1.Antagonistic action of Jiunaoning injection aganist oxygen/glucose-deprived and reperfusion injury-induced cultured rat cortical neurons

AIM: To determine whether Jiunaoning injection has protective effects on oxygen/glucose-deprived and reperfusion injury-induced neurons. METHODS: Various concentrations of Jiunaoning injection (0.5-5 mL/L) were used to observe their effects on cultured rat cortical neurons induced by oxygen/glucose-deprived and reperfusion injury in various time points. The neuronal metabolic rate and viability were assessed by using 3-(4,5-dimethylthiazol)-2, 5-diphenyl-tetra zoliumbromide (MTT) and lactate dehydrogenase (LDH) assay. RESULTS: Jiunaoning injection enhanced the neuronal metabolic rate in a dose-dependent manner in the range from 0.5 to 5 mL/L, and Jiunaoning injection (1.5-2.5 mL/L) enhanced the neuronal metabolic rate, decreased the cell death rate and depressed LDH leak rate significantly. CONCLUSION: Jiunaoning injection has an affirmative protective effect on oxygen/glucose-deprived and reperfusion-induced neuronal injury.
2.Effect of Methylprednisolone on expression of aquaporin 1 in lung tissue of pulmonary edema rats induced by adrenaline

Objective To observe the changes in the lung aquaporin 1 (AQP1) expression in adrenaline-induced pulmonary edema(PE),and the effect of Methylprednisolone (MP) on its expression.Methods Fifty Wister rats of 1-month old were randomly divided into 5 groups (10 rats in each group):control,adrenaline PE,MP A,MP B,and MP C groups,respectively.Control group animals were treated with 0.27 mL 9 g/L saline;PE group was given 2.7 mg/kg adrenaline (1 ∶ 1 000) by intraperitoneal injection;MP A,MP B and MP C groups rats were intraperitoneally injected 10 mg/kg,20 mg/kg,and 30 mg/kg MP intraperitoneally immediately after intraperitoneal injection of adrenaline,respectively.The morphology changes in the lungs were observed with HE staining,and lung wet/dry weight (W/D) was measured.The levels of AQP1 mRNA,AQP1 protein,and AQP1 distribution in the lung tissues were detected by using real time-polymerase chain reaction,Western blot,and immunohistochemical method.Results (1)PE group exhibited a faster breathing rate,and double lung volume increased significantly;there was a visible hemorrhagic distribution in the lung surface and cross section,endotracheal filled with white or pink foam liquid.(2) The W/D of rats in PE group was higher than that of the control group (6.50 ± 0.53 vs.4.59 ± 0.36,P < 0.05).(3) Pathological grading of PE group (3.80 ± 0.42) increased significantly compared with that of the MP A group (3.30 ± 0.48),MP B group (2.30 ± 0.68) and MP C group (1.20 ± 0.42),and there were significant differences (all P < 0.05).(4) Immunohistochemistry showed that the expression of AQP1 in PE group (1.20 ± 0.79) was reduced compared with that of the control group (4.20 ± 1.03),and there were significant differences (all P < 0.05).(5) The levels of AQP1 mRNA and AQP1 protein (0.12 ± 0.43 and 0.20 ± 0.04) were significantly lower than those of the control group (0.90 ± 0.32 and 0.60 ± 0.15),and there were significant differences (all P < 0.05);compared with PE group,AQP1 mRNA and AQP1 protein of each group with MP treatment showed the highest values (MP A group:0.17 ±0.06 and 0.32 ±0.04,MP B group:0.39 ±0.13 and 0.37-±0.09,MP C group:0.61 ±0.21 and 0.44 ± 0.07) (all P < 0.05).Conclusion The expression of AQP1 reduced in adrenaline-induced PE rats.MP could improve the expression of AQP1,and significantly ameliorate the PE and bleeding.
3.The effect observation of comprehensive intervention on postoperative pain of patients with rectal disease

Objective To explore the effect of comprehensive intervention on postoperative pain of patients with rectal disease. Methods A total of 200 patients with postoperative pain after the treatment of anorectal perianal disease from May 2015 to May 2016 were randomly divided into two groups with 100 cases each. The control group was treated with drugs and usual nursing, the observation group were adopted drugs and comprehensive nursing intervention. The improvement of pain, psychological states and the quality of sleep were compared between two groups. Results The VAS pain scores at 4, 6, 12, 24, 48 h after treatment was (2.1 ± 0.6), (3.3 ± 0.4), (3.5 ± 0.3), (2.3 ± 0.5), (1.9 ± 0.5) points in the observation group, and (3.0 ± 0.5), (5.1 ± 0.6), (6.2 ± 0.6), (5.7 ± 0.8), (5.8 ± 0.5) points in the control group, and the difference was statistically significant (t=8.539-38.806, P < 0.05). The Self-rating Anxiety Scale was (20.32 ± 6.16) points in the observation group, and (35.58 ± 7.43) points in the control group, and the difference was statistically significant (t=41.188, P<0.05). The sleep quality, the amount of sleep , sleep time, sleep efficiency of Pittsburgh Sleep Quality Index Questionnaire scores was (0.91±0.28), (0.86±0.2), (0.83±0.27), (0.59±0.31), (0.62±0.27), (0.58±0.41), (4.39±1.79) points in the observation group, and (1.61± 0.88), (1.32 ± 0.75), (1.59 ± 0.89), (1.34 ± 0.58), (1.36 ± 0.45), (1.29 ± 0.86), (8.51 ± 3.55) points in the control group, and the difference was statistically significant (t=4.557-17.740, all P<0.05). Conclusions The comprehensive intervention on postoperative pain relief in patients with anal disease is significant, it is beneficial to relieve the pain response, improve sleep quality, and achieve physical and psychological comfort, and has a positive effect to clinical.
4.Clinical comparison of two microinvasive surgery for giant benign prostatic hyperplasia

