1.IMMUNOHISTOCHEMICAL INVESTIGATION OF THE CHANGES OF PANCREATIC ISLET CELLS DURING LIVER REGENERATION FOLLOWING PARTIAL HEPATECTOMY

50 adult albino rats were divided randomly into 3 groups, i, e. partial hepatectomy group, sham-operation group and normal control group. Specimens of tail portion of pancreas were taken at 1, 2, 3, 6 and 14 days after operation respectively. Paraffin sections were made and stained with immunohistochemical PAP method to demonstrate insulin-containing B cells,C-peptide-containing B cells,glucagon-containing A cells and somatostatin-containing .D cells. The changes in hormone secretion activity of B, A and D cells have been observed during liver regeneration period. Based on the different amount of immunoreactive product, the endocrine activity of islet cells can be divided into 3 grades: +, ++, and +++. Percentage of each grade of islet cells was calculated and taken as the parameter of endocrine activity of islet cells. Insulin release activity of B cells increased after 1-3 days of operation, and insulin biosynthesis and/or storage enhanced 3-6 days after. Glucagon release activity of A cells raised gradually during 1-3 days after operation, and somatostatin released activity of D cells declined at the same time. After 6 days, endocrine activity of A cells returned to normal level, but release of somatostatin of D cells elevated. Endocrine activity of islet cells all recovered to normal level 14 days later. The changes of endocrine activity of B cells and A cells were closely related to and parallel with regeneration process of liver. The changes of D cells suggest that it probably be a reflection of regulation action on endocrine activities of B cells and A cells.
2.HISTOLOGICAL AND HISTOCHEMICAL CHANGES OF GRANULAR CONVOLUTED TUBULES OF SUBMANDIBULAR GLAND DURING LIVER REGENERATION FOLLOWING PARTIAL HEPATECTOMY OF RATS

50 adult rats divided into 3 groups, i. e. partial hepatectomy, sham-operation and normal control groups. The submandibular glands (SMG) were removed at different intervals (1-14 days) after operation, and stained with histological and histochemical method. On the 1-3 days after operation, especially on 3 days, some changes were found in granular convoluted tubule (GCT) cells of SMG. The secretory granules in the GCT cells were decreased in number. The reactions of PAS, -SS- group, PTAH were weaker than those of the sham-operation group, but the reactions of content in tubule system of SMG were raised. The reaction of SDH was stronger than those of the sham-operation group and the activities of NE and AcP decreased. After 7 days all the changes gradually restored, except the activities of ASDH increased and NE decreased continuously. After 14 days, all the changes became nearly normal. In addition, the reactions of RNA, G6Pase and 5-Nase in GCT cells of hepatectomized rats did not show apparent changes. The results suggested that, the changes of SMG GCT cells following partial hepatectomy seem, to be closely related with regenerative process of liver.
3.Effects of Radix Ginseng and Radix Notoginseng formula on secretion of vascular endothelial growth factor and expression of vascular endothelial growth factor receptor-2 in human umbilical vein endothelial cells.

Objective: To investigate the effects of Radix Ginseng and Radix Notoginseng formula on secretion of vascular endothelial growth factor (VEGF) and expression of vascular endothelial growth factor receptor-2 (VEGFR-2) in human umbilical vein endothelial cells (HUVECs) in vitro. Methods: HUVECs were cultured in vitro. Bovine basic fibroblast growth factor (bFGF) at concentration of 320 U/mL and Radix Ginseng and Radix Notoginseng formula at concentrations of 0.1, 0.2 and 0.4 mg/mL were used to culture with HUVECs. And HUVECs in blank control group were cultured with culture solution only. After 24-hour culture, the content of VEGF in supernatant was detected by enzyme-linked immunosorbent assay and the expression of VEGFR-2 was detected by immunocytochemical staining and Western-blotting. Results: Radix Ginseng and Radix Notoginseng formula at 0.4 mg/mL, the same as bFGF, increased VEGF content in the HUVEC supernatant and the number of VEGFR-2-positive HUVECs. Expression of VEGFR-2 protein in high-dose Radix Ginseng and Radix Notoginseng formula group was up-regulated as compared with the blank control group. Conclusion: Radix Ginseng and Radix Notoginseng formula can promote HUVEC proliferation and secretion of VEGF, as well as the expression of VEGFR-2 protein, which may be one of the mechanisms of Radix Ginseng and Radix Notoginseng formula in promoting angiogenesis.
4.The effects of an instant haw beverage on lipid levels, antioxidant enzyme and immune function in hyperlipidemia patients.

OBJECTIVETo determine the effectiveness of an instant haw beverage in regulating lipid disturbance, enhancing antioxidant enzyme activity and immune function.
METHODSData was collected from 60 hyperlipidemic subjects. In this crossover design, each subject randomly received either the instant haw beverage (100 ml corresponding to 3 g of haw powder or 30 g of fresh haw fruit plus the carrier-guar gum plus some starch) or placebo (guar gum 1.5 g plus some starch as the carrier of the beverage) twice daily. Each supplementation lasted 31 days with a 28-day washout period between treatments.
RESULTSThe instant haw beverage significantly reduced total serum cholesterol (9.6%), triglyceride (12.1%), LDLC (18%) while significantly increased SOD activities (7.5%). The placebo was shown to have positive results in some of the lipid profiles, though the effects of the instant haw beverage demonstrated greater significance. Serum triglyceride levels were significantly decreased and SOD activity significantly increased only as subjects were supplemented with the instant haw beverage while no significant changes were seen with placebo.
CONCLUSIONSupplementation with the instant haw beverage positively affects blood lipid profile, antioxidant status and immune function in individuals with hyperlipidemia.

