Object To study the HPLC fingerprints and establish a sensitive and specific method for controlling the quality of Radix Paeoniae Rubra (RPR) Methods The HPLC method was applied for quality assessment of RPR All 18 samples collected from different places were determined A Hypersil C 18 column (200 mm?4 6 mm, 5 ?m) was used with the mobile phase being acetonitril 0 025 mol/L H 3PO 4: THF (9 5∶90 5∶1 25), flow rate being 0 8 mL/min, detecting wavelength being 254 nm, the column temperature being at room temperture Results This method had a good repeatability and reproducibility, and the ratio of peaks' area of different habitat samples were different Conclusion The method is suitable to differentiate RPR form different sources conveniently, and can be used as a quality control method for this herb
2.An in vivo study of the inhibitory effect of erythromycin on wear particles induced osteolysis

[Objective]In present study an in vivo wear particles induced mouse calvarial osteolysis model was used to investigate the inhibitory effect of erythromycin over wear particles induced osteolysis.[Method]Twenty-four male eight-week-old C57BL/J6 mice were allocated into 4 groups: PMMA group receiving 30 mg PMMA particles implantation onto the calvariae,PMMA+2EM group receiving 30 mg PMMA particles implantation and EM 2mg/kg ip injection,PMMA+10EM group receiving 30 mg PMMA particles implantation and EM 10mg/kg ip injection,control group receiving sham operation.Seven days later,mice calvariae were harvested for pathology analysis.[Result]Middle suture area in the control group was 0.079 mm2?0.011 mm2,in PMMA group 0.335 mm2?0.129 mm2.In PMMA+2EM group the area was 0.094 mm2?0.019 mm2,PMMA+10EM group was 0.091 mm2?0.028 mm2.Osteoclasts number in osteolysis area in the control group were 5.3?1.0,in PMMA group 19.2?5.3,PMMA+2EM group was 6.6?1.1,PMMA+10EM group 6.1?1.9.Compared with control group,PMMA particles induced significant osteolysis(P
3.Investigation of Phthalates in Disposable Plastic Products

Objective To explore the kinds and levels of phthalates leaching from disposable plastic products. Methods Samples of peritoneal dialysis solution, blood preservative solution, infusion instruments, preservative film, disposable plastic bags and water in plastic bottles were analyzed for phthalates by RP-HPLC after liquid-liquid extraction and/ or solid phase extraction. Results Di(2-ethylhexyl) phthalate (DEHP) was leached from all medical instruments, the maximum level of which reached 77.51?g/ L. Di-n-butyl phthalate was leached from disposable plastic bags, the level of which reached 91.45?g/ kg. Phthalates were not found in samples of preservative film and water in plastic bottles. Conclusion As DEHP leaching from the medical instruments might directly enter the human body, attention should be paid to its health hazards.
4.THE EFFECT OF STRESS-RELAXATION PLATE FIXATION ON THE REMODELING OF THE CORTEX UNDER PLATE

Objective To explore the influence of stress-relaxation plate(SRP) fixation on the remodeling of cortex under plate. Methods Twenty-eight New Zealand rabbits were used in this study, the bilateral tibia were osteotomized in the middle and fixed with SRP (experimental group) and rigid plate (control group) respectively. The scanning electron microscopy was used to observe the bone remodeling process from 2 to 48 weeks after operation. Results There was cortex osteoporosis beneath plate in different degree in both experimental and control groups before 8 weeks, it showed as the disorganization of collagen fiber structure and formation of resorption cavities. In comparison, the osteoporosis degree in experimental group showed milder than that of the control group. After 12 weeks, the resorption cavities became smaller, and the structure of collagen fibers became regular with the alignment parallel to the long axis of cortex. In contrast to the experimental group, the bone osteoporosis under plate of control group exacerbated continuously. Conclusion Without removal of the bone plate, SRP fixation not only reduce the degree of plated bone osteoporosis, but also make the osteoporosic bone return to normal.
5.Effects of Stress- Relaxation Plate on the Disorganization and Reparation of Regional Bone Structure

ObjectiveTo study the influence of stress- relaxation plate on disorganization andrepair of the cortex under the plate. MethodsA washer made of viscoelastic polyethylene was placedbetween the screw and the screw hole of a conventional stainless rigid plate(RP) to form a stress- relax-ation plate(SRP). Both SRP and RP were applied to the osteotomized New Zealand rabbit tibia, thefracture healing process of either fixing with SRP or RP(control) was put under comparative study bypolarized light microscopy, in situ hybridization of collagen mRNA and immunohistochemistry tech-nique from 2 to 36 weeks postoperatively. ResultsThe study of plated bone remodeling showed thatthe degree of cortex osteoporosis beneath the plate was similar between the SRP and RP group 12 - weekpostoperatively. The organization of the bone structure in the SRP group happened later and milder thanthat of the RP group, and the repair process began 12 weeks after implantation. So the resorption cavi-ties became smaller and the structure of collagen fibers became well oriented along, and the osteoblastslying on the surface of resorption cavities expressed and synthesized type Ⅰ collagen 8 to 12 weeks afterimplantation. The changes above increased significantly in most cavities by 36 weeks. No expression andsynthesis of any kind of collagen could be observed during 12 to 36 weeks after implantation in the RPgroup. ConclusionWithout removal of the bone plate, the SRP fixation not only reduces the degreeof plated bone osteoporosis, but also makes the disorganized bone structure return to normal by the ex-pression and synthesis of type Ⅰ collagen mRNA of osteoblasts lying on the surface of resorption cavities.
6.Toxicokinetics of Phthalates in Male Rabbits

