1.Relationship between endometrial stem/progenitor cells and gynecological diseases
Clinical Medicine of China 2017;33(1):93-96
The existence of stern/progenitor cells in the endometrium was postulated many years ago,but the first functional evidence was published in 2004.The study of endometrial stem/progenitor cells had opened up an active field in the subsequent 10 years.Human endometrium has immense regenerative capacity and bilayer structure,in which the upper functionalis is shed at menses and regenerates from the remaining basalis in the subsequent cycle, these characteristics had been the motivation for investigators to identify and characterize endometrial stem/progenitor cell populations. The discovery of specific markers for endometrial stem/progenitor cells has enabled the examination of their role in endometrial proliferative disorders, including endometriosis, adenomyosis and Asherman's syndrome.

4. Preparation and quality evaluation of two types of colchicine ethosome patches
Chinese Traditional and Herbal Drugs 2013;44(17):2380-2385
Objective: To optimize the two types of colchicine ethosome patches using uniform design and to investigate their in vitro release, transdermal properties, stability, and stimulus. Methods: Two kinds of colchicine ethosomes made of soybean lecithin (SL) and hydrogenated phosphatidy (HP) were prepared using injection method. By the DPS 7.0 sotfware, the uniform design was used to optimize the patch components of colchicines, polyacrylic resin IV, succinic acid, triethyl citrate, and azone in patch pressure-sensitive adhesive, with its initial adhesion and hold tack as indexes. The colchicine ethosomes patches of SL and HP were prepared, respectively. The automated dissolution test analyzer and improved Franz diffusion cell were adopted to compare the in vitro release and transdermal properties of the two types of colchicine ethosomes patches, respectively. Moreover, the preparation stability under different light, humidity, and high temperature conditions was studied, and the skin irritation test for rabbits was carried out. Results: The optimized colchicine ethosome patches by the uniform design adhere with the viscous standards in Chinese Pharmacopeia 2010. The in vitro release of the three types of patches were fitted with the Higuchi equation and their release rates increased in turn, respectively, which were (3.1331 ± 0.3272), (2.8724 ± 0.3481), and (2.3802 ± 0.2134) μg/(cm2·h1/2). Their in vitro transdermal releases were fitted with the zero-class equation and increased in turn, respectively, which were (0.9390 ± 0.0731), (0.8775 ± 0.0908), and (0.6167 ± 0.0816) μg/(cm2·h). During the storage, the HP patches had better stability than the SL patches, especially with the stable color. In the skin irritation test for rabbits, there was no erythema and oedema reactions in 48 h. Conclusion: The HP patches have obviously higer in vitro release rate and transdermal rate, and for its better stability of preparation, the HP patches are superior to the SL ones.

Objective The newly developing gene therapy method and dominant negative mutants were bein g used as new promising HBV therapy method, and a dominant negative mutant of HB V X g ene we have reported in our previous report has some effects both on HBV replica tion and expression in transient expression, but the effects were interfered by persistent secretion of HBV in HepG 2 2.2.15 cell line in the experiment. To mak e sure the effects of dominant negative mutant of HBx gene, we established a HBx DN stably expressing cell clone, and evaluated the effects of HBx dominant negat ive mutant on HBV gene expression. Methods The prev HBx-GFP dominant mutant and the plasmids pRev Xwt, pRev GFP which contain the wild type X gene or GFP gene then transfected into HepG 2 2.2.15 cells by liposome. The HBsAg, HBeAg by in media were as sayed by RIA and HBV-related RNA were assayed by Northern blot. Results The pRev HBx-GFP, GFP and wild type X constructs can be effectively expressed in HepG 2 2.2.15 cells. The stable expressed HBx -GFP can significantly reduce HBeAg, HBeAg in media and the HBV-related RNA in HepG 2 2.2.15 cells, but not for pRev Xwt and pRev GFP. Conclusions The dominant negative mutant pRev HBx-GFP can significantly inhibit the HBV gen e expression. It also suggested that X gene may be one promising target for HBV gene therapy.
