1.Application and effect assessment of the model of clinical nursing path management

Objective To explore the application effect of the model of clinical nursing path management in clinical nursing. Methods A comparative study was conducted of the clinical nursing paths respectively with regard to patients who had received thyroid adenoma resection and with regard to patients who had received hysteromyomectomy. Results In the experimental group involving clinical nursing path management, the average length of stay was shortened, pre operation days were reduced, and the average hospitalization expenses were lowered while the satisfaction rate of patients with thyroid adenoma rose by 2.18%, reaching 98.96% and that of patients with hysteromyoma rose by 2.87%, reaching 99.55%. At the same time, clinical nursing path management also enhanced the job satisfaction of nurses. Conclusion It is viable to adopt the model of clinical nursing path management in clinical nursing in China.
2.Micro-invasive evacuation for treatment of cerebral hemorrhage

Objective To evaluate the dfect of micro-invasive evacuation for treatment of cerebral hemorrhage.Methods Eighty patients with cerebral hemorrhage were treated by the micro-invasive surgery.Results The hematoma was evacuated between 1~5 days in 55 cases,7 dyas in 24 cases and 14 days in 1 case.The total effeetive rate was 81%.Seyen cases died.Modified rankin scale(MRS)0~1 was found in 21 cases,MRS 2~3 was found in 44 cases,MRS 4~5 in 8 cases.Conclusion Compared with the medical conservative treatment,the microinvasive evacuation for treatment of brain hemorrhage could significantly reduce patients'hospitalization time,morbidity and mortality.At the same time indication are relatively extensive,complications are less.
3.The expression of CIP2A and c-Myc and their correlation analysis in cervical carcinoma tissues

Objective To investigate the expression of CIP2A and c‐Myc and their correlation in cervical carcinoma .Methods We detected CIP2A and c‐Myc expression using immunohistochemistry in 72 samples of cervical carcinoma and 12 samples of normal cervix tissues .Correlation between proteins and clinicopathologic features and relation between CIP2A and c‐Myc expression were analyzed .Results The positive expression of CIP2A and c‐Myc in cervical carcinoma tissues were 52 .8% and 56 .9% ,respec‐tively .While their positive expression in normal cervix tissues were 8 .3% and 25 .0% ,respectively .The differences had statistical significance(χ2 =8 .169 ,P=0 .004 ;χ2 =4 .208 ,P=0 .040 ,respectively ) .Clinicopathological analysis suggested that CIP2A and c‐Myc protein expression were associated with histopathological differentiation and clinical stage(P<0 .05) in cervical carcinoma ,but the protein expression was not related to age ,lymph node metastasis and pathological type .CIP2A was significantly positive correla‐ted with c‐Myc protein in cervical carcinoma(r=0 .673 ,P=0 .001) .Conclusion The high‐expression of CIP2A is correlated with malignant clinicopathologic characteristics ,and CIP2A is positively associated with c‐Myc ,suggesting that CIP2A may promote tumor initiation and development through maintaining c‐Myc protein in cervical carcinoma .
4.The effects and mechanisms of Gankyrin silencing on reversing the cisplatin resistance of human ovarian cancer SKOV3/DDP cell line

