1.The role of miR-132 in primary cortical neuron ischemic injury induced by oxygen-glucose-deprivation

Objective To investigate the role of miR-132 in oxygen-glucose-deprivation (OGD) induced ischemic injury model using primary cortical neurons.Methods Cultured primary cortical neurons were exposed to oxygen-glucose-deprivation (OGD)for 2 h followed by 24 h reperfusion.The cells were randomly divided into sham operation group (Sham),OGD group,lentivirus control group (LV-control),miR-132 low expression group (LV-anti-miR-132),and miR-132 overexpression group (LV-miR-132).CCK-8,real-time PCR,and Western blot were used to study the effects and mechanisms of miR-132 on primary cortical neurons injured by OGD.Results The expression levels of miR-132 were significantly reduced in the OGD-induced ischemic injury model (P<0.05).The reduced expression of miR-132 aggravated OGD injury and significantly reduced cell viability in OGD-induced primary cortical neuron ischemic injury.On the contrary,the increased expression levels of miR-132 reduced OGD injury and increased cell survival.Western blot results showed that overexpression of miR-132 upregulated the levels of synapsin-1 and PSD95.Conclusion Overexpression of miR-132 can improve OGD-induced ischemic injury and increase the survival of primary cortical neurons by upregulating the synapsin-1 and PSD95.

Objective To establish a reliable pretreatment method for the detection of mequindox and its metabolite in pork luncheon meat by ultra-performance liquid chromatography/triple qudrupole tandem mass spectrometry. Methods Samples were extracted with ethyl acetate, and the results of purification and enrichment by PAX and PEP solid-phase extraction columns were analyzed. Acetonitrile/methanol (3:11) - 0.1% formic acid water was used as the mobile phase, and Shimadzu Inertsil ODS-3-column (3µm, 2.1 × 100mm) chromatographic columns were used for qualitative and quantitative analysis using the multi-reaction detection positive ion mode. Results The results showed that PEP cartridge had good recovery rate. The detection limit of mequindox was 0.10µg/kg, and limit of quantitation was 0.30µg/kg. The average recoveries for spiked levels of 0.33, 0.83, and 1.65µg/kg were 127%, 72.0%, and 60.1%, respectively. The detection limit of 2-quinoxalinecarboxylic acid was 0.10µg/kg, and limit of quantitation was 0.40µg/kg. The average recoveries for spiked levels of 0.42, 1.05, and 2.1µg/kg were 125%, 99.0%, and 60.9%, respectively. Conclusion This method is suitable for the determination of mequindox and its metabolite 2-quinoxalinecarboxylic acid in luncheon meat.

5.Observation of Therapeutic Efficacy of Oxcarbazepine Combined with Levetiracetam in the Treatment of Cognitive Dysfunction in Adult Patients with Temporal Lobe Epilepsy

OBJECTIVE:To observe therapuetic effica cy of oxcarbazepine combined with levetiracetam in the treatment of cognitive dysfunction in adult patients with temporal lobe epilepsy. METHODS :According to inclusion and exclusion criteria ,83 adult patients with temporal lobe epilepsy were collected from neurology department of the First Affiliated Hospital of Kunming Medical University during January 2018 to October 2019,and then divided into control group (39 cases)and observation group (44 cases). Control group was given Oxcarbazepine tablet with initial dose of 8-10 mg/(kg·d),morning and night ,increased by 5-10 mg/(kg·d)each week according to patient ’s condition ,the maximal dose was 45 mg/(kg·d),lasting for 3 months with the lowest effective treatment dose. Based on treatment of control group ,observation group was additionally given Levetiracetam tablet with initial dose of 5-10 mg/(kg·d),morning and night ,increased by 5-10 mg/(kg·d)each week according to the severity of disease,the total dose was not more than 2 000 mg/d,lasting for 3 months with the lowest effective treatment dose. The event-related potential P 300 at Pz site in 2 groups was measured before and after treatment (including the incubation period of N 1, P2,N2 and P 3 waves,amplitude of P 3 wave). Webster ’s Adult Intelligence Scale [WAIS-RC ,including verbal IQ (VIQ), performance IQ (PIQ),full scale IQ (FIQ)scores] and Clinical Memory Scale [CMS ,converting to memory quotient (MQ) score] were adopted. The occurrence of ADR was recorded. RESULTS :Totally 5 patients of control group and 7 patients of observation group fell off. Finally ,a total of 71 patients participated in the entire study (34 cases in control group and 37 cases in observation group ). Before treatment ,there was no statistical significance in P 300 at Pz site ,WAIS-RC and CMS between 2 groups (P>0.05). Compared with before treatment ,incubation period of N 2 and P 3 waves was shortenedsignificantly in 2 groups,and amplitude of P 3 wave wasincreased significantly after treatment (P<0.05);VIQ,FIQ and MQ scores were increased significantly (P<0.05). The incubation pe riod of P 2 wave was shortened significantly,and PIQ score was increased significantly in observation group (P< 0.05). Compared with control group ,the shortening degree of incubation period of P 2,P3 waves,rising degree of amplitude of P 3 wave and rising degree of VIQ ,PIQ,FIQ,MQ scores were more bigger after treatment in observation (P<0.05). In the control group,there was 1 case of slight rash ,1 case of conscious asthenia ;in the observation group ,there was 1 case of nausea and vomiting,1 case of conscious asthenia ,and 1 case of emotional instability ;no serious ADR was found in 2 groups. CONCLUSIONS:Oxcarbazepine combined with levetiracetam can improve cognitive dysfunction in adult patients with temporal lobe epilepsy ,and the treatment effect is significantly better than oxcarbazepine monotherapy.

