1.Clinical and imaging analysis for 162 cases of moyamoya disease

Objective To investigate the clinical manifestations and imaging features of moyamoya disease (MMD).Method The clinical manifestations and the results of CT angiography ( CTA ) and digital subtraction angiography (DSA) examinations of 162 cases diagnosed with M MD were analyzed retrospectively.Results In 162 patients with MMD, 132 cases (81.48% ) had stroke, in which cerebral infarction and transient ischemic attack (TIA) were 20 cases, intraventricular hemorrhage were 48 cases, cerebral hemorrhage were 52 cases, subarachnoid hemorrhage (SAH) were 10 cases and acute subdural hemorrhage were 2 cases; 8 cases(4.94%) had simple migraine and 22 cases(13.58%) had simple epilepsy.All the patients were with typical imaging findings: ( 1 ) The central branch of the internal carotid artery,bilateral internal carotid artery clinoid segment and proximal arterial stenosis or occlusion was the main performance of MMD.(2) Abnormal vascular network in the skull base was the most feature of MMD changes.(3) The performance of collateral circulation.Conclusions CTA and DSA can be used for the diagnosis of MMD patients.DSA is the gold standard for diagnosis of MMD.
2. A preliminary study on the origin of neonatal intestinal microbiota during cesarean section

OBJECTIVE: To explore the composition of microbial communities in vagina and intestine of the mother,the placenta and the neonatal meconium after cesarean section and to analyze the origin of neonatal intestinal microbiota.METHODS: Samples of intestine,vagina and placenta and neonatal meconium from 4 women who underwent cesarean section in Yan'an Hospital of Kunming City in October 2016 were collected.The high-throughput sequencing technology was used to sequence the 16 S rRNA gene,and the composition of the microbial communities and the relationship among the samples were analyzed.RESULTS: Firstly,comparing the number of OTUs in different samples,it was found that the number in placenta was the highest,followed by the intestine and vagina,and the least was in the neonatal meconium.Secondly,each sample was analyzed by PCA clustering,and it was found that the neonatal meconium was affected least by individual differences but the vagina was affected most.The intestinal and placental microbial communities had certain similarities.Finally,comparing the microbial community composition of each sample,it was found that the highest abundance in the neonatal meconium and placenta was Proteobacteria,and in the intestine and vagina it was the Firmicutes.CONCLUSION: During the caesarean section,maternal microbiota transfer may be from the mother's intestine to the placenta and then to the infant's intestine.
3.Determination of mequindox and its metabolite in pork luncheon meat by ultra-performance liquid chromatography/triple qudrupole tandem mass spectrometry

Objective To establish a reliable pretreatment method for the detection of mequindox and its metabolite in pork luncheon meat by ultra-performance liquid chromatography/triple qudrupole tandem mass spectrometry. Methods Samples were extracted with ethyl acetate, and the results of purification and enrichment by PAX and PEP solid-phase extraction columns were analyzed. Acetonitrile/methanol (3:11) - 0.1% formic acid water was used as the mobile phase, and Shimadzu Inertsil ODS-3-column (3µm, 2.1 × 100mm) chromatographic columns were used for qualitative and quantitative analysis using the multi-reaction detection positive ion mode. Results The results showed that PEP cartridge had good recovery rate. The detection limit of mequindox was 0.10µg/kg, and limit of quantitation was 0.30µg/kg. The average recoveries for spiked levels of 0.33, 0.83, and 1.65µg/kg were 127%, 72.0%, and 60.1%, respectively. The detection limit of 2-quinoxalinecarboxylic acid was 0.10µg/kg, and limit of quantitation was 0.40µg/kg. The average recoveries for spiked levels of 0.42, 1.05, and 2.1µg/kg were 125%, 99.0%, and 60.9%, respectively. Conclusion This method is suitable for the determination of mequindox and its metabolite 2-quinoxalinecarboxylic acid in luncheon meat.

Objective To explore the correlation between 4 tag single nucleotide polymorphisms (Tag SNP) sites (rs7721799, rs32897, rs7718461, rs10062367) of corticotropin releasing hormone binding protein (CRHBP) and schizophrenia and aggressive behavior in the Yunnan Han population. Methods Case-control correlation analysis was used to establish a complex amplification system. Improved multiplex ligase detection reaction (iMLDR) technology was used to detect the genotypes of 4 SNP sites of CRHBP gene of 163 Han schizophrenic patients (including 81 patients with aggressive behavior, 82 patients without aggressive behavior) and 345 healthy Han individuals, which were analyzed statistically by SPSS 19.0, Haploview 4.2 and PHASE 2.1 software. Results There was no correlation between the 3 SNP sites of CRHBP gene and the onset of schizophrenia except for the rs7718461 site (P>0.05). The relative risk of aggressive behavior of patients carrying GG or GA genotype at rs7718461 site were 4.903 times higher than those carrying AA genotype (P<0.05). Conclusion The CRHBP gene may not be associated with the occurrence of schizophrenia in Yunnan Han population, but AA genotype of rs7718461 may reduce the risk of aggressive behavior in schizophrenia patients.