Background and purpose: Ovarian cancer is the common gynecological cancer, and the drug resistance of anti-tumor drug was one of major reasons for therapy failure, some studies considered that there is a closed relationship between Gankyrin and drug resistance. In this study, we investigated the effects and mechanisms of Gankyrin silencing on reversing the cisplatin resistance of ovarian cancer drug-resistant SKOV3/DDP cell line. Methods:The expression of Gankyrin in SKOV3 and SKOV3/DDP cells was measured by real-time PCR assay, MTS assay was employed to determine the effect of Gankyrin on SKOV3/DDP sensitivity to cisplatin, apoptosis rate and intracellular concentration of rhodamine-123 (Rh-123) were determined by lfow cytometry, the expression of multi-drugs resistant protein MDR1, Caspase-3/8, Survivin and Bcl-2 were determined by Western blot and real-time PCR. The phosphorylation of AKT and expression of p53, NF-κB and PTEN were analyzed by Western blot assay. Results:The expression of Gankyrin was increased in SKOV3/DDP cells, Gankyrin silencing was able to increase the cisplatin sensitivity of SKOV3/DDP. Before and after gene silencing, the reverse folds (RF) to cisplatin were 1.81 and 2.45, respectively, the intracellular levels of Rh-123 were 1.73 and 2.42 fold, the apoptosis rates were 2.23 and 4.23 fold,the expressions of MDR1, Survivin and Bcl-2 were downregulated, the mRNA expressions of MDR1 were 62.8%and 21.6%, the mRNA expressions of Survivin were 24.5%and 10.3%, the mRNA expressions of Bcl-2 were 47.5%and 18.4%, the levels of Caspase-3/8, p53 and PTEN were elevated, phosphorylation of AKT and expression of NF-kB were downregulated compared with control group. Conclusion:Gankyrin silencing was able to reverse the cisplatin resistance of SKOV3/DDP cells by inhibiting the drug eflfux and promoting cell apoptosis, the PTEN/AKT/NF-κB/p53 may be the key pathway.
5.RESEARCH ON SPONTANEOUS DIFFERENTIATION OF STEM CELLS AND RELATED GENE EXPRESSION PROFILE

Objective To investigate the expression of endometrial developing-related genes,the endometrium-like structure and cells during spontaneous differentiation of hESCs. Methods (1) Embryoid bodies were cultured in suspension for 14 days,fixed in 10% neutral-buffered formalin and embedded in paraffin.Estrogen receptor(ER) was detected by immunocytochemical staining.(2) hESCs were cultured in a 35 mm plastic dish and differentiated spontaneously for 10 days.The expression of ER and Vimentin/Keratin was detected by double immunofluorescence histochemistry.(3) hESCs were cultured in a 35 mm plastic dish and differentiated for 5 days.The expressions of endometrial developing-related genes including Wnt4,Wnt7a,Wnt5a,Hoxa10,Hoxa11 and ER were detected by RT-PCR. Results (1) The endometrium-like structures were detected in 14-day-old EBs and some were positive for ER staining.(2) Some spontaneously differentiated cells from hESCs for 10 days were positive for ER together with Vimentin/Keratin.(3)Wnt7a,Wnt4,Hoxa10,Hoxa11 and ER were detected by RT-PCR in hESCs which were differentiated spontaneously for 5 days.Conclusion During the spontaneous differentiation of hESCs,some cells are liable to differentiate into endometrial stem cells and endometrium-like cells.However,further identifications of the whole development process are needed to be done in the future.
6.Construction of eukaryotic expression system of a cell surface molecule——human CD40 ligand

Objective To construct human eukaryotic expression system expressing T cell surface molecule CD40L and identify the expression product in SCCL cell line h446. Methods The expression vector pcDNA3.1-CD40L was constructed by recombinant DNA technology, then transfected into h446 cell line of SCCL by lipidosome methods, and the expressed membrane protein was identified by flow cytometry. Results The pcDNA3.1(+)-CD40L was constructed successfully and the coding region was inserted into the vector correctly. The membrane protein was observed by flow cytometry on transfected h446 cell line of SCCL. Conclusion Human CD40L was expressed successfully in eukaryotic system, which is very important in the anti-tumor vaccination study with a recombinant human CD40L in clinic.
7.The relationship between the degree of intracranial and extracranial arterial stenosis and the plasma level of homocysteine in patients with ischemic cerebrovascular disease

