2.Urodynamic and Histological Changes in a Sterile Rabbit Vesicoureteral Reflux Model.

This study aimed to investigate pressure changes of renal pelvis and histological change of kidneys in a surgically induced sterile rabbit vesicoureteral reflux (VUR) model. Five rabbits served as a control group, 7 as the sham-operated group, and 8 served as the VUR group. Three weeks later, urodynamic studies were performed, and histological examinations evaluated degree of inflammation, fibrosis, and tubular damage in the kidneys. At a low infusion rate, renal pelvic pressure in the VUR group was stable until late filling phase and then increased slightly. At a high infusion rate, the renal pelvic pressures of the sham-operated and control groups were stable until late filling phase and then increased slightly, whereas the renal pelvic pressure in the VUR group steadily increased from mid filling phase. Focal thinning of the tubular epithelium and interstitial widening were observed in certain cortical areas of refluxing kidneys, without inflammatory cell infiltration. Obvious changes in the mean diameters of distal tubules and extracellular matrix volume fractions were observed in two highly refluxing kidneys. High pressure reflux with bladder instability may result in renal cortical changes.

3.Evaluation of renal function in children with moderate or severe hydronephrosis after operation.

OBJECTIVETo evaluate postoperative renal function in children with congenital moderate or severe hydronephrosis.
METHODS99mTc-labeled diethylenetriaminepenta-acetic acid scintigraphy was performed in 50 children with unilateral moderate or severe hydronephrosis to determine postoperative renal function. We also analyzed the factors influencing renal function recovery.
RESULTSAverage postoperative renal function in 50 cases was 40.62% +/- 10.09%. Among them, 32% of patients had nearly normal renal function and differentiated renal function reached up to 45%. Average preoperative and postoperative renal function in 25 cases was 23.89% +/- 11.65% and 39.33% +/- 8.59% respectively and the increase of renal function was about 15.44% +/- 11.18% (P = 0.0003). Renal parenchyma thickness was negatively correlated with postoperative renal function (r = -0.62, P = 0.0009). The follow-up period was positively correlated with postoperative renal function (r = 0.58, P = 0.0026). The patients' age had no correlation with renal function recovery (r = -0.05, P = 0.80). Recovery of renal function in hydronephrosis with extrarenal pelvis was greater than that in hydronephrosis with intrarenal pelvis (P = 0.016).
CONCLUSIONSPostoperative renal function in children with moderate or severe hydronephrosis can recover to normal. Recovery of renal function was more obvious in hydronephrosis with thinner renal parenchyma, longer follow-up period and extrarenal pelvis.

5.Research on building method of spleen kidney Yang deficiency diarrhea rats model.

Spleen kidney Yang deficiency (SKYD) diarrhea is a common syndrome in tranditional Chinese medicine (TCM). Until now, there is not an ideal SKYD diarrhea rat model for the research. In this study, we compared single factor way (method I, injecting hydrocortisone and gavaging Sennae Folium) with compound factors way(method II, gavaging adenine, improper diet, exhaustion, and gavaging Sennae Folium) on establishing SKYD diarrhea rat model. After modelling, diarrhea index, D-xylose excretory rate, NOS/cGMP signal transduction system, organ index and histopathology examination were used to evaluate the two ways. The results showed that, compared with health group, all the assessment criterias of method I and method II had significant differences (P < 0.01, 0.05). In addition, the index such as diarrhea index, NOS/cGMP signal transduction system, organ index (kidney, testis and thymus) and histopathology examination had significant differences (P < 0.01, 0.05) between method I and method II. In conclusion, the compound factors modelling method better conforms to the symptom of diarrhoea model caused by SKYD. This new modelling method provides a basis for studying on TCM astringents warming and tonifying the spleen and kidney, relieving diarrhea.

6.Related reproductive issues on male autosomal dominant polycystic kidney disease.

