Objective To study the effect of H_2O_2 on the cultured melanocytes and the relationship between oxidative stress and vitiligo.Method Melanocytes were cultured with H_2O_2,the proliferation and melanogenesis of melanocyte were analysed by MTT and NaOH assay.Result H_2O_2 can inhibite the proliferation and melanogenesis of melanocytes,and the inhibition was concentration and-time-dependent.Conclusion Oxidative stress can inhibite the proliferation and melanogenesis of melanocytes,but oxidative damage can not induce vitiligo.

Objective Through researching the name,etiology,pathogenesis and therapeutic methods and prescriptions of the breast cancer in ancient medical documents to provide the theoretical basis and syndrome differentiation thinking for the treatment of breast cancer clinically.Methods By disease name searching,content analysis was used to screening the ancient literature that associated with breast cancer.The pathogenesis,treatment and medicine used of breast cancer in these ancient literatures were summarized.Results The stagnation of Qi and phlegm,blood stasis,and vital deficiency are important factors for breast cancer formation,while the deficiency of vital qi and viscera deficiency are important pathogenesis of breast cancer.Prevention before the onset of disease,eliminating pathogenic factors at the early period of the disease,supplement vital qi and eliminating pathogenic vital qi in the middle stage,and supplementing vital qi at the late stage of the disease should be emphasized.Meanwhile soothing liver to regulate mode and depression should also be paid attention.Conclusion The recognition of breast cancerby TCM and advantage of TCM in treating this disease need further literature researches.

Objective To study the role of psychological factors in the onset of psoriasis,to investigate the role of monoamine substances in the course of psychological factors-associated psoriasis,and to reveal the psychoneurologic pathogenesis of psoriasis.Methods The psychological factors were assessed with life events scale and questionnaire of A character.The serum levels of norepinephrine(NE),dopamine(DA)and5-hydroxyindoleacetic acid(5-HIAA)were detected with high-performance liquid chromatography(HPLC).Results It was shown that there was an overall tendency of weak A character and moderate depression and anxiety in psoriasis patients.The concentrations of NE,DA and5-HIAA were higher in psoriatic patients than those in normal controls(P
4.The curative effect of music-regulated laser therapy and the quality of life of patients with primary hyper-tension

Objective To assess the therapeutic effect of music- regulated laser therapy on mild and moderate primary hypertension (PH) so as to provide clinical evidence for the development of an effective non-drug therapy for hypertension. Methods One hundred mild and moderate PH patients were divided into obser-vation ( n = 50 ) and control groups ( n = 50 ) according to their stage of hypertension. There were two phases of treatment with 6 weeks in each phase for a total of 12 weeks. In the first phase, patients in the observation group received music-regulated laser therapy while the control group received drug therapy. In the second phase, the treatments were reversed, with the observation group receiving the drugs. The blood pressure and quality of life (QOL) of all the patients was evaluated after each phase. Results Before therapy there were no significant differences in average blood pressure or QOL between the groups. After the first phase, systolic blood pressure (SBP) and diastolic blood pressure (DBP) had been reduced significantly in both groups, but the decrease was significantly greater in the control group. The score on each item of the QOL evaluation was not significantly different between the two groups. After the second phase, blood pressure had decreased further in the observation group, but no significant change was observed in the control group, and there was no significant difference in blood pressure between two groups. The scores on each item of the QOL evaluation were not significantly different between the two groups, but average somatic symptoms, healthy and pleasant feelings, task perform-ance and life satisfaction were significantly ameliorated in both groups. Conclusions Music-regulated laser therapy can lower the blood pressure of mild and moderate PH patients effectively. Compared with antihyperten-sion drugs, music-regulated laser therapy provides a weaker effect in lowering blood pressure, but plays an e-quivalent role in improving the QOL of patients.
5.Biocompatibility of carboxymethyl chitosan membranes with human skin melanocytes

