1.Arsenic disulfide induced apoptosis and concurrently promoted erythroid differentiation in cytokine-dependent myelodysplastic syndrome-progressed leukemia cell line F-36p with complex karyotype including monosomy 7.

OBJECTIVEAcute myeloid leukemia progressed from myelodysplastic syndrome (MDS/AML) is generally incurable with poor prognosis for complex karyotype including monosomy 7 (-7). Qinghuang Powder (, QHP), which includes Qing Dai (Indigo naturalis) and Xiong Huang (realgar) in the formula, is effective in treating MDS or MDS/AML even with the unfavorable karyotype, and its therapeutic efficacy could be enhanced by increasing the Xiong huang content in the formula, while Xiong huang contains > 90% arsenic disulfide (As2S2). F-36p cell line was established from a MDS/AML patient with complex karyotype including -7, and was in cytokine-dependent. The present study was to investigate the effects of As2S2 on F-36p cells.
METHODSCell proliferation was measured by an 3-(4,5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide (MTT) assay. Cell apoptosis was identified by Annexin V-staining. Cell viability was determined by a propidium iodide (PI) exclusion. Erythroid differentiation was evaluated by the expression of cell surface antigen CD235a (GpA).
RESULTSAfter treatment with As2S2 at concentrations of 0.5 to 16 μmol/L for 72 h, As2S2 inhibited the proliferation of F-36p cells. The 50% inhibitory concentrations (IC50) of As2S2 against the proliferation of F-36p cells was 6 μmol/L. The apoptotic cells significantly increased in a dose-dependent mannar (P<0.05). The cell viabilities were significantly inhibited by As2S2 dose-dependent in a dose-dependent manner (P<0.05). Significant increases of CD235a-positive cells were concurrently observed (P<0.05) also in a dose-dependent manner.
CONCLUSIONSAs2S2 could inhibit proliferation and viability, induce apoptosis, and concurrently promote erythroid differentiation dose-dependently in F-36p cells. As2S2 can inhibit proliferation and viability, induce apoptosis, and concurrently promote erythroid differentiation in cytokine-dependent MDS-progressed human leukemia cell line F-36p with complex karyotype including -7. The data suggest that QHP and/or As2S2 could be a potential candidate in the treatment of MDS or MDS/AML even with unfavorable cytogenetics.

2.Effectiveness of Mao-bushi-saishin-to in Treating Common Cold Syndrome. Controlled Comparative Study Using the Sealed Envelope Method.

To investigate the effectiveness of Maobushisaishin-to (traditional Japanese herbal medicine; Tsumura TJ-127) in treating the common cold, a clinical comparison between Maobushisaishin-to and a general common cold drug was conducted using the sealed envelope method. The study involved 83 patients in the TJ-127 group and 88 patients in the general cold drug group. No differences in age, gender or the period from the onset of the disease to the beginning of treatment were observed between the two groups.
The results indicated greater than moderate improvement in 81.9% of the TJ-127 group, compared with 60.3% of the compound cold drug group (p<0.01). Further analysis of symptom diaries kept by the patients indicated that TJ-127 provided more rapid relief for symptoms such as fever, feeling feverish, coughing and phlegm than did the general cold drug. No side effects were observed for the TJ-127. These results suggest that TJ-127 is effective in the treatment of the common cold.
3.Questionnaire Survey on the Pharmacy Students’ Attitudes and Opinions following the Dementia Supporter Education Lecture

Although pharmacy students receive professional education regarding dementia, it is the current state that many pharmacy students cannot learn the countermeasures to treat people with dementia. We conducted the “Otani dementia supporter education lecture” which improved a lecture by Caravan Mate targeting pharmacy students, and subjected the students to a questionnaire survey after the lecture. One hundred and forty-nine students participated and there were most percentages of third grades by 40.9%. After attending the lecture, most of the students opined that one can help a patient with dementia by understanding the patients’ feeling. It was the answer obtained by the lecture for the students to ensure that a pharmacist responds to consultations and requests from their family with a dementia patient. The first information that the students additionally wanted to know about dementia pertained to a method for the early detection of dementia. The results of text mining on the correspondence for patient with dementia showed that categories of “dementia patient,” “correspondence,” “understanding the patients’ feeling,” and “tact,” independently had strong mutual relations. Moreover, the results of text mining on the feedbacks obtained for the lecture showed that categories such as “dementia patient,” “When close person and one were,” “A chance was obtained,” and “correspondence” independently had significant mutual relations. Students who participate in this lecture understand that it is necessary to contact with dementia patients appropriately and feel that there is a need to become a presence that can support dementia patients as a pharmacist in the future. Therefore, this lecture was effective in motivation to correspondence to a person with dementia.
