1.Assessment of effectiveness of individual rehabilitation therapy in nonspecific low back pain

Objective To explore the effectiveness of individual rehabilitation therapy in patients with nonspecific low back pain.Methods A total of 50 patients with nonspecific low back pain ( NLBP) between January 2013 and January 2015 were enrolled in this study.Individual rehabilitation therapy was performed based on their different pathogenesis and risk factor of low back pain.Visual analogue score (VAS),Oswestry disability index ( ODI) and Japanese orthopaedic association ( JOA) were used for the assessment of effectiveness of individual rehabilitation therapy.Results Compared with pre-rehabilitation, VAS and ODI scores were significantly decreased at two-week after rehabilitation(P<0.05),and were further decreased at six-week after rehabilitation(P<0.01).The mean score of JOA of 50 patients before rehabilitation was (10.83 ±2.94) score,which was decreased to (24.37±7.55 )score at six-week after rehabilitation(P<0.05),with 12 patients were cured,32 were improved obviously,6 were improved moderately.The rate of efficacy was 100%.Conclusion Individual rehabilitation therapy was an effective option for NLBP,with significant decrease of pain.
2.Prostate health index in predicting the results of prostate biopsy for prostate cancer: a meta-analysis.

OBJECTIVETo systematically evaluate prostate health index (PHI) in predicting the results of prostate biopsy in the diagnosis of prostate cancer.
METHODSWe retrieved the literature relevant to the value of PHI in predicting prostate cancer at prostate biopsy published from the inception to February 2014 in the databanks of PubMed (1966 - 2014), CNKI (1982 - 2014), VIP (1989 - 2014), and Cochrane Library (1999 - 2014). According to the inclusion and exclusion criteria and retrieval strategies, we extracted the data, evaluated the quality of the included literature, and performed meta-analysis using the Meta-Disc 1.4 software.
RESULTSTotally, 64 articles were identified, of which 52 were excluded and 12 included with 1 430 cases of prostate cancer and 2 159 normal or BPH controls. There was heterogeneity among the included studies. Based on the random effect model, in predicting the results of prostate biopsy in the diagnosis of prostate cancer, serum PHI exhibited a pooled sensitivity of 55.1% (95% CI: 0.525 - 0.577), a pooled specificity of 71.5% (95% CI: 0.695 - 0.734), a positive likelihood ratio (P-LR) of 2.379 (95% CI: 1.922 - 2.943), a negative likelihood ratio (N-LR) of 0.515 (95% CI: 0.428 - 0.619), a diagnosis odds ratio (DOR) of 5.268 (95% CI: 3.870 - 7.170), a summary receiver operating characteristic curve--area under the curve (SROC AUC) of 0.7578, and a Q * index of 0.6999.
CONCLUSIONSerum PHI plays a role of auxiliary diagnosis in detecting prostate cancer and can be used in the prediction of the results of prostate biopsy.

3.Construction of recombinant expression plasmid pLenti6/V5-D-TOPO(R)-TβRⅡDNglytk and production of the lentivirus in the immunotherapy for prostate cancer

Objective Our hypothesis was if we rendered host' s cytotoxic tumor lymphocytes insensitive to TGF-β,these immune cells could be able to overcome the TGF-β mediated immunosuppression and reject the tumor.We aimed to develop a lentivirus mediated gene transfer program incorporating a herpes simplex virus thymidine kinase(HSV-tk)in our dominant negative TGF-β type Ⅱ receptor(TβRⅡDNglytk)expression vector.So we first need to construct the lentiviral pLenti6/V5-D-TOPO vector containing TβRⅡDNglytk and produce the recombinant lentivirus as the transfection vector.Methods PCR were used to amplify the genes TβRⅡDN and HSV-tk from the respective plasmids.Then the genes were linked by recombinant PCR technology to construct the fusion gene TβRⅡDNglytk and control vector TRANSglytk.According to the operation manual from the Invitrogen company,TOPO cloning technology was used to construct the plasmids of pLenti6/V5-D-TOPO(R)-TβRⅡDNglytk and pLenti6/V5-D-TOPO(R)-TRANSglytk.Both of the constructed plasmids were verified by sequencing.ViraPowerTM Lentiviral System and 293 FT cells provided by Invitrogen were used to produce the recombinant lentivirus vector,the tillers of the lentivirus were determined by 293 cells.Results The construction of the plasmids of pLenti6/V5-D-TOPO(R)-TβRⅡDNglytk and pLenti6/V5-D-TOPO(R)-TRANSglytk were completed succefully.The DNA sequencing results showed that both the plasmids were constructed correctly.Using them we successfully produced infectious lentivirus vectors with appropriate tilters.Conclusions Using TOPO cloning technology in the construction of the plasmids of pLenti6/V5-D-TOPO(R)-TβRⅡDNglytk and pLenti6/V5-D-TOPO(R)-TRANSglytk and recombinant PCR in the fusion genes is feasible.Recombinant PCR combine with TOPO cloning technology can be a simple,highly efficient and rapid way to construct lentiviral vector.The production of infectious lentivirus with appropriate tilters using 293FT is suitable and feasible.The construction of pLenti6/V5-D-TOPO(R)-TβRⅡDNglytk and production of the infectious lentivirus will lay a foundation for immunotherapy of prostate cancer.
4.Detection of serum hyper sensitive C-reactive protein level in patients with acute ischemic stroke and its clinical significances

