1.Effects of Sarcandra glabra extract on immune activity in restraint stress mice.

OBJECTIVETo study the protective effect of Sarcandra glabra extract (SGE) on immune system in restrained mice.
METHODThe male C57BL/6 mice were randomly divided into normal control group, stress control group, 125, 500 mg x kg(-1) SGE group. The spleen lymphocyte suspensions of each group were prepared. The parameters of spleen T cells subsets, NK cell and NKT cell proportion and number was detected by Flow cytometry.
RESULTSGE regulated the balance of T cell subsets, increased the percent of NK cells and NKT cell proportion and number in restrained mice.
CONCLUSIONSGE has immunologic protective effect in restrained mice probably via the amelioration of immune cells proportion and number.

3.KIR and allogeneic hematopoietic cell transplantation - review.

The profound graft-versus-leukemia (GVL) effect presented after allogeneic hematopoietic cell transplantation (allo-HSCT) has been evidenced. In contrast to T cell mediated GVL, natural killer (NK) cells recognize target cells and introduce GVL effect by using an integration of activating and inhibitory receptors. This review has summarized current literatures from 2001 - 2003 on human killer cell immunoglobulin receptors (KIR) and other NK cell receptors involved in recognition of tumor targets and the polymorphism of KIR genes of donor/recipient pairs of related and unrelated Allo-HSCT. KIR epitope mismatch may facilitate engraftment and reduce leukemia relapse post transplant by mediating lysis of recipient's cells and introducing GVL effect under the condition of KIR epitope mismatch. Clinical roles of KIR in Allo-HSCT and immunotherapy are discussed. Technologic approach in allogeneic reactive NK cells introduction, identification and selection in vitro, development of inhibitory receptor blockade as well as up-regulation of activating NK cells may significantly enhance GVL immune response. Further investigation on the regulation of KIR inhibitory receptors enables to design novel strategy in cancer immunotherapy over the forthcoming decade.

4.Meta-analysis on effect of Grifola frondosa polysaccharide in regulating in vivo immunoregulatory function on animal disease models.

The study aimed to explore the in vivo immunoregulatory function of Grifola frondosa polysaccharide( GFP) on animal disease models. Databases of PubMed,Embase,Web of Scinece,CNKI,CBM and Wan Fang Data were searched from the date of their establishment to February 2018. Two reviewers independently screened included studies and evaluated their quality by using SYRCLE's risk of bias tool. R software was used to analyze the data. Finally,20 animal experiment studies were included. According to Metaanalysis. For cellular immunity,GFP could effectively enhance the proliferation of effect or T cells,natural killer cells and macrophages in mice. The percentage of CD4+T cells( MD = 1. 89,95% CI [0. 94,2. 83],P < 0. 000 1),CD8+T cells( MD = 8. 46,95% CI[5. 93,11. 00],P<0. 000 1),NK cells( MD= 2. 67,95% CI [0. 23,5. 11],P= 0. 03),and macrophages( MD= 14. 09,95% CI[0. 84,27. 34],P= 0. 04) were all higher than those in control group. For humoral immunity,GFP could increase the secretion of TNF-α and INF-γ. The secretion of TNF-α( SMD = 15. 92,95% CI [9. 07,22. 76],P<0. 000 1) and INF-γ( SMD = 5. 34,95% CI[3. 42,7. 26],P<0. 000 1) were all higher than those in control group. In conclusion,GFP could regulate immunologic function by enhancing the proliferation activity of immune cells( CD4+T cells,CD8+T cells,NK cells and macrophages) and the secretion of immune factors( TNF-α and INF-γ) . However,it is necessary to further standardize the selection of specific surface markers of immune cells and the administration of GFP,in order to reduce the heterogeneity among the studies. At the same time,more attention shall be paid to experimental design,implementation and full report,especially to the establishment and implementation of animal experimental registration system,so as to improve the transparency and quality of the whole process of animal experimental research,enhance the value of basic research ultimately,and provide a reliable theoretical basis for the transformation of basic research into clinical research.

5.Chemistry of polysaccharide Lzps-1 from Ganoderma lucidum spore and anti-tumor activity of its total polysaccharides.

AIMTo study the structure and anti-tumor activity of polysaccharide from Ganoderma lucidum spore treated with microwave.
METHODSDEAE-cellulose and Sephadex G-50 column chromatography were used to isolate and purify the polysaccharide whose structure was characterized by using chemical and spectral methods.
RESULTS AND CONCLUSIONOne polysaccharide, named Lzps-1 was obtained from the water extract, with its molecular weight estimated by HPGPC to be 8000. Its structure was investigated to be glucan. The total polysaccharides, Lzps processed antitumor activity against sarcoma 180 and Lewis lung cancer in mice and enhanced the NK cell activity. Lzps-1 is obtained for the first time from Ganoderma spore Lzps has anti-tumor activity.

6.Comparative Quantitative Analysis of Cluster of Differentiation 45 Antigen Expression on Lymphocyte Subsets.