5.A comparison of clinical characteristics between acute fatty liver of pregnancy and hemolysis, elevated liver enzymes and low platelets syndrome

Objective:To compare and analyze the clinical characteristics between acute fatty liver of pregnancy (AFLP) and the hemolysis, elevated liver enzymes and low platelets (HELLP) syndrome.Methods:This is a retrospective cohort study. The clinical data of 13 cases with AFLP and 34 cases with HELLP syndrome were collected from three tertiary referral centers in Yunnan (the First Affiliated Hospital of Kunming Medical University, the Second Affiliated Hospital of Kunming Medical University, and Yan'an Hospital of Kunming City) from January 2016 to December 2021. The patients were diagnosed to AFLP and HELLP syndrome according to the Swansea criteria and the Tennessee classification system. The general characteristics, clinical features, laboratory results within 24 hours after admission, complications, maternal and neonatal outcomes were compared to analysis the differences between the two groups.Results:① Maternal characteristics: compared with HELLP syndrome group, AFLP group had lower body mass index (BMI) and blood pressure at admission (both P < 0.01). ②Clinical features: the most common symptoms in AFLP patients were skin jaundice, abdominal pain, nausea and vomiting, edema. The main manifestations of patients with HELLP syndrome were albuminuria, hypertension, edema, headache. Some patients had multiple symptoms concurrently. ③ Laboratory results: compared with HELLP syndrome group, the levels of platelet count (PLT), total bilirubin (TBil), direct bilirubin (DBil), γ-glutamyl transferase (γ-GGT), alkaline phosphatase (ALP), total bile acid (TBA), serum creatinine (SCr) and international standardized ratio (INR) in AFLP group were significantly increased within 24 hours after admission [PLT (×10 9/L): 107.69±51.13 vs.76.71±43.25, TBil (μmol/L): 121.60 (83.20, 170.00) vs.15.25 (7.22, 29.05), DBil (μmol/L): 86.50 (58.60, 104.00) vs. 4.30 (2.22, 10.10), γ-GGT (U/L): 87.00 (37.00, 127.00) vs. 41.00 (19.00, 64.42), ALP (U/L): 199.10 (109.00, 349.20) vs. 125.50 (90.50, 155.25), TBA (μmol/L): 51.50 (16.20, 117.40) vs. 4.15 (2.02, 6.95), SCr (μmol/L): 155.80 (129.00, 237.00) vs. 79.00 (65.43, 113.70), INR: 1.28 (1.17, 1.63) vs. 0.94 (0.88, 1.08), all P < 0.05], prothrombin time (PT) was significantly prolonged [seconds: 16.10 (14.50, 19.20) vs. 12.40 (11.43, 13.40), P < 0.05]. The level of blood glucose (GLU), fibrinogen (FIB) and the activity of antithrombin Ⅲ (ATⅢ) decreased significantly [GLU (mmol/L): 5.18±1.33 vs. 6.33±1.19, FIB (g/L): 1.96±1.46 vs. 3.81±1.58, ATⅢ (%): 40.61±25.84 vs. 66.39±24.11, all P < 0.05]; ④ Complications: compared with HELLP syndrome group, the incidence of patients with hypoglycemia [30.77% (4/13) vs. 0% (0/34)], acute liver failure [53.85% (7/13) vs. 5.88% (2/34)], acute renal insufficiency [69.23% (9/13) vs. 8.82% (3/34)], coagulopathy [76.92% (10/13) vs. 38.24% (13/34)], disseminated intravascular coagulation (DIC) [53.85% (7/13) vs. 5.88% (2/34)], and multiple organ dysfunction syndrome (MODS) [53.85% (7/13) vs. 5.88% (2/34)] were significantly higher in AFLP group (all P < 0.05). ⑤ Maternal and neonatal outcome: all patients delivered after admission. The total length of hospital and intensive care unit stay were significantly longer in the AFLP group than in the HELLP syndrome group [days: 17.00 (11.00, 25.00) vs. 9.00 (7.00, 12.00), 12.00 (4.00, 22.00) vs. 3.91 (0, 7.00), both P < 0.01]. Two AFLP patients died, including one due to intracranial venous thrombosis and one due to multiple organ failure and cardiopulmonary arrest. There were no deaths in the HELLP syndrome group. Conclusions:There are significant differences in maternal characteristics, laboratory results and complications between AFLP and HELLP syndrome. TBil, γ-GGT, SCr, FIB, INR and ATⅢ activity may help to distinguish the two diseases.
6.The relative location between the mandibular third molar and inferior alveolar canal studied by CBCT

Objective: To determine the positional relationship between impacted mandibular third molar(IMTM) and mandibular canal(MC) by CBCT. Methods: 664 IMTMs with root apexes contacted to MCs on panoramic radiograph were further examined by CBCT,the exact positional relationship between IMTMs and MCs was observed and analyzed by I-Dixel software. Results: The 664 cases were divided into lowly(6. 3%),intermediately(60. 7%) and highly(33. 0%) impacted groups by the depth of IMTMs in mandibulae showed on CBCT images. The distance(mm) from the root apexes of IMTMs to the intact superior wall of MCs(n = 329) in lowly, intermediately and highly impacted cases were 1. 39 ± 1. 38,1. 28 ± 1. 03 and 1. 79 ± 1. 54 respectively(P < 0. 05). MCs on the buccal side,lingual side,under the IMTMs and between the root apexes were found in 49. 8%,12. 0%,36. 6% and 1. 5% of the cases; the MC wall deffect was found in 65. 8%,27. 5% and 95. 0% of the cases with the MCs under,on buccal and lingual side of the IMTMs, respectively(P < 0. 001). Conclusion: CBCT examination is necessary for the determination of the positional relationship between MC canal and IMTM with the root apex cotacted or overlapped to MC.
