Objective To study the effects of different concentrations of antisense oligodeoxynucleotides (ASODN) on human pancreatic cancer cell line PaTu8988s. Methods Human pancreatic cancer cell line PaTu8988s in exponent growth stage are adopted. We observe the effect of different time point on the PaTu8988s cell at 12、24、48 and 72 hour. Results The inhibitory rate on PaTu8988s cell line is 42.25%、66.29%、69.55%、74.58% and 66.20%、91.43%、98.18%、98.33% for ASODN concentrations of 50 ?g/ml and 100 ?g/ml at 12、24、48 and 72 hour, respectively. Conclusion The inhibitory effect of ASODN began from 12 hour and becomes more obvious at 48~72 hour. The higher the concentration of ASODN, the earlier the peak of inhibited rate.
3.Mechanism of omeprazole in inducing gastric epithelium apoptosis

Objective To study whether omeprazole induces gastric epithelial cells apoptosis and analyse the possible mechanism. Methods The cell cycle and apoptosis ratio of SGC 7901 cell line were determined by flow cytometry in the presence of omeprazole, meanwhile, TUNEL positive cells from gastric mucosa were counted after incubated with omeprazole for 72 hours. Expression of p53、 GADD 45 、 p15,16,18,19 and bcl 2 were stained by immunohistochemical assay and ATPase activity was tested. Results significant alteration with decreasing percentage of G 1 phase and increasing percentage of G 2 phase of SGC 7901 cell line could be detected after incubating 24 hours with omeprazole at double clinical doses . An apoptosis peak appeared in flow cytometry after incubating 72 hours with omeprazole. However, no genetic products such as p53, GADD 45 , p15,16,18,19 and bcl 2 were seen. Concentrations of ATPase were markedly increased from 0.326 0.387 before incubated with omeprazole to 0.054 0.103 after incubated with omeprazole. OBJECTIVETo explore the characteristics of Helicobacter pylori (H. pylori) antigen in serum and to evaluate its clinical diagnostic value.
METHODSEnzyme-linked immunosorbant assay (ELISA) was developed to detect the soluble H. pylori antigen (S-Hp) and circulatory specific H. pylori antigen immunocomplexes (Hp-IC) in serum.
RESULTSThe positive rate of S-Hp was 90.91% from 66 patients with H. pylori infection, which was much greater than 0% found in 28 controls (P < 0.001). Moreover, its concentration closely reflected the number of H. pylori in the gastric mucosa layer. We also found that Hp-IC existed bound with IgG and/or IgA in patients with positive S-Hp. However, there is no evidence to show the concentration of S-Hp reduced significantly in followed-up subjects after effective therapy.
CONCLUSIONSThese methods as newly and noninvasive complementary tools can be used for clinical diagnosis of H. pylori infection. In addition, S-Hp and Hp-IC may be of importance in H. pylori pathogenesis.

6.Pathological analysis of 602 renal biopsy results from a hospital in Zhejiang Province

Objective To exploring the distribution characteristics of renal disease pathology spectrum in Zhejiang Provincial Hospital of Traditional Chinese Medicine.Methods The pathological data of 602 patients who underwent renal biopsy at Zhejiang Provincial Hospital of Traditional Chinese Medicine from January 2016 to June 2021 were collected and retrospectively analyzed.Results ①Among the 602 patients,there were 319 males and 283 females,with a male to female ratio of 1.13∶1.The patient's age was 9-78 years old,average(44.44±15.22)years old.There are statistical differences in the gender ratio and age distribution of various pathological types.②There were 471 cases of primary glomerular disease(PGD),117 cases of secondary glomerular disease(SGD),5 cases of tubulointerstitial nephritis,and 9 cases of hereditary kidney disease.③The most common PGD is immunoglobulin(Ig)nephropathy(242 cases),followed by membranous nephropathy(110 cases).④The most common SGD was diabetes nephropathy(42 cases),followed by lupus nephritis(22 cases).Conclusion PGD is the most common pathological type of renal biopsy,with IgA nephropathy and membranous nephropathy being the most common.Diabetes nephropathy is the most common SGD.In recent years,the incidence rate of diabetes nephropathy has increased significantly.
7. Association of polycyclic aromatic hydrocarbon exposure and CYP1A1 gene and enzyme activity of peripheral blood monocyte cells in coke oven workers

Objective:
To explore the association between polycyclic aromatic hydrocarbons (PAHs) exposure and cytochrome P450 CYP1A1 expression at gene and enzyme activity levels in the peripheral blood monocyte cells in coke oven workers, and to provide a certain basis for the biological monitoring of health damage in coke oven workers.
Methods:
We surveyed 118 coke oven workers and 63 controls (energy power workers in the same company) using self-designed questionnaire, determined their post-shift urinary 1-hydroxypyrene (1-OH-Py) concentration using high performance liquid chromatography (HPLC)-fluorescence detector method. We also isolated the peripheral blood mononuclear cell (PBMC) from fasting venous blood, and detected DNA damage using comet assay, CYP1A1 mRNA level using the real-time fluorescent quantitative PCR (FQ-PCR), and EROD activity using spectrophotometry. Statistical analyses including one-way analysis of variance and multiple linear regressions were used to analyze the association of urinary 1-OH-Py and CYP1A1 mRNA level and EROD activity.
Results:
Compared to the control group, the urinary 1-OH-Py concentration and PBMC DNA tail moment were significantly increased in coke oven workers (