Objective To study the relationship between the degree of intracranial and extracranial arterial stenosis and the plasma level of homocysteine ( Hcy ) in patients with ischemic cerebrovascular disease .Methods 96 patients with ischemic cerebrovascular disease were checked by DSA .The distribution of stenosis in the studied cerebral arteries was analyzed .According to the degree of stenosis ,all 96 patients were divided into some groups ,at the same time Hcy level was detected ,and the correlation between Hcy level and intracranial and extracranial artefial stenosis were analyzed .Results 35 of the 96 patients had normal findings in angiography ,and 61 cases had occlusive lesions of different degrees .The mean plasma Hcy concentration in patients with intracranial and extracranial arterial stenosis was 19.26 (14.79~26.13)μmol/L,which was significantly higher than 11.60(8.8~15.11)μmol/L in the patients with no arterial stenosis (P<0.05).There was a significant positive correiation between Hcy level and cerebral artery stenosis degree(r=4.126,P<0.05).Conclusion The plasma Hcy level was positively correlated with intracranial and extracranial arterial stenosis in patients with ischemic cerebrovascular disease ,incidence and nar-row-degree of cerebral artery stenosis are all along with the increasing of Hcy level .
8.Effective analysis of organ donation intervention in community residents

Objective We tried to explore the effects of organ donation intervention on organ donation knowledge,attitude and willingness of community residents and analyze its promotive factors,thereby to provide valuable basis to increase organ donation rate.Methods 354 community residents in Hengyang city of Hunan province were randomly collected and received organ donation intervention.The scores of organ donation knowledge,attitude and willingness,living organ (bone marrow and kidney) donation willingness rate,and supporting relative organ donation status were compared before and after intervention.We also investigated the factors that could enhance organ donation.Results In comparison with before intervention,organ donation knowledge and willingness scores(t value was 4.46 and 5.13),bone marrow donation willingness rate (x2 value was 9.27) and supporting relative organ donation proportion(x2 value was 10.74) were increased,but organ donation attitude score was decreased in community residents.However,organ donation intervention had no effect on kidney donation willingness rate.Community residents recognized that economic compensation (43.68%),organ distributing fair (23.85%) and consummate organ donation regime (11.21%) were the most important to facilitate organ donation.Conclusions Organ donaton intervention can notably improve organ donation knowledge,attitude and willingness of community residents.Moreover,giving determinate economic compensation,ensuring organ distributing fair and ameliorating organ donation regime may play key roles in relieving current organ shortage crisis.

10.Clinical Observation of Oxytocin Combined with Compound Leonurus japonicus Oral Solution in the Treat-ment of Hemorrhage after Artificial Abortion

OBJECTIVE:To observe therapeutic efficacy of Compound Leonurus japonicus oral solution in the treatment of hemorrhage after artificial abortion and its effect and safety on normal menstruation recovery. METHODS:100 cases of artificial abortion selected and randomly divided into treatment group and control group,with 50 cases in each group. Control group re-ceived oxytocin intramuscularly after surgery,10 U/time,12 h/time,for 3 times;treatment group was additionally given Com-pound L. japonicus oral solution,2 piece/time,3 times/d,for 5 days,on the basis of control group. Metrorrhagia duration,hemor-rhage amount,normal menstruation recovery and duration of abdominal pain and ADR were all observed in 2 groups. RESULTS:The metrorrhagia duration [(5.38±0.95)d] of treatment group was significantly lower than that [(8.58±1.32)d] of control group, with statistical significance(P<0.05);hypomenorrhea cases(24 cases)of treatment group was higher than those(9 cases)of con-trol group;menorrhagia cases(8 cases)was lower than those(24 cases)of control group;with statistical significance(P<0.05). The normal menstruation recovery time [(27.34±3.87)d] of treatment group was significantly shorter than that [(40.13±2.58)d] of control group,with statistical significance(P<0.05);the duration of abdominal pain [(2.85±0.89)d] in treatment group was sig-nificantly shorter than [(5.02 ± 1.33)d] in control group,with statistical significance (P<0.05).Obvious ADR was not found in both groups. CONCLUSIONS:Compound L. japonicus oral solution is effective in the treatment of hemorrhage after artificial abor-tion,can significantl shorten metrorrhagia duration and hemorrhage amount,recover normal menstruation early,improve the symp-toms of abdominal pain after operation and is easy to take and good safety.