Objective To explore the level of hypersensitive C-reactive protein (hs-CRP)in the patients with acute ischemic stroke(AIS)and its clinical significance.Methods The BNⅡhighly sensitive automatical scattering turbidimetric immunity analy-zer was adopted to detect serum hs-CRP level in 64 cases of AIS and contemporaneous 32 individuals with healthy physical examina-tion as the control group.The data of AIS cases were retrospectively analyzed.Results The positive rate of hs-CRP was 53.3% in the AIS group and 9% in the control group with statistically significant difference between them(P <0.05);the overall levels of hs-CRP in the AIS group was in inconformity to the normal distribution,the median was 4.26mg/L,which in the control group was (0.99 ± 0.72 )mg/L,there was statistically significant difference between them (P < 0.05 );the average level of hs-CRP was (5.03±1.51)mg/L in the AIS male group and (2.79 ± 1.09)mg/L in the AIS female group,the difference between them had sta-tostical significance(P <0.05).Conclusion The serum hs-CRP level in the AIS patients is higher than that in the healthy crowd with statistical difference(P <0.05).The age and sex may be the causes influencing serum level of hs-CRP in the AIS patients.The average age in the AIS patients with positive hs-CRP is higher than that with negative hs-CRP.Middle-aged and elderly people with hypertension and diabetes are much more dangerous than healthy people in getting AIS.

6.Effects of MT01 on expressions of TLR9,TRAF6 and IL6 in periodontal tissue during experimental tooth movement of rats

Objective:To investigate the changes of expression levels of TLR9 and IL-6 in the periodontal tissue during the experimental tooth movement of the rats, and the effecst of the MT01 on the expression of TLR9,TRAF6 and IL-6 in periodontal tissue,and to clarify its related mechanisms.Methods:Thirty Wistar rats were randomly divided into MT01 intervention group(n=6) and non-MT01 group(n=24).Force of 0.49 N was applied to move the upper first molars mesially. The rats in Non-MT01 intervention group were sacrificed on the days 3,7,14 and 21, and the rats in MT01 intervention group were all sacrificed on the day 7. RT-qPCR was used to detect the expression levels of TLR9,TRAF6 and IL-6 mRNA in maxillary first molar alveolar bone in each group.Results:The expression levels of IL-6 and TLR9 mRNA in loaded side were higher than those in control side(P<0.05), and reached the maximum levels on the day 7(P<0.01);with the interpose of MT01, the expression levels of TLR9 and TRAF6 mRNA were lower than control side(P<0.01).Conclusion: MT01 could down-regulate the expression levels of TLR9 and TRAF6 during orthodontic tooth movement and eventually resists the inflammation during the tooth movement.

Objective: To construct the lentiviral plenti6/V5-D-TOPO vector containing TβR Ⅱ Dnglytk and control vector by developing a lentivirus mediated gene transfer program incorporating a herpes simplex virus thymidine kinase(HSVtk) in the dominant negative TGF-β type Ⅱ receptor (TβR Ⅱ Dnglytk) expression vector. Conclusion: Using TOPO cloning technology in the construction of the plasmids of plenti6/VS-D-TOPO -TβR Ⅱ Dnglytk and plenti6/VS-D-TOPO(R)-TRANSglytk and recombinant PCR in the fusion genes are feasible. The recombinant PCR combined with ToPe cloning technology can be the simple, highly efficient and rapid way to construct lentiviral vector and construction of plenti6/V5-D-TOPO(R)-TβR Ⅱ Dnglytk, which will lay a foundation for tumor immunotherapy.
8.Expression and significance of vasculogenic mimicry and matrix metalloproteinase-2 in prostate cancer

