This research was based on a questionnaire that was conducted in 599 doctors from three tertiary hospitals in Guangzhou. The results indicate that four factors affect doctors' innovative ability with different degrees. Relevant measures are suggested

BACKGROUND: Obesity is serious complex disease determined by multiple genetic factors. Body mass index (BMI) is a common phenotype for obesity. Although some candidate genes have been shown to be related with BMI variation, the underlying substantial relationship remains poorly understood.OBJECTIVE: To investigate whether estrogen receptor-α (ER-α) Pvull genetic polymorphism is associated with BMI variation.METHODS: In total 390 premenopausal women from a local population of Nanchang City in Jiangxi Province People's Hospital.Body weight and height were measured. All participants were genotyped at the ER-α Pvull locus using polymerase chain reaction-restriction fragment length polymorphism.RESULTS AND CONCLUSION: The ER-α Pvull genotype frequencies of PP, Pp and pp were 0.133, 0.485 and 0.382,respectively. The distribution of ER-Pvull genotypes was in accordance with Hardy-Weinberg equilibrium (HWE) (P > 0.05).There was no significant correlation between ER-α Pvull genetic polymorphism and BMI variation (P = 0.338). These findings indicate that ER-α Pvull genetic polymorphism does not greatly affect BMI variation of premenopausal women in Nanchang region.
3.Empirical Study on the Effects of Different Dosage of Grassleaf Sweetflag Rhizome on Central Nervous System

OBJECTIVE:To study the effect of different dosage of grassleaf sweetflag rhizome on central nervous system.METHODS:The effects of different dosage of grassleaf sweetflag rhizome on the hours of sleep and hypoxia live time of mice were observed,and the effects of which on water contents of brain tissues,apoptosis,endothelin(ET),malonaldehyde(MDA),erythrocuprein(SOD),glutathione peroxidase(GSH-Px),etc.in rats with cerebral ischemia were observed as well.RESULTS:Compared with the control group,different dosage of grassleaf sweetflag rhizome could shorten the sleeping time of mice and prolong the live time of hypoxia mice;high dosage of grassleaf sweetflag rhizome could remarkably decrease the water content of brain tissue and MDA level and inhibit the apoptosis of brain tissue cells while increase the activity of SOD and GSH-Px.CONCLUSION:High dosage of grassleaf sweetflag rhizome has the strongest protective effects to central nervous system,the effects of middle and low dosage of grassleaf sweetflag rhizome are lower than that of the high dosage.

Objective To detect CD_(36) expressions in polycystic ovary (PCO), and to explore its correlation with local androgen and insulin at transcription level. Methods From August 2002 to February 2003, 12 patients with asymmetric PCO, 15 primary or secondary infertile patients without endocrine disorders and 8 polycystic ovary syndrome (PCOS) with bilateral PCO were recruited. Extraction of follicular fluid and detection of testosterone(T), dehydroepiandrosterone sulfate (DHEAS), insulin (INS) and androstenedione (A_2) were performed. Relative CD_(36) mRNA expression level of human ovarian inner thecal cells was analyzed by auto image analysis system (IAS) after RT-PCR. BACKGROUND: Body mass index(BMI)is a commonly used phenotype for obesity,which is determined by multiple genetic factors.OBJECTIVE: To investigate whether osteocalcin(also known as bone Gla protein,BGP)Hind Ⅲ genetic polymorphism is associated with BMI variation.METHODS: A total of 390 premenopausal women from a local population of Nanchang City were selected.Body weight and height were measured.All participants were genotyped at the BGP Hind Ⅲ locus using polymerase chain reaction-restriction fragment length polymorphism(PCR-RFLP).RESULTS AND CONCLUSION: The BGP genotype frequencies of HH,Hh and hh were 0.077,0.408 and 0.515,respectively.The distribution of BGP Hind Ⅲ genotypes was consistent with Hardy-Weinberg equilibrium(P > 0.05).The BGP Hind Ⅲ were significantly associated with BMI(P=0.002),which could explain about 5.47% of BMI variation.On average,BMI of individuals with HH genotype was the highest[(22.81±0.73)kg/m2],individuals with Hh genotype was intermediate[(21.50±0.53)kg/m2],while individuals with hh genotype was the lowest[(20.23±0.63)kg/m2].Therefore,carriers of HH and Hh genotypes had,respectively,approximately 12.75% and 6.28% higher BMI than carriers of the hh genotype.To our best knowledge,this is the first study reporting the association of BGP Hind Ⅲ genetic polymorphism and BMI in healthy premenopausal women.

The effects of ketamine on transient outward potassium current (I(to)) of isolated human atrial myocytes were investigated to understand the mechanism of part of its effects by whole-cell patch-clamp. Atrial myocytes were enzymatically isolated from specimens of human atrial appendage obtained from patients under going cardiac valve displacing. Ito is recorded in voltage-clamp modes using the patch-clamp technique at room temperature. Currents signals were recorded by an Axopatch 200B amplifier with the Digidata 1322A-pClamp 9.0 data acquisition system. Ketamine decreased I(to) of human atrial myocytes in a dose-dependent manner. The current-voltage curve was significantly lowered, 30, 100, 300, and 1000 micromol x L(-1) ketamine decreased respectively I(to) current density about (13.62 +/- 0.04)%, (38.92 +/- 0.05)%, (72.24 +/- 0.10)% and (83.84 +/- 0.05)% at the potential of 50 mV, with an IC50 of 121 micromol x L(-1). The I(to) activation curve, inactivation curve and the recovery curve were not altered by ketamine. So, ketamine concentration-dependently decreased I(to) of human atrial myocytes.
7.Changes of IL-1, IL-2 and TNF-α in response to lappaconitine in rats with traumatic brain injury