Objective To study the biocompatibility of carboxymethyl chitosan (CMCS) membrane with melanocytes from healthy human skin,and to investigate the feasibility to transport and carry melanocytes by using CMCS membrane.Methods CMCS membrane was prepared by a casting method combined with a glutaraldehydebased cross-linking method.Melanocytes were isolated from the foreskin of healthy men,and subjected to primary culture and subculture.The third-passage melanocytes were classified into two groups to be cultured on the CMCS membrane (test group) or traditional culture plates (control group).Methyl thiazolyl tetrazolium (MTT) assay was performed to evaluate the proliferative activity of melanocytes,and a sodium hydroxide-based lysis method to determine melanin content.HMB45 staining was conducted,and tyrosinase activity was estimated for melanocytes.Results Inverted microscopy showed that melanocytes were evenly distributed on the CMCS membrane with a normal shape.The melanocytes adherent to the CMCS membrane stained positive for anti-HMB45 monoclonal antibody.The growth curve of the melanocytes on the CMCS membrane,which was obtained from MTT assay,demonstrated that CMCS membrane could support the normal growth of melanocytes.No significant difference was observed between the test group and control group in melanin content (0.083 ± 0.015 vs.0.066 ± 0.008,t =2.38,P > 0.01) or tyrosinase activity (0.234 ± 0.083 vs.0.241 ± 0.061,t =0.23,P > 0.05).Conclusion CMCS membrane can maintain the normal biological activity of melanocytes and have good biocompatibility with skin melanocytes.
6.Evidence summary of health workers′ self-protection during aerosol-generating procedures in patients with acute respiratory infectious disease

Objective:To evaluate and summarize the best evidence of health workers respiratory protection during aerosol-generating procedures in patients with acute respiratory infectious diseases.Methods:We searched EBM Guidelines, Essential Evidence Plus, Dynamed, UpToDate, JBI, BMJ, Clinical Key, Cochrane Library, NICE, AARC, PubMed, EMbase, CKNI, Wanfang to collect related literature including guidelines,evidence summary, recommended practices, standards, consensus and systematic reviews.Results:A total of 20 articles were included, including 10 guidelines, 1 consensus, 2 standards, 2 evidence summaries, and 5 systematic reviews. 24 best evidences including aerosol-generating procedures, training of protective equipment using, principles of protective equipment using, protective equipment using, aerosol operating environment control and medical staff self-monitoring were summarized.Conclusion:The study integrated the best evidence of healthcare workers respiratory protection during aerosol-generating procedures in patients with acute respiratory infectious disease. It is recommended to combine the current status of institutional protection resources and clinical practice experience to promote the conversion of the best evidence to clinical practice.
7.Patterns of Frequency Domain Indexes of Surface Electronic Signals of Adolescent Idiopathic Scoliosis Patients' Paraspinal Muscles

Objective To investigate the patterns of frequency domain indexes of surface electronic signals of adolescent idiopathic scoliosis (AIS) patients' paraspinal muscles by using surface electromyographic (sEMG) techniques .Methods 25 AIS patients enrolled, 7 males, 18 females, ages range from 11 to 21 years old. All of the enrolled patients undertook the Biering Sorensen test(BST) and the object-lifting test, a Finland made type ME3000P sEMG instrument was applied to record the electronic activities of paraspinal muscles(convex /concave) of all subjects, and the frequency domain indexes: median frequency(MF),mean power frequency(MPF), zero crossing rate(ZCR)were analyzed.Results The wave amplitudes and scales of paraspinal muscles electronic frequency domain indexes(MF, MPF, ZCR) were lower than the other tested positions when recorded at the zone of apex vertebrae, and MFslope, MPFslope and ZCRslope all showed a linear degressive tendency as the exercise time was extended.Conclusion Paraspinal muscles at the zone of apex vertebrae have low fatigue durabilities and more likely to be exhausted. sEMG ought to be one of the objective examinations used to evaluate the differences of electronic activities of paraspinal muscles(convex /concave) of AIS patients, and may have a promising value in clinical practice.
8.Application of DNA microarray in the identification of Candida spp. and mutations of ERG11 gene resulting in fluconazole resistance

