6. Protective mechanism of Qizhi hypoglycemic tablets on eye diseases in streptozotocin-induced diabetic rats
Journal of International Pharmaceutical Research 2019;46(10):761-768
Object: To investigate the protective effect of Qizhi hypoglycemic tablets on eye diseases in strepto- zotocin-induced diabetic rats and explore its possible mechanism. Methods: Streptozotocin was used to prepare diabetic model in rats. The diabetic model rats were given Qizhi hypoglycemic tablets(780 mg/kg),metformin hydrochloride tab- lets(200 mg/kg)or glibenclamide tablets(1.5 mg/kg)via intragastric administration once a day for three months, and the dynamic changes in eye diseases were investigated. The examined index included blood glucose level, lens opacity, serum levels of aspartate aminotransferase(AST)and alanine aminotransferase(ALT),retina expression of collagen, matrix metalloproteinase-2(MMP-2)and tissue inhibitor of matrix metalloproteinase-1(TIMP-1). Network pharmacology was used to analyze protective mechanism. Results: Compared with the normal group, the model rats showed significant hyperglycemia(P<0.01),polydipsia,polyphagia and polyuria(P<0.01)as well as the decreased body mass(P<0.01), increased serum ALT and AST level(P<0.01),decreased the retinal Collagenexpression(P<0.01),decreased: TIMP-1(P<0.01)and increased lens opacity(P<0.01). Compared with the model group, metformin hydrochloride tab- lets and glibenclamide tablets could improve hyperglycemia(P<0.01),polyphagy,polyuria and emaciation(P<0.01), but had no significant effect on the degree of retinal turbidity(P>0.05). Compared with the model group,Qizhi hypogly- cemic tablets also had hypoglycemic effect(P<0.05),which could improve hyperglycemia, polyuria and emaciation (P<0.01),significantly reduce the degree of retinal turbidity(P<0.01)and up-regulate the expression of retinal Colla- genand TIMP-1(P<0.01). According to the network pharmacology analysis,Qizhi hypoglycemic tablets could regu- late the cell proliferation and apoptosis via VEGF and MAPK signals. Conclusion: Qizhi hypoglycemic tablets could im- prove the ophthalmic disease in diabetic rats, which might be a promising medicine for the treatment of diabetic ophthal- mic disease.
7. Studies on high-efficiency loading, sustained-release and permeability as transdermal delivery of graphene oxide to docetaxel composites
Chinese Traditional and Herbal Drugs 2019;50(12):2830-2834
Objective: To synthetize the new-type GO-DEX-β-CD/DOC and Fe3O4/GO-Na/DOC inclusion compound, and study its high-efficiency loading, sustained-release and permeability as transdermal delivery for docetaxel (DOC) composites. Methods: The concentration of DOC was determined by high efficiency liquid chromatography. The high-efficiency loading, sustained-release and permeability as transdermal delivery of GO-DEX-β-CD/DOC and Fe3O4/GO-Na/DOC were studied, and the encapsulation efficiency and drug loading of them were determined by centrifugation. The GO-DEX-β-CD/DOC and Fe3O4/GO-Na/DOC were applied onto the female mice skin in vitro and in vivo to develop the permeability of them. Results: The encapsulation efficiency and drug loading of Fe3O4/GO-Na/DOC were higher than GO-DEX-β-CD/DOC, and its slow release property and permeability as transdermal delivery were better. The results showed that the accumulation permeation amount was (22.512 ± 0.715) μg after Fe3O4/GO-Na/DOC being applied over 90 h, DOC concentration in skin reached a peak at 15 min by the application of Fe3O4/GO-Na/DOC. After 5 h of administration, DOC concentration in the blood of female mice reached (76.886 ± 1.232) μg/mL. Conclusion: The preparation techniques of Fe3O4/GO-Na/DOC was feasible with better sustained release and transdermal effect, which had a promising application prospect.
8.Effect of maternal hypothyroidism on fetal glucose metabolism
Chinese Journal of Endocrinology and Metabolism 2015;(6):561-564
[Summary] The theory of fetal origins of adult disease (FOAD) is now widely accepted by researchers who hold the opinion that adult degenerative and metabolism diseases have close relationship with the environment of fetal development inside and outside the womb. Some studies have proved that maternal hypothyroidism can negatively affect the glucose metabolism of their offsprings. However, the whole mechanism is not clear yet. Insufficient thyroid hormone during pregnancy was proved to slow down the formation of fetal pancreatic cytoskeleton, to decrease the proinsulin gene transcription, and to modulate series of cytokines and enzymes which are related to glucose dependent insulin secretion. Thyroid hormone receptor is also considered to be partially responsible for the relation between low thyroid hormone and β cell insufficiency. However, more studies in vivo should be carried out to prove this hypothesis. Epidemiologic studies have suggested that type 2 diabetes and low birth weight can be different phenotypes of the same genotype. The definite mechanism of maternal hypothyroidism in influencing fetal β-cell function should be studied by further investigation.
