A qualitative and quantitative analysis of alprazolam in biological samples by TLC scan is described in this paper. The distribution of alprazolam in poisoned rabbits was studied. It is showed that 4 hours after a dose of 21mg/kg, the concentration of the drug in liver, spleen, kidney; lungs, heart,brain, blood,bile and urine of poisoned rabbits were 19. 6 ? 6. 1, 3.31?0. 5, 3. 5? 0. 3, 0. 4? 0.1, 0. 4 ?0. 1, 1. 6 ? 1. 8, 4. 0 ? 1. 3, 20. 4 ? 8.0 and 8. 6 ? 2. 4ug/ml. Alprazolam is not well-distributed in poisoned rabbits. Blood, bile, urine and liver are better samples for analysis of alprazolam poisoning victim.

OBJECTIVE:To isolate and identify the major constituents-steroidal saponins from Di'ao xinxuekang.METHODS:The constituents were separated and purified using normal-pressured and pressurized silica gel column chromatography technology,and the structures were identified based on the physico-chemical property and spectral analysis.RESULTS:Two steroidal saponins were obtained from Di'ao xinxuekang and their structures were identified as dioscin(1)and pseudoprodioscin(2).CONCLUSION:Compound 1 and 2 were demonstrated to be two major constituents in Di'ao xinxuekang.This finding is of great importance for the quality control and evaluation of Di'ao xinxuekang(Chinese Medicine).

Objective To study the ability of adhesion, proliferation and differentiation of human adipose mesenchymal stern cells,which were cultured in PLGA,and thus to provide a basis for their in vivo implantation.Methods PLA was mixed with PGA in a ratio of 1∶ 1,and PLGA was obtained by insert moldling with organic solvents.In vitro, human adipose mesenchymal stem cells were adhered to PLGA.The attachment and the proliferation of human adipose mesenchymal stem cells in PLGA were analysed by scanning electron microscope,and the morphological change of the cells was investigated before and after induction.Results Human adipose mesenchymal stem cells were able to attach, proliferate and secrete extracellular matrix in porous PLGA (pore size:100～400 pm) with porosity of 85 %, and to differentiate into round lipoblast-like cells under the induction of adipose medium.After 14 days, the particulate of PLGA was overlaid by extracellular matrix .Conclusion Human adipose mesenchymal stem cells have the capacity to adhere to PLGA and proliferate, indicating that PLGA can be used as a carrier for human adipose mesenchymal stem cells in adipose tissue engineering.

Objective To study the mutagenic effect of reclaimed water in Tianjin. Methods Reverse phase C-18 solid-phase extraction (RP-C18SPE) was used for the extraction of target compounds from water samples. Ames test was used to detect mutation. Results PAHs were detected in both low-water and high-water period,including fluorene,phenanthrene,naphthalene,acenaphthylene and acenaphthene. In the high-water period, the total concentration of 16 kinds of PAHs was 1 777.9 ng/L and 1 380.1 ng/L in untreated reclaimed water and treated reclaimed water respectively, in the low-water period,it was 2 225.1 ng/L and 1 827.1 ng/L respectively. Ames test showed that both untreated reclaimed water and treated reclaimed water could induce mutation of TA98 and TA100 strains with a dose-response relationship. Conclusion The reclaimed water used in Tianjin still contains PAHs of a certain level, which have mutagenic effect.

AIM: To study the effect of partial liquid ventilation on the ultrastructure of pulmonary surfactant(PS) system in endotoxin-induced acute lung injury METHODS: The effect of partial liquid ventilation on the ultrastructure of PS system in endotoxin-induced acute lung injury was observed with electronmicroscope histochemistry. RESULTS: There was ultrastructure impairment of PS system in endotoxin-induced acute lung injury, the pulmonary surfactant layer was discontinuous, lamellar bodies and plasmosomes in type Ⅱ pneumonocytes vacuolated, and a few of them were even necrotized and disrupted into the alveolar space. The pulmonary surfactant layer was still continuous, the vacuolation of lamellar bodies and plasmosomes in type Ⅱ pneumonocytes was little with partial liquid ventilation. CONCLUSION: Partial liquid ventilation can lessen the impairment of PS system in endotoxin-induced acute lung injury in rats.

Object To compare the content in corilagin of Phyllanthus urinaria L. harvested from ten different regions of China in different months. Methods The content of corilagin was determined by UV spectrophotometry, detective wavelength is 270 nm. Results The content of corilagin in P. urinaria from Hainan and Guizhou province was the lowest, 0 58%. The highest content was given by the plant from Xi'an zone, Shanxi province, 1 78%. The best harvest time is in August and September, content of corilagin is 1 24%. Conclusion The difference of the corilagin contents in P urinaria . harvested from different regions and in different harvest time in the same region is great.

