1.BMP9 Regulates Osteogenic/Odontogenic Differentiation of Immortalized Stem Cells from the Apical Papilla via the ERK5 Pathway

Objective The aim of this study was to investigate the effects of the ERK5 pathway on bone morphogenetic protein?9(BMP9)?regu?lated osteogenic/odontogenic differentiation of immortalized stern cells from the apical papilla(iSCAP). Methods BMP9 was introduced into the iSCAP by using recombinant adenoviruses,and the P?ERK5 protein expression was measured via western blotting. Then,the osteogenic/odontoblas?tic changes were analyzed by alkaline phosphatase(ALP)staining and Alizarin red staining,and the expression of osteogenic/odontoblast?associat?ed genes,such as Runx2,OCN,OPN,and DMP1 was measured by RT?PCR. Results BMP9 could up?regulate the phosphorylation of ERK5 in iSCAP. After using BIX02189,which was an inhibitor of ERK5,the phosphorylation of ERK5;the activity of ALP;the expression of Runx2, OCN,OPN,and DMP1;and the calcium deposition were all significantly inhibited. Conclusion The ERK5 signaling pathway plays an important role in BMP9?regulated osteogenic/odontogenic differentiation of iSCAP.

4.Influence of Ginsenoside-Rbl on cardiomyocyte tumor necrosis factor level in rats with adriamycin-induced chronic heart failure

Objective: To explore whether relieving effect of Ginsenosides-Rb1 (Gs-Rb1) on chronic heart failure (CHF) induced by adriamycin is related to improving levels of tumor necrosis factor (TNF) receptor (TNFR) and ligand TNF-α or not. Methods: Adriamycin-induced CHF model rats were randomly divided into adriamycin group (n=15, received adriamycin 1 μmol/L) and Gs-Rb1 group (n=17, received Gs-Rb1 70 mg•kg-1•d-1 based on adriamycin group), another 10 healthy Wistar rats were selected as normal control group. Meanwhile, cultured cardiomyocytes of neonatal rats were accordingly divided into normal control group, adriamycin group and adriamycin+Gs-Rb1 group. After intervention, echocardiography, levels of TNFR/TNF-α mRNA and protein were measured and compared among three groups. Results: 1. In vivo: Compared with normal control group TNFR-1 [protein(0.13±0.02)kDa, mRNA(0.19±0.05)bp],TNFR-2[protein(0.24±0.01)kDa, mRNA(0.13±0.02)bp],TNF-α [protein(0.11±0.01)kDa, mRNA(0.26±0.05)bp], the levels of TNFR-1 [protein(0.67±0.04)kDa,(0.51±0.04)kDa;mRNA(0.81±0.03)bp,(0.49±0.05)bp],TNFR-2 [protein(0.61±0.05)kDa, (0.47±0.03)kDa,mRNA(0.28±0.03)bp,(0.18±0.04)bp] and TNF-α [protein(0.28±0.04)kDa, (0.20±0.03)kDa, mRNA(0.67±0.05)bp,(0.45±0.04)bp] significantly rose(P<0.05 or <0.01) in adriamycin group and Gs-Rb1 group; and those levels of TNFR-1, TNFR-2, TNF-α protein and mRNA of Gs-Rbl group were significant reduction than those of adriamycin group; 2. In vitro: The results were similar, those of adriamycin group and Gs-Rb1 group were also significant rise than those of normal control group(P<0.05 or <0.01),those of Gs-Rbl group were significant reduction than those of adriamycin group(P<0.05 or <0.01). Conclusion: TNFR/TNF-α expression may play a certain role in the process of adriamycin-induced CHF; improving effect of Gs-Rb1 on adriamycin-induced CHF may be related to regulation of TNFR/TNF-α.
5.The study of application of the intra-aortic balloon pump in the patients with renal insufficiency during cardiac surgery

Objective To discuss the protective effect of intra-aortic balloon pump(IABP) on renal function in patients with chronic renal insufficiency undergoing cardiac operations.Methods From May 2010 to May 2012,17 patients with moderate renal insufficiency who underwent cardiac surgery using cardiac pulmonary bypass(CPB) were randomized to divide into IABP group and control group.IABP was inserted before the cardiac operation,the support was maintained perioperatively in IABP group.In control group the IABP was never used during the operation.The clinical parameters related to renal function including the output of urine,the serum creatinine levels were documented perioperatively.Results The serum creatinine levels were increased obviously post-operation in both groups.But the degree of increase was significantly smaller in IABP group.Conclusion To patients with moderate renal insufficiency undergoing CPB cardiac operations,perioperative application of IABP is beneficial to protect renal function and may help to improve the outcome.