Objective To compare the clinical effect between retropubic extraperitoneal laparoscopic prostatectomy with prostatic urethra preservation and transurethral resection of the prostate (TURP) for giant benign prostatic hyperplasia (BPH).MethodsThe clinical data and follow-up of 128 cases of giant BPH were analyzed retrospectively.Seventy-two cases underwent TURP (TURP group) and 56 cases underwent retropubic extraperitoneal laparoscopic prostatectomy with prostatic urethra preservation ( laparoscopic group ).The operation time,blood loss,gland mass excision,bladder washing time,catheterization time,hospital stay,hospital cost,international prostate symptoms score (IPSS) and quality of life questionnaires (QOL),maximum flow rate (MFR),residual urine volume (RUV) were compared between two groups.ResultsThe bladder washing time,catheterization time,hospital stay in laparoscopic group were less than those in TURP group[0 d vs.(2.8 ± 1.2) d,(2.3 ± 0.6) d vs.(5.2 ± 1.5) d,(4.2 ± 0.5) d vs.(7.5 ±0.5) d],gland mass excision in laparoscopic group was more than that in TURP group [(100.2 ±25.4) g vs.(85.6 ± 15.5) g],there were significant differences between two groups(P< 0.05).There was no significant difference in the operation time,blood loss,hospital cost between two groups (P > 0.05).There were significant differences in IPSS,QOL,RUV and MRF before and after 3,6 months treatment for two groups [ laparoscopic group:(9.1 ± 3.4),(7.5 ± 2.5 ) scores vs.(27.5 ± 5.8) scores,( 1.8 ± 1.1 ),( 1.6 ± 0.8)scores vs. (5.5 t0.5) scores,(26.5 ± 11.5),(22.4 ± 12.6) ml vs. (145.0 ±48.0) ml,(17.6 ±8.4),(20.2 ± 5.4) ml/s vs.(8.3 ± 3.5) ml/s;TURP group:(9.2 ± 3.8),(7.8 ± 2.2) scores vs.(28.5 ± 5.4) scores,( 1.9 ± 1.2),( 1.7 ± 0.6) scores vs.(5.0 ± 0.5 ) scores,(28.5 ± 12.9),(23.0 ± 11.7) ml vs. ( 155.0 ± 47.0) ml,( 17.8 ± 9.2),( 19.8 ± 4.5 ) ml/s vs.(7.2 ± 3.2 ) ml/s ] (P < 0.01 ),but there was no significant difference between two groups (P > 0.05).ConclusionsThe clinical effect of two microtrauma surgery are good.Laparoscopic technique is a feasible treatment option for patients suffered from giant BPH for which has the benefit of a quicker recovery,shorter hospital stay,less complications,no bladder washing.
5.Effects of endothelin-1 on the proliferation of human HCT-116 cells

Objective To investigate the effects of endothelin-1 on the proliferation of HCT-116 cells. Methods The expression of endothelin-1 and endothelin receptors A and B in the HCT-116 cells was detected by RT-PCR and immunohistochemistry method.The effects of endothelin-1 at different concentrations and action time points on HCT-116 cells were measured by MTT assay and were analyzed by variance analysis.Results The expression of endothelin-1 and endothelin receptors A and B at the gene and protein levels in HCT-116 cells was detected.Endothelin-1 had significant effects on the proliferation of HCT-116 cells.The proliferation peak of HCT-116 cells was reached when the concentration of endothelin-1 was 1×10-7mol/L and the action time was 48 hours.Conclusions Endothelin-1 promotes proliferation of HCT-116 cells in a dose-and time-dependent manner,and the process is mediated by endothelin receptors.
6.Clinical Assessment of Fluorescence in Situ Hybridization for Prenatal Diagnosis of Chromosomal Abnormalities