BACKGROUND: Since the recent introduction of radioimmunotherapy (RIT) using antibodies against cluster of differentiation (CD) 45 for the treatment of lymphoma, the clinical significance of the CD45 antigen has been increasing steadily. Here, we analyzed CD45 expression on lymphocyte subsets using flow cytometry in order to predict the susceptibility of normal lymphocytes to RIT. METHODS: Peripheral blood specimens were collected from 14 healthy individuals aged 25-54 yr. The mean fluorescence intensity (MFI) of the cell surface antigens was measured using a FACSCanto II system (Becton Dickinson Bioscience, USA). MFI values were converted into antibody binding capacity values using a Quantum Simply Cellular microbead kit (Bangs Laboratories, Inc., USA). RESULTS: Among the lymphocyte subsets, the expression of CD45 was the highest (725,368+/-42,763) on natural killer T (NKT) cells, 674,030+/-48,187 on cytotoxic/suppressor T cells, 588,750+/-48,090 on natural killer (NK) cells, 580,211+/-29,168 on helper T (Th) cells, and 499,436+/-21,737 on B cells. The Th cells and NK cells expressed a similar level of CD45 (P=0.502). Forward scatter was the highest in NKT cells (P<0.05), whereas side scatter differed significantly between each of the lymphocyte subsets (P<0.05). CD3 expression was highest in the Th and NKT cells. CONCLUSIONS: NKT cells express the highest levels of CD45 antigen. Therefore, this lymphocyte subset would be most profoundly affected by RIT or pretargeted RIT. The monitoring of this lymphocyte subset during and after RIT should prove helpful.

8.Extranasal T/NK-cell lymphoma presenting as intestinal diverticulum.

A case of intestinal angiocentric T/NK-cell lymphoma in a 58-year-old man is reported. The patient presented initially with panperitonitis because of perforation of sigmoid colon diverticulum. He underwent segmentectomy of involved bowel. Histologically, the intestinal wall showed diffuse infiltration of medium or large size lymphoma cells with angiocentric growth and necrosis. The lymphoma cells were CD56+, CD45RO+, CD3+, CD4-, CD8-, CD20-, and CD30- in paraffin sections with germline configuration of TCR-gamma gene, consistent with T/NK-cell lymphoma. Further staging revealed splenomegaly. Intestinal angiocentric T/NK cell lymphoma represents a distinct etiology of diverticulum with perforation.

9.Preliminary study on effect of ganhuang injection on reject reaction of xenograft.

OBJECTIVETo study the effect of Ganhuang Injection (GHI) on reject reaction of xenograft.
METHODSRT-PCR technique was used to detect the activated relevant gene expression in porcine endothelial cells (PEC), and 51Cr releasing method was used to test the killing and adhesion action of NK cells.
RESULTSThe normal human serum and human NK-92 cell could up-regulate the mRNA expressions of E-selectin and IL-1 alpha gene in PEC, showing the PEC activating action, GHI could inhibit these activated gene expressions. Cyto-toxic experiment showed that GHI could also inhibit the cytotoxicity of NK cell on PEC dose-dependently, which was in accord with its inhibition on adhesive action of NK on PEC.
CONCLUSIONGHI could inhibit not only the PEC activation in hyperacute rejection, but also the function of NK cells in delayed xenograft rejection.

10.Effect of Xiaochaihu decoction and different herbal formulation of component on inhibiting H22 liver cancer in mice and enhancing immune function.

OBJECTIVETo observe the effects of Xiaochaihu decoction and different herbal formulation of componenton on inhibiting H22 mouse solid liver cancer and on the immune function, for approaching the dose-effect relationship and compatibility mechanism.
METHODTransplanted H22 liver the cancer to mice, filling on high, moderate, low dose of mice in the xiaochaihu decoction drug (27, 13.5, 6.75 g x kg(-1)), 5-FU intraperitoneal injection (20 mg x kg(-1)) as positive control. After being administered for 10 days, we determined the cancer weight, body weight, spleen index and thymus gland index for studying the dose-effect relationship. And we studied compatibility rule of Xiaochaihu decoction and the different herbal formulations of its components by using the same method. Mice in the Xiaochaihu decoction group (13.5 g x kg(-1)) and Ginseng-Chinese Date-Licorice Root apozem group (5 g x kg(-1)) were given the medicines for 10 days to observe the weight of tumor, body weight, spleen index, the NK cell activity, proliferation of the T lymphocytes and IL-2 level.
RESULTGrowth of H22 mouse liver cancer was markedly inhibited in high, moderate, low dose group; the inhibiting rates were 49.66%, 48.52%, 36.91%, respectively; the optimal administration dose was moderate. In the study of compatibility rule, only inhibiting rates of Ginseng-Chinese Date-Licorice Root apozem group and Xiaochaihu decoction group exceed 30%; the inhibiting rates were 41.55% and 43.65%. As compared with normal group, spleen index of three dose groups and the different herbal formulation groups was evidently increased (P < 0.01); thymus gland index of the different herbal formulation groups was significant decreased (P < 0.05); thymus gland index and body weight of 5-FU group were evidently decreased (P < 0.001). As compared the model group, the NK cell activity, proliferation of the T lymphocytes and IL-2 level of Xiaochaihu decoction were remarkably increased (P < 0.001); the NK cell activity and IL-2 level of Ginseng-Chinese Date-Licorice Root group were remarkably increased (P < 0.05); the NK cell activity and proliferation of the T lymphocytes and IL-2 level of 5-FU group were remarkable decreased (P < 0.01). As comparing with the normal control group ,body weight of 5-FU group was evidently decreased (P < 0.05).
CONCLUSIONXiaochaihu decoction and Ginseng-Chinese Date-Licorice Root group can markedly inhibit the growth of H22 mouse solid liver cancer and improve the immune function of tumor-bearing mice. The inhibiting effect probably closely related with improving the tumor-bearing host immune function. There is a hint that compatibility Ginseng-Chinese Date-Licorice Root of strengthening body resistance is the core of inhibiting effect.
