1.Preparation, characterization and cytology study of Pluronic-PEI micelles.

To prepare and characterize Pluronic-PEI micelles as a drug/gene delivery system.We used the low-molecular-weight PEI as a cross-linking agent to prepare the Pluronic-PEI micelles. The particle size, zeta potential and critical micelle concentration (CMC) were measured by dynamic light scattering (DLS) and pyrene fluorescence probe. The cytotoxicity, transfection efficiency and the impact on the intracellular ATP and P-gp levels of Pluronic-PEI micelles were investigated at the cellular level.Pluronic-PEI micelles were successfully prepared with a suitable particle size (120-180 nm), zeta potential (+6-+9 mv), and a good ability to carry the drug/gene. Anstudy showed that Pluronic-PEI had low cytotoxicity, and the P123-PEI600 possessed high gene transfection efficiency and could downregulate the intracellular ATP and P-gp levels.Pluronic-PEI is a good drug/gene delivery system, and P123-PEI600 is an ideal vector, which may be used in the combination therapy for reversing multidrug resistance.
2.Expression and clinical significance of DNAJB11 in epithelial ovarian cancer.

To study the expression of DNAJB11 protein in epithelial ovarian cancer tissues and its clinical significance.Immunohistochemistry was used to examine DNAJB11 expressions in 105 tissue specimens of ovarian epithelial carcinoma, 23 normal ovarian tissues, 17 tissues of benign tumor, and 13 tissues of of borderline tumor. The correlations between protein expression and clinicopathological factors were analyzed by Chi square test.The correlations between protein expression and survival were analyzed by Kaplan-Meier and Cox regression.Positive expression of DNAJB11 protein was observed in 0.0% in normal ovary and benign tumor, 7.69% in borderline tumor, and 78.10% in epithelial ovarian cancer, respectively. Positive expression of DNAJB11 protein was significantly higher than the rest of the ovarian tissues and normal ovarian tissues (<0.001).Higher expression of DNAJB11 was more prevalent in tissues from patients with advanced FIGO stages, high serum CA125, poor histological differentiation, and serous cancer. Kaplan-Meier curves revealed that higher expression of DNAJB11 was significantly associated with poor disease-free survival and overall survival.Multivariate survival analysis revealed that strong positive expression of DNAJB11 was an independent prognostic factor for disease-free survival and overall survival.DNAJB11 may play a role in tumorigenesis and progression of epithelial ovarian cancer.Strong positive expression of DNAJB11 was an independent prognostic factor in epithelial ovarian cancer.
3.Synthesis of BODIPY photosensitizers and their photodynamic effect on cancer cells.

To design and synthesize photosensitizers with different substituents and to identify its physicochemical characteritics and photodynamic effect on cancer cells.Two kinds of BODIPY photosensitizers BPOI and BPCI were synthesized through condensation reaction between aldehyde and reactive hydrogen of pyrrole, followed with electrophilic substitution reaction. Physicochemical properties were characterized byH NMR, FT-IR and UV-visible absorption spectra and fluorescence emission spectra. The ability to produce reactive oxygen species was detected by BPDF and DCFH-DA. Photodynamic therapy effect on rat glioma C6 cellswas determined by MTT method.Two kinds of BODIPY photosensitizers BPOI and BPCI were successfully synthesized with different substituents, which were confirmed byH NMR, FT-IR. Both materials had low toxicity and could be readily taken up by tumor cells. The ability of synthesized photosensitizers to produce reactive oxygen species was strongly influenced by solvent polarity when the substituent was electron-donating group, while no effect was found when the substituent was electron-withdrawing group.Photosensitizer BPOI with electron-donating substituent produces reactive oxygen species with a slow rate in a highly polar environment, while greatly enhanced this effect in a low polarity environment, which is expected to be used for environmental-selective photodynamic therapy in tumor cells.
4.Application of traditional Chinese medicine(TCM) traceability system based on TCM quality characteristics and HACCP system.

It is crucial to establish a complete set of traditional Chinese medicine(TCM) quality traceability management process system, in order to stabilize the pricing order of TCM market and reconstruct the transmission path of TCM quality signals. In this study, we reviewed the mature experience of food and drug supervision at home and abroad, analyzed the quality characteristics of TCM, and put forward that the quality control of TCM products can learn from the hazard analysis and critical control point(HACCP) system in food safety quality control. This study points out that the HACCP system provides not only technical guidance for the traceability management of TCM, but also ideas for improving the quality of TCM products and the safety risk control of TCM. The application of the HACCP system in TCM quality control can help establish an international dialogue platform for TCM and help realize the modernization and internationalization of TCM industry.

5.Situation analysis and standard formulation of pesticide residues in traditional Chinese medicines.

Chinese Pharmacopoeia provides nine pesticide Maximum Residual Limits(MRLs) of traditional Chinese medicines(TCMs), The number of pesticides used in production are far more than those listed in pharmacopoeia. The lack of the standards make it's hard to reflect the real situation of pesticide residues in TCMs correctly. The paper is aimed to analyze the data of pesticide residues in TCMs from 7 089 items in 140 reports, and judging the exceedance rate of pesticides in TCMs using the MRLs of European pharmacopoeia,which is widely accepted in many countries. The results show that:①Pesticide residues in 18 kinds of TCMs are higher than MRLs,while in 137 kinds are below MRLs, such as Atractylodis Macrocephalae Rhizoma, Menthae Haplocalycis Herba and Fritillariae Thunbergii Bulbus. The average exceedance rate of all TCMs is 1.72%. The average exceedance rates of organochlorine, organophosphorus and pyrethroid are 2.26%, 1.51%, 0.37%,respectively. ②The average exceedance rate of pesticides is 2.00%, and the exceedance rate is more than 5%, accounting for 8.33%, the exceedance rate is between 1%-5%, accounting for 18.75%. the exceedance rate is between 0%-1%, accounting for 18.75%. The remaining 29 kinds of pesticides were not exceeded, accounting for 60.42%.Some reports like Greenpeace's organization exaggerated the pesticide residues in TCMs.But the pesticide residue question is still worthy of attention, so we proposed to amend the Chinese Pharmacopoeia pesticide residues standards, to increase the pesticide species of traditional Chinese medicine in production on the basis of retaining the existing types of pesticide residues, to strengthen the system research of pesticide residues in TCMs, providing a basis for making standard and promoting import and export trade in TCMs.
