This article analyzes the inspiration of the thought of "to govern by non-intervention" in modern hospital management from the following four respects. They are the objects of establishing healthy career, setting up the managing method of "go with the nature", the people-oriented flexible management pattern, and the leadership art of "To govern by non-intervention". It is beneficial for hospitals to take their social responsibilities, establish systemic management systems, explore human resources and improve the managers' qualifications. With all the above approaches accomplished, the management art of "To govern by non-intervention" will be achieved.
2.Polysaccharide from Gynostemma entaphyllum and its protection effect on plasmid DNA damage

AIM: To analyse monosaccharides of gynostema polysaccharide from Gynostemma pentaphyllum gathered from Pingli County,Shanxi Province and study the protecting effect of polysaccharide on plasmid DNA damage. METHODS: Gynostema polysaccharide was extracted with hot distilled water.After dialysis and deproteinization,polysaccharide in trifluoroacetic acid solution was hydrolyzed by 1-phenyl-3-methyl-5-pyrazolone,and then determined by HPLC.Tolerance of gynostema polysaccharide to the UV-photolysis of H_2O_2 was used to investigate the protecting effect. RESULTS: Polysaccharide consisted of mannose(Man),rhamnose(Rha),glucuronic acid(GlcUA),galacturonic acid(GalUA),glucose(Glc),galactose(Gal),xylose(Xyl) and arabonose(Ara).And it had the effect on protecting plasmid DNA from damage,showing a concentration dependent velationship. CONCLUSION: The method is simple and sensitive,the result is precise and believable.It provides scientific data for the use of Gynostemma pentaphyllum.
3.Testicular microlithiasis and its relation to varicocele on ultrasound findings of symptomatic men

Objective To study testicular microlithiasis(TM)on scrotal ultrasound findings in re-lation to the incidence of varicocele in males with scrotal symptoms.Methods Ultrasound finding of 8351 cases aged 40 days to 88 years with scrotal symptoms were reviewed.Results TM was found in 170 pa-tients(2.04%)and varicocele Was found in 1449 patients(17.35%)of symptomatic individuals.Varicocele Was found in TM and non-TM were 15.88%(27/170)and 17.38%(1422/8181)respectively,statistical anal-ysis revealed that there was no close relationship between TM and varicocele(x2=0.26,P>0.05)in symp-tomatic population. Conclusion TM is a comnlofl disease and has no direct correlation with varicocele.
4.Establishment of a new method based on nucleic acid functionalized GO for the rapid detection of Salmonella typhimurium carrying SSeC gene

Objective To establish a simple, efficient and low-cost method for the detection of Salmonella typhimurium carrying SSeC gene. Methods In this study, a nano-biosensor ( FAM-P/GO) was successfully established based on the noncovalent assembly of carboxy-fluorescein ( FAM)-labeled probe and graphene oxide ( GO) . The target gene at different concentrations and SSeC gene-harbored bacterium sam-ples were detected by the FAM-P/GO nano-biosensor to evaluate its sensitivity. The specificity of the estab-lished nano-biosensor was evaluated by using DNAs with mismatched base pairs and single-stranded DNAs ( ssDNAs) extracted from various species. Results The established strategy for SSeC gene detection showed a good linear range of 0. 05-1. 0 μmol/L (R2=0. 992 1) with a lower limit of 0. 05 μmol/L. Moreover, the lower detection limit for target bacterium samples was 103 CFU/ml and the fluorescence intensity increased linearly with the concentration from 103 CFU/ml to 108 CFU/ml. The signal-to-noise ( S/N) of the experi-mental group was much greater than that of the control group, which indicated that the establish method was highly specific. Conclusion The FAM-P/GO nano-biosensor was successfully established in this study, which provided a new and possible way for the rapid detection of Salmonella typhimurium harboring SSeC gene.
5.Clinical value of contrast-enhanced TVS in the assessment of invasion depth of endometrial cancer

Objective To discuss the diagnostic value of contrast-enhanced transvaginal sonography TVS in the diagnosis of myometrial invasion grade of endometrial cancer. Methods Total twenty-eight cases were studied by contrast-enhanced TVS,which were proved pathologically endometrial cancer in our hospital. The diagnosis of myometrial invasion grade of endometrial cancer by contrast-enhanced TVS were compared with pathologic results based on FIGO. Results The total coincidence of contrast-enhanced transvaginal sonography in detecting the depth of myometrial invasion was 78.6%. In the evaluation of no myometrial invasion,it showed the sensitivity of 71.4%,specificity of 85.7% and coincidence of 62.5%. In evaluation of superficial myometrial invasion,the sensitivity,specificity and coincidence was 73.7% ,77. 8% ,87. 5% ,respectively. The sensitivity in detecting deep invasion was 100. 0% ,the specificity was 96.0% ,while coincidence was 75.0%.There was no significant difference among the coincidence of various depth of myometrial invasion by contrast-enhanced TVS. Conclusion Contrast-enhanced TVS was valuable in the diagnosis of myometrial invasion grade of endometrial cancer.
6.miR-101 inhibits growth and invasion of ovarian cancer cells by targeting DNMT3A

