1.Characterization of gene expression and adipokine secretion during differentiation of primary cultured human omental preadipocytes

Stromal preadipocytes from human omental adipose tissues were isolated with enzyme-digesting method,and cultured for inducing differentiation.Preadipocyte factor-1 was abundant during preadipoeyte stage,whereas lipoprotein lipase and PPARγ2 mRNA were rapidly up-regulated at early stage of differentiation and CCAAT/enhancer binding protein expression was increased rather late.In addition,resistin expression and secretion was primarily detectable in preadipocytes,leptin was gradually increased during adipogenesis,whereas adiponectin appeared specifically in mature adipocytes.Insulin-like growth factor-Ⅰ secretion was remarkably detected in both preadipocytes and mature adipocytes.
2.Relation between Body Height and Combined Length of Manubrium and Mesosternum of Sternum Measured by CT-VRT in Southwest Han Population

Objective To establish the linear regression equation between body height and com bined length of manubrium and mesosternum of sternum m easured by CTvolum e rendering technique (CT-VRT) in southw est H an population. Methods One hundred and sixty subjects, including 80 m ales and 80 fem ales w ere selected from southw est H an population for routine CT-VRT(reconstruction thickness 1 m m ) ex-am ination. The lengths of both manubrium and mesosternum w ere recorded, and the com bined length of manubrium and mesosternum was equal to the algebraic sum of them . The sex-specific linear regression equations between the com bined length of manubrium and mesosternum and the real body height of each subject w ere deduced. Results The sex-specific sim ple linear regression equations between the com bined length of manubrium and mesosternum (x3) and body height (y) w ere established (m ale:y=135.000+2.118x3 and fem ale:y=120.790+2.808x3).Both equations show ed statisticalsignificance (P<0.05) w ith a 100% predictive accuracy. Conclusion CT-VRTis an effective m ethod for m easurem ent of the index of sternum . The com bined length of manubrium and mesosternum from CT-VRTcan be used for body height estim ation in southw est H an population.
3.Application of~(125)I seed permanent plantation in osseous metastases

Objective To evaluate the value of ~(125)I permanent plantation in treating osseous metastases.Methods Twenty-two patients with osseous metastases were accepted radioactive seeds ~(125)I permanent plantation.The curative effect was appraised according to the degree of ostalgia relieving and the changing of the radiology imaging in patients.Results Accepted radioactive seeds ~(125)I permanent plantation,relief of pain was obtained and the effective rate is 91%(20/22).However none of the patients showed severe side-effect.Among 32 lesions in 22 cases followed-up by CT in 2 months,4 obtained CR, 18 obtained PR,10 NC and 0 PD.The responsive rate was 68.7%.Conclusion ~(125)I permanent plantation procedure can be a safe and effective method in treating osseous metastases and obtaining good clinical effects with minimal damage and few comnlications.

5.Cardiac troponin T and I: application in myocardial injury and forensic medicine.

The Cardiac Troponin T and I are highly cardiac specific biochemical markers of myocardial injury. They are very sensitive markers to detect all kinds of myocardial injury, and are able to distinguish myocardial injury and skeletal injury. Furthermore, They are independent predictor of future cardiac events. Such markers are now widely used in the clinic practice. It is prospective to use them in Forensic Medical Science.

6.Mitochondrial apoptotic signaling pathway in neurons following brain injury induced by hypoxia.

Impairment of neuronal mitochondria following hypoxia of brain not only result in nerve cell's energy-deprivation and dysfunction, mitochondria also play key roles in apoptosis of neurons. A central step being the release of cytochrome c (cyt c) across the outer mitochondrial membrane into the cytoplasm through opening of the mitochondrial permeability transition pore. Releasing of cytochrome c induce to downstream consequences of specific caspase activation. The antiapoptotic and proapoptotic members of the Bcl-2 family regulate mitochondrial activities relevant to apoptotic signaling by influencing the realaseing of cyt c.

METHODS:
One hundred and sixty subjects, including 80 males and 80 females were selected from southwest Han population for routine CT-VRT (reconstruction thickness 1 mm) examination. The lengths of both manubrium and mesosternum were recorded, and the combined length of manubrium and mesosternum was equal to the algebraic sum of them. The sex-specific linear regression equations between the combined length of manubrium and mesosternum and the real body height of each subject were deduced.
RESULTS:
The sex-specific simple linear regression equations between the combined length of manubrium and mesostenum (x3) and body height (y) were established (male: y = 135.000+2.118 x3 and female: y = 120.790+2.808 x3). Both equations showed statistical significance (P < 0.05) with a 100% predictive accuracy.
CONCLUSION
CT-VRT is an effective method for measurement of the index of sternum. The combined length of manubrium and mesosternum from CT-VRT can be used for body height estimation in southwest Han population.

