1.The effect of chinese medcine Xiao Bi Ling(XBL) on chondrocyte prliferation in vitro

Objective In order to study the effect on chondrocyte proliferation and the mechanism of treating osteoarthritis(OA) by XBL,the experimant is carried out.Method The first- subcuiture chondrocytes are implanted into two 96- foramen culture plates and sixteen culture flasks.After 24 hours in cubation 8 groups are divided into as follows:No 1 group(control)5% serum DMEM No,2~ 7 groups(XBL)5% serum,0.5% 、 1% 、 2% 、 4% 、 8% 、 16% XBL- DMEM are added respectively.NO 8 group no cell incubation.After culture of 24 and 48 hours ,two plates are detected for viablilty and proliferation respectively by MTT method(Light absorption value A),cell total protein is assessed by comassie Brilliatnt G- 250 colormetric assay.Result 2% ~ 8% XBL can promote the proliferation and protein synthesis of chondrocyte.Conclusion XBL can protec degradation from Matrix metallo protzinase and decrease the impairment of chondrocyte.
2.Incidence of depression in patients with advanced malignancy and its effect on quality of life

Objective To investigate the incidence of depression in patients with advanced malignancy and its effect on quality of life.Methods Patients with advanced malignancy were evaluated with Beck depression and quality of life questionnaires.The correlation of Beck depression scoring with quality of life was analyzed.Results A total of 230 in-patients with advanced malignancy were enrolled in this study and 213 copies of valid questionnaire were collected.95.3 % (203/213) patients had depression.The Beck depression scoring had positive correlation with body,role,mood,cognition,society,vomiting,pain,dyspnea,agrypnia and appetite (all P < 0.05),and had negative correlation with total health factors (r =-0.296,P < 0.01).Conclusion Depression is prevalent in patients with advanced malignancy and correlated with quality of life.Therefore,psychological interventions are needed to relief depression and improve quality of life.
3.Evidence summary of health workers′ self-protection during aerosol-generating procedures in patients with acute respiratory infectious disease

Objective:To evaluate and summarize the best evidence of health workers respiratory protection during aerosol-generating procedures in patients with acute respiratory infectious diseases.Methods:We searched EBM Guidelines, Essential Evidence Plus, Dynamed, UpToDate, JBI, BMJ, Clinical Key, Cochrane Library, NICE, AARC, PubMed, EMbase, CKNI, Wanfang to collect related literature including guidelines,evidence summary, recommended practices, standards, consensus and systematic reviews.Results:A total of 20 articles were included, including 10 guidelines, 1 consensus, 2 standards, 2 evidence summaries, and 5 systematic reviews. 24 best evidences including aerosol-generating procedures, training of protective equipment using, principles of protective equipment using, protective equipment using, aerosol operating environment control and medical staff self-monitoring were summarized.Conclusion:The study integrated the best evidence of healthcare workers respiratory protection during aerosol-generating procedures in patients with acute respiratory infectious disease. It is recommended to combine the current status of institutional protection resources and clinical practice experience to promote the conversion of the best evidence to clinical practice.
4.Expression of WT1 gene in acute leukemia and its significance

Objective To explore the relationship between WT1 and prognosis of patients with acute myeloid leukemia (AML) or acute lymphoblastic leukemia (ALL), and to evaluate the possibility of WT1 as a potential marker for monitoring the minimal residual disease (MRD). Methods Bone marrow mononuclear cells from 58 patients with primary AML, 32 patients with primary ALL, 40 patients with AML-complete remission (CR), 28 patients with ALL-CR and 31 patients with trilineage hyperplasia (control group) were collected. Real-time fluorescent quantitative PCR method was used to detect the expression of WT1 in all patients. The expression threshold of WT1 in each group was established. WT1 copy number/ABL copy number ratio×100%denotes the relative expression level of WT1 gene. Results Median relative expression level of WT1 in the control patients was much lower than that in primary AML patients [0.026%(0-0.240%) vs. 20.880 % (3.550 %-48.500 %), Z=-7.74, P<0.000 1]. Relative expression level of WT1 between AML-CR patients [0.102%(0-5.380%)] and primary AML patients had significant difference (Z=-8.34, P<0.000 1). Moreover, the relative expression level rate of the first course in AML patients with higher WT1 expression level (>20.880 %) was 60.7 % (17/28), while the CR rate was 76.7 % (23/30) in those with lower WT1 expression. WT1 expression was increased dramatically in recurrent AML patients. Relative expression level of WT1 was significantly higher in primary ALL patients [0.350 % (0.021 %-10.780 %)] compared with that in the control group Z=-2.58, P<0.05. There was no significant difference in relative expression level of WT1 between ALL and ALL-CR patients [0.038 % (0-2.800 %), P=0.065]. Conclusion WT1 expression level in AML patients is relatively high, which could be used as an effective index of prognosis evaluation and MRD monitoring for AML patients, but not for ALL patients.