AIM: To determine the contents of corilagin in Erodium stephanianum Willd. from different collection month and to define the best collection time for this herb. METHODS: The contents of corilagin in each samples of E. stephanianum were determined by HPLC. RESULTS: The linear regression of corilagin was obtained in the range of 0.22～1.10?g. The average recovery was 98.6% with a RSD of 0.63%. CONCLUSION: The content of corilagin in E. stephanianum collected in August is the highest among the samples throughout the collection months.

AIM:To explore whether cyclosporine A (CsA) can regulate the expression of nometastatic gene23 H1 type (nm23-H1) in human choriocarcinoma Bewo cells,in order to seek new proof of treating trophoblast diseases.METHODS:The Bewo cells were divided into two groups. The vehicle control group,and the CsA group with different concentrations from 10-2 ?mol/L to 10 ?mol/L. The effect of CsA on the transcription of nm23-H1 was determined by reverse transcription-polymerase chain reaction (RT-PCR) after cultured for 48 h and protein level of nm23-H1 was determined by In-cell Western after cultured for 72 h.RESULTS:Compared to the vehicle group,CsA significantly downregulated the transcription and translation of nm23-H1 in a dose-dependent manner from 10-2 ?mol/L to 10 ?mol/L,and the inhibition reached its top when concentration of CsA was 1.0 ?mol/L (P

Objective: To evaluate mid-term outcomes for the application of homograft valve conduits in right ventricular outlfow reconstruction in patients with congenital heart disease. Methods: We retrospectively studied 122 patients who received right ventricular outlfow reconstruction by homograft valve conduits application in our hospital from 2007-10 to 2014-07. The patients were divided into different sets of groups, by surgical procedure: Ross group,n=38 and Non-Ross group,n=84; by median age: ≤6 years group,n=61 and >6 years group, n=61; by the type of valve conduits: Aortic homograft group,n=21 and Pulmonary homograft group,n=101; by the diameter of conduits: ≤19 mm group,n=31 and >19 mm group,n=91. The relationships between pre-operative conditions, different types of conduits and diameters to the prognosis were analyzed; the post-operative death, re-operation, free homograft valve conduits failure rates were followed-up in all patients. Results: The average follow-up time was (35.4 ± 22.2) months and 2/122 (1.6%) patients died during that period, the overall free conduits failure rates at 1, 5 and 7 years post-operation were 94.2%, 81.2% and 75.4% respectively. The free conduits failure rates in Pulmonary homograft group at 1, 5, 7 years post-operation were 96.2%, 86.1%,79.9% and in Aortic homograft group were 80.0%, 59.7%, 59.7% respectively,P=0.011; in Ross group were 96.4%, 89.0%, 89.0% and in Non-Ross group were 91.3%, 78.3%, 67.1% respectively,P=0.045. While the age, conduits diameter, cyanosis and re-operation had no statistical meaning to free conduits failure rates, allP>0.05. Conclusion: Application of homograft valve conduits had good mid-term outcomes in right ventricular outflow reconstruction in patients with congenital heart disease, while the long-term effects should be further emphasized in clinical practice.

OBJECTIVE： To confirm the efficacy and safety of latanoprost in treating glaucoma.METHODS： In a double blind， randomized control clinical trial， we compared the efficacy and adverse drug reactions of once daily topically applied 0.005％ latanoprost with those of twice daily 0.5％ timolol for 12 weeks in patients with open angle glaucoma or ocular hypertension.RESULTS： The study included 46 patients（22 pts.randomized to latanoprost treatment， 24 pts.to timolol） ， 46 patients remained at the end of the study.Comparing with baseline diurnal intraocular pressure（IOP） ， the IOP reduction（mean±standard deviation） achieved with latanoprost（7.86±2.39） mmHg， （31.1％， P<0.001），and timolol（6.24±2.58）mmHg （24.9%，P<0.001），the difference between the two groups（1.62mmHg） being significant（P<0.01）. Two patients treated with latanoprost had foreign body sensation. No other ocular and systemic adverse reactions related to the two drugs were found. CONCLUSION: The results of this study demonstrated that 0.005% latanoprost applied once daily is well tolerated and more effective in reducing IOP than 0.5% timolol applied twice daily. Thus, latanoprost has the potential for becoming one of the ideal antiglaucoma drugs.
