1.Protective effect of isorhamnetin on oxidative stress injury of HaCaT cells induced by H 2O 2

Objective:To investigate the protective effect of isorhamnetin on oxidative stress injury of HaCaT cells induced by H 2O 2. Methods:HaCaT cells were cultured in vitro and treated with different concentrations of H 2O 2 (300, 600, 900, 1 200 μmol/L) for 12 h. Cell proliferation activity was detected by cell counting kit-8 (CCK-8) assay; SOD activity was detected by superoxide dismutase (SOD) kit and malondialdehyde (MDA) content was detected by MDA assay. The oxidative stress model was established by the selection of suitable H 2O 2 concentration. HaCaT cells were pretreated with isorhamnetin at different concentrations for 12 h, and cell survival rate was detected by CCK-8 method to determine the safe concentration of isorhamnetin for subsequent experiments. HaCaT cells were pretreated with safe concentration of isorhamnetin for 12 h, and H 2O 2 was used to interfere with HaCaT cells for 12 h. Cell proliferation activity, SOD activity and MDA content were detected. Results:With the increase of H 2O 2 concentration, the cell survival rate decreased gradually, the SOD activity decreased gradually and MDA content increased gradually. Compared with the control group, the survival rate of 600, 900 and 1 200 μmol/L H 2O 2 groups was statistically significant ( P<0.05); The SOD activity and MDA content of H 2O 2 groups (300, 600, 900, 1 200 μmol/L) were significantly different from those of the control group ( P<0.05). The oxidative stress model of HaCaT cells was established by 600 μmol/L H 2O 2. HaCaT cells treated with 20, 40, 60, 80 and 100 μmol/L isorhamnetin for 12 h showed no cytotoxic effect. 20, 40 and 60 μmol/L isorhamnetin was selected for subsequent experiments. Compared with H 2O 2 groups, the cell proliferation activity in 40 and 60 μmol/L isornetin groups was significantly increased [(72.21±5.11)%, (76.08±4.91)%, P<0.05], SOD activity increased (19.81±0.38, 20.52±0.52, 15.45±3.13, P<0.05) and MDA content decreased (35.94±0.31, 22.04±0.26, 19.26±1.36, P<0.05). Conclusions:The flavonoid isorhamnetin has a protective effect on oxidative stress injury induced by H 2O 2 in HaCaT cells, suggesting that isorhamnetin may be a potential drug component in the treatment of vitiligo.
2.The image noise effect on the results of Gamma knife dosimetry parameters test.

In order to analyze the image noise effect on the results of Gamma knife dosimetry parameter test, we tested the dosimetry parameters of the Gamma knives according to GBZ 168-2005. Radiological protection standards of X (gamma)-ray stereotactic radiosurgery for head treatment. Dose analysis software was applied to examine the testing film before and after image denoising, and SPSS 11.0 software was used for statistical analysis. The results showed that there was a significant difference in the results of the maximum deviation between radiation field size and its nominal value (t = 7.600, P < 0.01) and the radiation field's penumbra region width of collimators also had significantly different sizes (t = 5.334, P < 0.01) before and after image denoising. This study indicated that the image noise could influence the results of testing Gamma knife dosimetry parameters, so as to cause deviations.

3.Efficacy of modified Park method in blepharoplasty

Objective:To investigate the modified application of orbicularis oculi muscle-levator aponeurosis composite tissue flap fixation technique (modified Park method) in double-eyelid surgery.Methods:From June 2019 to March 2021, 823 patients included 92 males and 731 females, aged from 18 to 45 years with mean (31.4±7.9) years, in which 142 patients with slightly ptosis were selected to receive the treatment in the Department of Plastic Surgery of the Affiliated Hospital of Nangjing University of TCM. The symmetry of bilateral double eyelids after operation was evaluated by the third party. The marginal reflex distance (MRD1) was measured before and 6 months after operation, and the difference between preoperative and postoperative MRD1 (ΔMRD1) was calculated. And the postoperative complications were counted. The patients were followed up from 6 to 12 months after surgery to evaluate the long-term results.Results:The patients were followed up for 4-22 months (mean 10 months) after operation. And the incisions of all the 823 cases had primary healling. Most patients had smooth upper eyelid crease, upturned lashes and dynamic fold. The skin tended to spread evenly without dimple. The symmetry evaluation by the third party showed that there were 764 patients with symmetry and basic symmetry after operation, the symmetry rate was 92.8%; 59 patients with asymmetry, the asymmetry rate was 7.2%. The data of MRD1 increased significantly 6 months after operation, from (3.25±0.72) mm to (3.64±0.61) mm, and the ΔMRD1 was (0.38±0.76) mm ( t=-20.41, P<0.05). The difference between the two groups was statistically significant. The double eyelid line became shallow in 8 cases and disappeared completely in 3 cases. The double eyelid curve of the patients was natural and smooth after the operation. And there was no obvious sunken scar and pretarsal fullness. Conclusions:The " modified Park technique" blepharoplasty can obtain a more firm, symmetrical and natural appearance. And the mild ptosis could be corrected by this method. The anatomical structure can be identified clearly, and the surgical results are highly repeatable. It is worth promoting.
4.Protective effects of isorhamnetin against H 2O 2-induced mitochondrial structural and functional damage in HaCaT cells

