1.Value of ADAM12-S in early diagnosis of ectopic pregnancy of pregnant women

Objective To determine the values and variation tendency of a disintegrin and metalloprotease 12-S(ADAM12-S)of early pregnancy, and discuss the value of ADAM12-S to the ectopic pregnancy in the first trimester.Methods 150 early pregnancy cases (from the 5th to 10th week of pregnancy)were selected in Huanggang Women &Children's Healthcare Hospital.There were 102 cases of normal pregnancy,48 cases of ectopic pregnancy.The AutoDELFIA immunoassay platform was used to measure the value of ADAM12-S in the serum.Results Of the 102 normal pregnancies, the values of ADAM12-S in 5th-10th week of pregnancy linearly increase with gestationalage.The correlation coefficient was 0.9217 (P<0.05).While the values of ADAM12-S in the ectopic pregnancy group were significantly lower than the normal pregnancy group and not linearly increased with gestationalage.The correlation coefficient was 0.2817.There was statistically significant difference between the two groups (P<0.05).Conclusion The values of ADAM12-S is stable in the first trimester of normal pregnancy,while it is much more lower in abnormal pregnancy.ADAM12-S has great significance in the diagnosis of ectopic pregnancy of the first trimester.
2.Study on the activation effects to T lymphocytes by gastric cancer cell-dendritic cell fusion vaccines

Objective To study the activation effects to cytotoxic T lymphocytes by gastric cancer cell-dendritic cell(DC) fusion vaccines and to provide theoretical data for biological therapy of gastric cancer based on(anti-tumor) fusion vaccines.Methods(1)Peripheral blood mononuclear cells from gastric cancer patients were separated and co-cultured with granulocyte macrophage colony stimulating factors,interleukin-4 and tumor necrosis factor-? to generate mature dendritic cells.(2)The dendritic cells and SGC7901 cells were fusioned by use of polyethylene glycol,and the pure fusion cells were screened out and cultured with HAT and HT(selective) culture systems.(3)The ability of fusion cells to activate the cytotoxic T lymphocytes was(investigated) by using MLA method.Results(1)Mature dendritic cells were gained from gastric cancer(patients)′ peripheral blood mononuclear cells by co-culturing with granulocyte-macrophage colony stimulating factors,interleukin-4 and tumor necrosis factor-?.(2)Dendritic cells and SGC7901 cells could be fusioned by PEG and could get pure fusion cells by culture with HAT/HT selective culture systems.(3)The fusion vaccines could strongly activate cytotoxic T lymphocytes,and there were significent differences between the(fusion) vaccine group and the control group.(P
3.Study on the induction culture and phenotypes of dendritic cells originating from peripheral blood of gastric cancer patients

Objective To analyse the morphological and phenotypic characteristics of dendritic cells obtained by induction of peripheral blood monocytes of gastric cancer patients and provide an experimental basis of (dendritic) cells for the biological treatment of gastric cancer. Methods Peripheral blood monocytes of gastric cancer patients were co-cultured with GM-CSF, IL-4 and TNF-? to obtain the dendritic cells. The (morphological) characteristics of the dendritic cells were observed by light and electron microscopes and the (phenotypes) were studied by flow cytometry. Results Mature dendritic cells were oblained seven days after (induction) culture. These cells showed numerous typical dendritic protuberances on their surface.The (phenotypes) of the cells increased gradually with their maturation and reached stable levels seven days later. Conclusions Mature dendritic cells could be obtained from peripheral blood monocytes of gastric cancer (patients) after induction culture with GM-CSF,IL-4 and TNF-?. The obtained cells had typical morphological and phenotypic characteristics. These cells could be used for further research in the biological treatment of (gastric) cancer.
4.The phenotype changes of SGC7901 gastric cancer cell-dendritic cell fusion vaccine

Objective To obtain SGC7901 gastric cancer cell-dendritic cell fusion vaccines by cell fusion technology. The phenotype changes of the fusion cells were detected to provide experimental data for the following research work. Methods The SGC7901 gastric cancer cells and dendritic cells were induced to be fused by PEG and the pure fusion cells were obtained by selective culture with the HAT/HT culture system. The phenotype changes of the fusion cells were detected by flow cytometer. Results The fusion cells had special morphologic characteristic and the phenotypes of the fusion cells were remarkably higher than the parental dendritic cells. Conclusion The SGC7901 gastric cancer cell-dendritic cell fusion cells kept special morphologic characteristic and their phenotypes were significantly higher than the parental dendritic cells, which may be the cellular base for their stronger biological effects.
5.Cytotoxic effects of gastric cancer cell-DC fusion vaccines

