1.Detection of human papillomavirus DNA genotypes in different populations and its clinical significance

Objective To analyze the detection results and clinical significance of human papillomavirus (HPV) DNA genotypes in the patients with condyloma acuminate (CA)and high risk (HR)population of CA and normal population of physical examination (PE).Methods 527 cases of CA patients (CA group),208 cases of HR (HR group)and 197 cases of normal PE (PE group)in the outpatient were selected.The human HPV DNA genotypes in 932 samples of different population were examined with HybriMax;the positive detection rates,the preponderance types of HPV and the multiple genotype infection in three groups were analyzed.Results The HPV positive rate in CA group was 98.29% and the preponderance types were HPV 6,11,33,and 52 genotypes.The HPV positive rate in HR group was 26.44% and the preponderance types were HPV 6,11,33,and 66 genotypes.The HPV positive rate in PE group was 5.58% and the preponderance types were HPV 6 and 11 genotypes.The difference in the positive detection rates of HPV between HR and PE groups had statistical significance (P < 0.05 ). The positive rates of multiple genotype infection in CA group and HR group were 36.43% and 5.77%,respectively,and there was significant difference (P < 0.01 ); the positive rate of double infection was the highest (18.41%), followed by the positive rate of triple heavy infection (11.20%) and the positive rate of septuple heavy infection (0.38%).Conclusion The HPV 6 and HPV 11 of low-risk HPV genotypes are the most common genotypes in different population. The HR group and PE group are all asymptomatic but the positive detection rate of HPV in HR group is higher than that in PE group. The positive rate of HPV multiple genotypes infection in CA group is higher than that in HR group,and the double infection or triple heavy infection are the most common infection.
2.Re-emergence of Schistosomiasis in Dali City after the Criteria of Transmission Control Were Met

Objective To analyze the epidemic situation on schistosomiasis in five years after reaching the criteria of transmission control in 1995 in Dali City. . Methods. Epidemiological data of schistosomiasis during 1995-2000 were collected and analyzed. . Results . Stool positive rate in the population was increasing yearly, new cases of infection including cases under 14 years old were detected each year. Acute infection occurred during 1996-1998 consecutively. Stool positive rate in cattle was unstable. Snail-ridden area increased significantly in 1996 and spots with infected snails stayed at a higher level during 1997-2000. . Conclusion . Schistosomiasis in Dali city has been re-emerging though it was once deemed under transmission control. Possible reasons include a low drug-taking rate though distributed, frequent movement of cattle population, and less activity on snail control.
3.Application of stable prosthesis in total knee replacement in elderly patients with rheumatoid arthritis

Objective To evaluate the short-middle term clinical effects of the fixed platform stabilized prosthesis in the treatment of the elderly patients with rheumatoid arthritis(RA).Methods The clinical data of 53 cases elderly RA patients who were admitted to the People's Hospital of Liuzhou from January 2009 to January 2013 and were treated with fixed platform and stable prosthesis in 62 knees were retrospective analyzed.Results Fifty-three patients were followed up for 24.0-43.0 months with average (25 ± 2.1)months.At the last follow-up,the degree of knee motion was improved significantly compared with the preoperative((116.0±6.0) ° vs.(46.0±7.0)°),the difference was significant(t=35.323,P<0.01).The HSS score was significantly improved compared with the preoperative ((85.3±7.6) points vs.(50.2±11.3)points),the difference was significant (t=4.002,P<0.05).At the last follow-up,35 cases (66.0%) of excellent,11 cases (20.8%) of good,and 7 cases (13.2%) of normal.After the operation,all patients obtained obvious improvement in joint pain,swelling,dysfunction,deformity and so on.Conclusion The fixed platform stabilized prosthesis is simple,effective,and less postoperative complications,and it is a better choice for the elderly patients with high flexion activity.
4.Research progress on the pharmacological effects and pharmacokinetics of linarin

Linarin, as a natural coumponent belongs to flavonoid glycoside, is widely existed in herbal plants such as chrysanthemum indicum and Mongolian flower, which has a variety of pharmacological effects, such as anti-inflammation, anti-cancer, liver protection, analgesia, antipyretic, anti-oxidation, anti-apoptosis, sedation and sleep, neuroprotection, preventing and treating hypertension, treatingdiabetes, preventing and treating osteoporosis, whitening, skin care and sunscreen. It is difficult to dissolve in water and has poor oral efficacy, but when combined with different substances or combined (forming phospholipid complex), its bioavailability can be improved, so as to improve its pharmacological efficacy.
5.Surgical treatment for lung cancer patients with poor pulmonary function.