Autosomal dominant polycystic kidney disease (ADPKD) is a most common inherited renal disease, about 50% with a family history, although the exact etiology not yet clear. To date, ADPKD, a multisystem disorder without effective preventive and therapeutic means, has been shown to be detrimental to human health. Recent studies show that severe oligoasthenozoospermia, necrospermia, immotile sperm, azoospermia, epididymal cyst, seminal vesicle cyst, and ejaculatory duct cyst found in male ADPKD patients may lead to male infertility, though the specific mechanisms remain unknown. Structural anomaly of spermatozoa, defect of polycystin, mutation of PKD genes, and micro-deletion of the AZF gene could be the reasons for the higher incidence of abnormal semen quality in male ADPKD patients. Assisted reproductive techniques can increase the chances of pregnancy, whereas the health of the offspring should be taken into consideration. This article presents an overview of reproductive issues concerning infertile male ADPKD patients from the perspective of the morbidity, pathophysiological mechanism, diagnosis, and management of the disease.

8.Survey of acupuncture and moxibustion for clinical treatment of renal diseases.

In order to understand survey of medication combined with acupuncture and moxibustion for clinical treatment of renal diseases, clinical application and the mechanisms of acupuncture and moxibustion for treatment of renal diseases were summarized by electric retrieval of literature from 1982 to 2007. It is indicated that acupuncture and moxibustion can increase human immunity, reduce urinary protein, improve renal function, antagonize the side-effects of glucocorticoid hormones, etc. and medication combined with acup-moxibustion has the advantages of convenience, lower cost, safety, no adverse effects, etc.

9.Changes of renal cortex lipid peroxidation in renal injury induced by cisplatin in rats.

AIMTo study the relativity of the renocortical lipid peroxidation with renal tubules structure damage in renal injury induced by cisplatin in rats.
METHODSFemale Wistar rats were randomly divided into NS group, CDDP(I) group, CDDP(II) group and CDDP(III) group. All rats were injected via the tail vein with NS or cisplatin and NS qd in five days. The changes in content of Scr, BUN and MDA, the activity of SOD and GSH-Px of the renal cortex were measured. Alkaline phosphatase of renal tubular epithelia was stained by histochemistry and the slices of renal cortex were observed.
RESULTSThe contents of Scr and BUN of CDDP groups were significantly higher than those of NS group (P < 0.01). The content of renocortical MDA was significantly higher than that of NS group (P < 0.05). The activities of renoconical SOD and GSH-Px were lower than those of NS group (P < 0.05). The content of MDA, activities of renocortical SOD and GSH-Px with the content of Scr and BUN were significantly correlative. Alkaline phosphatase of renal tubular epithelia cells was losed largely and renal tubular epithelia cells were denaturative and necrotic partly in sections.
CONCLUSIONThe damage of renal cortex was correlative with its lipid peroxidation. The injury of renal cortex became heavier with cisplatin dose increased.

10.The investigation of macrophage infiltration in the early phase of ischemic acute renal failure in mice.

BACKGROUND/AIMS: Inflammation plays a key role in ischemic acute renal failure (ARF). The present study investigated the infiltration of macrophages in the early phase of ischemic ARF in mice. METHODS: Ischemic ARF was induced by renal clamping for 22 min, while the control mice underwent sham surgery (no clamping). The serum creatinine and blood urea nitrogen (BUN) levels were measured in the control and post-ischemia mice. Immunofluorescence staining was used to measure the number of CD 11b-positive cells in the kidney tissue sections to determine the amount of post-ischemic macrophage infiltration. Lipo-Cl2MBP (clodronate) for macrophages depletion was injected via a tail vein 5 d before ischemia induction and again 2 d before ischemia induction. RESULTS: The study found that the post-ischemia mice had higher levels of serum creatinine and BUN at 16 and 24 h compared to the controls. Immunofluorescence staining showed there were more macrophages in the post-ischemic tissue at 2, 8, 16 and 24 h compared to the control tissue, and that most of these macrophages were located in the outer medulla. The mice treated with clodronate prior to ischemia induction were found to have lower levels of serum creatinine compared to those mice that weren't treated with clodronate. CONCLUSIONS: There was significant infiltration of macrophages from the early phase of ischemic ARF, and this peaked at 16-24 h. Macrophage depletion using clodronate was protective against ischemic ARF.
