1.HSV AMPLICON-MEDIATED NEUROTROPHIN-3 TRANSDUCTION PREVENTS MOUSE SPIRAL GANGLION NEURONS FROM DAMAGE

Objective To determine whether NT 3 overexpression could abrogate DDP toxicity in vitro . Methods We constructed a herpes simplex virus(HSV) amplicon vector to transduce a c myc tagged version of rat NT 3. In the HSVnt 3myc vector, the chimeric neurotrophin cDNA was placed under the transcriptional control of the CMVIE promoter. The resultant vector was packaged utilizing the recently developed helper virus free method. Results Transduction of NT 3myc in cultured mouse cochlear explants at a multiplicity of infection(MOI) of 0 25 resulted in production of NT 3 up to 3?g/L over a 48 hours period. The mouse cochlear explants were transduced with HSVnt 3myc or HSVmiap(control vector expressing the reporter gene, murine intestinal alkaline phosphatase) for 48 hours and the exposed to cisplatin for 48 hours.The cochlear explants transduced with HSVnt 3myc had a significantly greater number of SGN survival than the control group. NT 3 powerfully enhanced the process length and the density of SGNs.Conclusion These data demonstrate that the neurotrophic cDNA transduction via HSV amplicon helper free virus system can attenuate the ototoxic action of DDP on organotypic culture. The potency of NT 3 in protecting spiral ganglion neurons from degenerating suggests that neurotrophins might be useful for the prevention or treatment of hearing disorders. [
2.Review and Analysis of Processing Technology and Active Ingredients of Medicinal Leech

Theancientandmodernprocessingmethods,activeingredientresearch,andqualitycomparisonbeforeandafterthe processing of medicinal leech were reviewed in the paper. Combined with the results of anticoagulation test on Whitmania pigra Whit-man, the article drew a conclusion that different varieties of medicinal leech had different active ingredients of anticoagulation, Whitma-nia pigra Whitman had active ingredients of anticoagulation with promising heat stability, and the processing technology should be dealt with according to the clinic use.
3.Comparison Study on Fingerprints of Liposoluble Components Between Salvia Yunnanensis Roots and Salvi-a Miltiorrhiza Roots

To study the fingerprints of liposoluble components in Salvia yunnanensis roots collected from different habi-tats, and compare with the fingerprints of Salvia miltiorrhiza. Methods: The fingerprints were determined by HPLC and the results were analyzed by a similarity evaluation system for chromatographic fingerprints of TCM ( Version 2004 A) . Results:The fingerprints of different samples of Salvia yunnanensis roots showed high similarity. And the characteristic peaks in the fingerprints of the liposoluble components were basically the same as those of Salvia miltiorrhiza, while the contents of several main effective components were lower than those in Salvia miltiorrhiza. Conclusion: The liposoluble components in Salvia yunnanensis roots show relatively high similarity with those in Salvia miltiorrhiza, however, there are obvious differences in the contents of the main constituents.
4.GC Fingerprints of High-yield Pinellia ternata from the West of Hubei Province

OBJECTIVE:To establish a fingerprint analysis method of Pinellia ternata from the west of Hubei province.METHODS:The samples of Pinellia ternata from different areas harvested at different time and processed by different methods were analyzed by GC method to establish their fingerprint data.RESULTS: The fingerprints of Pinellia ternata have been established using GC data of the chromatographic peaks of five phytosterols from various samples,and these samples had a similarity of between 0.877 and 0.999.CONCLUSION: The fingerprints of phytosterols of Pinellia ternata are stable thus can be used as a new method for the quality control of Pinellia ternata.