7.Effects of chronic hypoxia on expression of NOSⅢ mRNA induced by acute hypoxia in pulmonary artery endothelial cells

AIM and METHODS: In situ hybridization and image-analysis system were used to investigate the expression of NOSⅢ mRNA in cultured porcine pulmonary artery endothelial cells (PAEC) under normoxic or chronic hypoxic conditions. RESULTS: The expression of NOSⅢ mRNA were increased by 48%, 125%, 119% respectively in the 2nd, 4th, 6th subculture of normoxic PAEC after expouse to acute hypoxia for 12 hours while in chronic hypoxic group,the expression of Ⅲ NOS mRNA were increased by 92%, 245%, 565% separately. The increase was more apparent in chronic hypoxic group than in normoxic group and became more significant following the prolonging duration of hypoxia. CONCLUSION: Chronic hypoxia increased the expression of NOSⅢ gene in PAEC in response to acute hypoxia. Nitric oxide may play an important role in the decrease of hypoxic pulmonary vasoconstriction caused by chronic hypoxia.
8.A comparative study on three devices in treatment of complete dislocation acromioclavicular joint

[Objective]To compare the results of 3 methods to treat complete dislocation of acromioclavicular joint.[Method]Three different surgical techniques were employed: K-wire tension band(in group A of 26 cases),clavicular hook plate(in group B of 34 cases) and percutaneous screw and K-wire(in group C of 32 cases).There were 78 males and 14 females,and all the cases were acute complete dislocation of acromioclavicular joint.[Result]Eighty-six cases were followed up with an average period of 19 months(13~31 months).Acording to Karlsson's standards,the rate of excellent and good in the three groups was 73.1%,94.1%,93.8% respectively.The mean oprate time was 35.6min,38.6min,28.1 min respectively.The average blood loss was 43.6ml,48.3ml,7.5ml respectively.The average incision length was 8.8cm,8.9cm,0.7cra cm respectively.The complications were 20,9,8 respectively.[Conclusion]Percutaneous coracoclavicular screw and acromioclavicular K-wire fixation has advantages of less opration time and blood loss,cosmetic scar,and a low complication rate,good clinical results.It is believed to be a better method for the treatment of acromioclavicular joint dislocation.
9.The relationship between preoperative albumin level and postoperative acute kidney injury following off- pump coronary artery bypass surgery

Objective To investigate the relationship between preoperative albumin level and postoperative acute kidney injury(AKI)following off-pump coronary artery bypass(OPCAB)surgery;to confirm albumin cut-off value associated with AKI. Method This was a retrospective cohort study. data from patients who accepted OPCAB surgery in last two years in our hospital and AKI was diagnosed according to KIDGO-AKI criteria. Results Generalized additive model and multivariable logistic regression analysis have shown that the higher preoperative albumin level is related with the lower the incidence of AKI after OPCAB surgery. By generalized additive model,multivariable logistic regression analysis and ROC curve,we confirmed the albumin cut-off value associated with AKI between 39~40 g/L. Conclusion For patients undergoing OPCAB surgery,the higher preoperative albumin level ,the lower the incidence of AKI. In order to reduce the incidence of AKI in OPCAB surgery patients,preoperative albumin level higher than 40 g/L should be maintained.
10.Firing properties and classification of MVN neurons in rats.

In order to know the effects of caloric stimulation on neuronal firing in medial vestibular nuclei (MVN) by middle ear irrigation, the middle ear was irrigated with ice (4 degrees C), hot (44 degrees C), and warm (37 degrees C) water, and the firing rate of MVN neuron was extracellularly recorded. The results showed that the firing rate of MVN neuron was changed by caloric stimulation, and the majority of MVN neurons showed excitation by irrigation with hot water and inhibition by ice water (type A). The neuronal firing was recovered immediately after the cessation of the stimulation. I It was concluded that the neuronal firing rate in MVN was changed by caloric stimulation in middle ear cavity. The response was different in various neurons.