Objective To investigate the relationship among stressful life events ,social support and depression through researc‐hing a large number of Chinese army men ,then discovered the social psychological mechanism of the depression .Methods 11 ,400 officers and soldiers were tested with three scales ,the center for epidemiologic studies depression scale ,life events scale and social support scale .The differences between groups were determined with the variance ,SPSS 18 .0 software and AMOS7 .0 were used to analyse and build model .In addition ,we built the path model of life events and social support on depression with AMOS 7 .0 soft‐ware .Results (1)There is a significant positive correlation between Chinese soldiers depression levels and life events (r=0 .382 , P<0 .01) .(2) There is a significant negative correlation between Chinese soldiers depression levels and the social support (r= -0 .379 ,P<0 .01) .(3) It was established the path model of life events ,the social support and depressive symptom level .Con‐clusion Depressive symptom is related to the military life events ,which may predict the occurrence of the depression .Moreover ,so‐cial support ,a protective factor for mental health ,can relieve the depression of officers and men .
6.The effect and safety analysis of arsenic trioxide combined with all-trans retinoic acid in treating patients with acute promyelocytic leukemia

ObjectiveTo observe the effect and safety of arsenic trioxide (ATO) combined with all-trans retinoic acid (ATRA) in treating patients with acute promyelocytic leukemia (APL).Methods Eighty-three cases with APL treated for the first time were divided into two groups by random digits table method:observation group with 48 cases was received combination induction treatment of ATO and ATRA,control group with 35 cases was treated with combination induction treatment of ATRA and chemotherapy.The clinical effect and adverse reaction between two groups were compared.ResultsThe effective rate and early death rate were 100.0%( 48/48 ) and 0 in observation group,97.1%(34/35 ) and 2.9%( 1/35 ) in control group,which had no significant difference between two groups(P > 0.05 ).The incidences of bone marrow suppression,infection,liver and kidney damage,cardiac toxicity and gastrointestinal symptoms were 8.3% (4/48),10.4% (5/48),12.5% (6/48),6.2% (3/48) and 18.8% (9/48) in observation group,while 97.1%(34/35),65.7%(23/35),45.7%(16/35),37.1%(13/35) and 100.0%(35/35) in control group,which had significant differences between two groups (P < 0.05).ConclusionCombination treatment of ATO and ATRA in APL has an obvious effect and few adverse reaction,which can be applied in clinic.
7.The characteristics of the fear emotion enhanced regulation of the soldiers with different mental quality levels

ObjectiveTo investigate the characteristics of autonomic nervous response of armymen with different mentalqualityuse the enhanced emotion regulationstrategies during fear emotion processes.Methods Mental Quality Questionnaire of Armymen was used to select participants from 818 armymen,screening out 30 armymen with low mental quality,30 armymen with moderate and 30 armymen with high.With elicitation of fear emotion,the participants adopted enhance emotion regulation strategies (cognition emphasis and emotion catharsis),and their blood volume pulse( BVP),skin conductance( SC ),normalized high frequency spectral component(HFnorm),standard deviation of all normal intervals(SDNN) and the recovery time of BVP and SC were recorded.Results ( 1 ) The different mental quality of army men activity in the autonomic physiological changes were not significant differences in resting state.(2) When using the cognition emphasis,the SC of different mental quality of armymen were reduced,and low mental quality of armymen( ( -0.67 ± 1.05 )μ.Ω),moderate mental quality of armymen ( ( - 0.49 ± 0.75 ) μΩ) had larger SC than high mental quality of armymen ( ( - 2.16 ± 2.18 ) μΩ ) ;high mental quality of armymen( ( - 2.17 ± 8.96) nU ) had lower HFnorm than the control( ( 6.96 ± 7.08 ) nU ).When using emotion catharsis,high mental quality of armymen ( ( 1.95 ± 6.58 ) nU ) had lower HFnorm than the control( (6.96 ±7.08)nU).(3)When using the cognition emphasis,low mental quality of armymen ((65.50 ±43.43 )s) had longer recovery time of BVP than high mental quality of armymen( (26.50 ± 25.72)s).When using emotion catharsis,low mental quality of armymen( (71.50 ± 48.36)mm Hg)had longer recovery time of BVP than high mental quality of armymen ( ( 21.00 ± 35.50 ) mm Hg) ; low mental quality of armymen ( ( 103.50 ± 38.01 ) s)had longer SC than moderate( (63.50 ±48.59)s) and high mental quality of armymen( (61.50 ±33.09) s).Conclusion The ability to regulate sympathetic in low mental quality group is weaker than the high mental quality group,although sympathetic can be suppressed when enhanced emotion regulation are adopted.

