1.Primary pharynx synovial sarcoma: a case report.

A case of primary pharynx synovial sarcoma was reported in this paper. A 15-year-old male patient experienced painless pharyngeal swelling that gradually proliferated for 1 month. Special examination showed an 8 cm × 4 cm × 3 cm tumor located in the left pharynx and the supratonsillar crypt. Imaging tests revealed an irregular mass on the left side of the oropharynx and an unclear boundary. Immunohistochemical examination yielded the following results: epithelial membrane antigen (+), cytokeratin (CK)19 (+), CD7(+), vimentin (+), CK10(-), E-cadherin (+), B-cell lymphoma-2 (-), CD2 (-), CD10 (-), CD138 (+), CD99 (+), leukocyte common antigen (+), and Ki-67 (20%+). This condition was pathologically diagnosed as primary pharynx synovial sarcoma.

2. Application of Quantitative Analysis of Multi-components by Single Marker in Quality Control of Chinese Medicinal Materials Containing Saponins

Due to the multi-component and multi-target features of Chinese medicinal materials (CMMs),multiple active components could be more reasonably represent the quality of CMMs compared with the single-component QC mode. However,it is still difficult to apply the multi-component QC mode because of the instability, high cost and inaccessibility of reference substances of CMMs. Saponins are glycosides with aglycones of triterpene or spirostane and widely distributed in plants. Saponins are also the major active constituents of many CMMs,with multi-effects of inhibiting tumors,regulating the immune system,inhibiting virus,preventing and treating cardiovascular diseases. Therefore,rational and effective control of the quality of CMMs containing saponins is of great significance for ensuring the clinical safety and efficacy of such CMMs and related products. The quantitative analysis of multi-components by single marker (QAMS) can use only one reference substance to achieve the simultaneous monitoring of multiple components in CMMs,and make up the weaknesses of multi-component QC mode, and has been well developed and validated in the QC and evaluation of CMMs for more than ten years since it was put forward. And now it has been widely used in the QC of CMMs containing saponins. Based on the investigation of QAMS theory and literatures in the past decade,studies on the QC of CMMs and related preparations containing triterpenoid saponins and steroidal saponins by QAMS were summarized and discussed systematically. In addition,some possible problems were analyzed and interpreted,in order to provide reliable basis for more QC of CMMs and reference for the continuous use and in-depth development of this method in the research of CMMs.
3.Effect of pneumoperitoneum on renal function after robotic-assisted laparoscopic kidney transplantation

Objective To investigate the effect of pneumoperitoneum pressure during robotic-assisted kidney transplantation (RAKT) on the function of the transplant kidney. Methods The data of 243 kidney transplant recipients were retrospectively analyzed and divided into open kidney transplantation (OKT) group (n=105) and RAKT group (n=138). The RAKT group was further divided into 13 mmHg group (n=67) and 7 mmHg group (n=71) based on pneumoperitoneum pressure. The donor information, recipient's preoperative general data, intraoperative data, and postoperative recovery of the three groups were compared. In the RAKT group, the renal artery, segmental artery, interlobar artery, and venous flow velocity of the transplant kidney were measured using laparoscopic ultrasound. Results There was a statistically significant difference in donor types among the groups (P<0.05), while other donor information and recipient's preoperative general data showed no statistically significant differences (all P>0.05). There were no statistically significant differences in serum creatinine and complications at 30 days and 1 year postoperatively among the groups (all P>0.05). The OKT group and 7 mmHg group had more intraoperative urine output than the 13 mmHg group. Both RAKT groups had less intraoperative blood loss and shorter hospital stays than the OKT group, and longer operation times than the OKT group (all P<0.05). There were no statistically significant differences in operation time, intraoperative blood loss, and hospital stay between the two RAKT groups (all P>0.05). The vascular flow velocity of the transplant kidney decreased at 13 mmHg compared to 7 mmHg pneumoperitoneum pressure, but the differences were not statistically significant (all P>0.05). Conclusions Controllable pneumoperitoneum pressure has a limited impact on the vascular flow velocity of the transplanted kidney. RAKT is a safe and effective surgical method under appropriate pneumoperitoneum pressure, and choosing a lower pneumoperitoneum pressure is more conducive to the early recovery of renal function postoperatively.
4.Progress of research on the role of Atoh1 gene in the regeneration of mammalian auditory hair cells.

Atoh1 gene encodes a helix-loop-helix transcription factor which is involved in the generation and differentiation of mammalian auditory hair cells and supporting cells, and regulation of the proliferation of cochlear cells, therefore plays an important role in the pathogenesis and recovery of sensorineural deafness. This study reviews the progress of the Atoh1 gene in hair cell regeneration, with the aim of providing a reference for the study of hair cell regeneration gene therapy for sensorineural deafness.

5.Molecular cloning, bacterial expression analysis and functional characterization of pathogenesis-related PR10-2 gene in Panax notoginseng.