OBJECTIVETo summarize surgical treatment of lung cancer patients with poor pulmonary function.
METHODSFrom 1991 to 1999, 181 lung cancer patients with poor pulmonary function underwent operation. The correlation between the results of preoperative pulmonary functional tests and the postoperative cardiopulmonary complications was analyzed by Chi-square test (chi(2)).
RESULTSIn 181 patients, pneumonectomy was done in 43, lobectomy in 118, partial lung resection in 16 and exploration in 4. The postoperative complication and mortality rates of the resection group were 42.3% (75/177) and 7.9% (14/177). The cardiopulmonary complication rates were 25.6%, 48.3%, 31.3% in pneumonectomy, lobectomy and partial lung resection. The morbidity and mortality rates of 8 patients who received preoperative chemotherapy and/or radiotherapy were 75.0% and 37.5%. The morbidity and mortality rates of 12 patients who had had a previous history of thoracotomy were 66.7% and 33.3%. In the present series, the 1-, 3- and 5-year survival rates were 71.1%, 42.2% and 31.1%. The 5-year survival rates of patients with stage I, II and III lesions were 55.0%, 25.0% and 0.
CONCLUSIONPreoperative spirometry is an important evaluation test for lung cancer patients with poor pulmonary function. It should be evaluated in combination with other pulmonary function tests such as CO(2) diffusion and cardiopulmonary excise tests, etc whenever possible. Patients with a history of thoracotomy, chemotherapy and radiotherapy should be carefully evaluated before operation to avoid high morbidity and mortality. Stage I and II lung cancer patients with poor pulmonary function can undergo operation if they have been well managed preoperatively and well taken care of with nursing care perioperatively.

6.OLC1 protein levels in plasma of patients with non-small cell lung cancer and its clinical application.

OBJECTIVEThe aim of this study was to detect the plasma concentration of OLC1 (overexpressed in lung cancer 1) protein as a potential cancer biomarker, and evaluating its clinical application value in the diagnosis of non-small cell lung cancer (NSCLC).
METHODSWe prepared OLC1 antibody with OLC1 full length protein, in 5-6-week old Bal B/c mice. Each mouse was immunized four times at a dose of 15-30 µg antigen protein, and the interval between two consecutive immunizations was two weeks. Antibody screening was made by ELISA and Western blot, and a double antibody sandwich ELISA kit was developed. We used this established ELISA kit to detect the plasma concentration of OLC1 protein in 281 NSCLC patients and 92 gender- and age-matched healthy controls. Area under the receiver operating characteristic curve (AUC) was used to evaluate the detection efficacy of OLC1.
RESULTSWe obtained 11 OLC1 monoclonal antibodies and successfully established the ELISA kit to detect the plasma concentration of OLC1 with a detection range from 1.95 ng/ml to 62.50 ng/ml. OLC1 concentration in the case group (124.69 ng/ml) was significantly higher than that in the control group (67.07 ng/ml, P < 0.001). In the scenario of distinguishing NSCLC from control group, AUC result was 0.69. When the cut-off was set at 67.72 ng/ml, the sensitivity and specificity was 84.4% and 51.1%, respectively. In term of distinguishing early lung cancer (IA) from normal controls, the AUC, sensitivity and specificity were 0.68, 77.8% and 54.4%, respectively.
CONCLUSIONThe plasma concentration of OLC1 protein is significantly elevated in NSCLC patients. OLC1 may be as a potential cancer biomarker applied in clinical diagnosis.