6.Research advances in mesenchymal stem cells from various sources in treatment of liver fibrosis

Mesenchymal stem cells (MSC) are derived from the mesoderm, and exhibit multipotential differentiation. MSC can alleviate liver fibrosis and improve liver function through paracrine cytokines and cell-cell interactions, but their therapeutic mechanism remains unknown. MSC are derived from various tissues, such as bone marrow, umbilical cord tissue, umbilical cord blood, peripheral blood, and adipose tissue. However, there are different advances in MSC with various sources in experimental studies and clinical treatment of liver fibrosis. This paper summarizes the possible therapeutic mechanisms of MSC in alleviating liver fibrosis, as well as the advances in experimental studies and clinical application of MSC with various sources. MSC transplantation is a new therapeutic method for liver fibrosis and has a broad application prospect in this field.
7.Clinical issues in the diagnosis and treatment of tuberculosis patients with positive antibody to hepatitis C virus

Tuberculosis patients with positive antibody to hepatitis C virus (TB-HCV patients) are often seen in clinical practice, and these TB-HCV patients include those with HCV infection. That makes the clinical management and diagnosis/treatment of TB-HCV difficult. This article introduces the prevalence of TB-HCV around the world, and analyzes the potential issues in the diagnosis and treatment of TB-HCV patients, such as drug-drug interactions, drug-induced liver injury, HCV reactivation, and TB reactivation. Through this review, it is recommended that the management should be strengthened and the appropriate therapeutic regimen should be selected in the diagnosis and treatment of TB-HCV patients.
8.Arrhythmia induced by clozapine in patients with schizophrenia: a Meta-analysis

BackgroundClozapine belongs to atypical antipsychotic drugs that has shown significant efficacy in treating schizophrenia. However, clozapine can induce arrhythmias in patients. Currently, there is a lack of systematic research on the types and incidence of clozapine-induced arrhythmias. ObjectiveTo explore the types and incidence of arrhythmias in patients with schizophrenia induced by clozapine through Meta-analysis, so as to provide references for clinical treatment. MethodsOn October 31, 2022, a search was conducted through PubMed, Embase, Cochrane Library and China National Knowledge Infrastructure (CNKI) to collect relevant literature on clozapine-induced arrhythmia in patients with schizophrenia. Joanna Briggs Institute (JBI) was used to evaluate the quality of the included literature. Meta-analysis was conducted using Review Manager 5.4. ResultsA total of 12 studies were included, involving 42 490 schizophrenia patients taking clozapine. The incidence of arrhythmias caused by clozapine was 23% (95% CI: 0.16~0.29). Among them, the incidence of tachycardia was 25% (95% CI: 0.15~0.35), atrial arrhythmia was 6% (95% CI: -0.02~0.14), ventricular arrhythmia was 7% (95% CI: -0.02~0.16), conduction block was 2% (95% CI: 0~0.04), QTc prolongation was 4% (95% CI: -0.04~0.12) and T-wave changes was 23% (95% CI: 0.16~0.30). ConclusionSchizophrenia patients taking clozapine may increase their risk of developing arrhythmia, with tachycardia being the most prevalent. 9.Protective effect of Epothilone D against traumatic optic nerve injury in rats.

OBJECTIVE:
To investigate the therapeutic effect of Epothilone D on traumatic optic neuropathy (TON) in rats.
METHODS:
Forty-two SD rats were randomized to receive intraperitoneal injection of 1.0 mg/kg Epothilone D or DMSO (control) every 3 days until day 28, and rat models of TON were established on the second day after the first administration. On days 3, 7, and 28, examination of flash visual evoked potentials (FVEP), immunofluorescence staining and Western blotting were performed to examine the visual pathway features, number of retinal ganglion cells (RGCs), GAP43 expression level in damaged axons, and changes of Tau and pTau-396/404 in the retina and optic nerve.
RESULTS:
In Epothilone D treatment group, RGC loss rate was significantly decreased by 19.12% (P=0.032) on day 3 and by 22.67% (P=0.042) on day 28 as compared with the rats in the control group, but FVEP examination failed to show physiological improvement in the visual pathway on day 28 in terms of the relative latency of N2 wave (P=0.236) and relative amplitude attenuation of P2-N2 wave (P=0.441). The total Tau content in the retina of the treatment group was significantly increased compared with that in the control group on day 3 (P < 0.001), showing a consistent change with ptau-396/404 level. In the optic nerve axons, the total Tau level in the treatment group was significantly lower than that in the control group on day 7 (P=0.002), but the changes of the total Tau and pTau-396/404 level did not show an obvious correlation. Epothilone D induced persistent expression of GAP43 in the damaged axons, detectable even on day 28 of the experiment.
CONCLUSION
Epothilone D treatment can protect against TON in rats by promoting the survival of injured RGCs, enhancing Tau content in the surviving RGCs, reducing Tau accumulation in injured axons, and stimulating sustained regeneration of axons.

10.Short-term effect of double bundle reconstruction of posterior cruciate ligament with total internal short tendon technique.

OBJECTIVE:
To explore the method and curative effect of double bundle reconstruction of posterior cruciate ligament with total internal short tendon technique.
METHODS:
From October 2010 to June 2018, 50 patients with simple posterior cruciate ligament rupture were admitted, including 35 males and 15 females, ranging in age from 20 to 45 years old, 16 with acute injury and 34 with old injury. The posterior cruciate ligament (PCL) was reconstructed by double bundle technique with the whole internal short tendon. The two ends of the transplanted ligament were fixed by titanium plate suspended with adjustable loop. The Lysholm knee score and IKDC score were used to evaluate the clinical effects.
RESULTS:
All the 50 patients were followed up, and the duration ranged from 6 to 60 months, with a mean of (28.84±9.52) months. At the latest follow up, the knee joint activity returned to normal range. The median value of Lysholm knee score was 54 before operation and 100 after operation. According to the IKDC score, there were 0 case of grade A, 0 case of grade B, 20 cases of grade C and 30 cases of grade D before operation;29 cases of grade A, 19 cases of grade B, 1 case of grade C and 1 case of grade D after operation;the difference was statistically significant (<0.01).
CONCLUSION
Arthroscopic double bundle reconstruction of posterior cruciate ligament with short internal tendon is safe and reliable, with less tendon transplantation, more stable fixation, and more bone reserve, which is beneficial for healing and renovation. The short term effect is positive.