The surgical treatment of hepatocellular carcinoma have advanced greatly in recent years: associating liver partition and portal vein ligation for staged hepatectomy can bring hope and relief to patients with advanced liver cancer and less future liver remnant; the application range of laparoscopic hepatectomy and robotic hepatectomy were amplified; hepatectomy following anatomic or non-anatomic direction should be decided by particular situations; the precise preoperative assessment of liver reserve function ensured the success of extended hepatectomy; the further discussion of United Network for Organ Sharing criteria for liver transplantation made the appearance of University of California at San Francisco, Up-to-seven and Hangzhou criteria; bridge therapy can decrease tumor progression and the dropout rate from the liver transplantation waiting list; downstaging treatment is used in selected patients with more advanced liver cancer who are beyond the accepted transplant criteria to acquire the chance of liver transplantation and increase survival rates.

9.Long Non-Coding RNA TUG1 Promotes Proliferation and Inhibits Apoptosis of Osteosarcoma Cells by Sponging miR-132-3p and Upregulating SOX4 Expression.

PURPOSE: Long non-coding RNA taurine upregulated gene 1 (TUG1) is reported to be a vital regulator of the progression of various cancers. This study aimed to explore the exact roles and molecular mechanisms of TUG1 in osteosarcoma (OS) development. MATERIALS AND METHODS: Real-time quantitative PCR was applied to detect the expressions of TUG1 and microRNA-132-3p (miR-132-3p) in OS tissues and cells. Western blot was performed to measure protein levels of sex determining region Y-box 4 (SOX4). Cell viability was assessed using XTT assay. Cell apoptosis was evaluated using flow cytometry and caspase-3 activity detection assays. Bioinformatics analysis and luciferase reporter experiments were employed to confirm relationships among TUG1, miR-132-3p, and SOX4. RESULTS: TUG1 was highly expressed in human OS tissues, OS cell lines, and primary OS cells. TUG1 knockdown hindered proliferation and induced apoptosis in human OS cell lines and primary OS cells. Moreover, TUG1 inhibited miR-132-3p expression by direct interaction, and introduction of miR-132-3p inhibitor partly abrogated the effect of TUG1 knockdown on the proliferation and apoptosis of OS cells. Furthermore, SOX4 was validated as a target of miR-132-3p. Further functional analyses revealed that miR-132-3p inhibited proliferation and induced apoptosis of OS cells, while this effect was greatly abated following SOX4 overexpression. Moreover, TUG1 knockdown suppressed proliferation and promoted apoptosis by upregulating miR-132-3p and downregulating SOX4 in primary OS cells. CONCLUSION: TUG1 facilitated proliferation and suppressed apoptosis by regulating the miR-132-3p/SOX4 axis in human OS cell lines and primary OS cells. This finding provides a potential target for OS therapy.

10.Development and reliability, validity analysis of overgeneral autobiographical memory questionnaire

Objective:To develop overgeneral autobiographical memory questionnaire(OGMQ) and test its reliability and validity.Methods:Through literature search, expert opinions and group discussion the initial items of the questionnaire were formed. A random cluster method was adopted to select 1 911 college students for the preliminary test.Totally 1 840 valid questionnaires were obtained, and 920 of which were assessed for exploratory factor analyses and 920 of which were assessed for confirmatory factor analyses and reliability and validity analysis. Thirty subjects with depression and 30 normal subjects were selected to test its criterion validity.Results:The questionnaire consisted of 19 items, two dimensions (construction, elaboration) and four factors (spatial-temporal factor, control factor, context factor, emotion factor). Exploratory analysis extracted 4 factors and 19 items, KMO (Kaiser-Meyer-Olkin) = 0.905, Bartlett test: χ 2=4 728.134, df=171, P<0.001.Confirmatory analysis showed that the factor loads of spatio-temporal factor and control factor in the construction dimension were 0.770 and 0.539 respectively, while the factor loads of context factor and emotional factor in the elaboration dimension were 0.911 and 0.800 respectively. The questionnaire had good internal consistency reliability (Cronbach α=0.870), structural validity (χ 2/ df=3.550, RMSEA=0.053, GFI=0.944, CFI=0.970, NFI=0.958, IFI=0.970) and criterion validity ( r(OGMQ, AMT)=0.870, r(OGMQ, BDI)=0.507), and it was able to effectively distinguish between depressed and normal groups (AUC=0.795). Conclusion:The OGMQ has satisfactory validity and reliability, and meets the needs of psychometrics, which can be used in depression-related research and practice.