ObjectiveTo observe the effect of lappaconitine (LA) on brain water content and serum IL-I,IL-2 and TNF-α levels in rats with traumatic brain injury (TBI) as well as its cerebral protective function.MethodsA total of 24 male SD rats were involved in the study and randomly divided into control group,TBI group and TBI +LA group,with eight rats per group.The rats in the TBI group and TBI + LA group were inflicted with fluid percussion injury ( FPI ).The rats in the TBI + LA group were treated with LA (4 mg/Kg/d,ip,for 10 consecutive days).The neurological score,brain water content and serum IL-I,IL-2 and TNF -α concentrations were detected at time points including TO ( before FPI ),T1 (one day after FPI),T2 (five days after FPI) andT3 (10 days after FPI).Results At each time point after FPI,the neurological dysfunction was observed in both the TBI group and TBI + LA group.The neurological dysfunction was gradually alleviated from TI to T3 in the FPI + LA group,which showed significant lower neurological score as compared with the TBI group (P <0.05 or 0.01 ).The brain water content in the TBI group and TBI + LA group was significantly higher than that in the control group at each time point after FPI.Meanwhile,the water content of the TBI + LA group was significantly lower than that of the TBI group ( P < 0.01 ).The serum IL-1,IL-2 and TNF-alpha concentrations in the TBI group and TBI + LA group were significantly higher than those in the control group at each time point after FPI,and the serum IL-I,IL-2 and TNF-αt concentrations of the TBI + LA group were significantly lower than those of the TBI group ( P < 0.01 ).Conclusions LA exerts cerebral protective effects of TBI rats by relieving the neurological dysfunction and cerebral edema and reducing the serum IL-1,IL-2 and TNF-α concentrations.
8.Model construction of rat coronary artery smooth muscle cell endoplas-mic reticulum stress induced by thapsigargin

AIM: To investigate the primary culture method for coronary artery smooth muscle cells (CASMCs), and to establish the endoplasmic reticulum stress ( ERS) model in CASMCs of SD rats.METHODS:CASMCs were cultured by tissue explant method .The morphological characteristics were observed under optical micro-scope.The marker proteins of CASMCs , including α-SMA and SM-MHC, were identified by immunofluorescence tech-nique.The protein expression levels of BiP and CHOP , the marker molecules of ERS, were determined by Western blot . RESULTS:The spindle-shaped CASMCs climbed out from the edge of coronary artery tissues after 6 d, and formed the typical hill and valleygrowth pattern of CASMCs at 9~10 d.The result of immunofluorescence technique showed that α-SMA and SM-MHC were positively expressed .The results of Western blot showed that the protein expression of BiP and CHOP in TG ( 1 and 2 μmol/L ) treatment groups was increased compared with control group .Compared with control group, the protein expression of BiP and CHOP was significantly increased after 1 μmol/L TG treatment for 24 and 48 h. CONCLUSION:CASMCs can be successfully cultured by tissue explant method .ERS model of CASMCs was established by 1 μmol/L TG treatment for 24 h.
9.Investigation of rumination level and its influential factors among psychiatry nurses who go through work place violence

Objective:To investigate the level of rumination and its influential factors among Chinese psychiatry nurses who go through work place violence.Methods:In this study, 150 psychiatry nurses were recruited from Affiliated Brain Hospital of Guangzhou Medical University (specialize in psychiatry), via the combination of convenient sampling and snowball sampling. Chinese Event-related Rumination Inventory (C-ERRI) was applied in the survey.Results:The total score of C-ERRI was (22.11±9.62) points, and the scores of intrusive rumination subscale and deliberate rumination subscale were (12.99±5.58) and (9.12±6.01) points, respectively. Multiple Linear Regression Analysis show that assault frequency in recent year ( B=-3.195, P<0.01) and whether got injury in the recent assault ( B=8.591, P<0.01) were predictors of deliberate rumination, which account for 26.8% variance of the equation. Gender (male) ( B=-2.415, P<0.01), Injury frequency in recent year ( B=2.864, P<0.01) and whether got injury in the recent assault ( B=8.949, P<0.01) were predictors of intrusive rumination. They account for 36.0% variance of the equation. Conclusion:In this study, the level of rumination among Chinese psychiatry nurses was low. Their rumination style was deliberate rumination.
10.Sacroiliac joint injury treated with oblique insertion at anatomical points: a randomized controlled trial.

OBJECTIVETo compare the effects of oblique insertion at anatomical points and conventional acupuncture for sacroiliac joint injury.
METHODSEighty patients were randomly divided into an observation group and a control group, 40 cases in each one. In the observation group, oblique insertion therapy at anatomical points was used, and the 9 points of equal division (anatomical points) marked by palpating the anatomical symbol were treated as the insertion acupoints. In the control group, conventional acupuncture was applied, and perpendicular insertion was adopted at Huantiao (GB 30), Zhibian (BL 54) and Weizhong (BL 40), etc. In the two groups, the! treatment was given once a day and 5 times per week. Ten treatments were made into one course and two courses were required. The clinical effects, the changes of visual analogue scale (VAS) and Oswestry dysfunctional index. (ODI) before and after treatment were observed in the two groups.
RESULTSThe total effective rate of the observation group was 90.0% (36/40), which was better than 72.5% (29/40) of the control group (P < 0.05). After treatment, the results of the VAS and ODI of the two groups were apparently declined (both P < 0.01), and those in the observation group were decreased more obviously (both P < 0.01).
CONCLUSIONThe effect of oblique inser-tion at anatomical points for sacroiliac joint injury is superior to that of conventional acupuncture, which can effectively relieve pain and improve the disfunction.