7.Improvement effect of curcumin combined with fecal bacteria transplantation on mice with ulcerative colities induced by DSS

Objective:To discuss the improvement effect of curcumin combined with fecal bacteria transplantation on the mice with dextran sulfate sodium(DSS)-induced ulcerative colitis(UC),and to clarify the related mechanism.Methods:Fifty mice were randomly divided into control,model,curcumin,fecal bacteria transplantation,and combination groups.Except for the mice in control group(given distilled water),the mice in the other groups were given distilled water containing 2%DSS to establish the UC models.The mice in curcumin group were gavaged with 0.4 mL of 60 mg·kg-1 curcumin solution once per day for 10 d;the mice in fecal bacteria transplantation group underwent enema with 0.2 mL of fecal bacteria suspension once per day for 10 d;the mice in combination group received the enema of 0.2 mL fecal bacteria suspension and gavaged with 0.4 mL of 60 mg·kg-1 curcumin solution.At the end of the experiment,the disease activity index(DAI)and colon macroscopic damage index(CDMI)of the mice in various groups were calculated;the morphology of colon tissue of the mice in various groups was detected by HE staining;the levels of interleukin(IL)-1β,tumor necrosis factor-α(TNF-α),IL-4,and IL-10 in colon tissue of the mice in various groups were detected by enzyme-linked immunosorbent assay(ELISA)method;the expression levels of occludin and zonula occludens-1(ZO-1)mRNA and proteins in colon tissue of the mice in various groups were detected by real-time fluorescence quantitative(RT-qPCR)and Western blotting methods.Results:The intestinal mucosal epithelial structure of the mice in control group was intact and continuous with regular glandular arrangement and without inflammatory cell infiltration or ulceration;the intestinal mucosal epithelial structure of the mice in model group exhibited loss of colonic mucosal epithelium,disordered glandular arrangement,reduced goblet cells,congestion and edema in mucosal and submucosal layers,and extensive infiltration of inflammatory cells with widespread small ulcers;the intestinal mucosal epithelial structure of the mice in curcumin,fecal bacteria transplantation,and combination groups exhibited relatively intact colonic mucosal epithelial structures with reduced inflammatory cell infiltration and ameliorated mucosal and submucosal congestion and edema.Compared with control group,the DAI and CDMI of the mice in model group were increased(P<0.05),the levels of IL-1β and TNF-α were increased(P<0.05),the levels of IL-4 and IL-10 were decreased(P<0.05),and the expression levels of occludin and ZO-1 mRNA and proteins were decreased(P<0.05);compared with model group,the DAI and CDMI of the mice in curcumin,fecal bacteria transplantation,and combination groups were decreased(P<0.05),the levels of IL-1β and TNF-α were decreased(P<0.05),the levels of IL-4 and IL-10 were increased(P<0.05),and the expression levels of occludin and ZO-1 mRNA and proteins were increased(P<0.05).Compared with curcumin group and fecal bacteria transplantation group,the DAI and CDMI of the mice in combination group were decreased(P<0.05),the levels of IL-1β and TNF-α were decreased(P<0.05),the levels of IL-4 and IL-10 were increased(P<0.05),and the expression levels of occludin and ZO-1 mRNA and proteins were increased(P<0.05).Conclusion:Curcumin combined with fecal bacteria transplantation can ameliorate the pathological damage in colonic tissue of the UC mice,inhibit the secretion of inflammatory factors,and promote the repaiment of intestin mucosa.
8.A Review of the Application of Propensity Scoring Method in Traditional Chinese Medicine Research

Propensity score can improve the efficiency and accuracy of clinical trials, and has been widely used in many fields such as comparison of efficacy evaluation, exploration of disease occurrence factors, and monitoring of adverse drug reactions. At present, the propensity scoe has application prospects in traditional Chinese medicine (TCM) monotherapy intervention, complex intervention and integrated Chinese and Western medicine intervention research, and it is a good auxiliary tool for real-world study. By analysing the cha-racteristics of clinical research in TCM, the authors believed that, firstly, the application of propensity score can acce-lerate the progress of clinical efficacy evaluation research in TCM, contribute to the collection of high-level clinical evidence, and promote the development and promotion of TCM therapies; Secondly, under the background of big data resources, propensity score has good application prospects in assisting TCM prescription analysis, drug safety and adverse reaction monitoring, and large-scale cohort studies in TCM; thirdly, in the process of clinical practice, propensity scoring method may face problems such as inappropriate method selection, incorrect judgement or measurement of confounding factors, and unreasonable interpretation or extrapolation of results.