Objective To study the toxicokinetics of phthalates in male rabbits. Methods Concentrations of DEHP or DBP in plasma of 6 healthy male rabbits were determined by RP-HPLC after constant rate infusion of 0.5 g/kg DEHP or DBP(IV). The toxicokinetic parameters were computed by 3P87 program. Results The toxicokinetic model of DEHP and DBP were both first-order elimination and two-compartment model with constant rate infusion. The main toxicokinetic parameters of DEHP were as follows: distribution phose t1/2(?)=0.101 h; elimination phase t1/2(?)=12.701 h; CLs=0.013 g?kg-1?h-1. The main toxicokinetic parameters of DBP were as follows: t1/2(?)=0.441 h; t1/2(?)=31.311 h; CLs=0.021 g?kg-1?h-1. Conclusion DEHP and DBP were both first order elmination and two-compartment medol with constand rate intusion(IV). DEHP and DBP could be rapidly eliminated in male rabbits.
7.NKT cells in peripheral blood of patients with esophageal carcinoma and its clinical significance

Objective To explore the expression, as well as the proportion of subsets of the natural killer (NK) T cells in peripheral blood of patients with esophageal carcinoma before and after surgery, and provide ideas and experimental basis for immune treatment through tests. Methods Using CD3, CD56, CD4 and CD8 antibody to study the NKT cells and its subsets. Peripheral NKT cell subsets in 59 patients with esophageal carcinoma were analysed by flow cytometer. Results With the ratio of CD+3 CD+56 CD+8/CD+3 CD+56 CD+4 gradually increased, the proportion of phase Ⅲ- Ⅳ patients with esophageal carcinoma is gradually decreased between the groups. One-way ANOVA analysis showed that there were differences between the groups (P <0.05). The ratio of CD+3 CD+56 CD+8/CD+3 CD+56 CD+4 was in Non-linear related to CD+3 CD+56 CD+4NKT cells; CD+3 CD+56 CD+8; NKT cells were positively correlated with CD-3 CD+56 NK cells and negatively correlated with CD+3 T cells.Conclusion The ratio of CD+3 CD+56 CD+8/CD+3 CD+56 CD+4 may relate to tumor burden, and is helpful to determine the extent of disease in patients with esophageal carcinoma and prognosis.
8.Effect of melittin on apoptosis and necrosis of U2 OS cells

OBJECTIVE: To study the effect of melittin on apoptsis and necrosis of osteosarcoma cell line U2 OS in vitro. METHODS: Osteosarcoma cell line U2 OS was treated with melittin. The growth and proliferation was observed by MTT assay and cell counting, and the necrosis was estimated by Trypan blue staining. The cell apoptsis, Fas and Apo2. 7 expression were detected by cytometer. RESULTS: The data showed that melittin could inhibit the proliferation of U2 OS dose-dependently at 16 and 64 mg/L. Cell apoptsis was detected by cytometer, when the cells were treated by 16 mg/L and 32 mg/L of melittin respectively, and the percentages of Fas and Apo2. 7 positive cells were increased. CONCLUSION: Melittin inhibits the proliferation of osterosarcoma cell line through up-regulating Fas expression and inducing apoptsis.
9.Application of chemical pattern recognition to quality control of Radix Paeoniae Alba

Objective With generalization and steadiness,a new evaluation model by Integrating Non Linear Features extraction algorithm with artificial neural networks(ANN) used for pattern recognition of quality control of Radix Paeoniae Alba was proposed in this paper.Methods The HPLC data from 29 samples with different quality were proceeded with nonlinear kernel principal component analysis(KPCA) and an improved Back propagation algorithm of ANN.The extract characteristics was fed into BP neural networks as input elements for pattern recognition.In the meantime,the processing data,the optimal numbers of hidden layers,the numbers of hidden nodes,excitation functions,and over-fitting,etc. were discussed wholly so that standardization networks was designed without jamming.Results As recognition ratio was 100%,the pattern recognition of quality control of Radix Paeoniae Alba was established successfully by trained networks and predicted results.Conclusion Integrating KPCA algorithm with ANN for pattern recognition of quality control of Radix Paeoniae Alba has been proved to be available.
10.Experimental study of effect of stress-relaxation bone plate on fracture healing.

OBJECTIVE: To study the change of the stress shi elding rate of stress-relaxation plate in vivo and its influence on fracture he aling. METHODS: The diaphyses of bilateral tibias in 70 New Zealand ra bbits were osteotomized and fixed with stress-relaxation plates (SRP, the SRP g roup) and rigid plates (RP, the RP group), respectively. The fracture healing pr ocess in these 2 groups was investigated by radiography, light and polarized l ight microscopy and biomechanical test at 2 to 48 weeks postoperatively. RESULTS: Early after fixation the stress shielding rate was abo ut 70% in both groups. While in the SRP group the stress shielding rate decrease d gradually as time passed, which was significantly lower than that of the RP gr oup (P<0.05) by the end of the 8th postoperative week, and stabilized at the level of about 27% at 36-48 weeks after fixation. Abund ant external callus associated with the formation of cartilaginous callus could be observed in the SRP group at 2-4 weeks postoperatively. The transformation o f the callus into the lamellar bone began at 8-12 weeks, the collagen gradually arranged in order, and the mechanical nature of the united bone was gradually s trengthened, too. In the RP group, the external callus was scarce at the early s tage of fracture healing, and the callus remodeling at the late stage of fractur e healing was dominated by bone absorption. The ultimate bending strength (UBS) was only 57.95% of that of the normal by 48 weeks. CONCLUSIONS: The decrease of the stress shielding rate of SRP i n vivo was well interrelated with the time of fixation. The application of SRP c ould promote the callus formation and bone reconstruction thus to favor the reco very of the mechanical strength of the united bone.