Objective To investigate the performance of DNA microarray in identifying 6 common Candida spp. and validating ERG11 mutations resulting in fluconazolc-resistance in Candida albicans. Methods Oligonucleotide probes were designed and synthesized targeting the species-specific sequence in the internal transcribed spacer 2 (ITS2) region of rDNA of Candida albicans, Candida tropicalis, Candida glabrata, Candida dubliniensis, Candida parapsilosis and Candida krusei, as well as 6 sequences embracing the following mutations respectively in ERG11 gene leading to fluconazole-resistance, i.c., T541C, A 1090G, C1361T, G1537A, G1547A, and T1559C, then arranged onto a chip. Twelve 50-base-pair oligonucleotides were artificially synthesized based on the above specific sequences, and utilized to hybridize with the DNA microarray. Thirty-lbur Candida strains, including 29 C. albicans, 1 Candida tropicalis, 1 Candida glabrata,1 Candida dubliniensis, 1 Candida parapsilosis and 1 Candida krusei, were detected with microarray. Genomic DNA was extracted from these tested strains and underwent multiple PCR for the amplification of ITS2 region and ERGI 1 gene. Sequencing was performed to analyze the sequence of ERG11 in 29 strains of C. albicans and the results were compared with those of DNA microarray hybridization. Results Multiple PCR successfully produced ITS2 fragment of 307-415 bp from all the 34 strains, as well as ERG11 fragment of 1712 bp from 29 C. albicans strains. DNA microarray hybridization offered the same results in species identification of the 34 strains with their given information, as well as in mutation detection of the 29 strains of C. albicans with ERG11 sequencing results. Also, the 6 synthesized oligonucleotides containing the muta- tions were identified precisely as T541C, A1090G, C1361T, G1537A, G1547A, and T1559C, and the 6 species specific oligonucleotieds were identified correctly as C. albicans, C. tropicalis, C. glabrata, C. dubliniensis, C. parapsilosis and C. krusei Both the sensitivity and the specificity of the microarray were 100%. Conclu- sion DNA microarray is a quite reliable method to identify Candida spp. and fluconazole resistance-associ- ated mutations in the ERG11 gene of C. albicans.
9.The effect and safety analysis of arsenic trioxide combined with all-trans retinoic acid in treating patients with acute promyelocytic leukemia

ObjectiveTo observe the effect and safety of arsenic trioxide (ATO) combined with all-trans retinoic acid (ATRA) in treating patients with acute promyelocytic leukemia (APL).Methods Eighty-three cases with APL treated for the first time were divided into two groups by random digits table method:observation group with 48 cases was received combination induction treatment of ATO and ATRA,control group with 35 cases was treated with combination induction treatment of ATRA and chemotherapy.The clinical effect and adverse reaction between two groups were compared.ResultsThe effective rate and early death rate were 100.0%( 48/48 ) and 0 in observation group,97.1%(34/35 ) and 2.9%( 1/35 ) in control group,which had no significant difference between two groups(P > 0.05 ).The incidences of bone marrow suppression,infection,liver and kidney damage,cardiac toxicity and gastrointestinal symptoms were 8.3% (4/48),10.4% (5/48),12.5% (6/48),6.2% (3/48) and 18.8% (9/48) in observation group,while 97.1%(34/35),65.7%(23/35),45.7%(16/35),37.1%(13/35) and 100.0%(35/35) in control group,which had significant differences between two groups (P < 0.05).ConclusionCombination treatment of ATO and ATRA in APL has an obvious effect and few adverse reaction,which can be applied in clinic.
10.A case of cutaneous phaeohyphomycosis caused by Phialophora verrucosa

A 16-year-old male presented with a 11-year history of progressively enlarging erythema and crusting on the right cheek.Physical examination revealed an irregularly shaped,sharply marginated,dark erythematous patch sized 6 cm x 10 cm and plaques with mild verrucous proliferation.There were strip-like scar at the margin of lesions and multiple ulcers measuring 0.5 to 1 cm in diameter with firm crusts.No small jellycolored nodules were observed.Direct microscopy of multiple scrapings under the crusts showed many light brown,septate,branching and irregular hyphae.Olivaceous-black woolly colonies grew at 25 C and 35 C on Sabouraud's dextrose agar and potato dextrose agar; flask-shaped conidiogenous cells with funnel-shaped collarettes and ellipsoidal conidia arranged in flower-like shape were observed microscopically.PAS staining showed numerous septate and branching hyphae,pseudohyphae and yeast-like cells.There was a 99.73% similarity in the species-specific rDNA sequence between the isolate and phialophora verrucosa standard strain CDC-B2152.The patient was diagnosed with cutaneous phaeohyphomycosis caused by Phialophora verrucosa.The lesion subsided after treatment with amphotericin B and itraconazole,but recurred after drug withdrawal.Itraconazole and terbinafine were administered for the retreatment of this patient.
