1.Effect of isorhynchophylline on head-shakes behavior and levels of monoamine neurotransmitter in model rats with Tourette syndrome

Objective To explore the effect of rhynchophylline and isorhynchophylline on headshakes behavior and levels of monoamine neurotransmitter in model rats with Tourette syndrome.Methods 40 Wistar rats were randomly divided into DOI-induced head-shakes rats (HSR group),haloperidol group,rhynchophylline group and isorhynchophylline group with 10 in each group.The inhibitory effects of rhynchophylline and isorhynchophylline were estimated by observing the HSR behavior.Dopamine (DA) and 5-hydroxytryptamine(5-HT) in the rat striatum were detected by the enzyme-linked immunosorbent assay (ELISA) method.The 5-HT2A receptor mRNA expression in prefrontal lobe cortex of the rats was measured by real-time PCR.Results Compared with HSR group,the head shakes of the rats in haloperidol group and isorhynchophylline group were significantly decreased(P<0.01),and no change of head-shakes number was observed in rhynchophylline group (P>0.05).There was no significant difference of head-shakes number between the haloperidol group and isorhynchophylline group(P>0.05).Compared with HSR group,DA levels in the rat striatum were significantly decreased in isorhynchophylline group and haloperidol group((152.35± 5.80) μ~L vs (111.19±4.30) μg/L,(152.35±5.80) μg/L vs (126.42±3.17) μg/L,P<0.01),while DA levels in the rat striatum in rhynchophylline group were not changed ((152.35±5.80) μg/L vs (142.71±5.51) μg/L,P>0.05).There was no significant change of 5-HT2A receptor mRNA expression in rat prefrontal lobe cortex in every group(P>0.05).Conclusion Isorhynchophylline may have an inhibitory effect on rats with DOI-induced HSR.Isorhynchophylline may decrease the DA levels in the rat stratum with DOI-induced HSR.Rhynchophylline has no significant inhibitory effect on head-shakes behavior and DA levels in the rat stratum with DOI-induced HSR.
3.Assessment of coronary artery by prospective ECG-triggered 256 multi-slice CT on children with congenital heart diseases

Objective To investigate the image quality and radiation dosage of the prospective ECG-triggered 256 multi-slice computer tomography (MSCT) angiography in accessing coronary artery (CA) on children.Methods Coronary arteries of ninety-three children were evaluated using prospective ECG-triggered 256-MSCT with the same system setting.Seventy-four patients had the heart rate records and a four-point scoring system was applied to study the capability of MSCT in detecting CA.The signal,noise and contrast-to-noise ratio (CNR) were analyzed to investigate the association of image quality with age (n =74).Then volumetric CT dose index (CTDIvol),dose-length product (DLP) and effective dose (E) were utilized to study the association of radiation dosage with age (n =93).Results On 74 patients with heart records,the detection rate for original,proximal,middle,distal and all 11 segments of CA was 100%,97%,92%,78% and 91%,respectively.No influence of age was found on their detection (all P>0.05).E of group A,B,C and D were 1.8,1.3,0.8 and 0.7 mSv,respectively.A strong negative correlation was found between E and age (r =-0.803,P < 0.01).Higher DLP was found on elder patients,though no correlation was found between ages vs.DLP(r =0.124,P > 0.05).No correlation was found between noise vs.age (r =0.041,P > 0.05).Significantly larger signals,CNR and E(t =3.386,2.073,3.825,P < 0.05) were found on younger patients (age < 3 years).Conclusions Prospective ECG-triggered 256-MSCT has considerable performance for the evaluation of coronary arteries on children.However,children under the age of 3 are not recommended for this examinary,but for examination on children ≥ 8-year old,the tube current could be further needed.
4.Relationship between the infection of human herpesvirus 6 and oral squamors cell carcinoma

Objective: To study the relationship between human herpes vi rus 6 (HHV-6) infection and oral squamors cell carcinoma. Methods: The serum anti-HHV-6 antibody titers from 16 cases of oral squamors cell carcinoma (OSCC) and 16 control subjects were detected by indirect immunofluores cence assay. HHV-6 DNA in peripheral blood mononuclear cells was amplified by P CR and the specificity was confirmed by Southern-blot hybridization with an int ernal probe oligonuclotide. An immunohistochemical staining was used to detect H HV-6 antigen in OSCC tissues. Results: Positive expression of s erum HHV-6 IgG antibody was found in all the cases of OSCC and in 12 of the 16 controls. 5.Factors influencing therapy decision in patients with severe pelvic organ prolapse