Objective To study the clinical monitoring of tacrolimus (FK506) area under the curve (AUC) of concentration-time after the first oral dose in kidney transplant recipients. Methods Sixteen kidney transplant recipients were treated with anti-lymphocyte globulin (ALG) and methyprednisolone (MP) for 3 days after operation.Then FK506 capsules were given orally at the same dose,0.075 mg/kg,on the third day.The pharmacokinetic monitoring of FK506 were conducted as follows.FK506 concentrations were measured by ELISA at 0.5,1.0,1.5,2.0,3.0,5.0,8.0,12.0 hours after the first oral dose. The data of FK506 pharmacokinetics were calculated using 3P87 pharmacokinetic procedures and SPSS 8.0. Results AUC of concentration-time of the first dose ranged from 44.40 ?g?h -1 ?L -1 to 158.01 ?g?h -1 ?L -1 (mean, 92.23?34.97 ?g?h -1 ?L -1 ). 7.Quantitative Analysis of Glucose Injection Content by Near Infrared Spectroscopy

Objective: To quantitatively analyze the content of glucose in glucose injection by near infrared spectroscopy to control the quality of the product.Methods: A quantitative model was established by near infrared reflectance spectroscopy and the injection came from pharmaceutical enterprises with different concentrations of glucose and the solution samples came from laboratories with different concentrations of glucose.The liquid sample accessories were selected, a quantitative model was established by a partial least squares method, and the effect of environmental temperature on the model was studied as well.Results: According to the quantitative model, the correlation coefficient reached up to 0.999 3, and RMS deviation (RMSECV) was 0.077 4.In the verification test, the results of various liquid formulas containing glucose were similar to those of the laboratory results.The prediction error was less than 5% within the temperature range of 20-35℃.Conclusion: The model of near-infrared partial least squares (PLS) can accurately predict the content of glucose in glucose products without sodium chloride, and can be used for the quality control of glucose intermediates and the finished products in the large infusion manufacturers
8.IEF Electrophorogram of protein in Rhizoma Pinelliae

Objective To discriminate Rhizoma Pinelliae from its relative species by electrophoro-gram of protein.Methods Using isoelectrofocusing(IEF) technology to compare the electrophorogram of tuber of Pinellia ternata produced in different habitats and from its relative plants then determining the pH values.Results The IEF electrophorogram of Rhizoma Pinelliae was stable with eight clear characteristic protein bands and had obvious differences from the electrophorogram of the tubers of its relative plants.Conclusion The IEF electrophoresis of Rhizoma Pinelliae can be used as the distinguishing basis.The PI value of the characteristic protein of P.ternana offers the basic parameter for separation and purification of the protein in Rhizoma Pinelliae.
9.Studies on Adsorption Technology with Macroporous Resin of Piceid from Rhizoma Polygoni cuspidati

Objective To study the adsorption technology of piceid on macroporous resin. Methods The extraction condition of piceid, the types of macroporous resin, the concentration of the sample of Rhizoma Polygoni cuspidati extract, the flow velocity of samples during adsorption were investigated and the leak curve was drawed to ascertain the optimal sample quantity. Results Resin D101 gave a better adsorption performance in the following technological conditions:the concentration of the sample of Rhizoma Polygoni cuspidati extract was 0.7774 mg/mL, the flow velocity of sample during adsorption 3BV/h, the optimal adsorption capacity of the resin was 10.88 mg/mL. Conclusion The procedure was simple, feasible and can be applied to industrial production.
10.Discussion on identification and phylogenetic relationships of medicinal plants in Selaginellaceae by NIRS

Objective:to study and solve some difficult problems in identification of medicinal plants in Selaginellaceae, and research the phylogenetic relationships among them.Methods:All the spectra were collected by using a Fourier transform near-infrared spectrometer equipped with a fiber-optic probe and a InGaAs detector for reflectance measurements.All the samples were scanned from 9 000 cm-1 to 4 000 cm-1,and each sample spectrum was obtained as an automatic mean of 32 times.The samples were measured 5 times.The average spectra obtained were transformed to first-order derivative spectra,and the latter were computerized with OPUS/INDENT operation software to conduct cluster analysis.Results:According to the information obtained from spectrograms and dendrograms,two types of S.heterostachys and two types of S.delicatula were identified as the same species respectively,and the variable type of the latter should be treated as a variety.S.tamariscina and S.pulvinata have very close relationship,indicating it is reasonable that they were applied as the same medicinal materials Juanbai.There is close phylogenetic relationship between S.moellendorffii and S.delicatula.This is also accord with the fact that they were used as the same folk herbal medicine.Conclusion:It shows that NIRS can be conveniently and quickly used for authentication of the medicinal plant species,resolving some correlative difficult problems,and inducing certain relationships between interspecies.