7.Mental health and influencing factors among residents under standardized residency training during the epidemic of COVID-19

ObjectiveTo analyze the mental health status and the influencing factors among the residents taking standardized residency training during the outbreak and stable period of COVID-19, so as to provide theoretical basis for their mental health education. MethodsOn February 8 to February 10 and April 11 to April 13, 2020, Symptom Checklist 90 (SCL-90), Beck Srivastava Stress Inventory (BSSI) and Simplified Coping Style Questionnaire (SCSQ) were distributed in online chat groups involving all grades of residents taking standardized residency training in the Second Affiliated Hospital of Kunming Medical University. Through two rounds of questionnaire survey, non-probability sampling method was used to obtain survey samples, and their mental status were analyzed. ResultsA total of 159 valid questionnaires were collected in the first round, and 99 valid questionnaires in the second round. The first survey showed that the total score of SCL-90 was (117.69±37.74) and the detection rate of positive symptoms in SCL-90 was 25.8%, and mainly dominated by obsession, fear and interpersonal sensitivity. In the second survey, the results showed that the total score of SCL-90 was (127.19±51.44), and the main positive symptoms included obsession, depression and interpersonal sensitivity, with a positive detection rate of 30.3 %. The first survey found that the mental health status had significant differences among residents of different grades (χ2=7.46, P<0.05), furthermore, the results indicated that lower grade was a risk factor while non-singleton was the protective factor of mental health status (P<0.05), and SCL-90 total score was positively correlated with score of negative coping styles (r=0.45, P<0.01). The second survey also classified lower grade as risk factor and non-singleton as protective factor (P<0.05), and SCL-90 total score was positively correlated with study stress, economic pressure, interpersonal relationship, clinical practice and negative coping styles (r=0.52, 0.46, 0.55, 0.54, P<0.05 or 0.01). ConclusionResidents under standardized residency training have obvious mental health problems during the outbreak of COVID-19, and the problems become more serious during the stable period of COVID-19.

The bile duct is the channel for bile transport in the liver, and the lining epithelial cells in the bile duct have functional and morphological heterogeneity and are the target of various bile duct diseases. In addition to bile metabolism and secretion, biliary epithelial cells are also involved in tissue damage and repair. Biliary epithelial cells have an immune barrier function and can secrete different proinflammatory factors and chemokines. Under the stimulation of endogenous and exogenous factors, biliary epithelial cells present immune reactivity and initiate immune response in the host. This article reviews the current status of research on bile duct lesions and related liver diseases.
9.Prognostic evaluation of transjugular intrahepatic portosystemic shunt in treatment of cirrhotic portal hypertension

Transjugular intrahepatic portosystemic shunt (TIPS) is an important method for the treatment of cirrhotic portal hypertension; however, its clinical application is limited by postoperative complications such as liver failure and hepatic encephalopathy. This article summarizes and analyzes the research advances in the clinical value of serological markers, clinical indices, and scoring systems in prognostic evaluation of TIPS. Indices can be selected or comprehensive evaluation can be performed based on clinical situations, in order to improve patients’ postoperative survival rate.