Base on the transcriptome analysis and RT-PCR techniques,a pathogenesis-related protein 10 gene was isolated from Panax notoginseng root and named as PnPR10-2. Bioinformatics and phylogenetic trees analysis revealed that open reading frame (ORF) of PnPR10-2 was 465 bp in length,encoding 154 amino acids,containing one typical conserved domain of pathogenesis related protein Bet v I family, and showed high similarity with that from P. ginseng. The recombinant expressed plasmid pET32a(+)-PnPR10-2 was expressed in Escherichia coli BL21. The expression conditions were optimized and it could be expressed well in soluble and inclusion body protein. Purified PnPR10-2 recombinant protein from the supernatant of cells was used to analysis the pathogen resistance activity by paper method. The purified recombinant protein could inhibit typical root rot disease pathogen (Fusarium solani and Cylindrocarpon destructans)growth evidently, we conjecture that PnPR10-2 may participated in defense response of P. notoginseng resistance to root rot disease pathogen.
6.Survey and analysis of tooth extraction anxiety of dental patients.

OBJECTIVEThis study aims to understand the current status and analyze the influencing factors of tooth extraction anxiety of dental patients.
METHODSThe General Factor Questionnaire and Modified Dental Anxiety Scale (MDAS) were used to survey and analyze tooth extraction anxiety of 800 dental patients. Single-factor and multi-factor analyses were used for statistical data analysis.
RESULTSA total of 768 qualified questionnaires were received out of 800, the efficiency was 96%. The mean MDAS score was 10.21±3.29. The prevalence percentage of dental anxiety was 14.7%. Significant statistical differences of the incidence of dental anxiety were observed with regard to sex, age, tooth extraction time, accompanying relatives and friends, before tooth extraction, tooth extraction experience for family and friends, pain tolerance, mental health state before tooth extraction, and self-reported oral health status (P<0.05. By contrast, no significant statistical differences in the incidence of dental anxiety were observed with regard to educational level, tooth extraction type, national life experience, profession, and personality (P>0.05). Logistic regression analysis indicated that the risk factors of dental anxiety were sex, accompanying relatives and friends, mental health state before tooth extraction, and self-rated oral health status.
CONCLUSIONSThe prevalence percentage of tooth extraction anxiety is high for dental patients. Gender, age, health satisfaction, tooth extraction time, accompanying relatives and friends, before tooth extraction, tooth extraction experience for family and friends, pain tolerance, mental health state before tooth extraction, and self-reported oral health status are the influencing factors of dental anxiety.
7. Clinical analysis of 83 cases of perineal Ⅲ-Ⅳ degree laceration

OBJECTIVE: To investigate the reasons and prevention and treatment measures of acute third-and fourthdegree obstetric lacerations.METHODS: A retrospective analysis of 83 cases of acute third-and fourth-degree obstetric lacerations in the First Affiliated Hospital of Kunming Medical University from 2010 to 2016 was performed.RESULTS:(1)The incidence of 3 rd or 4 th degree laceration was 0.394%(83/21 083),which was 0.433%(72/16 620)in primiparas and 0.246%(11/4463)in multiparous women,there being a statistical difference(P<0.05).(2)The incidence of 3 rd or 4 th degree laceration was significantly increased in the group with episiotomy than without episiotomy[0.583%(48/8229)vs.0.272%(35/12 854),P<0.05].(3)The incidence of 3 rd or 4 th degree laceration was significantly increased in the group with Forceps delivery than without Forceps deliery[3.120%(37/1186)vs.0.231%(46/19 897),P<0.05].(4)In patients with forceps delivery,the incidence of 3 rd or 4 th degree laceration was 2.911%(28/962)in episiotomy group and 4.018%(9/224)in without episiotomy group,there being a significant difference(P<0.05).(5)The incidence of 3 rd or 4 th degree laceration was significantly increased in the macrosomia group than in the non-macrosomia group[1.225%(11/898)vs.0.357%(72/20 185),P<0.05].(6)The incidence of 3 rd or 4 th degree laceration was significantly increased in the prolonged second stage group than in the normal second stage group[2.381%(24/1008)vs.0.294%(59/20 075),P<0.05].CONCLUSION: The incidence of third-and fourth-degree obstetric lacerations is increased in primiparous women,episiotomy,forceps delivery,forceps delivery without episiotomy,big fetal weight and prolonged second stage.
8.Protective effect of against ethanol-induced gastric ulcer and its mechanism.

