1.An in vitro study of trophic activity on facial motoneuron by soluble substances derived from Schwann cells of rats

Objective: To compare trophic effects of soluble substances derived from Schwann cells (SC) of neonatal and Wallerian degenerating segments of rats on facial motoneuron (FMN) cultures. Methods: Serum-free conditioned media of Schwann cell cultures (SC-CM) from facial and sciatic nerves of neonatal and Wallerian degenerating segments in adult rats were individually collected and concentrated by ultra-filtration with molecular weight cut-off at 30 000 and 10 000. The growth activities of FMNs in vitro were determined by means of MTT assay under the condition of serum-free medium added with different components of concentrated SC-CMs (SC-CMCs). The absorbance values were then statistically analyzed. Results: Survival and growth rate of FMN cultures in four kinds of SC-CMCs were significantly higher than that in media both with serum and non-serum and the difference between SC-CMCs was not statistically significant (P>0.05). Neurotrophic molecules were predominantly protein or peptide components with relative molecular weight larger than 30 000 and their trophic activity was positively related to total protein concentration in SC-CMCs. Conclusion: There were soluble trophic molecules with relative molecular weight larger than 30 000 for survival and neurite growth of FMN cultures in media with SC-CMCs derived from facial and sciatic nerves of neonatal rats and with SC-CMCs derived from Wallerian degenerating facial and sciatic nerves of adult rats. It might be reasonable to choose SC from sciatic nerves of rats on account of the findings from SC cultures on facial motoneurons.
2.Effects of BUSHEN YIZHI DECOCTION on interspace explore learning and memory in rat model with Alzheimer's disease

Object To observe the improving effects of BUSHEN YIZHI DECOCTION (BSYZD) on interspace explore learning and memory in rat model with Alzheimer's disease. Methods Eighty 15-month Wistar rats were induced by ip D -galactose for four weeks and injection of basal nucleus of Meynert with ibotenic acid (IBO) to make AD model, then randomly divided into AD model group, Hup-A treated group, BSYZD (high dose, 12 g/kg?d) treated group and BSYZD (low dose, 6 g/kg?d) treated group, and also normal aged and young groups. After treating for four weeks, Morris water maze was used to assess the improvement of rat interspace explore learning and memory. Results In the interspace explore experiment, the significant differences were observed between the model, Hup-A treated groups and normal aged, young groups (P0.05). Conclusion BSYZD possesses a certain preventive effects on interspace explore learning and memory in AD rat model.
3.Epidemiological survey of oral submucous fibrosis in Xiangtan City, Hunan province, China

Objective To understand oral submucous fibrous (OSF) because the hair cause of disease and the quantitative corresponding measures,do a good job in OSF prevention and control work.Methods The quantitative cluster sampling,according to the diagnostic criteria of the development of the Pindborg,yuhu in Xiangtan city of different types of 57 units of 11 046 people to chew areca cause OSF epidemiological investigation.Results OSF 335 diagnosis example,the prevalence rate of 30.33‰,4 cases were oral cancer,oral cancer coexist rate was 11.94‰; All OSF patients had a history of betel nut,no chewing betel nut (containing the cigarette,wine,and spicy aficionados) were not found in patients with OSF; Of OSF prevalence in the chewing hobby was no differences in sex and age in the crowd; OSF prevalence of high and low with length of fixed number of year of average chewing betel nut dose and chewing betel nuts were closely related(r =0.28828,P < 0.01) ; OSF prevalence was different from eating betel nut additive that had a very significant difference.Different hobbies compatibility with standardized test,7 incidence group had 6 group without significant difference,but people only eat chili can (in the control group,1 329) and outsiders no OSF patients (control group,698).Conclusions Survey results confirm that chewing betel nut is the main factor of Xiangtan people of OSF,and OSF carcinoma prevalence is lower than abroad.
4.Efficacy and safety of lamivudine in the patients of hepatitis B virus-associated glomerulonephritis treated by prednisolone with leflunomide

Objective To investigate the efficacy and safety of lamivudine in the patients of hepatitis B virus-associated glomerulonephritis (HBV-GN) treated by prednisolone with leflunomide. Methods 41 HBV-GN patients treated by prednisolone and leflunomide were enrolled in this study. According to the presence of the indications of antiviral treatment, the patients were divided into the prevention group and the treatment group received lamivudine treatment, and another 15 patients with chronic hepatitis B were treated with lamivudine as the control group. The biochemical, virological and serological responses during the process of the treatment and the serum levels of IFN-γ and IL-4 were determined. Results No significant differences of biochemical, virological and serological responses in the prevention and treatment groups compared with the control group at 48 weeks after treatment. Levels of the IFN-γ and IFN-γ / IL-4 decreased significantly in the prevention group and treatment groups compared with the control group (P < 0.05, respectively). No serious adverse reactions occurred in all patients. Conclusion The antiviral treatment of lamivudine lead to good curative effect and security in HBV-GN patients received hormones combined with leflunomide treatments, but leflunomide with prednisolone inhibited the production of IFN-γ and removal of hepatitis B virus.
5.Effects of Bushen Yizhi Decoction on Somatostatin-like Neurons in Hippocampal Gyrus of Rat Models with Alzheimer's Disease

Objective To observe the effects of Bushen Yizhi Decociton (BYD) on somatostatin-like (SS-like)neurons in hippocampal gyrus of rat models with Alzheimer's disease(AD). Methods Fifteen-month_old AD rat models were established by intraperitoneral injection of D-galactose for 4 weeks combined with ibotenic acid injection into bilateral nucleus basalis of Meynert. AD model rats were randomly allocated to AD model group(Group C), Hup-A treatment group(Group D) and BYD treatment groups (Group E and Group F,treated with high dosage and low dosage respectively), and 10 normal aged rats (Group B)and 10 normal youth rats (Group A)served as the normal control groups. The methods of immunohistochemistry and dig_labeling c-DNA probe in situ hibridization were used to detect the number of SS-like immunoreactive positive and SS mRNA expressed positive neurons in hippocampal CA1 and CA3 fields and the dentate gyrus.
