoting osteogenesis around implants.

METHODSRecombinant adenovirus Ad-APN-EGFP containing adiponectin (APN) was constructed by DNA sequencing and restriction enzyme digestion. Rat bone marrow stem cells (BMSCs) were transfected in vitro. Transfection efficiency and APN mRNA expression were tested. Animal models of rat femoral epiphysis and hydroxyapatite (HA)-coated implants were established. The Ad-APN-EGFP adenovirus at 10 μL was injected into the defect around HA-coated implants in the experimental group, whereas the same volume of phosphate-buffered saline was injected into the defect in the control group. Osteogenesis around implants was observed by hematoxylin-eosin staining four weeks after implantation.

RESULTSAd-APN-EGFP was successfully constructed by DNA sequencing and restriction enzyme digestion. The transfection efficiency of Ad-APN-EGFP to BMSCs was up to ≥ 90%. APN mRNA expression of BMSCs transfected with Ad-APN-EGFP was higher than that of the control group. Osteogenesis in the experimental group was more evident than that in the control group in vivo.

CONCLUSIONAd-APN-EGFP could be transfected into BMSCs and express APN mRNA at a high level. Ad-APN-EGFP could improve osteogenesis around implants in vivo.

Objective To explore the phonological characteristics of lateral misarticulation in children with functional articulation disor-ders and the rehabilitation approaches for them. Methods From June, 2013 to December, 2014, 44 children with lateral misarticulation were assessed and the phonological characteristics of lateral misarticulation were analyzed. They accepted targeted rehabilitation training. The cor-relation was analyzed between lateral misarticulation and age as well as severity of disorder. Results The frequency of lateral misarticulation was the most in Blade-palatals (75.00%), and then the dentals (65.91%), lingua-palatals (56.82%) and blade-alveolar consonant (20.45%). Thirty-eight cases were cured and 4 cases improved after 1 to 5 courses of rehabilitation. The overall cure rate was 86.36%(38/44), and it was 100%(11/11), 86.67%(13/15), 77.78%(14/18) in those with mild, moderate and severe disorder, respectively. There was no significant correlation between lateral misarticulation and age (r=0.100, P=0.752) as well as severity (r=0.257, P=0.092). Conclusion Lateral misarticu-lation in children with functional articulation disorders mainly occurs on blade-palatals, dentals and lingua-palatals. The targeted speech re-habilitation training is beneficial.