: To investigate the protective effect of (FD) against ethanol-induced gastric ulcer and its mechanism. : Human gastric epithelial GES-1 cells were divided into normal control group, model control group, FD 95% alcohol extract group, FD 50% alcohol extract group and FD decoction extract group. Gastric ulcer was induced by treatment with 1% ethanol in GES-1 cells. The cell proliferation was detected with MTT method in each group. Sixty SD rats were randomly divided into normal control group, model control group, ranitidine group and low-dose, medium-dose, high-dose FD 95% alcohol extract groups (150, 300, 600 mg/kg). The corresponding drugs were administrated by gavage for The gastric ulcer model was induced by intragastric administration of anhydrous ethanol. The gastric ulcer area and ulcer inhibition rate of rats were measured in each group; the degree of gastricmucosal damage was observed by scanning electron microscopy; the levels of tumor necrosis factor (TNF)-α, interleukin (IL)-6, IL-1β in serum and the content of malondialdehyde (MDA), superoxide dismutase (SOD), glutathione (GSH), catalase (CAT) in gastric tissues were detected by ELISA method. : 95% alcohol extract of FD had the strongest protective effect on proliferation of GES-1 cells. In animal experiments, compared with the normal control group, a large area of ulcers appeared on the gastric mucosa in the model control group, while the ulcer areas of the FD groups and ranitidine group were significantly smaller than that of the model control group (all <0.05). Compared with the model control group, FD groups and ranitidine group significantly reduced the levels of TNF-α, IL-1β, IL-6 in serum and the MDA content in the gastric tissues, and increased the activity of SOD, CAT and GSH in gastric tissues (all <0.05). : The 95% alcohol extract of FD can reduce the levels of TNF-α, IL-1β and IL-6 in serum and the content of MDA in gastric tissues, and increase the activity of SOD, CAT and GSH in gastric tissues to achieve the protective effect against gastric ulcer.

9.Methylation status and expression of TWEAK gene promoter region in peripheral blood of patients with rheumatoid arthritis.

OBJECTIVE:
To explore the relationship between tumor necrosis factor like weak inducer of apoptosis (TWEAK) gene and the pathogenesis of rheumatoid arthritis (RA) by detecting the DNA methylation level, mRNA expression level and serum protein concentration of TWEAK gene in peripheral blood.
METHODS:
The MassARRAY method was used to detect the DNA methylation level of the TWEAK gene in the peripheral blood of 112 RA patients and 86 matched healthy volunteers. The real-time quantitative polymerase chain reaction method was used to detect the mRNA expression level of the TWEAK gene in the peripheral blood of the subjects. The enzyme-linked immunosorbent assay method was used to detect the serum TWEAK protein concentration of the subjects. The TWEAK gene DNA methylation level, mRNA expression level and serum protein concentration between the RA group and the healthy control group were compared, and the relationship between it and the degree of disease activity analyzed.
RESULTS:
The overall DNA methylation level of TWEAK gene and the DNA methylation levels of CpG_11, CpG_17.18.19.20, CpG_40.41.42 site in the RA group were higher than those in the healthy control group (P=0.002, P=0.01, P=0.006, P=0.002, respectively). The DNA methylation level of CpG_55.56 site in the high disease activity group was higher than that in the medium and low disease activity group (P=0.041). The expression level of TWEAK gene mRNA in the peripheral blood of the RA group was lower than that of the healthy control group (P=0.023). The expression level of TWEAK gene mRNA in the high disease activity group was lower than that in the medium and low disease activity group (P=0.035). The serum TWEAK protein concentration of the RA group was not significantly different from that of the healthy control group (P=0.508), but it was positively correlated with the mRNA expression level (r=0.482, P < 0.001).
CONCLUSION
The TWEAK gene is closely related to the onset and progression of RA, and its hypermethylation state may be one of the epigenetic mechanisms regulating its low mRNA expression, and it can be used as one of the important indicators for clinical monitoring and evaluation of RA.

10.Investigation on Demodex infections among university students in Kunming City

Objective To investigate the prevalence of Demodex infection among students in Kunming Medical University, and identify the factors affecting Demodex infections, so as to provide the evidence for the development of the strategy for the prevention of Demodex infections. Methods A total of 1 463 students from Grade 2014 who studied Medical Parasitology in Kunming Medical University were included in the survey. Demodex was examined in students’facial skin using the cellophane tape method, and the species was identified using microscopy. The students’gender, ethnicity, place of origin and skin type were captured using a questionnaire survey. Results The overall prevalence of Demodex infections was 19.07% (279/1 463) on the facial skin among the university students, and a higher prevalence was seen in girls (21.16%, 183/865) than in boys (16.05%, 96/598) (χ2 =5.965,P <0.05).TheprevalenceofDemodex infectionswas18.33%(66/360)amongminorethnicstudents,andnoethnicity-specific prevalence was seen (P > 0.05). Demodex folliculorum was the predominant species, with a prevalence of 50.54% (141/279), and mild infections were predominant among all infections (96.77%, 270/279), without severe infections seen. Multivariate nonconditional logistic regression analysis revealed that gender and roommates with Demodex infections were risk factors of Demodex infections, and the infection was not associated with ethnicity, place of origin or skin type. There were only 2.53% (37/1 463) of the subjects understanding the knowledge pertaining to the prevention and control of Demodex infection. Conclusions A relatively low prevalence of Demodex infection is detected in the facial skin of students from Kunming Medical University, and Demodex infection is associated with gender and roommates with Demodex infections. Health education pertaining to the prevention of Demodex infections is suggested to be intensified among university students.