Objective To investigate the factors influencing therapy decision of surgery or pessary in patients with severe pelvic organ prolapse (POP). Methods Totally 419 cases ofⅢtoⅣdegree POP patients were studied retrospectively. Patients were divided into surgery and pessary groups according to their own choice. Clinical characters were compared such as age, body mass index (BMI), age of onset and disease duration, POP stage, complications. Results 67.5%(283/419) patients were in the surgical group and 32.5%(136/419) patients in the pessary group. Patients in surgical group had higher BMI [(25.1 ± 3.5) versus (23.8±2.6) kg/m2], elder age of onset [(62±12) versus (57±11) years old], longer disease duration [(5± 8) versus (11±11) years] and higher POP staging of middle compartment and less cardiac disease [20.1%(57/283) versus 30.9% (42/136)] than those in pessary group, all had significant difference (P<0.05). Logistic regression analysis on the above factors showed a statistically significant difference between two groups, BMI, disease duration and POP staging of middle compartment were independent factors (OR=1.141, 0.932, 1.389;all P<0.01). Conclusions Patients with higher BMI, higher POP staging of middle compartment and less cardiac disease tended to choose surgery. Patients with younger age of onset and longer disease duration tended to choose pessary. Factors as age, POP staging of anterior and posterior compartment, history of POP surgery, complicated with hypertension and diabetes, showed no influence on treatment choice.
6.The α7 nicotine acetylcholine receptor regulates sensitivity of regular chemotherapeutic agent of cholangiocarcinoma

ObjectiveTo study the effect of α7 ( α7 AChR) agonist nicotine on regulating sensitivity of regular chemotherapeutic agent in cholangiocarcinoma cells,and explore the possible target.MethodsThe effect of nicotine and α-BTX pretreatment on the survival ability of cholangiocarcinoma cells was investigated when applied with 5-FU by using MTT and Flat cloning formation experiment.ResultsApplied with 5-FU,in various con centrations nicotine stimulating group( 10-3 g/L,10-4 g/L,10-5 g/L ),the survive rate of QBC939 was 128%,124%,118%,while that in α-BTX stimulating group and combined stimulation group was 92%,94%,93%,92%,respectively.The cloning formation ability of nicotine- stimulating group (6.2 ± 0.40) was significantly higher than α- BTX stimulating group (3.2 ± 0.20 ),combined stimulation group ( 3.2 ± 0.20 ) and control group ( 3.4 ±0.33).ConclusionNicotine can prevent chemotherapy-induced apoptosis,and improve cholangiocarcinoma cell survival via α7 nicotine acetylcholine receptor in vitro.

A growing number of studies show that drug and non-drug means can be activated phosphatidyl inositol-3-kinase serine/threonine kinase(PI3K/Akt signaling pathway)in the beginning of reperfusion, and make its downstream fork protein transcription factor O3a (FoxO3a) phosphorylation, and then promote cell survival and reduce the apoptosis so as to alleviate the symptoms of myocardial ischemia.In this paper,the role of PI3K/Akt and FoxO3a effector in myocardial ischemia was reviewed, and illustrated the role of PI3K/Akt/FoxO3a signaling pathway in myocardial ischemia.
9.Comparison of casting precision of two kinds of commercially available gypsum-bonded investment with different setting and thermal expansion characteristics.

OBJECTIVETo investigate expansion characteristics of two kinds of commercial investment (Cristo balite Micro II and Cristo Quick), and their effects on cast accuracy of a dental Ag-Pd casting alloy.
METHODSTwo kinds of investment were mixed at the ratio of 1:3. The setting and thermal expansion curves of the investment were recorded according to the requirements of ISO7490 and expansion rate calculated, respectively. Plate-like wax-patterns with pre-marked points were invested in casting rings (lined with ceramic fiber liners with thickness of 0.45 mm). Castwell M.C dental alloy were melted with gas flame and cast with a centrifugal casting machine. The differences between the castings and the wax patterns at corresponding points were calculated to represent the dimensional changes of the investment.
RESULTSThe setting, thermal and total expansion rates were 0.68, 1.18, 1.86 for Cristo balite Micro II, and 0.94, 1.03, 1.97 for Cristo Quick. The setting and thermal expansion rates were statistically significant between two kinds of investment (P < 0.05), while not for the total expansion rates (P > 0.05). Effect of different investment on the precision of castings was not statistically significant (P > 0.05).
CONCLUSIONWithin the scope of this study, gypsum-bonded investment with similar total expansion rates but different expansion characteristics has no effect on the casting precision of Ag-Pd dental alloys.

10.The expression and activity of caspase-8 in the process of vitamin E succinate-induced apoptosis in human gastric carcinoma SGC-7901 cells.

OBJECTIVETo investigate the role of caspase-8 in vitamin E succinate (VES)-induced apoptosis in human gastric carcinoma cells.
METHODSThe expression and activity of caspase-8 were measured by the Western Blot method with fluorescence and DAPI staining.
RESULTSVES treatment induced the expression and activity of caspase-8 in a dose-dependent manner; the apoptosis ratio induced by VES was significantly decreased in SGC-7901 cells treated with caspase-8 inhibitor.
CONCLUSIONCaspase-8 plays an important role in SGC-7901 cell apoptosis induced by VES.