Objective:To evaluate protective effects of isorhamnetin on mitochondrial structure and function in HaCaT cells under oxidative stress.Methods:HaCaT cells served as the research object, and were divided into 4 groups: control group receiving conventional culture, isorhamnetin group treated with 60 μmol/L isorhamnetin, H 2O 2 group treated with 600 μmol/L H 2O 2, and isorhamnetin + H 2O 2 group pretreated with 60 μmol/L isorhamnetin for 12 hours followed by medium replacement and 12-hour treatment with 600 μmol/L H 2O 2. Flow cytometry was performed to detect cellular reactive oxygen species (ROS) levels, transmission electron microscopy to observe mitochondrial ultrastructure, confocal fluorescence microscopy to evaluate mitochondrial membrane potential, real-time fluorescence-based quantitative PCR (qRT-PCR) to determine the mitochondrial DNA copy number, and adenosine triphosphate (ATP) assay kit was used to determine the mitochondrial ATP content. One-way analysis of variance was used for comparisons among multiple groups, and least significant difference- t test for multiple comparisons. Results:Oxidative stress was provoked in HaCaT cells after the treatment with H 2O 2. Compared with the control group, the H 2O 2 group showed significantly increased ROS levels (10 725.0 ± 845.8 vs. 1 708.0 ± 69.4, t = 18.40, P < 0.001), but significantly decreased fluorescence intensity of mitochondrial membrane potential, mitochondrial ATP content, and expression of ND-1 (a characteristic gene of mitochondrial DNA) ( t = 4.58, 4.48, 6.11, P = 0.010, 0.010, 0.003, respectively). After the pretreatment with isorhamnetin followed by H 2O 2 treatment, the isorhamnetin+ H 2O 2 group showed significantly decreased ROS levels (7 640.0 ± 922.7) compared with the H 2O 2 group ( t = 4.27, P = 0.013), but significantly increased fluorescence intensity of mitochondrial membrane potential, mitochondrial ATP content, and expression of ND-1 compared with the H 2O 2 group ( t = 4.59, 4.58, 5.61, P = 0.010, 0.010, 0.005, respectively). Under the electron microscope, the mitochondrial structure was clearer and more complete in the isorhamnetin+ H 2O 2 group than in the H 2O 2 group; there was slight or no swelling of mitochondrial cristae, and no vacuolization of mitochondria in the isorhamnetin+ H 2O 2 group; in addition, autophagosomes engulfing damaged mitochondria were observed in the isorhamnetin+ H 2O 2 group. Conclusion:Isorhamnetin may reduce ROS levels by inducing autophagy, and has a protective effect against the H 2O 2-induced mitochondrial structural and functional damage in HaCaT cells.
5.Clinical validation of the 2012 classification criteria for early rheumatoid arthritisin: a domestic multi-center cohort

Objective To evaluate the value of 2012 classification criteria for early rheumatoid arthritis (ERA),2010 American College of Rheumatology/European League Against Rheumatism (ACR/EULAR) classification criteria,and 1987 ACR classification criteria in the diagnosis of early rheumatoid arthritis (RA).Methods Patients who had at least one swollen and tender joint with disease duration no more than 2 years,and age more than 16 years were enrolled.The patients were diagnosed as RA or other non-RA by 2 experienced rheumatologists.The clinical and laboratory parameters were recorded.The sensitivity and specificity of three RA classification criteria were compared by McNemar test,The areas under the receiver operating characteristic curve (ROC) curve (AUC) of each RA classification criteria were analyzed using MedCalc software.Results Atotal of 310 patients were enrolled in this study,including 182ERA and 128 non-RA.The sensitivity(88.5%) of ERA criteria was much higher than that of the 1987 ACR criteria (45.6%,x2=75.013,P<0.05),and not significantly different with the 2010 ACR/EULAR criteria (91.8%,X2=1.042,P>0.05).The specificity of ERA criteria (91.4%) of 2010 ACR/EULAR criteria (87.5%,x2=1.8,P>0.05) was similar to that of the 1987 ACR criteria (96.1%,x2=3.1,P>0.05).The AUC of ERA criteria was 0.962 [95%CI(0.934,0.980)],which was slightly better than that of the 2010 ACR/EULAR criteria 0.959 [95%CI(0.931,0.978)],Z=0.380,P=0.7038,and much higher than that of the 1987 ACR criteria 0.885 [95%CI (0.845,0.919)],Z=4.517,P<0.01.Conclusion Overall evaluation,the diagnostic value of ERA criteria is better than 1987 ACR and 2010 ACR/EULAR criteria in early rheumatoid arthritis.Compared to 2010 ACR/EULAR classification criteria,ERA criteria is more simple and practical.