8.Inhibition of growth and proliferation of Hep-2 cells by targeting human telomerase reverse transcriptase mRNA using RNA interference technology.

OBJECTIVETo investigate the effect of RNA interference by targeting human telomerase reverse transcriptase (hTERT) mRNA in the larynx cancer cell line, Hep-2.
METHODSThe primary structures of hTERT cDNA were found in GenBank. Then the structure analysis were done according to RNAi strategy which determined the specific base sequences to design shRNA plasmid. Two types of plasmid, pshRNA1 and pshRNA2, involved in fluorescein gene were synthesized based on the specific base sequences. Control pshRNA3, a random sequence, and control pshRNA4, without additional specific sequence were also constructed. Cells were treated daily with pshRNA1-4 or normal culture medium respectively. The pshRNA1-3 was identified by electrophoresis. After administration of pshRNA1-4, fluorescence expression was detected by confocal microscopy, the expression of hTERT of the transfected cells was determined by Western blotting, telomerase activity was measured by TRAP-PCR ELISA, cell viability was determined by MTT assay, morphological changes and apoptosis were examined by inverted microscope and TUNEL respectively.
RESULTSThere was a 400 bp balteum in pshRNA1-3 after cut by SalI, which was identical with the size of the objective gene. Many cells presented green fluorescence after being treated by pshRNA1-4, but there are much more dead green fluorescent cells in the pshRNA1 and pshRNA2 group. hTERT protein and telomerase activity was significantly decreased after treated by pshRNA1 or pshRNA2. It was observed that treatment with pshRNA1 or pshRNA2 in the presence of a valid transfection reagent could reduce cell viability of Hep-2 cells within 96 h (P < 0.01). Under the same culture conditions, cells grew more sparsely and the number of apoptotic cell increased significantly.
CONCLUSIONSshRNA plasmid directed against human telomerase reverse transcriptase can effectively transfect Hep-2 cells. shRNA targeted hTERT gene can significantly inhibit the growth and proliferation of Hep-2 cells, which results in apoptotic cell death. RNA interference may be a promising strategy for the treatment of laryngeal cancer.

9.Role of serotonergic neurons in dorsal raphe nuclei in regulation of sleep.

AIMTo investigate the roles of serotonergic neurons in dorsal raphe nuclei (DRN) in sleep.
METHODSStereotaxic, microinjection and polysomnography (PSG) were used in the experiment.
RESULTSMicroinjection of L-glutanate (L-Glu) into the DRN decreased slow wave sleep (SWS) and paradoxical sleep (PS), and increased wake (W). Microinjection of kainic acid (KA) and p-chlorophenylalanine (PCPA) respectively into the DRN, SWS and PS were promoted, and W was reduced.
CONCLUSIONSerotonergic neurons in dorsal raphe nuclei involved in the regulation of sleep. Sleep was reduced when the serotonergic neurons were excited, and when the neurons were inhibited. sleep was increased

10.Clinical analysis of myoepithelial carcinoma of head and neck.

OBJECTIVETo evaluate clinical feature, diagnosis, treatment and prognosis of myoepithelial carcinoma (MC) in the head and neck.
METHODSClinical data of 11 patients which were confirmed by pathology and immunohistochemistry in Cancer Center, Sun Yat-sen University from Jan. 1992 to Dec. 2006, were reviewed. There were 5 cases in parotid gland, 1 in hard palate, 1 in maxillary sinus, 1 in pharyngeal recess, 1 in bucca cavioris, 1 in scalp, and 1 in gingiva. Male female ratio is 4 to 7. The median age at diagnosis was 37 years (range: 14 - 60 years).
RESULTSAll cases were operated, 4 underwent surgery alone, 2 underwent surgery plus adjuvant radiotherapy, 2 received surgery plus adjuvant chemotherapy, 3 underwent surgery plus adjuvant chemoradiation. After operation, 8 cases occurred local recurrence and 4 cases occurred distance and lymph nodes metastasis. There was spindle cell type in 5 cases, clear cell type, plasmacytoid cell type in 2 cases, epithelioid cell type, mixed type in 1 case. The median follow-up time was 40 months. AS to the last follow-up time, 8 patients died.
CONCLUSIONSThe characteristics of the tumor were rapidly enlarging, invading the surrounding regions, high rates of lymph node metastasis, high rates of distance metastasis. It was easily recurrence if the surgical excision was not radically for MC, and the prognosis was poorly. MC was a sort of malignant tumor. Radical operation must be advocated initially. Chemotherapy and radiotherapy may be effective after operation.