Objective To find out if an unique blood mode-vasculogenic mimicry (VM) exist in prostate cancer and analyze its relationship with prognosis. Methods The age of the patients was 59 to 72 years,with a mean age of (66.7 ± 11.0) years.All had been confirmed as prostate cancer by biopsy.The level of serum PSA was 15.6 to 76.7 μg/L,with a mean level of (34.6 ± 1.7 ) μg/L.A histochemical and immunohistochemical dual staining method for PAS-CD31 and MMP-2 in 96 prostatic adenocarcinomas were conducted to explore if VM exist in prostate cancer,describe its morphology and distribution.The different expression of MMP-2 in VM positive and VM negative tissue were.The clinical data were collected and analyzed to explore the relationship between disease progression and VM. Results Among 96 cases,24 cases were found VM positive,the others were negative.The boundary membrane of VM was PAS positive,which was continuous or incontinuous. A positive correlation between VM density and high Gleason score were found.The tPSA were higher in the VM positive cases than that in VM negatives.The VM positive patients had poorer prognosis than that of the negatives.The MMP-2 expression was found correlated with VM positive (rs =0.60,P < 0.01 ). Conclusions VM exists in the prostate cancer as an independent structure,and it is more common in the high-grade cancer.The progression-free survival of VM positive patients could be worse than the negative.MMP-2 and VM could be the indicators of poor prognosis.
9.Comparative studies of casein phosphopeptide-amorphous calcium phosphate and fluoride products in inhibiting enamel demineralization.

OBJECTIVETo evaluate casein phosphopeptide-amorphous calcium phosphate (CPP-ACP) and several fluoride products in inhibiting enamel demineralization.
METHODSSeventy healthy premolars extracted before orthodontic treatment were divided into seven groups (A: CPP-ACP; B: CPP-ACP + fluoride mouth rinses; C: fluoride mouth rinses; D: fluoride glass ionomer protection; E: fluoride resin binder; F: fluoride varnish; G: control group). There were ten samples in each group. The teeth were dipped into an artificial caries solution ten minutes at a time, then applied separately with CPP-ACP, CPP-ACP + fluoride mouth rinses, fluoride mouth rinses, fluoride glass ionomer protection, fluoride resin binder and fluoride varnish on labial enamel. The samples were dipped into an artificial saliva solution (37°C). Then all samples were examined with atomic force microscope (AFM) at the end of first, second and third monthes.
RESULTSAt the end of the first month, surface roughness in group D [(114 ± 1) nm] was significantly lower than that in group G[(172 ± 9) nm]. At the end of the second month and the third month, significant difference was found in surface roughness between group G and the rest of groups (P < 0.05); between group B and the rest of groups; and between group C and groups (D, E and F).
CONCLUSIONSCPP-ACP and fluoride could both inhibit enamel demineralization in vitro. Solid fluoride (groups D, E and F) had better results than liquid fluoride (group C).

10.Animal experiment and clinical study of Argon plasma coagulator in simple enucleation for small renal cell carcinoma

Objective To evaluate the feasibility and clinical effect of Argon plasma coagulator in simple enucleation for small renal cell carcinoma. Methods On the basis of successful performing the animal experience of coagulating therapy on the wound tissue during partial nephrectomy with Argon plasma coagulator in rabbit models, 10 cases of simple enucleation for small renal cell carcinoma with Argon plasma coagulator were accomplished. Results Both with the standard of stopping bleeding of wound tissue by Argon plasma coagulator and with the standard of re-spraying the wound tissue for 2 s after stopping bleeding using Argon plasma coagulator, the depth of wound tissue necrosis without blocking the renal pedicle is deeper than that with blocking the renal pedicle(P=0. 012 and P=0. 002, respectively).If the wound tissue was re-sprayed for 2 s after stopping bleeding by Argon plasma coagulator, the depth of the wound tissue necrosis without blocking the renal pedicle was deeper than that just with blocking the renal pedicle(P=0. 007 and P=0. 002,respectively). In the part of application in clinical, all procedures were successfully completed. The mean operative time was 163 min (range, 100-210 min) and mean blood loss was 230 ml (range, 100-400 ml). Drainage tube was pulled out 1 month after operation in 1 case for being allergic to absorbable hemostatic gauze, and the mean pulling drainage tube out time in others was 4. 2 d (range, 3-5 d). During a mean follow-up of 22 months (range, 10-38 months), no local tumor recurrence and distant metastasis was found. Conclusion Argon plasma coagulator can be used in simple enucleation for small renal cell carcinoma, and the clinical effectiveness is ideal.