Objective To study on the cytotoxic effects in vivo and in vitro of the T lymphocytes which were activated by the DC-gastric cancer cell fusion vaccines. Methods The mononuclear cells were separated from the peripheral blood of gastric cancer patients and co-cultured with granulocyte-macrophage colony stimulating factors, interleukin-4 and tumor necrosis factor-? to generate mature dendritic cells. The human gastric cancer SGC7901 cells and the dendritic cells were fusioned by using polyethylene glycol, and the pure fusion cells were screened out by culturing with HAT and HT selective culture systems. The mixture of SGC7901 and DC without fusion and the T lymphocytes served as controls. The ability of fusion cells activated by T lymphocytes to kill SGC7901 gastric cancer cells was investigated by MTT in vitro. The anti-tumor effects were evalutated by detecting the growth of the new planted tumors in the nude mice, the apoptosis and proliferation of the planted cancer cells, the pathological changes of the tumor tissues and the prohibitive rate for planted tumors before and after the application of the fusion cell vaccine. Results The mature dendritic cells were obtained from peripheral blood mononuclear cells of gastric cancer patients successfully. Dendritic cells and SGC7901 cells were fusioned and the pure fusion cells were got. Objective To provide experimental basis for the study of SGC7901 gastric cancer cell-dendritic cell fusion vaccine. Methods The fusion of SGC7901 gastric cancer cells and dendritic cells was induced by PEG. We obtained pure fusion cells by selective culture with HAT/HT culture systems. The growth curve characters, clone abilities, abilities to irritate T lymphocytes and the ability to grow were detected. Results The fusion cells had weaker proliferative abilities and clone abilities with increased abilities to irritate T lymphocytes. The CPM value of T lymphocytes were 15083?231、 9608?83、 4214?135、 3020?28 respectively when they were co-cultured with the fusion cells on different proportions. Conclusion SGC7901 gastric cancer cell-dendritic cell fusion cells keep strong abilities to irritate T lymphocytes while inable to grow into new planted tumors in immunodeficiency animals.

8.Identification of the related substances in docetaxel injection by LC-MS/MS.

The related substances in docetaxel injection were identified by LC-MS/MS. Ethyl acetate was used to extract the injection to remove the pharmaceutical excipients. HPLC separation was carried out on a Hedera ODS-2 column (150 mm x 2.1 mm, 5 microm) with a mobile phase consisting of acetonitrile - 0.1% acetate acid aqueous solution (40: 60). Electrospray ionization source was set in the positive mode for the LC-ESI-MS/MS, and the ion monitoring modes were full scan and product ion scan. According to the mass spectra of the related substances, the fragment profiles were explained, and the chemical structures were elucidated. Docetaxel and its main related substances were well separated. Nine related substances in docetaxel injection were detected by LC-MS/MS. Their chemical structures were proposed, and four of them were identified in the docetaxel injection for the first time. The established LC-MS/MS method is effective in the separation and identification of the related substances in docetaxel injection. The test results are useful for its quality control.

9.Inhibitive effects of gastric cancer cell-dendritic cell fusion vaccine on tumor cell proliferation cycle.

OBJECTIVETo study the inhibitive effects of gastric cancer cell-dendritic cell fusion vaccine on tumor cells of proliferation cycle.
METHODSPeripheral blood mononuclear cells were separated from gastric cancer patients and co-cultured with granulocyte macrophage colony stimulating factors(GM-CSF), interleukin-4(IL-4) and tumor necrosis factor-alpha(TNF-alpha) to generate mature dendritic cells. The dendritic cells and SGC7901 cells were fused by polyethylene glycol, and the pure fusion cells were screened out by selective culture systems. The inhibitive effects of gastric cancer cell-dendritic cell fusion vaccines on tumor cell proliferation cycle in vivo and in vitro were detected by flow cytometry.
RESULTSTreated with the fusion vaccine in vitro, the percentages of G(0)/G(1), S and G(2)/M cells of tumor cells were (76.77+/- 4.38)%, (16.50+/- 2.90)% and (6.73+/- 1.59)% respectively. There were significant differences in the percentages of different cell cycle tumor cells between the tumor cells treated with the fusion vaccine and those co-cultured with dendritic cell or T cells alone(P< 0.01). The proliferative index of the tumor cells treated with the fusion vaccine was 23.34+/- 3.51, significantly lower than those co-cultured with dendritic cell and controls (P< 0.05).
CONCLUSIONSFusion vaccines can affect cell cycle of the tumor cells, thus inhibit tumor cell proliferation and growth.

10.Health risk factors and presenteeism: a look into employees at two enterprises in China

Objective To analyze the effects of health risk factors on presenteeism in enterprise employees.Methods A hospital in Jinan of Shandong province and a publishing enterprise in Beijing were selected as target settings of this cross-sectional study. Questionnaire survey was carried out to collect information on demographic data,health condition and presenteeism,and multiple logistic regression was used to analyze the influence of health risk factors on labor-related presenteeism.Results Insufficient physical activity was found in 42% percent of the participants,and those with heavy-loaded pressure,overweight or obesity,high blood pressure and unhealthy diet habit accounted for 46%,41%,40% and 34%,respectively. Proportions of employees with presenteeism varied with age ( x2 =10.1665,P =0.0377),occupation ( x2 =35.3579,P < 0.05 ) and education level ( x2 =50.5377,P < 0.05 ).Female employees ( P =0.0323,OR =1.728,95% CI 1.047 to 2.850),graduates ( P < 0.05,OR =6.159,95% CI 2.510 to 15.109),overweight or obesity individuals ( P =0.0076,OR =1.673,95% CI 1.147 to 2.44) and adults with poor self-perception of health ( P =0.0049,OR =2.284,95% CI 1.285 to 4.060)seemed to be more likely to be presenteeism.Conclusions Overweight or obesity and poor self-perception of health may be associated with labor-related presenteeism.