Objective:To investigate the clinical efficacy of fluorescence in situ hybridization(FISH)in prenatal diagnosis of chromosomal aneuploidy.Methods:FISH and karyotyping analysis were done in 120 samples of amniotic fluid from pregnant women.Results:FISH analysis results were consistent with those of karyotyping,the dectecting rate can reach 100%for chromosomal aneuploidies.There was no significant difference in the preference for FISH or karyotying for prenatal diagnosis when facing high matemal age,multi-indications and other factors(P>0.05).FISH was better when biochemical datafor down's syndrome were positive (P=0.029),however karyotyping was better when there was abnormal fetal ultrasound scan(P=0.000).Conclusions:FISH can detect the chromosomal aneuploidy quickly and accurately,especially when biochemical data for down's syndrome were positive.FISH could be the first choice for prenatal diagnosis in the third trimester of high-risk pregnancy.
7.The effect of infrared spectral analysis in analyzing of the chemical composition of renal staghorn ;calculi and its relationship with urinary tract infections

Objective To explore the effect of infrared spectral analysis in analyzing of the chemical composition of renal staghorn calculi and its relationship with urinary tract infections. Methods From June 2014 to June 2016, the clinical data of 186 patients with renal staghorn calculi were collected. The stone composition were analyzed by infrared spectroscopy and traditional chemical titration, and the stones infection were detected by microbial analysis system. The relation between stones infection, urinary tract infection and stone composition were analyzed. Results The results of infrared spectroscopy and traditional chemical titration in detecting renal staghorn calculi ingredient had no significant differences (P>0.05). In 186 patients, 56 patients (30.11%) was in infected group, and 130 patients (69.89%)was in non-infected group. The abnormal urine rate, urinary tract infection rate, medistream urine positive infection rate and cotton swabs positive infection rate in infected group were was significantly higher than those in non-infected group: 73.21%(41/56) vs. 50.77%(66/130), 19.64%(11/56) vs. 3.85%(5/130), 50.00%(28/56) vs. 6.15%(8/130), 67.86%(38/56) vs. 8.46%(11/130), P<0.01. The carbonate apatite stones rate and six water magnesium ammonium phosphate rate in infected group were significantly higher than those in non-infected group: 21.43%(12/56) vs. 5.37%(7/130), 57.14%(32/56) vs. 2.31%(3/130), P<0.01. The calcium oxalate rate and uric acid rate in non-infected group were significantly higher than those in infected group:50.00%(65/130) vs. 5.36%(3/56), 24.62%(32/130) vs. 1.79%(1/56), P<0.01. Conclusions Analysis of staghorn calculi ingredient caused by urinary bacterial infection with infrared spectroscopy is simple, reliable and easy to operate. It is important for postoperative infection prevention.
8.Effect of "Shen Yan Yi Qi Solution" on renal function and histopathology of rats suffering from an experimental nephrotoxic serum nephritis

AIM: To investigate the therapeutic effects of "Shen Yan Yi Qi Solution",a chinese Medicine,on nephrotoxic serum nephritis in the rat. METHODS: Nephrotoxic serum nephritis model of the rat was set up by the Clarke's method. RESULTS: Proteinuria and renal insufficiency occurred in all rats after nephrotoxic serum (NTS) injection,but symptoms in treatment group were reduced. At 2nd week,the typical linear deposition of IgG along the glomerular basement membrane (GBM) was observed by fluorescence microscopy. Mesangial hyperplasia and crescent formation in glomeruli were also observed by light microscopy. In treatment group,all the above lesions were alleviated. CONCLUSION: It demonstrated that "Shen Yan Yi Qi Solution" might alleviate the pathological change of glomeruli and improve renal function.

AIM: To observe the changes of glycemia and serum cholesterol and triglyceride in diabetic nephropathy rats and therapeutic effects of Xiaoke Keli Ji. METHODS: 3/4 nephrectomy was adopted firstly, three weeks later streptozotocin(STZ) was administered intraperitoneally to establish diabetic nephropathy model in rats. Animals were divided into four groups:model group, Xiaoke Keli Ji treatment group, positive control group and sham group. Changes of serum sugar and serum creatinine, cholesterol and triglyceride were examined at 1, 2, 3, 4, 5 weeks after STZ injection. Renal tissue samples were adopted at 6th week and studied by light microscopy. RESULTS: Model group demonstrated different degree of glomerular sclerosis. Lesions in treatment group were alleviated. Objective:To explore any effect of transcranial magnetic stimulation and peripheral magnetic stimulation of the mylohyoid muscle on dysphagia among stroke survivors.Methods:Sixty stroke survivors with dysphagia were randomly divided into a control group, a peripheral magnetic stimulation group, a central magnetic stimulation group, and a central + peripheral magnetic stimulation group, each of 15. In addition to routine swallowing training, the subjects were given the appropriate magnetic stimulation daily, five times a week for two weeks. Before and after the intervention, swallowing was evaluated using the Functional Oral Intake Scale (FOIS), Penetration-Aspiration Scale (PAS) and a functional dysphagia scale (FDS).Results:The average FDS, PAS, and FOIS scores of all four groups had improved significantly after the treatment. Improvement significantly greater than in the control group was observed in the average FOIS, FDS and PAS scores of the other three groups. The peripheral + central magnetic stimulation group showed the greatest average improvement.Conclusion:Combining mylohyoid muscle magnetic stimulation with magnetic stimulation of the cerebral cortex can significantly relieve dysphagia. It is more effective than conventional swallowing rehabilitation or either magnetic stimulation alone.