Background and purpose:miR-101 has been reported to be down-regulated in gastric cancer, colorectal cancer, breast cancer as well as prostate cancer acting as a tumor suppressor gene. However, its function in ovarian cancer is still unknown. The aim of this study was to investigate whether miR-101 can suppress cell growth and invasion of ovarian cancer cells by targeting DNMT3A, so as to reveal molecular mechanism to inhibit ovarian cancer. Methods:Quantitative real-time palymerase chain reaction (qRT-PCR) method was employed to detect the expression of miR-101 in ovarian cancer and cancer adjacent normal ovarian tissues. SKOV3 cells were transfected with miR-101 mimics, and DNMT3A siRNA was transfected as a positive control. Then Western blot was used to detect the expres-sion of DNMT3A protein regulated by miR-101 in SKOV3 cells. The growth and invasion ability of SKOV3 cells were evaluated by MTT and Transwell invasion assays.Results:qRT-PCR showed that miR-101 was down-regulated in ovarian cancer tissues. Western blot showed that the level of DNMT3A protein was inhibited by restored miR-101 or knock-down of DNMT3A in SKOV3 cells. Following transfection of miR-101 mimics or knock-down of DNMT3A for 48, 72 and 96 h respectively, MTT assay showed that theD values were signiifcantly lower than the control group, (P<0.05). After transfection of miR-101 mimics or knock-down of DNMT3A for 36 h, Transwell invasion assay showed that the numbers of cells through the basement membrane was (105±7) and (107±13), respectively, which are signiifcantly different from the control group (213±11), indicating invasion of SKOV3 cells signiifcantly slowed down (P<0.05).Conclusion:miR-101 suppresses cell growth and invasion by targeting DNMT3A in ovarian cancer.
7.Reproduction of reference method for serum glucose detection and comparison with routine glucose kits

Objective To reproduce and validate the hexokinase reference method for glucose detection and compare other 5 routine glucose kits with this reference method. Methods The CDC hexokinase reference method for glucose detection was established and the performance was validated through testing a standard reference material (SRM) and participating in the IFCC ring-trial for reference laboratories for glucose evaluation. The CLSI EP 9-A2 protocol was used to compare the 5 routine glucose kits with the hexokinase reference method. Forty serum samples were analyzed by 5 routine kits and the hexokinase reference method. Results When SRM 965a was determined by the reference method,the bias of level 2 and level 3 were 0. 93%, -0. 23% respectively. The results for IFCC ring trial were within the accepted range. For the 5 routine kits, the confidence intervals of the predictive bias at the medical decision point Xc (Xc = 6. 11 mmol/L) were all within the range of defined acceptable error (10%) and the range of biological variation bias (6.9%). Conclusions The hexokinase reference method for serum glucose was reproduced in our lab. The serum glucose results measured with 5 routine kits were different from results detected with the reference method, but the bias was acceptable,and it will not affect the detection results.
8.Changes of cardiac function and myocardial apoptosis in rabbits after limb ischemia reperfusion

Objective To investigate the changes of cardiac function and myocardial apoptosis after limb ischemia reperfusion in rabbits.Methods Thirty rabbits were randomly divided into 3 groups:sham-operated control group (group SC),ischemia group (groupⅠ,8 hours of bilateral hind limb ischemia) and ischemia reperfusion group (group IR,4 hours of bilateral hind limb ischemia and 4 hours of reperfusion).Immunohistochemical studies and TUNEL were done to evaluate cell apoptosis.The SOD activity and MDA content in myocardial tissue and plasma LDH activity were determined. The left ventricular function were observed by physiology recording instrument.The cardiac histopathologic changes in experimental rabbits were observed.Results In group IR,the apoptosis index and expression of Bax were significantly increased compared with the other two groups.Similarly,ischemia reperfusion reduced SOD activity and enhanced LDH activity and MDA level,and the left ventricular function decreased in group IR.There was a positive correlation among the MDA content and the LDH activity,the expression of bax,the number of apoptosis cells.Under light microscope, myocardial impairment was found in group IR.Conclusion The myocardial injury occured and the left ventrieular function decreased after limb ischemia reperfusion,and they were associated with oxidative damage and apoptosis.
9.Clinical analysis of 41 cases with the late onset of systemic lupus erythematosus

Objective To investigate the clinical manifestations and laboratory findings of patients with the late onset systemic lupus erythematosus(LOSLE) and the elderly onset SLE. Methods Forty one patients with SLE onset beyond the age of 50 years(including 20 patients SLE onset beyond the age of 60 years) were identified and compared with 41 patients who had SLE onset before the age of 50 (General SLE). Patients in the group of general SLE were selected by 1 to 1 individual matching according to sex, course of diseases, date of admission. The presenting clinical features, laboratory findings, incidence rate of complication, efficacy of glucocorticoid and the prognosis of patients in two groups of LOSLE and general SLE were compared and analyzed. In addition, 20 cases of elderly onset SLE were also compared and analyzed. Results There were significant differences in symptoms between groups of LOSLE and General SLE. 10.Strain Screening for Bioconversion of Glycerol to Lactic Acid and Optimization of Culture Medium

A strain, Escherichia coli AC-521, which could efficiently covert glycerol to lactic acid, was isolated from soil samples. A series of morphological and biochemical characteristics and sequence analysis of 16S rDNA reveal that it belongs to Escherichia coli. Through orthogonal experiment of L16(45), the optimum medium compositions were determined as initial glycerol 70 g/L, yeast extract 4 g/L, peptone 7 g/L, (NH4)2SO4 10 g/L, and K2HPO4 2.5 g/L. The 5 L fermenter fed-batch fermentation was performed under optimal conditions, giving 74.5 g/L lactic acid with a yield of 0.87 mol/mol glycerol after 80 hours.
